  • The effective components of Korean ginseng showing the lipid-peroxide depressing activity were isolated. From the ether-soluble acidic fraction of fresh ginseng three phenolic acids were obtained. Salicylic acid and vanilic acid exhibited the potent antioxidant activity, where p-hydroxycinnamic acid did not.

  • Aliphatic esters of protocatechuic acid (PA, 1), vanillic acid (VA, 9) and gallic acid (GA, 18) were prepared and their anti-thrombotic effects were evaluated in the mouse model of thrombosis. The aliphatic groups included methyl, ethyl, n-propyl, i-propyl, n-butyl, i-butyl, n-amyl and cyclohexyl. n-Amyl ester of PA (7), i-propyl and cyclohexyl esters of VA (13 and 17 respectively) and ethyl ester of GA (20) treatment significantly lowered the death rate and increased the recovery from paralysis due to the thrombotic challenge. From the limited analogs available, it was tentatively concluded that the structural conformation, where carboxy oxygen (=O or -O) of the carboxyl group (COOH) at $C_1$ and the oxygen function at $C_3(either\; OH\; or\; OCH_3)$ are closely situated, is favorable for the esters of PA, VA and GA to be more antithrombotic.

  • A pyridyl alkaloid, 3-pyridylcarbinol (1) and benzoic acid derivatives, 4-hydroxy benzoic acid (2), 4-hydroxyactophenone (3), vanilic acid (4), and benzoic acid (5) were isolated from the rhizomes of Anemarrhena asphodeloides on the basis of spectroscopic and physicochemical analyses including 1D- and 2D- NMR techniques as well as by comparison of their data with the published values. Compounds 1 - 5 were isolated for the first time from this plant source.

  • Administration of an MeOH extract of Pharagmites communis on hyperlipidemic rats produced a significant decrease in blood triglyceride. The fatty acid mixtures, $\beta$ -sitosterol and its glucoside, p-coumaric acid, vanilic acid, ferulic acid, p-hydroxybenzaldehyde and sucrose were isolated. Sterol and p-coumaric acid were shown to be effective in reducing the elevated level of triglyceride. The wet weight and the level of triglyceride in epididymal adipose tissue increase following administration of the methanol extract in rats fed the purified diets at the 1% level for 4 weeks. This results suggest that the methanol extract may accelerate the accumulation of lipid in adipose tissue.

Hesperetin Ameliorates Inflammatory Responses in Lipopolysaccharide-stimulated RAW 264.7 Cells via p38 MAPK and ERK1/2 (마우스 대식세포 RAW 264.7 세포주에서 hesperetin에 의한 p38 MAPK와 ERK1/2를 통한 염증반응 조절)

  • In a previous study, we isolated 11 different kinds of compounds from ethyl acetate fractions of lees (jubak) which is a by-product of Korean traditional wine production. These compounds were identified as caffeic acid, coumaric acid, D-mannitol, ferulic acid, hesperetin, hesperidin, naringenin, naringin, sinapic acid, syringic acid, and vanilic acid. To evaluate their anti-inflammatory activities in an in vitro model, nitric oxide (NO) production was measured in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells after the treatment of these cells with each compound. Among the various chemicals, hesperetin and naringenin showed the highest inhibition of NO production in the LPS-activated RAW 264.7 cells. Hesperetin was chosen for further study because of its strong anti-inflammatory activity and because the mechanisms underlying its anti-inflammatory properties still remain unclear. Our results showed that hesperetin dramatically inhibited NO production in a dose-dependent manner as compared with in an LPS-only treated group, without affecting cell viability. In addition, hesperetin reduced the protein expression of the pro-inflammatory gene inducible nitric oxide synthase (iNOS) in a dose-dependent manner, whereas it did not affect cyclooxygenase-2 (COX-2) expression. Furthermore, hesperetin inhibited phosphorylation of p38 mitogen- activated protein kinase (MAPK) and extracellular signal regulated kinase (ERK) 1/2, whereas it did not affect phosphorylation of c-jun N- terminal kinase (JNK). The results indicated that hesperetin regulated the LPS-induced inflammatory response by suppressing p38 MAPK and ERK1/2 signaling. Overall, our results may help to understand the mechanisms underlying the anti-inflammatory activity mediated by hesperetin.

  • The 12 compounds were isolated from MeOH extract of Magnolia biondii and their structures were identified as seven lignans, two phenolics, one coumarin, and two flavonoid compounds, respectively. Among these constituents, tiliroside (3), kaempferol-7-methyl ether (4), 4-hydroxybenzoic acid (5), vanilic acid (6), and scopoletin (9) were isolated from Magnolia biondii for the first time.

Effects of Vaniltic Acid on the Cell Viability and Melanogenesis in Cultured Human Skin Melanoma Cells Damaged by ROS-Induced Cytotoxicity

  • The purpose of this study was to examine the effect of vanillic acid on the cell viability and melanogenesis in melanocytes damaged by reactive oxygen species (ROS). The human skin melanoma cells (SK-MEL-3) were cultured with various concentrations of hydrogen peroxide $(H_2O_2)$. The cell viability for $H_2O_2$-induced cytotoxicity or vanillic acid against $H_2O_2$ was measured by XTT assay in these cultures. For the effect of vanillic acid on the melanogenesis, the tyrosinase inhibitory activity was measured by colorimetric assay at a wavelength of 490 nm, and melanin synthesis activity were assessed after cells were cultured in the media with or without various cencentrations of vanillic acid. In this study, $H_2O_2$ decreased cell viability dose- and time-dependent manners and $XTT_{50}$ was determined at a concentration of 80 ${\mu}M$, $H_2O_2$. Vanillic acid increased the cell viability dose dependently in human skin melanoma cells damaged by $H_2O_2$-induced cytotoxicity. In the tyrosinase inhibitory activity, vanillic acid supresssed tyrosinase activity in dosedependent manner, and also decreased significantly melanin synthesis activity compared with $H_2O_2$-treated group. From these results. It is suggested that $H_2O_2$-mediated cytotoxicity was highly by the toxic criteria of Borenfreund and Puerner and also, vanillic acid has the protective effect on ROS-induced cytotoxicity and melanogenesis in these cultures.

