  • The current environmental problem is that environmental pollution is accelerating due to the generation of large amounts of waste and indiscriminate consumption of energy. Fossil fuels, a representative energy production fuel, are burned in the process of producing energy, generating a large amount of greenhouse gases and eventually causing climate change. In addition, the amount of waste generated worldwide is continuously increasing, and environmental pollution is occurring in the process of waste treatment. One of the methods for simultaneously solving these problems is the energy recovery from and reduction of organic wastes. Sewage sludge generated in sewage treatment plants has been treated in various ways since ocean disposal was completely prohibited, but the amount generated has been continuously increasing. Since the sewage sludge contains a large amount of organic materials, it is desirable to recover energy from the sewage sludge and reduce the final discharged waste through anaerobic digestion. However, most of the excess sludge is a mass of microorganisms used in sewage treatment, and in order for the excess sludge to be anaerobically digested, the cell walls of the microorganisms must be destroyed first, but it takes a lot of time to destroy the cell walls, so high rates of biogas production and waste reduction cannot be achieved only by anaerobic digestion. Therefore, the pre-treatment process of solubilizing excess sludge is required, and the thermal solubilization process is verified to be the most efficient among various solubilization methods, and high rates of biogas production and waste reduction can be achieved by anaerobic digestion after destroying cell walls the thermal solubilization process. In this study, when pretreating TS 10% thickened excess sludge through a thermal solubilization system, a study was conducted on solubilization characteristics according to retention time and operating temperature variables. The experimental variables for the retention time of the thermal solubilization system were 30 minutes, 60 minutes, 90 minutes, and 120 minutes, respectively, while the operating temperature was fixed at 160℃. The soulbilization rates calculated through TCOD and SCOD derived from the experimental results increased in the order of 12.11%, 20.52%, 28.62%, and 31.40%, respectively. And the variables according to operating temperature were 120℃, 140℃, 160℃, 180℃, and 200℃, respectively, while the operating retention time was fixed at 60 minutes. And the solubilization rates increased in the order of 7.14%, 14.52%, 20.52%, 40.72%, and 57.85%, respectively. In addition, TS, VS, T-N, T-P, NH4+-N, and VFAs were analyzed to evaluate thermal solubilization characteristics of thickened excess sludge. As a result, in order to obtain 30% or more solubilization rate through thermal solubilization of TS 10% thickened excess sludge, 120 minutes of retention time is required when the operating temperature is fixed to 160℃, and 170℃ or more of operating temperature is needed when the operating time is fixed to 60 minutes.

  • A study was conducted to determine the effect of alcohol-fermented feedstuff formulated with byproducts on the fermentation characteristics and dry matter disappearance in the rumen. Dietary treatments were either a soybean curd-based alcohol-fermented feedstuff(AFS) and brewery grain-based alcohol-fermented feedstuff(AFB). The AFS and AFB are composed of 50% commercial beef cattle feed, 50% soybean curd dreg, 5% molasses and 0.5% yeast, and 25% commercial beef cattle feed, 25% brewery grain, 25% soybean curd dreg, 25% corn grit, 5% molasses and 0.5% yeast, respectively. The ruminally cannualted Korean cattle were utilized to investigate the change of ammonia, pH alcohol, volatile fatty acids, and DM digestibility at 0, 2, 4, 8 and 12 hr after feeding. The rumen ammonia concentrations were significantly lower in AFS and AFB with incubation time, especially at 6 hr incubation(AFS, 0.7 mg/dl; AFB, 1.5 mg/dl; control 2.5 mg/dl). Lower rumen pH was observed in AFS and AFB during the early stage of incubation, but no significant difference was found at late stage of incubation. The total VFA concentrations were not affected by diet treatments at 2 hr incubation time, but the concentration significantly decreased after that. The dry matter disappearance was significantly lower in AFS and AFB during the early stage of incubation. However, the dry matter disappearance of AFS and AFB was similar to that of control during the late stage of incubation. It is concluded that the industrial byproducts such as soybean curd dreg and brewery grain were effective materials to make an alcohol fermented feedstuffs and resulted in better fermentation characteristics in the rumen when both were applied to Hanwoo.

