  • Electron microscopic examination of the liver and heart tissues of methylene blue-treated rats before gamma-irradiation was observed in this study. 1. It was observed severe alteration and degeneration of organelles: accumulation of glycogen particles, severe swollen mitochondria, and broken endoplasmic reticulum in liver tissue of saline-treated rat(control) opposed by emthylene blue-treated rat at 64 and 212 hours following gamma-irradiation. 2. Heart muscles of both methylene blue-treated and saline-treated rats showed no significant alterations, but it was observed that slightly elongated mitochondria with broken cristae and some of vacuoles as well as increased glycogen particles in sarcoplasmic reticulum at 212 hours following gamma-irradiation. 3. It may be considered that methylene blue greatly reduces the sensitivities of rats to gamma-irradiation.

  • Horsehair worms (Chordodes koreensis) develop as parasites in the bodies of grasshoppers, crickets, cockroaches, and some beetles. Chordodes koreensis is an accidental parasite of humans, livestock, or pets and poses no public health threat. The male of Chordodes koreensis in the later larval stage from canine vomitus was investigated by the scanning and transmission electron microscopy. In cross sections, the body wall is composed of four components namely epicuticle, cuticle, epidermis, and muscle layers. The parenchymal tissue fills the rest of the body and surrounds the visceral organs such as intestine, and ventral nerve cord but testes were not found. The epicuticle is a thin superficial layer whose surface shows rows of polygonal elevations called areoles. The cuticle has 17 layers of collagenous fibers spirally wound about the long axis of the worm. The section through the cuticle reveals the layers of large fibers cut obliquely lengthwise, alternating with layers of fibers sectioned obliquely crosswise. The layers of large fiber formed a double helix about longitudinal axis of the worm. The epidermis is a single layer. The muscles were interrupted by the nervous lamella in the only midventral portion. The medulla of muscle plate is composed of lightly stained cytoplasm, mitochondria, weakly developed endoplasmic reticulum, and glycogen granules. Between the medulla of a cell and the plasmalemma lies a broad cortical zone of myofilaments. The circular muscles are absent. The characteristic feature of the cytoplasm is that there was no content in peripheral mesenchyme, but was an abundance of large clear vacuoles which give the cytosome a foamy appearance. The nucleus of mesenchyme is not easily identified in our specimens.

  • In this study, the callus was induced and regenerated from the immature embryo and ultrastructural characteristics of developmental stages in Citrus junos SIEB, were investigated. The yellowish callus was induced by 5 to 6 week of culture of citrus. In proliferation callus after 6 weeks of culture, large vacuole was formed by fusion between adjacent small ones. In the non-embryogenic callus cultured for 12weeks, re-differentiated cells of callus showed the large nucleus with globular nucleus and amyloplast with large size of starches. In the embryogenic callus cltured for 14-16 weeks, the active exocytosis occurred in cells, secretory vesicles appeared on cell membrane and small particles from cytoplasm were released to intercelluar space. In the embryogenic callus cultured for 24 weeks, a sperical type of chloroplast bounded on cytoplasm by double membrane and typical grana was dispersed equally among matrix. In the normal plantlet after 26 weeks of culture, a lot of vessels and companion cells apperaed in the leaf cell of plantlet. In the normal plantlet after 30 weeks of culture, the immature leaf showed many small companion cells, sieve tubes and central vacuole. Also, the secondary vacuole protruded into the central vacuole and elongated chloroplasts near plasma membrane. In the matured plant habituated on the soil, palisada tissue composed of orderly arranged cells contained the nucleus in the center of the cell and large vacuoles on either side of the nucleus.

  • Chick embryos which have received a single injection of the organophosphate compound, malathion (0.1 mg, 0.5 mg, 1.0 mg or 2.0 mg in 0.05 ml of corn oil) via the yolk sac at certain times (2 daya, 4 days or 6 days after incubation) have been investigated. After 9 days of incubation, chick embryos were harvested to investigate the effects of malathion on the development of cerebrum morphologically and biochemically. The effects of simultaneous injection of malathion and nicotinamide were also compared. On ultrastructural examination, neurons in cerebral cortex showed to be inhibited in their differentiation by malathion; nuclear irregularity, swelling of endoplasmic reticulum, and cytoplasmic vacuoles were observed. On cytochemical study of acetylcholinesterase(AChE) by electron microscope, the positive reaction products of this enryme were localized at the membrane of nucleus and endoplasmic reticulum of neurons. Inhibition of AChE activty was severe in groups treated with relatively low doses, which was consistent with the results of spectrophotometric analysis. The activity of lactate dehydrogenase(LDH) of cerebrum in groups treated with malathion was higher than that of the control group. The nicotinamide adenine dinucleotide(NAD) content of chick embruo treated with malathioti decreased significantly, and nicotinamide coinjection raised the NAD level as compared with the control group, thus preventing malathion-induced momhological alteration. In conclusion, it is suggested that malathion changes the ultrastructure of differentiating neurons and alters some enzyme activities in chick embryo cerebrum, and the severity of which is consistently dose-or age-dependent.

