Hexavalent Chromium Reduction by Bacteria from Tannery Effluent

  • Chromium is generated from several industrial processes. It occurs in different oxidation states, but Cr(III) and Cr(VI) are the most common ones. Cr(VI) is a toxic, soluble environmental contaminant. Some bacteria are able to reduce hexavalent chromium to the insoluble and less toxic Cr(III), and thus chromate bioremediation is of considerable interest. An indigenous chromium-reducing bacterial strain, Rb-2, isolated from a tannery water sample, was identified as Ochrobactrum intermedium, on the basis of 16S rRNA gene sequencing. The influence of factors like temperature of incubation, initial concentration of Cr, mobility of bacteria, and different carbon sources were studied to test the ability of the bacterium to reduce Cr(VI) under variable environmental conditions. The ability of the bacterial strain to reduce hexavalent chromium in artificial and industrial sewage water was evaluated. It was observed that the mechanism of resistance to metal was not due to the change in the permeability barrier of the cell membrane, and the enzyme activity was found to be inductive. Intracellular reduction of Cr(VI) was proven by reductase assay using cell-free extract. Scanning electron microscopy revealed chromium precipitates on bacterial cell surfaces, and transmission electron microscopy showed the outer as well as inner distribution of Cr(VI). This bacterial strain can be useful for Cr(VI) detoxification under a wide range of environmental conditions.

The effect of Macmoondongtang in Rat Exposed to Cigarette Smoke (맥문동탕 흡연에 관한 효과)

  • Cigarette is the risk factor of Lung cancer, chronic obstructive pulmonary disease and ischemic heart disease. So I have studied the effect of Macmoondongtang in rat exposed to cigarette smoke. Thirty-six healthy rat subjects were divided into 6 groups-the control group(sample I), the groups of administration of Platycodi Radix(sampleII), Adenophorae Radix(sampleIII), Liriopsis Tuber(sampleIV), Maximowicziae Fructus(sample V) and Macmoondongtang(sample VI), and expose to cigarette per day for 12 days. 1. On level of leckocyte are significant all the group of sampleII, sampleIII, sample IV, sample V, sample VI on 6th day and singificant the groups of sampleIV, sample V on 12th day. 2. On level of hemoglobin are singificant the groups of sample IV, sample V, sample VI, on 6th day, and significant sample II, sample III, sample IV, sample V, sample VI on 12th day. 3. On platelet level are significant all the group of sampleII, sample III, sample IV, sample V, sample VI on 12th day. 4. On SOD activity are significant all the groups of sample IV, sample VI, on 6th day, and significant sampleII, sample III, sample VI on 12th day. According to the results, Macmoondogtang is effective to risk of cigarette smoke.

Induction of Oxidative Stress by Hexavalent Chromium in Human Bronchial Epithelial Cells (BEAS-2B) (배양 기관지 상피세포(BEAS-2B cells)에서 6가 크롬에 의한 산화적 스트레스)

  • Chromium compounds are widely used in diverse industries including pigment manufacturing, painting, metal plating and leather tanning. With the wide uses of chromium, various adverse effects of the compounds on the environment and human health have been reported. Among them, hexavalent chromium [Cr (VI)], which is a carcinogenic heavy metal, has been widely studies. Epidemiological investigations have shown that respiratory cancers had been found in workers who had been occupationally exposed to Cr (VI). In this study, cell toxicity and induction of reactive oxygen species (ROS) by Cr (VI) (1, 2, 4, $8{\mu}M$) in cultured human bronchial epithelial cells were investigated. Exposure of the cells to Cr (VI) led to cell death, ROS increase, and cytosolic caspase-3 activation. The ROS increase was related with the decreased level of GSH. Chromatin condensation and fragmentation were occurred by Cr (VI) when evaluated by DAPI staining or agarose gel electrophoresis of the extracted DNA. Expression of ROS related genes including glutathione S-transferase, heme oxygenase-1, metallothionein were significantly induced in Cr (VI) treated cells. This result suggests the toxicity in cultured cells by Cr (VI) was expressed through the apoptotic process with ROS induction.

