Exposure to Copper (II) Chloride Induces Behavioral and Endocrine Changes in Zebrafish (CuCl2 노출에 의해 유도되는 제브라피시의 행동과 내분비계의 변화)

  • The aim of this study was to investigate the effect of copper (II) chloride (CuCl2) on zebrafish. Zebrafish were exposed to various CuCl2 concentrations and subjected to different exposure times to determine the median lethal concentration (LC50) values. To evaluate stress responses, we measured whole-body cortisol levels and behavioral parameters using the open field test (OFT) or the novel tank test (NTT). The zebrafish were exposed to CuCl2 solution at concentrations of 1.5-150 ㎍/l or a vehicle for 1 hr before behavioral tests or sample collection for whole-body cortisol. The LC50 values were 30.3, 25.3, and 14.8 ㎍/l at 24, 48, and 96 hr, respectively. The NTT showed that mobility, velocity, and distance covered were significantly lower in zebrafish exposed to CuCl2 than in the control group (p<0.05), while the turn angle was significantly higher in zebrafish exposed to a CuCl2 concentration of 150 ㎍/l than in the control group (p<0.05). The OFT also showed that mobility, velocity, and distance covered were significantly lower and the turn angle and meandering were significantly higher in zebrafish exposed to all concentrations of CuCl2 than in the control group (p<0.05). The whole-body cortisol levels were significantly higher in zebrafish exposed to CuCl2 than in the control group (p<0.05). These results suggest that exposure to lethal CuCl2 concentrations induces an intense toxic and stress response in zebrafish, causing behavioral changes and increasing whole-body cortisol levels.

Study on the limitation of AVO responses shown in the seismic data from East-sea gas reservoir (동해 가스전 탄성파 자료에서 나타나는 AVO 반응의 한계점에 대한 고찰)

  • In the case of the deep reservoirs like the gas reservoirs in the East-sea, it is often difficult to observe AVO responses in CMP gathers. Because the reservoir becomes more consolidated as its depth deepens, P-wave velocity does not decrease significantly when the pore fluid is replaced by the gas. In this study, we analyzed the effects of Poisson's ratio difference on AVO response with a variety of Poisson's ratios for the upper and lower layers. The results show that, as the difference in Poisson's ratio between the upper and lower layers decreases, the change in the reflection amplitude with incidence angle decreases. To consider the limitation of AVO responses shown in the gas reservoir in East-sea, the velocity model was made by simulation Gorae V structure with seismic data and well logs. The results of comparing AVO responses observed from the synthetic data with theoretical AVO responses calculated by using material properties show that the amount of the change in reflection amplitude with increasing incident angle is very small when the difference in Poisson's ratio between the upper and lower layers is small. In addition, the characteristics of AVO responses were concealed by noise or amplitude distortion arisen during preprocessing. To overcome such limitations of AVO analysis of the data from deep reservoirs, we need to acquire precisely reflection amplitudes in data acquisition stage and use processing tools which preserve reflection amplitude in data processing stage.

Numerical Analysis of Bragg Reflector Type Film Bulk Acoustic Wave Resonator (수치적 계산을 이용한 Bragg Reflector형 탄성파 공진기의 특성 분석)

  • Bragg reflector type FBAR was fabricated on the Si(100) substrate. We measured a frequency response of the resonator at 5.2 GHz and analyzed it by numerical calculation considering actual acoustic losses of each layer in the structure. We fabricated nine layer Bragg reflector of W-SiO$_2$pairs using r.f. sputtering method and fabricated AlN piezoelectric and Al electrodes using pulsed dc sputtering. The return loss(S$_{11}$) of the fabricated Bragg reflector type FBAR was 12 dB at 5.38 GHz and the series resonance frequency(f$_{s}$) was 5.376 GHz and the parallel resonance frequency(f$_{p}$) was 5.3865 GHz. Effective electro-mechanical coupling constant (K$_{eff{^2}}$) and Quality factors(Q$_{s}$), the Figures of Merit of the resonator, were about 0.48% and 411, respectively. We extracted acoustic parameters of AlN piezoelectric and reflection coefficient of the Bragg reflector by numerical calculation. We could know that material acoustic impedance and wave velocity of AlN piezoelectric decreased for intrinsic value and the electromechanical coupling constant(K$_2$) value was very low owing to the poor quality of the AlN piezoelectric. Reflection coefficient of Bragg reflector was 0.99966 and reflection band was very wide from 2.5 to 9.5 GHz.