  • Pneumconiosis is a sort of pulmonary fibrosis consequent to the inhalation of the respirable dusts. Thus, the pathogenesis of silicosis have concentrated largely on the early response of alveolar macrophage and the later fibroblastic stimulation. But the role of the other cells and continuing cell injury in the pathogenesis has not been fully studied. And the chemical factors such as prostaglandin, fibroblast stimulating factor and inhibiting factor and chemotaxin are also participated in the mechanism of pulmonary fibrosis in silicosis. In order to clarify the role of alveolar cells and prostaglandin, we investigated the changes of the cellularities in bronchoalveolar lavage fluid and tissue pathology in the experimental silicosis with the time sequence. The experimental animals were divided into 3 groups; control group received only intratracheal injection of 0.5 ml saline, silica group received the intratracheal instillation of 40 mg silica with the same amount saline, and aspirin group received 450 mg/kg of aspirin after silica instillation. The results were as follows: 1) The total cells of bronchoalveolar lavage fluid in the silica group markedly increased in comparison with the control group, but there was no significant difference between the silica and aspirin groups. 2) The percentages of alveolar macrophages to the total number of cells in the silica group tended to be lower than those in the control group and also lower than those in the aspirin group at the 1st week after silica instillation. 3) The percentages of neutrophils to the total number of cells in the silica group were significantly higher than those in the control group during the entire period and also higher than those in the aspirin group at the 3rd day after silica instillation. 4) In the silica group, the percentages of lymphocytes to the total number of cells were increased 143 progressively with the time course and those were significantly higher than those in the control group from the 3rd week after silica administration. There were marked differences of lymphocyte percentages between the silica and aspirin groups at the 1st week after silica instillation. 5) The inflammatory change was observed in the rat lung at the 1st day after silica instillation. Also the silicotic nodule appeared in the silica group at the 1st week but we could not find out that nodule in the aspirin group at that time. The fibrotic changes in the rat lung tended to be increased progressively with the time course, therefore, the diffuse fibrotic pattern appeared in the whole field at the 20th week after silica instillation. 6) By the electron microscopy, there were gradual increases of phagosomes and vacuoles in the alveolar macrophage in the silica group as compared with the control group. These results suggest that the neutrophils and the lymphocytes have also participated in the pulmonary fibrosis even though the alveolar macrophage has a major role, and prostaglandin mediate the inflammation and pulmanary fibrosis in the experimental silicosis.

  • The purpose of this study was to investigate the structure and secretory function of the von Ebner's gland in parasympathetic or sympathetic nerve innervation. Sprague Dawley rats were sacrificed 3, 7, 10, 14, and 21 days after bilateral glossopharyngeal or hypoglossal nerve axotomy, respectively. The circumvallate papilla portion of the tongue was dissected and we observed morphological changes in the von Ebner's gland. The properties of glycoconjugate in the von Ebner's gland were investigated using nine biotinylated lectins (PSA, UEA I, GSL I $B_4$, ECL, DBA, SBA, HPA, SJA, or sWGA). Compared with the control group, cytoplasmic vacuoles appeared in the serous acini of the von Ebner's gland in the 3-day group, and the serous acini were significantly vacuolized and degenerated in the 10-day group after glossopharyngeal nerve axotomy. However, the structure of the von Ebner's gland did not change after hypoglossal nerve axotomy. In the control group, the von Ebner's glands secreted glycoconjugates containing ${\alpha}$-D-galactose, N-acetyl-D-galactosamine, and N-acetyl-D-glucosamine oligomer, and the amount of the secretion decreased significantly in the 10-day group after glossopharyngeal nerve axotomy. However, the amount of the glycoconjugate secretion did not change after hypoglossal nerve axotomy. Therefore, the results of this study suggest that the glossopharyngeal nerve containing parasympathetic nerve fibers is important for maintaining the structure of and secretory function in the von Ebner's gland in rats.

