Effect of Different Silages for TMR on In vitro Rumen Simulative Fermentation

  • In this study, the in vitro fermentation parameters of whole crop barley (WCBS-TMR) and Italian ryegrass (IRGS-TMR) silage total mixed rations were compared. A rice straw based diet (RSBD), which was a mixture of rice straw and concentrate (60:40), was used as the control. The feeds were incubated in buffered rumen fluid for 3, 6, 9, 12, 24, 48 and 72 hours at $39^{\circ}C$. At the end of each incubation period the following parameters were determined, total gas, pH, ammonia nitrogen ($NH_3$-N), volatile fatty acids (VFA) and then the acetate to propionate ratio (A/P) was calculated. The dietary treatments did not affect (p>0.05) the overall production of $NH_3$-N, gas, total VFA and all the individual VFA, with the exception of n-butyrate (p<0.001). The treatment diets significantly affected the A/P ratio (p<0.01). The control diet resulted in the lowest A/P ratios, followed by WCBS-TMR and lastly IRGS-TMR had the highest ratios. Gas production was not different between treatments, suggesting a probable similar level of digestibility when treatments are fed to animals. It can therefore be concluded from the present study that WCBS and IRGS are of almost an equivalent nutritional value when incubated in a TMR form. WCBS-TMR however resulted in lower A/P ratios than IRGS-TMR, which is indicative of a more energy efficient diet.

생육 후기 거대억새의 In vitro 반추위 발효특성 및 건물 소화율 (Effect of Mature Miscanthus sacchariflorus var. No. 1 on In Vitro Rumen Fermentation Characteristics and Its Dry Matter Digestibility)

  • 본 연구는 반추동물의 조사료 자원으로서 거대억새를 개발하기 위한 목적으로 수행되었다. 우리나라에서 새롭게 개발된 품종인 거대억새 1호를 완숙기 이후에 채취하여 in vitro 반추위 발효를 이용해 반추위내 pH, 암모니아태 질소, 가스발생량, 휘발성 지방산 생성량 및 건물소화율을 조사하였으며, 볏짚과 비교하여 평가하였다. 거대억새는 볏짚에 비하여 유의적으로 높은 반추위내 pH를 나타내었다(p<0.01). 암모니아태 질소의 경우 배양 12시간 이후에는 두 처리구간의 유의적인 차이를 나타내지 않았다 (p>0.05). 배양 6시간 이후 부터는 거대억새의 가스발생량이 볏짚에 비하여 유의적으로 낮게 나타났다 (p<0.05). 휘발성 지방산 생성량에 있어 acetate, propionate, butyrate, valerate 및 총생상량에서 볏짚이 거대억새보다 높게 나타났다. 그러나 iso-butyrate와 iso-valerate에서는 두 조사료원별 차이는 발견되지 않았다. 건물소화율에 있어 배양 12~24시간 사이의 거대억새 소화율이 볏짚에 비하여 유의적으로 나타났다. 결론적으로 거대억새의 이용성은 볏짚의 약 80% 수준인 것으로 나타났다.

Comparison of in vitro ruminal fermentation between different originated corn grains

  • This study was conducted to compare in vitro rumen fermentation characteristics among corn grains imported from America, Brazil, Argentina and Ukraine A and Ukraine B. Two Holstein steers, each surgically fitted with a ruminal cannula, consuming total mixed ration were used as rumen fluid donors. In vitro rumen fermentation experiments were performed in a completely random design which included a control (no corn) and treatments with 3.0 g of corn from different geographical origins, i.e., America, Brazil, Argentina, and Ukraine A and Ukraine B, respectively. Ruminal pH, ammonia-N, volatile fatty acid (VFA) and total gas production were measured at 0, 1, 3, 6, 12, 24 and 48 h post-incubation, respectively. No differences (p > 0.05) in mean ruminal pH appeared among the treated groups, however, ruminal pH patterns differed; i.e. corn treated groups had dramatically lower pH compared with control during the entire incubation period. Similarly, no different patterns between the groups in ammonia-N (p > 0.05) appeared until 6 h post-incubation. Unexpectedly, higher ammonia-N concentration for control than that for the corn treated groups appeared after 12 h post-incubation despite that for all groups increased. Total VFA was similar between the groups until 6 h post-incubation, but VFA after 12 h post-incubation was different (p < 0.05), i.e. VFA for corn from Argentina, Ukraine A, Ukraine B, and Brazil were comparatively higher than for America. Overall, data in this study showed that the corns of different origins may have different feed values to ruminants despite having similar chemical compositions.