Cr(VI) removal using Fe2O3-chitosan-cherry kernel shell pyrolytic charcoal composite beads

  • In this study, cherry kernel shell pyrolytic charcoal was synthesized (CKSC) and composite beads were obtained by blending this pyrolytic charcoal with chitosan and Fe2O3 nanoparticles (Fe-C-CKSC). Cr(VI) adsorption from aqueous solutions by Fe-C-CKSC composite beads and CKSC adsorbents was studied comparatively. The effects of Cr(VI) initial concentration, adsorbent dosage, contact time, pH and temperature parameters on Cr(VI) adsorption were investigated. Adsorption reached an equilibrium point within 120 min for CKSC and Fe-C-CKSC adsorbents. The maximum Cr(VI) removal was obtained at the initial pH value of 1.56 for CKSC and 2.00 for Fe-C-CKSC. The optimum adsorbent dosage was found to be 5 g/L for CKSC and 3 g/L for Fe-C-CKSC. Based on the Langmuir model, the maximum adsorption capacities were calculated as 14.455 mg/g and 47.576 mg/g for CKSC and Fe-C-CKSC, respectively. Thermodynamic and kinetic studies were performed. As a result of adsorption kinetics calculations, adsorption was found to be consistent with the pseudo second order kinetic model. Characterization of the synthesized adsorbents was performed by SEM, BET, FTIR and elemental analysis. This study has shown that low cost adsorbents CKSC and Fe-C-CKSC can be used in Cr(VI) removal from aqueous solutions.

Reduction of Hexavalent Chromium Collected on PVC Filters in Field Electroplating Process (현장 도금 공정에서 PVC 여과지에 채취된 6가 크롬의 환원)

  • Recently, pilot studies showed an evidence of reduction of airborne hexavalent chromium, Cr(VI), on PVC filter during air sampling and storage. However, the information on this in the field was limited. Thus, we studied the reduction behaviors of airborne Cr(VI) on PVC filters during sampling and storage at three field electroplating operations. Regression between sampling time and the reduction (ratio of Cr(VI) to total Cr concentrations) was not statistically significant (p>0.05). However, the reductions in samples collected for 240 ~ 340 minutes were significantly higher than those for 30 - 60 minutes. On the other hand, another experiment showed a good correlation (r=0.96) between sampling time and the reduction without an exceptional value. Storage temperature was not a factor affecting the reduction of Cr(VI) collected on PVC filter. The loss of Cr(VI) samples stored in alkali solution (2% NaOH/3% Na$_2$CO$_3$) was significantly lower than that stored in vial according to NIOSH method (p<0.05). Thus, dipping Cr(VI) samples into alkali solution was a storage method to minimize tile reduction.

Ionic Equilibria and Ion Exchange of Molybdenum(VI) from Strong Acid Solution

  • Ion exchange experiments of molybdenum(VI) from strong HCl and $H_2SO_4$ solution have been done to investigate the existence of anionic complexes. The concentration of HCl and $H_2SO_4$was changed from 1 to 9 M. From the data on the complex formation of molybdenum in aqueous solution, a new distribution diagram of Mo(VI) was constructed in the pH range from zero to 10. AG 1 X-8, an anion exchange resin, and Diphonix, a cation exchange resin were used in the loading experiments. Ion exchange results indicate that anionic complexes of Mo(VI) begins to form from 3 M HCl and 1 M $H_2SO_4$ solution and the tendency to form anionic complexes is stronger in HCl than in $H_2SO_4$ solution. Our results can be utilized in the analysis of Mo(VI) in strong acid solution and in the design of a process to separate Mo(VI).