Improvement of Seismic Performance of Long-span Bridges using Complex Dampers (복합감쇠기를 이용한 장대교량의 내진성능향상)

  • This paper presents a new vibration control method for long-span bridges using complex damper system. The new system presents simple mechanical configuration with oil and elasto-plastic dampers which have velocity and displacement dependency in vibration energy absorbing. This system can produce various damping forces according to the applied external forces by the velocity and displacement-dependent characteristics of the dampers. The oil damper dissipates vibration energy for relatively frequent and small amplitude like in the case for small to moderate earthquakes, whereas the elasto-plastic damper system works for rare and large amplitude vibration such as high seismic excitation. Thus, the proposed system exhibits the advantage of low cost with high performance since the roles of the two different dampers are effectively separated. A numerical model is established for the complex damper system, and the response characteristics and effectiveness of the proposed system are presented through numerical simulations. Numerical results show that the proposed complex damper system can significantly improve the seismic performance of long-span bridge structures with much more effective damping mechanism than single conventional passive damper systems.

Exposure to Dithiopyr Alters Swimming Performance Parameters in Zebrafish (Dithiopyr에 노출이 zebrafish의 유영 행동에 미치는 영향)

  • The aim of this study was to identify the effects of dithiopyr (DTP), a herbicide, on behavior in zebrafish. The toxicity of DTP has rarely been investigated in fish. In the present study, zebrafish were exposed to different concentrations of DTP in the range of 10-20 μM for 48 h in a test container, in order to measure the value of median lethal concentrations (LC50). Behavioral experiments were performed, including the novel tank test (NTT) and the open field test (OFT), to assess stress responses or locomotion. After exposure to the DTP solution at a sublethal concentration of 2.5–10 μM for 6 min, the behavior of the zebrafish was observed for 6 min. In the acute toxicity test, the LC50 value of DTP showed as 14.49 μM in the zebrafish. The NTT showed that the duration of immobility and the velocity were significantly increased by exposure at a concentration of 5 μM of DTP, compared with a control group (p<0.05). However, compared with the control group, DTP significantly decreased the distance moved and the frequency at the top of the tank, and significantly increased the turn angle and duration at the bottom, in a concentration-dependent manner (p<0.05). In addition, in the OFT, exposure to DTP significantly decreased the distance moved and velocity compared with the control group (p<0.05). Exposure to DTP also significantly increased the duration of immobility, the turn angle, and the meandering movement, in a concentration-dependent manner (p<0.05). Further, exposure to DTP at a low concentration elevated whole-body cortisol levels in the zebrafish. The results of this study thus suggest that DTP induces a toxic response and negative effects on behavior and the endocrine system in zebrafish.

Association between periodontal bacteria and degenerative aortic stenosis: a pilot study

  • Purpose: Although several reports have described the relationship between periodontal disease and cardiovascular disease, information about the association between periodontal disease and the progression of degenerative aortic stenosis (AS) is lacking. Therefore, we performed a retrospective, single-center, pilot study to provide insight into this potential association. Methods: Data from 45 consecutive patients (19 men; median age, 83 years) with mild or moderate degenerative aortic stenosis were analyzed for a mean observation period of 3.3±1.9 years. The total amount of Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis and titers of serum immunoglobulin G (IgG) against periodontal bacteria and high-sensitivity C-reactive protein (hs-CRP) were evaluated. Aortic valve area (AVA), maximal velocity (Vmax), mean pressure gradient (mean PG), and the Doppler velocity index (DVI) were evaluated. The change in each parameter per year ([ParameterLATEST-ParameterBASELINE]/Follow-up Years) was calculated from the retrospective follow-up echocardiographic data (baseline vs. the most recently collected data [latest]). Results: No correlation was found between the concentration of periodontopathic bacteria in the saliva and AS status/progression. The anti-P. gingivalis antibody titer in the serum showed a significant positive correlation with AVA and DVI. Additionally, there was a negative correlation between the anti-P. gingivalis IgG antibody titer and mean PG. The hs-CRP concentration showed positive correlations with Vmax and mean PG. Meanwhile, a negative correlation was observed between the anti-P. gingivalis IgG antibody titer and ΔAVA/year and Δmean PG/year. The hs-CRP concentration showed positive correlations with Vmax and mean PG, and it was significantly higher in patients with rapid aortic stenosis progression (ΔAVA/year <-0.1) than in their counterparts. Conclusions: Our results suggest that periodontopathic bacteria such as A. actinomycetemcomitans and P. gingivalis are not directly related to the status/progression of degenerative AS. However, inflammation and a lower immune response may be associated with disease progression.

