Male Reproductive System of Leucosia Anatum: Histological and Histochemical Analysis

  • Leucosia anatum, also known as the pebble crab, is a species of crab that belongs to the family Leucosiidae. Crabs are the most conspicuous and abundant components of the epibenthic macrofauna of the mangrove ecosystems. The present investigation was carried out with the objective of enlightening the reproductive system of L.anatum through histological and histochemical studies. Histological analysis of the epithelial cells of vas deferens, the luminal size and shape of the vas deferens was carried out, since the spermatophore material is secreted and molded by the vas deferens. Histochemical analysis of the blood plasma, testis, proximal vas deferens, middle vas deferens, distal vas deferens, seminal plasma and spermatophore was done to reveal the possible transformation of their secretory materials into the spermatophoric components. Biochemical components present in the blood plasma, testis, proximal vas deferens (PVD), median vas deferens (MVD) and distal vas deferens (DVD) regions seminal plasma and spermatophore of pebble crab were analyzed to find out the quantity of free sugars, proteins, and fats. Thus the current study focuses on the histological and histochemical analysis of Testis and Vas Deferens and their secretions in L.anatum.

Studies on the Adrenotropic Receptors of Vas Deferens of Ditrema temmincki Bleeker (척출(剔出) 망상어 정관(精管)에 있어서 Adrenotropic Receptor 성질(性質)에 관(關)한 연구(硏究))

  • The author studied the adrenotropic receptors of isolated vas deferens from Ditrema temmincki Bleeker, using adrenergic activators such as epinephrine, norepinephrine, isoproterenol and phenylephrine, and adrenergic blocking agents such as phenoxybenzamine and propranolol. The results are as follows: 1. The vas deferens was stimulated by epinephrine, norepinephrine and phenylephrine, but not affected by isoproterenol. 2. The excitatory effect of phenylephrine on the vas deferens was completely blocked by phenoxybenzamine, but more stimulated by propranolol. 3. The excitatory effects of epinephrine and norepinephrine were markedly reduced by phenoxybenzamine, but stimulated by propranolol. 4. The vas deferens pretreated with phenoxybenzamine and propranolol was not affected by epinephrine and norepinephrine. 5. The vas deferens was not affected by isoproterenol and also not affected by the pretreatment with either kind of blocking agent plus isoproterenol. 6. It seemed that the vas deferens had both alpha-excitatory receptor and beta-receptor, but it was difficult to detect the character of beta-receptor whether it was inhibitory or excitatory.

Morphine induced inhibition of the activities of accessory reproductive ducts in male rats

  • Adult male albino rats were treated with 0.5 mg and 0.75 mg morphine/100 g body weight intraperitoneally for 30 days. All the animals were autopsied on $31^{st}$ day. Epididymis and vas deferens were dissected out, weighed and processed for histological and biochemical studies. Morphine has caused a reduction in the weight of epididymis and vas deferens in both the doses of drug treated groups. The total cholesterol content is increased while protein, DNA and RNA contents and epididymal sperm counts are decreased. The acid phosphatase content is decreased 10.12 $\pm$ 0.11 in caput, 9.26 $\pm$ 0.30 in cauda of epididymis and in vas deferens 8.14 $\pm$ 0.15 in 0.5 mg treated groups and in 0.75 mg treated rats shows 9.52 $\pm$ 0.27 in caput, 9.14 $\pm$ 0.18 in cauda of epididymis and in vas deferens 7.84 $\pm$ 0.11is decreased, whereas alkaline phosphatase is increased. The surface epithelial cell height of these ducts is reduced and secretory activity is inhibited with the disruption of epithelial cell projections. The gravimetric and histometric changes of epididymis and vas deferens may be due to non-availability of androgens in morphine treated rats.

The Study on the Role of Prostaglandin in Contraction of Vas Deferens (정관운동에 있어서 prostaglandin 의 역할에 관한 연구)

  • Prostaglandin(PG) is ubiquitously distributed in most mammalian tissue and their actions are complicated. Especially in autonomic nervous system, there are evidences indicating that PGs act as neuromodulators i.e., PGs, which are released in the vicinity of autonomic neuroeffector junctions, influence the release and the response of the neurotransmitter. Present study was undertaken to elucidate the interrelationship between $PGF_{2\alpha}$ and adrenergic ${\alpha}_2-receptor$ function in electrical field stimulation induced contractile response of vas deferens in rat. Male rat, weighing 150{\sim}200\;g, was sacrificed and vas deferens was obtained. The isolated vas deferens strip was placed between two platinum electrodes in temperature controlled $(37^{\circ}C)$ muscle chamber containing Tyrode's solution and the electrical field stimulation(EFS) induced contraction was recorded with Grass Polygraph(Model 7) via force displacement transducer (FT .03, Grass). The results are summarized as follows: 1) Electrical field stimulation for 1sec( 1 msec, 40 cps) induced contraction of vas deferens was completely blocked by tetrodotoxin. 2) Bretylium caused marked inhibition of the EFS-induced contraction, hut tyramine and cocaine augmented the contraction. 3) EFS-induced contraction was inhibited or little affected in distal portion of vas deferens by norepinephrine or methoxamine, but the contraction was rather augmented by the ${\alpha}-agonists$ in proximal portion. 4) Clonidine inhibited the EFS-induced contraction proportionally to the concentration in distal portion, which was blocked by yohimbine pretreatment, but in the presence of $PGF_{2\alpha}$ the blockade by yohimbine was reversed. 5) Indomethacin pretreatment reduced the effect of clonidine, but addition of $PGF_{2\alpha}$ after washing-out the indomethacin caused the contraction to the control level. From these results it is suggested that PG synthesis is a necessary step and the PG itself has a permissive role in ${\alpha}_2-adrenoceptor$ action in rat vas deferens.