Degradation characteristics and intermediate study of tetracycline in aqueous system by liquid ferrate(VI) (Liquid ferrate(VI)에 의한 다양한 수중 환경에서의 tetracycline 분해특성 및 중간생성물 연구)

  • Tetracycline is one of the most commonly used antibiotics in domestic and foreign livestock industries to suppress the growth of pathogens. Tetracycline has been reported as a non-biodegradable compound. Therefore it has been not completely removed in the sewage treatment process. In this study, tetracycline was degraded using liquid ferrate (VI). Based on these experiments, the optimal water condition (pH and water temperature) were selected, appropriate liquid ferrate (VI) dosage was calculated, and finally the degradation pathway was estimated with the intermediate products detected by LC/MS/MS. All degradation experiments were completed within 30 seconds and the optimal condition was obtained in basic condition (pH 10) at room temperature (20℃). And the appropriate molar ratio between tetracycline and liquid ferrate (VI) was 12.5:1. Finally, 12 intermediate products were detected with LC/MS/MS and the degradation pathways and the degradation pathways and proposed the degradation pathways.

  • The chelating agent with $\beta$-diketo funtional group, 1-phenyl-3-methyl-4-acyl-pyrazolone-5-one, has been used in separating and extracting radionuclides in a waste solution. The derivatives of this pyrazolone compound, prepared by different acyl groups, were synthesized and examined to figure out the extracting ability for Uranium (VI) and Zirconium (IV). The product prepared with succinic anhydride, called succinyl pyrazolone, showed excellent extraction for uranium (VI) in a chloroform solvent system. This result indicates that acyl pyrazolones having carboxylic acid group as a functional group forming $\beta$-diketo functionality are very selective for uranium (VI) and generally other metal ions with high valency.

Efficient Removal of Sulfamethoxazole in Aqueous Solutions Using Ferrate (VI): A Greener Treatment

  • The aim of this research is to assess the use of high purity potassium ferrate (VI) for the efficient removal of sulfamethoxazole (SMX), one of the potential micro-pollutant found in aqueous waste. In addition, various parametric studies have enabled us to deduce the mechanism in the degradation process. The pH and concentration of sulfamethoxazole enable the degradation of pollutants. Moreover, the time-dependent degradation nature of sulfamethoxazole showed that the degradation of ferrate (VI) in presence of sulfamethoxazole followed the pseudo-second order kinetics and the value of rate constant increased with an increase in the SMX concentration. The stoichiometry of SMX and ferrate (VI) was found to be 2 : 1 and the overall rate constant was estimated to be 4559 L2/mmol2/min. On the other hand, the increase in pH from 8.0 to 5.0 had catalyzed the degradation of SMX. Similarly, a significant percentage in mineralization of SMX increased with a decrease in pH and concentration. The presence of co-existing ions and SMS spiked real water samples was extensively analyzed in the removal of SMX using ferrate (VI) to simulate studies on real matrix implication of ferrate (VI) technology.

Simultaneous Removal of Cd & Cr(VI) by Fe-loaded Zeolite in Column System (Fe-loaded zeolite를 이용한 칼럼 실험에서의 Cd & Cr(VI) 동시제거 반응성 평가)

  • Laboratory column experiment for simultaneous removal of Cd and Cr(VI) were conducted using newly developed material of Fe-loaded zeolite having both reduction ability and sorption capacity. The solution containing Cd and Cr(VI) was injected into the column and the breakthrough curves (BTCs) for the contaminants were observed at the effluent port. Cd breakthrough was not initialized until Cr(VI) breakthrough was completed. Therefore it could be concluded that overall efficiency of Fe-loaded zeolite should be determined by the reactivity for Cr(VI). The relative concentration of Cr(VI) BTC increased to the unit value while initial breakthrough was delayed and the propagation of breakthrough was slowed. In order to quantitatively describe the shape of Cr(VI) BTC, new parameters of ${\alpha}\;and\;{\beta}$ designated to be shape parameters, were defined and applied in contaminant transport concentration. These parameters were employed to represent the degree of initial breakthrough delay and the degree of breakthrough propagation, respectively. As initial contaminant concentration increased, ${\alpha}$ decreased, which indicated the delay of BTC's initiation. And as initial contaminant flow rate increased, ${\beta}$ decreased, which represented the faster propagation of the BTC. From these results, Fe-loaded zeolite was found to be an effective reactive material for PRBs against heavy metals having different ionic forms in groundwater. And it could be expected that as groundwater flows faster, the propagation of breakthrough would be faster and as contaminant concentration is higher, the initial point of breakthrough would appear earlier.