Feasibility Evaluation for Remediation of Groundwater Contaminated with Heavy Metal using Calcium Polysulfide in Homogeneous media (균질한 매질 내 Calcium polysulfide 주입에 따른 고농도 중금속 오염 지하수 정화 타당성 검토)

  • In this study, column tests using relatively uniform Jumunjin sand media were conducted to evaluate the feasibility of calcium polysulfide (CaSx, CPS) in removing high concentration of Zn2+ in groundwater. The injected CPS solution reacted rapidly with Zn2+ in artificial groundwater and effectively reduced Zn2+ by more than 99% through metal sulfide precipitation. Since the density (d = 1.27 g/cm3 ) of CPS solution was greater than that of water, CPS solution settled down rapidly while capturing Zn2+ and formed stable CPS layer similar to dense nonaqueous phase liquid. Mass balance analysis on Zn2+ in CPS solution suggested that CPS solution effectively reacted with Zn2+ to form metal sulfide precipitates except for high groundwater seepage velocity of 400 cm/d. With greater groundwater seepage velocity, injected CPS did not completely dissolve at the CPS-water interface, but a partially-misible CPS layer continuously moved and reacted with Zn2++ in the direction of groundwater flow. Since hydraulic conductivity (Kh) decreased slightly due to the generated metal precipitates in the inter-pores of media, injection of CPS solution should be optimized to prevent clogging. As evidenced by both XRF and SEM/EDS results, ZnS precipitates were clearly observed through the reaction between the CPS solution and Zn2+. Further study is warranted to evaluate the feasibility of CPS to remove high-concentration heavy metalcontaminated groundwater in complex and heterogeneous media.

Evaluation of the applicability of the surface wave method to rock fill dams (사력댐에서의 표면파 기법 적용성 평가 연구)

  • In current design practice, the shear wave velocity (Vs) of the core and rock-fill zone of a dam, one of the characteristics essential for seismic response design, is seldom determined by field tests. This is because the borehole seismic method is often restricted in application, due to stabilisation activities and concern for the security of the dam structure, and surface wave methods are limited by unfavourable in-situ site conditions. Consequently, seismic response design for a dam may be performed using Vs values that are assumed, or empirically determined. To estimate Vs for the core and rock-fill zone, and to find a reliable method for measuring Vs, seismic surface wave methods have been applied on the crest and sloping surface of the existing 'M' dam. Numerical analysis was also performed to verify the applicability of the surface wave method to a rock-fill dam. Through this numerical analysis and comparison with other test results, the applicability of the surface wave method to rock-fill dams was verified.

Analysis of Nocturnal Cold Air Flow Characteristics for Setting of Tropical Night Response Zone in Daegu (대구시 열대야 대응 구역 설정을 위한 야간 찬공기 유동성 분석)

  • Heat wave generation in cities is basically affected by global warming, but it is further exacerbated by the impact of artificial heat emission and heat accumulation in the city. In particular, the effects of urban heat waves directly affect the occurrence of tropical nights. Basically, however, the choice of countermeasures against tropical nights is very limited compared to the daytime heat wave response. The purpose of this study was to analyze the characteristics of cold air flow at night as a countermeasure against tropical nights in Daegu Metropolitan City and to suggest its spatial applicability. As a research method, the spatial characteristics (flow velocity, flow rate, flow direction and range) of cold air flow in Daegu were quantitatively analyzed using KLAM_21, a cold air flow analysis program. As a result of the analysis, it was found that cold air generation and flow in the surrounding mountains of Daegu Metropolitan City was very active, but the inflow was limited to the urban area, which has tropical nights. However, it has been shown that the flow of cold air flowing from the surrounding mountains is very active in some urban areas, so it has spatial conditions that are very effective in countering tropical nights. If these spatial conditions are used for the urban planning, it will be very useful to develop countermeasures for tropical nights.

Identification of damage states and damge indices of single box tunnel from inelastic seismic analysis (비탄성 지진 해석을 통한 박스 터널의 손상 상태 및 손상 지수 규명)

  • In a performance-based design, the structural safety is estimated from pre-defined damage states and corresponding damage indices. Both damage states and damage indices are well defined for above-ground structures, but very limited studies have been performed on underground structures. In this study, we define the damage states and damage indices of a cut-and-cover box tunnel which is one of typical structures used in metro systems, under a seismic excitation from a series of inelastic frame analyses. Three damage states are defined in terms of the number of plastic hinges that develop within the structure. The damage index is defined as the ratio of the elastic moment to the yield moment. Through use of the proposed index, the inelastic behavior and failure mechanism of box tunnels can be simulated and predicted through elastic analysis. In addition, the damage indices are linked to free-field shear strains. Because the free-field shear strain can be easily calculated from a 1D site response analysis, the proposed method can be readily used in practice. Further studies are needed to determine the range of shear strains and associated uncertainties for various types of tunnels and site profiles. However, the inter-linked platform of damage state - damage index - shear wave velocity - shear strain provides a novel approach for estimating the inelastic response of tunnels, and can be widely used in practice for seismic designs.