  • The effect of papaverine on the contractions induced by adrenergic neurotransmission in the isolated mouse vas deferens was investigated. Papaverine, $10^{-7}-10^{-5}$M, showed a dose-dependent and reversible inhibition on the induced contractions. When the frequency of stimulation was varied (2.5-20.0 Hz), the inhibitory effect tended to be marked at the lower frequencies.

The authors investigated the effects of $PGE_1$ on the action of sympathomimetics in the vas deferens of guinea-pig, comparing with those in the rat vas deferens, and also the action of $PGE_1$ on the motility of nerve-free smooth muscle of chick amnion. 2. In the isolated guinea-pig vas deferens, the actions of phenylephrine and norepinephrine were much potentiated by pretreatment with $PGE_1$. Futher, in the isolated hypogastric nerve-vas deferens preparation of guinea-pig, effects of phenylephrine, norepinephrine and tyramine on the contractile response of vas to the hypogastric nerve stimulation and to the transmural stimulation were also augumented especially in tension by $PGE_1$-pretreatment. 3. In the isolated hypogastric nerve-vas preparation of rat, both contractile responses to hypogastric nerve and transmural stimulation were slowly reduced by treatment with $PGE_1$ and the potentiated effect of phenylephrine or norepinephrine was not observed in spite of pretreatment with $PGE_1$. 4. The actions of phenylephrine and norepinephrine on the denervated vas deferens of guinea-pig were also enhanced by $PGE_1$ as it were in the intact vas deferens, but there was no significant effect by $PGE_1$ on the action of norepinephrine in the denervated rat vas deferens. 5. $PGE_1$ in low concentration $(10^{-8}g/ml)$ did not affect the spontaneous motility of nerve-free smooth muscle of chick amnion ($9{\sim}11$ th day incubated chick), but in large concentration $(5{\times}10^{-8}g/ml)$ it caused irregular and slightly inhibitory movement. Pretreatment with $PGE_1$ on chick amnion did not exert any change on the action of phenylephrine applied. However, the stimulatory action of physostigmine on the chick amnion was a little antagonized by the low concentration of $PGE_1$. 6. It might be summarized that there is species difference between the actions of $PGE_1$ on the vas deferens of guinea-pig and that of rat, and the action of $PGE_1$ on the guinea-pig vas deferens might be mediated by the other mechanism rather than by direct action on the vas musculature.

Pharmacological Studies on Human Vas Deferens -Coexistence of Adrenergic and Cholinergic Receptors, and Effect of Diazepam- (인체 정관의 약리학적 검색 -아드레날린성 및 콜린성 수용체의 공존과 Diazepam의 작용-)

  • This study aimed to investigate the autonomic innervations of human vas deferens and the effect of diazepam, a benzodiazepine sedative antianxiety drug, on the smooth muscle contractility of vas deferens. The specimens were obtained from healthy volunteers undergoing elective vasectomy with local anesthesia. The muscle preparation did not show any spontaneous contraction, but showed a good contraction induced by norepinephrine exerting the strongest response at $33^{\circ}C$. Phentolamine inhibited the norepinephrine-induced contraction concentration-dependently. Isoproterenol, a beta-adrenergic agonist evoked a considerable extent of contraction, and this contractile activity was antagonized by propranolol, a beta-adrenergic blocking agent. Acetylcholine induced a dashing contraction of the human vas deferens, and atropine, a muscarinic receptor blocking agent abolished the acetylcholine-induced contraction. Diazepam inhibited the norepinephrine-induced contraction in a concentration dependent manner. These results suggest that the smooth muscle of human vas deferens has cholinergic muscarinic and beta adrenergic receptors as well as the predominant alpha adrepergic receptor. Diazepam inhibits the motility, especially norepinephrine-induced contraction of human vas deferens.