Comparative Analysis of Performance and Microbial Characteristics Between High-Solid and Low-Solid Anaerobic Digestion of Sewage Sludge Under Mesophilic Conditions

  • High-solid anaerobic digestion of sewage sludge achieves highly efficient volatile solid reduction, and production of volatile fatty acid (VFA) and methane compared with conventional low-solid anaerobic digestion. In this study, the potential mechanisms of the better performance in high-solid anaerobic digestion of sewage sludge were investigated by using 454 high-throughput pyrosequencing and real-time PCR to analyze the microbial characteristics in sewage sludge fermentation reactors. The results obtained by 454 highthroughput pyrosequencing revealed that the phyla Chloroflexi, Bacteroidetes, and Firmicutes were the dominant functional microorganisms in high-solid and low-solid anaerobic systems. Meanwhile, the real-time PCR assays showed that high-solid anaerobic digestion significantly increased the number of total bacteria, which enhanced the hydrolysis and acidification of sewage sludge. Further study indicated that the number of total archaea (dominated by Methanosarcina) in a high-solid anaerobic fermentation reactor was also higher than that in a low-solid reactor, resulting in higher VFA consumption and methane production. Hence, the increased key bacteria and methanogenic archaea involved in sewage sludge hydrolysis, acidification, and methanogenesis resulted in the better performance of high-solid anaerobic sewage sludge fermentation.

Molecular Weight, Protein Binding Affinity and Methane Mitigation of Condensed Tannins from Mangosteen-peel (Garcinia mangostana L)

  • The objectives of this study were to determine the molecular weight of condensed tannins (CT) extracted from mangosteen (Garcinia mangostana L) peel, its protein binding affinity and effects on fermentation parameters including total gas, methane ($CH_4$) and volatile fatty acids (VFA) production. The average molecular weight ($M_w$) of the purified CT was 2,081 Da with a protein binding affinity of 0.69 (the amount needed to bind half the maximum bovine serum albumin). In vitro gas production declined by 0.409, 0.121, and 0.311, respectively, while CH4 production decreased by 0.211, 0.353, and 0.549, respectively, with addition of 10, 20, and 30 mg CT/500 mg dry matter (DM) compared to the control (p<0.05). The effects of CT from mangosteen-peel on in vitro DM degradability (IVDMD) and in vitro N degradability was negative and linear (p<0.01). Total VFA, concentrations of acetic, propionic, butyric and isovaleric acids decreased linearly with increasing amount of CT. The aforementioned results show that protein binding affinity of CT from mangosteen-peel is lower than those reported for Leucaena forages, however, the former has stronger negative effect on IVDMD. Therefore, the use of mangosteen-peel as protein source and $CH_4$ mitigating agent in ruminant feed requires further investigations.

양돈폐수로부터의 수소 생성 특성 (Characteristics of H2 Production from Swine wastewater)

  • The characteristics of $H_2$ production from livestock wastewater were investigated through an anaerobic acid forming process using Clostridium beijerinckii and the photosynthetic process using Rhodobacter sphaeroides. The submerged separation membrane was installed in the acid forming reactor, The photosynthetic process is composed of two reactors(photosynthetic reactor 1 and photosynthetic reactor 2) which is connected continually. The removal rate of COD and the production of volatile fatty acid(VFA) in the acid forming process were approximately 50% and 1000mg/L, respectively. The 70% of COD in the effluent of acid forming process was removed through the photosynthetic process. The production of $H_2$ in the photosynthetic reactor 1 and 2 was 50 and $25mLH_2/gVFA_{COD}$, respectively. The values of Y in acid forming reactor, photosynthetic reactor 1 and 2 was 0.2263, 0.0601 and 0.0393, respectively. The acetic acid and butyric acid produced in acid forming process were converted to $H_2$ by photosynthetic bacteria.

다양한 유기성 폐자원에서 바이오 수소 생성 연구 (Feasibility of fermentative bio-hydrogen production from different organic wastes)

  • 다양한 유기성 폐자원이 혐기성 소화 공정 수소 생성에 미치는 영향을 고찰하기 위해 혼합균주를 사용하여 혐기 발효 회분식 형태 연구하였다. 폐자원은 부패 과일 폐수로 각각 사과, 배, 옥수수가루 및 황산염이 함유된 유기폐수 (황산염 농도 <= 1000 mg/L)를 이용하였다. 최대 수소 생성(547.1 mL)은 부패된 사과 폐수에서 나타났으며, 유입 농도는 132.2 g COD/L이다. 황산염 함유 폐수 적용결과, 황산염 농도에 관계없이 수소가 발생하였고, 잔존황산염은 pH 5.3-5.5에서 96-98 %로 나타나 황산염 환원은 발생되지 않았다. 모든 실험 조건에서 유기산은 HBu > HAc > HPr로 발생하였다. 본 연구결과 다양한 유기 폐자원에서 발효공정을 통해 수소 발생이 가능한 것으로 사료된다.