A study on Resistant Substance to Pine Needle Gall Midge Among Phenolic Compounds in Pine Needles (소나무류(類) 침엽내(針葉內)에 함유(含有)된 phenolic compounds 중 솔잎혹파리의 저항성(抵抗性) 물질(物質) 조사(調査))

  • This study was carried out to study the resistant substances to pine needle gall midge and seasonal variation of phenolic compounds in pine needles among susceptible and resistant species to pine needle gall midge. Free and water soluble phenolic compounds contained in pine needles showed seasonal variations among pine species. But, catechol, vanillic acid and syringic acid including phenolic compounds showed no variation between pine species, even the amount of those component varied by the seasons. Salicylic acid in pine needles showed 140ppm in Pinus virginiana, 35ppm in Pinus thunbergii and 72ppm in hybrid pine, Pinus thunbergii${\times}$P. virginiana, but traces in Pinus densiflora on May and June, hatching season in larvae of pine needle gall midge. There was high level of salicylic acid in resistant species to pine needle gall midge but traces in susceptible species. As our previous researches, the necrosis rate of larvae of pine needle gall midge showed high level on the solution of phenolic compounds extracted from needles of P. virginiana and salicylic acid, and there was no gall formation in P. virginiana, so that salicylic acid in pine needles seems to have relation with those results. Therefore, it is estimated that salicylic acid is resistant substance to pine needle gall midge.

Quality characteristics of vinegar fermented with different amounts of black garlic and alcohol (흑마늘의 첨가량과 초기 알코올 농도를 달리한 발효 식초의 품질특성)

  • The quality characteristics of black garlic vinegar produced by acetic acid fermentation were analyzed over a 25 day period. Black garlic vinegar was produced using black garlic 3, 6, 9 and 12 folds with water, and initial alcohol concentrations of 6~12%. The pH values of the vinegar samples were in range of 3.85~4.46 after 25 days fermentation, and the sugar contents were highest after 9 days of fermentation ($12.00{\sim}13.50^{\circ}Brix$). The total polyphenol concents of vinegars were in the range of 20.65~52.04 mg/100 mL 25 days fermentation. Pyrogallol and vanillic acid were detected in the completely fermented vinegars, and their contents increased with black garlic content. Five organic acids (lactic acid, acetic acid, citric acid, succinic acid and propionic acid) were detected after 25 days of fermentation, with the total amount of organic acids being 5,327.34~6,307.64 mg/100 mL, and the total abetic acid contents beging the highest of the organic acids at 4208.90~5,570.29 mg/100 mL. After 25 days of fermentation, the total minerals contents were higher, and depending on the quantity of black garlic added. Of the minerals, the of potassium content was the highest (366.23~1,619.67 mg/L).

  • 꾸지뽕나무(Cudrania tricuspidata)는 뽕나무과(Moraceae)에 속하는 낙엽활엽 소교목이다. 우리나라와 중국, 일본과 같은 동아시아에 주로 분포하며 척박한 땅에서도 잘자라고 병충해에 강하다고 알려져 있다. 예로부터 꾸지뽕나무는 항암, 간보호, 눈을 밝게하는 작용이 있다고 동의보감과 신농본초경에 기록되어 있다. 우리나라는 오랫동안 된장 등 발효식품을 자주 접하고 섭취하여 왔다는 점을 고려하여 본 연구를 수행하였다. 발효균을 접종하면 항암활성, 면역체계 개선 등 다양한 생리활성 물질이 증가한다고 알려져 있다. 본 연구는 가시가 없고 잎이 커 작업성이 용이한 대품 품종을 2018년 9월에 전남 신안군에서 채취하여 분석 시료로 사용하였다. 항산화활성 측정은 프리라디칼(DPPH, ABTS) 소거능을 측정하여 농도(EC50)별 측정결과 $100{\mu}g/mL$ ext. 이하로 항산화 활성이 열매보다 잎이 높다는 것을 확인하였다. 황국균(Aspergillus oryae)을 꾸지뽕나무 잎과 열매에 접종시켜 페놀성화합물을 스크리닝 한 결과 기존에 발견되지 않은 Salicylic acid, Naringenin, Vanilic acid, Oxyresveratrol 등 기능성 물질이 발견되었고, 잎의 경우 36시간 발효물(355mg/g)은 무처리군(179mg/g)에 비해 2배정도 상승하였다. 열매의 경우 48시간 발효시켰을 경우(472mg/g)으로 무처리군(156mg/g)보다 3배정도 상승하였다. 발효를 통해 꾸지뽕나무 잎과 열매의 최적의 추출조건을 확립하고 생리활성 물질 분석을 이용한 효능탐색 등을 진행하였다. 향후 꾸지뽕나무를 활용한 식품 소재개발 등 사업화에 기초적인 자료를 제공하여 임업인의 새로운 소득품목 육성에 기어코자 한다.