Ultrastructural Changes of the Vas Deferens Epithelium by Season in a Slug Incilaria fruhstorferi (산민달팽이(Incilaria fruhstorferi) 수정관 내강 상피조직의 계절에 따른 미세구조적 변화)

  • A study on the ultrastructural changes In the epithellum of the vas deferens by season was conducted for the spdng and summer specimens of a slug Incilarfa fruhstoiferl. The vas deferens of the spdng spedmen was muscular tube about 0.4mm in diameter. Its lumen was divided into three flat grooves and the each groove was subdivided into two subbranches. The luminal epithellal celis of the Vas deferens which were irregular In shape showed strong methylenophilla in a double stain of methylene blue and basic fuchsln. The lumen of the vas deferens was filled with components strongly stained by methylene blue. The circular muscle layers surrounding the luminal epithellum of the vas deferens contained numerous granules arranged at regular intervals. The vas deferens of the summer specimen also was a thick muscular tube showing 0.4 mm in diameter. Its lumen was divided into four grooves but, the each of the grooves was not subdivided to form certain branclees unlikely to the spdng spedmen. The lining epfthelium of the lumen was consisted of simple ciliated columnar cells, irregular columnar cells and conical cells. The histological features were quiet different from those of the spring spedmen which showed irregular cell arrangement. According to electron microscopy the epithelium of the vas deferens in the spring specimen was composed of irregular columnar cells which had irregular shaped nuclei. The nuclei of the epitheilal cells were relatively large in comparison to their cytoplasm. The overall electric density of the cytoplasm was relatively high. The lumen of the vas deferens in the summer specimen was lined by a epfthelium with tail ciliated columnar cells and irregular cells. The unclei of the epithellal cells were long ellipsoid or irregular in shape. Both of the cytoplasm and the nuclei were showed low electric density. in consideration with the observable cell organelles were only ndoplasmic reticulum, lysosomes and microtutules, the cell organelles were poorly developed. The apical surfaces of the epithelial cells possessed brush borders with numerous microvilli and cilia with 9+2 arrangement of microtubules. The circular muscle layers surrounding the epithelium are usually thick and the degree of development of the circular muscle layers seems to be even in the both of the spring and summer specimens.

Alteration of ${\alpha}-Adrenoceptor$ by Cd-poisoning in Rat Vas Deferens and Tail Artery (카드뮴중독이 흰쥐 정관 및 꼬리동맥의 아드레나린성 ${\alpha}-$수용체에 미치는 영향)

  • It was aimed to elucidate the underlying mechanism which is elevated the systemic arterial blood pressure by cadmium(Cd). By using the isolated vas deferens and tail artery from Cd-poisoned rats, it was undertaken to see whether the ${\alpha}-adrenoceptor$ activity and prostaglandins action on the twitch response to electrical stimulation were altered. It was discussed in relation with sympathetic nerve activity. The response to electrical stimulation in vas deferens from Cd-poisoned rats was much enhanced, and frequency 50(Freq.50) was significantly lowered, comparing to control. However, the response of rat tail artery was attenuated by Cd. Inhibitory action of clonidine on the twitch response of vas deferens to electrical stimulation was decreased by Cd-poisoning. However, the increase in response in the presence of methoxamine was not affected. The inhibitory action of prostaglandin $E_2$ on the twitch response in the vas deferens but not in the tail artery was abolished or rather inverted by Cd-poisoning. In the tail artery from Cd-poisoned rats, the maximum contraction to methoxamine or clonidine was significantly decreased, however, $ED_{50}$ of both agonists was not changed. These results suggest that in vas deferens the autoreceptor of presynaptic membrane is inhibited by Cd-poisoning, by which mechanism the sympathetic activity is being enhanced and this alteration may be causally related to the elevation of arterial pressure.

  • This study was performed to understand the reproductive ecology of cephalopods, described the microanatomical structure of the male reproductive organs and spermatophore in the common squid, Todarodes pacificus, a major cephalopods in Korea. The common squid was gonochorism and had sexual dimorphism, the color of the reproductive organs reflected on the mantle and the presence of the hectocotylus. Male reproductive organs were composed of testis, primary vas deferens, spermatophoric gland (seminal vesicle), spermatophoric sac (Needham's sac) and secondary vas deferens. The male has specialized reproductive organs such as the spermatophore forming organ, spermatophoric gland. Testis was histologically a seminiferous tubule type. The primary vas deferens was a thin and inverted triangular spring-like form that connected from the rear of the testis to the spermatophoric gland. Inside, it was filled with sperm of basophilic in H-E stain. The spermatophoric gland is an irregular oval connected to the primary vas deferens and spermatophoric sac, and there were a number of tubular glands. The spermatophoric sac is a tubular structure located between spermatophoric gland and secondary vas deferens, and a number of spermatophores have been identified in the lumen. The secondary vas deferens was connected to the posterior of the spermatophoric sac and had a spermatophore inside. The spermatophore was a long, transparent tube about 22 mm long, with a sperm mass inside.