Effects of Replacing Lucerne (Medicago sativa L.) Hay with Fresh Citrus Pulp on Ruminal Fermentation and Ewe Performance

  • Two studies were conducted to determine the effects of replacing 30% (% in diet DM) of lucerne (Medicago sativa L.) hay with citrus pulp in Merino ewe diets: i) an in vitro study which measured ruminal fermentation; and ii) an in vivo study in which twelve Merino ewes pre- and post-lambing were fed experimental diets in a cross-over design over 120 days to evaluate effects on ewe performance (i.e. DM intake, average daily gain (ADG) and wool growth). In both the in vitro and in vivo studies, the control treatment consisted of lucerne (91.3% in diet DM), lupins (8.3% in diet DM) and phosphate (0.42% in diet DM), while the citrus pulp treatment consisted of lucerne (57.7% in diet DM), lupins (9.5% in diet DM), phosphate (0.48% in diet DM) and fresh citrus pulp (32.3% in diet DM). Data were analysed using the mixed model procedure of SAS. In the in vitro study, gas production, total volatile fatty acid (VFA) yield, proportion of propionic acid to total VFA and in vitro dry matter digestibility (IVDMD) were higher (p<0.02) in the citrus pulp treatment compared to the control treatment. In contrast, in vitro ammonia production, pH and the acetate to propionate ratio were lower (p<0.03) for the citrus pulp treatment compared to the control treatment. In the in vivo study, DM intake of ewes fed the citrus pulp diet was lower than their control ewe counterparts throughout both the pre- and post-lambing periods (928.9 vs. 1,115.0 g/d pre-; 1,285.0 vs. 1,620.3 g/d post-lambing, p<0.01), however ADG was similar (p = 0.12). Wool growth parameters and lamb performance did not differ (p>0.32) between treatments. In summary, the in vitro study demonstrated that the replacement of 30% of a lucerne diet with fresh citrus pulp improved total VFA yield, increased total gas production and improved IVDMD, while decreasing the production of ammonia, acetic acid and rumen pH. In addition, the in vivo study demonstrated that the replacement of 30% of a lucerne diet with fresh citrus pulp pre- and post-lambing decreased intake but did not affect ewe performance in terms of ADG and wool growth. These findings, of course, would be of significant interest to sheep producers endeavouring to control cost of feed ingredients whilst maintaining productivity.

Influence of Sunflower Whole Seeds or Oil on Ruminal Fermentation, Milk Production, Composition, and Fatty Acid Profile in Lactating Goats

  • This study aimed to investigate the effect of sunflower seeds, either as whole or as oil, on rumen fermentation, milk production, milk composition and fatty acids profile in dairy goats. Fifteen lactating Damascus goats were divided randomly into three groups (n = 5) fed a basal diet of concentrate feed mixture and fresh Trifolium alexandrinum at 50:50 on dry matter basis (Control) in addition to 50 g/head/d sunflower seeds whole (SS) or 20 mL/head/d sunflower seeds oil (SO) in a complete randomized design. Milk was sampled every two weeks during 90 days of experimental period for chemical analysis and rumen was sampled at 30, 60, and 90 days of the experiment for ruminal pH, volatile fatty acids (tVFA), and ammonia-N determination. Addition of SO decreased (p = 0.017) ruminal pH, whereas SO and SS increased tVFA (p<0.001) and acetate (p = 0.034) concentrations. Serum glucose increased (p = 0.013) in SO and SS goats vs Control. The SO and SS treated goats had improved milk yield (p = 0.007) and milk fat content (p = 0.002). Moreover, SO increased milk lactose content (p = 0.048) and feed efficiency (p = 0.046) compared to Control. Both of SS and SO increased (p<0.05) milk unsaturated fatty acids content specially conjugated linolenic acid (CLA) vs Control. Addition of SS and SO increased (p = 0. 021) C18:3N3 fatty acid compared to Control diet. Data suggested that addition of either SS or SO to lactating goats ration had beneficial effects on milk yield and milk composition with enhancing milk content of healthy fatty acids (CLA and omega 3), without detrimental effects on animal performance.

Comparisons of In vitro Nitrate Reduction, Methanogenesis, and Fermentation Acid Profile among Rumen Bacterial, Protozoal and Fungal Fractions

  • The objectives were to compare the ability of various rumen microbial fractions to reduce nitrate and to assess the effect of nitrate on in vitro fermentation characteristics. Physical and chemical methods were used to differentiate the rumen microbial population into the following fractions: whole rumen fluid (WRF), protozoa (Pr), bacteria (Ba), and fungi (Fu). The three nitrogen substrate treatments were as follows: no supplemental nitrogen source, nitrate or urea, with the latter two being isonitrogenous additions. The results showed that during 24 h incubation, WRF, Pr and Ba fractions had an ability to reduce nitrate, and the rate of nitrate disappearance for the Pr fraction was similar to the WRF fraction, while the Ba fraction needed an adaptation period of 12 h before rapid nitrate disappearance. The WRF fraction had the greatest methane ($CH_4$) production and the Pr fraction had the greatest prevailing $H_2$ concentration (p<0.05). Compared to the urea treatment, nitrate diminished net gas and $CH_4$ production during incubation (p<0.05), and ammonia-N ($NH_3$-N) concentration (p<0.01). Nitrate also increased acetate, decreased propionate and decreased butyrate molar proportions (p<0.05). The Pr fraction had the highest acetate to propionate ratio (p<0.05). The Pr fraction as well as the Ba fraction appears to have an important role in nitrate reduction. Nitrate did not consistently alter total VFA concentration, but it did shift the VFA profile to higher acetate, lower propionate and lower butyrate molar proportions, consistent with less $CH_4$ production by all microbial fractions.