  • The present study was designed in order to observe relationship between the endoplasmic reticulum and the Golgi complex during spermiogenesis of the long-fingered bat (Miniopterus schreibersi fuliginosus). The testes were obtained from adult bats and treated with the prolonged osmification or fixed with ferrocyanide reduced osmiun. In the Golgi phase, The Golgi complex shows an oval shape, and was composed of a cortex and a medullar enclosing acrosome. The Golgi vacuoles with electron-dense granules of crescent shape were fused with each other. The smooth endoplasrnic reticulum was scattered in all the area of the cytoplasm. In the cap phase, The Golgi complex was crescent in shape, and faced to a nucleus. Large and small vesicles were fused with each other, and then fused with a acrosomal vacuole. The rough endoplasmic reticulum was close to the large Golgi vacuole. In the acrosome phase, The Golgi complex was moved to behind of the acrosome face. Small vesicles were fused with an acrosome, and cisternae of the trans-face of Golgi complex was connected with an acrosome in the early acrosome phase. The smooth endoplasmic reticulum was distributed in the cytoplasm. The annulate lamellar was originated from a radial body-annulate lammellae complex. In the maturation phase, The Golgi complex with dilated cistrern appeared in the cytoplasm, and also, annulate lamellar was observed in the cytoplasm. The connection of the annulate lamellar with the cistern of radial body suggests that an annulate lamellar seems to be closely related to radial body. The smooth endoplasmic reticulum was scattered in the cytoplasm in the early Golgi phase, but annulate lamellar-radial body complex which might be a residual and disappearing form of the smooth endoplasmic reticulum appeared in the acrosome phase. The Golgi complex steadily remained in the late maturation phase when the endoplasmic reticulum began to disappear from the cytoplasm: the Golgi complex was still occurred after acrosome formation. The observations obtained in the present study, which was characterized by the presence of the Golgi complex in the late maturation phase, suggests that the Golgi complex may play an important role also even after the acrosome formation.

  • Human noroviruses (HuNoVs) are a leading cause of gastroenteritis outbreaks worldwide. However, the paucity of appropriate cell culture models for HuNoV replication has prevented developing effective anti-HuNoV therapies. In this study, first, the replication of the virus at various temperatures in different cells was compared, which showed that lowering the culture temperature from 37℃ significantly increased virus replication in Madin-Darby canine kidney (MDCK) cells. Second, the expression levels of autophagy-, immune-, and apoptosis-related genes at 30℃ and 37℃ were compared to explore factors affecting HuNoV replication. HuNoV cultured at 37℃ showed significantly increased autophagy-related genes (ATG5 and ATG7) and immune-related genes (IFNA, IFNB, ISG15, and NFKB) compared to mock. However, the virus cultured at 30℃ showed significantly decreased expression of autophagy-related genes (ATG5 and ATG7), but not significantly different major immune-related genes (IFNA, ISG15, and NFKB) compared to mock. Importantly, expression of the transcription factor FOXO1, which controls autophagy- and immune-related gene expression, was significantly lower at 30℃. Moreover, FOXO1 inhibition in temperature-optimized MDCK cells enhanced HuNoV replication, highlighting FOXO1 inhibition as an approach for successful virus replication. In the temperature-optimized cells, various HuNoV genotypes were successfully replicated, with GI.8 showing the highest replication levels followed by GII.1, GII.3, and GII.4. Furthermore, ultrastructural analysis of the infected cells revealed functional HuNoV replication at low temperature, with increased cellular apoptosis and decreased autophagic vacuoles. In conclusion, temperature-optimized MDCK cells can be used as a convenient culture model for HuNoV replication by inhibiting FOXO1 and providing adaptability to different genotypes.

  • This present study was designed to investigate by light microscope the morphologic changes in the uterine and vaginal epithelium of postpartum Korean native goats. Tissues were obtained for study on days 1, 3, 10 and 21 postpartum. The results obtained in this study were as followed; 1. Light microscopically, the height of the uterine epithelium was gradually decreased with the intervals and secretory profiles of apocrine were observed at between 1 and 10 days postpartum. The frequency of the light and dark cells with Masson's trichrom stain was high at 1 day postpartum an low at 21 days. A few of PAS positive cells were generally observed at 1 day postpartum, while PAS positive cells were not seen at 21 days. Numerous globule leucocytes were found between the epithelium and in the subepithelium at 1 day and thereafter moderate globule leucocytes were also found in the other periods. The intraepithelial vacuoles with crystalline structure appeared at 10 days postpartum. 2. The height of the vaginal mucosa was gradually decreased with the intervals but the highest layer was found at 3 days postpartum. The frequency of the mast cells was increased with times. At 3 and 10 days postpartum the shape of the surface-epithelium was cuboidal and the vacuolation of the epithelium was observed at 10 and 21 days.