  • We evaluated the effects of adding fibrolytic enzyme into ruminant diets on ruminal fermentation (in vitro) and lactational performances of dairy cows (in vivo). Through the in vitro experiment that was carried out with different contents of NDF (34, 38, 43%) in diets, digestibilities of NDF in the rumen appeared not significantly different by the addition of enzyme but were different by NDF content in diets showing higher digestibility in NDF 43% diet. It could be attributed by the relatively higher amount of hemicellulose in the current experimental diets than in conventional diets that might have been digested easily by the addition of fibrolytic enzyme in the rumen. The addition of fibrolytic enzyme tended to increase NDF digestibilities to a little extent both in 0.05 and 0.1% enzyme levels. Ruminal pH, NH3-N concentrations and VFA production in the rumen were not affected by the addition of fibrolytic enzyme. Activities of CMCase and xylanase were higher in enzyme treated diets of both NDF 34 and 38%. In particular, the activities of xylanase that slowly decreased from 0 to 12 hr but rapidly after 24 hr indicates that the major action of the enzyme in the rumen occurs in early period of incubation. Through an in vivo experiment, fibrolytic enzyme addition into the diets of dairy cows indeed affected lactational performance of milk yield. The cows fed enzyme treated diets produced 8% (1.9kg/d) more amounts of milk than with no enzyme addition. Milk composition of milk fat and protein was not affected by enzyme addition. Overall, the results of this in vivo study indicates that fibrolytic enzyme can be used to improve milk production in lactating cows. In respect that animals in different treatments of this study had the same amounts of intake, the increased milk yield with enzyme addition may be attributed to the improved utilization of nutrients in the digestive tract.

  • An experiment was conducted to evaluate condensed molasses solubles(CMS, a by-product from monosodium glutamate production) as a source of nitrogen for ruminant with particular reference to its effects on microbial protein synthesis. Four non-lactating dairy cows fitted with rumen cannulas were used in a 4 ${\times}$ 4 Latin square with 14-day periods. The four treatments were (1) basal diet consisting of barley straw ad libitum and 3 kg/d of rolled barley, (2) basal diet plus 200 gld molasses and 300 g/d water, (3) basal diet plus 200 g/d molasses, 100 g/d CMS and 200 g/d water, (4) basal diet plus 200 g/d molasses, 200 g/d CMS and 100 g/d water. Ruminal pH remained at high levels and showed little variation during the day between treatments. The concentration of total and individual VFA in the rumen was similar between treatments. There was no difference in the concentration of ammonia in the rumen between treatments, although the intake of nitrogen in molassesl CMS mixture treatments was higher than that of control and molasses treatment. But there was a suggestion of an increased synthesis of microbial protein with the higher level of inclusion of CMS when the allantoin/creatinine ratio was used as an index of microbial protein production(P <0.10).

  • In this study, the experiment was carried out to produce methane by applying Semi-Continuous Leachate Recirculation Anaerobic Digestion System fed with source separated food waste from school cafeteria. There were two systems and each system consisted of a bioreactor and a liquid tank. Each bioreactor had a screen near the bottom of the reactor. 2.5L of separated liquid was transferred to the liquid tank for 30min each day by using a tubing pump and the liquid from the liquid tank was pumped to the bioreactor at the upper of the bioreactor as soon as the transfer was ended. Through this circulation, the liquid having high concentration of VFAs was supplied to the top of bioreactor. At the beginning of the experiment, food waste/inoculum anaerobic sludge volume ratio was 2:8 that is 9g VS/L of OLR(Organic Loading Rate). Feeding was conducted every two weeks. Experimental results showed that the contents of moisture, combustible matter, ash were 65.91%, 32.73%, and 1.36%, respectively. Two different food waste loading were studied. The average organic loading rates were 3.51g VS/d for System A and 3.86g VS/d for System B, respectively. The average produced methane based on food waste fed to bioreactor were observed as $6.30m^3CH_4/kgVS{\cdot}d$ for system A and $4.94m^3CH_4/kgVS{\cdot}d$ for System B, respectively.

  • This study was conducted to investigate the effects of the addition of activated charcoal (AC) and oak charcoal on the ruminal fermentation characteristics, nutrient utilization in Korean native goats. Compared to reference diet, digestibility of dry matter, and crude protein in AC and charcoal diets tended to incraese. However, any tendency in ruminal degradation of crude fat was not observed. Ruminal degradation of NDF in AC diets tended to be more increased than that in non-AC diet. Although it was not significant, ADF tended to be increased in AC and charcoal diets. AC and charcoal did not affect the ruminal pH and ammonia-N. Concentration of total VFA and butyrate tended to be increased by adding AC and charcoal(p<0.05). Although it was not significant, acetate/propionate ratio tended to decreased in AC and charcoal diets. Although there appeared some beneficial effects in adding AC and charcoal to ruminant diets in this study, more works could be needed with AC before we can make clear conclusion on use of AC and charcoal in the ruminant diets.

  • An in vitro study was conducted to examine the effects of the addition of activated charcoal (AC) on the ruminal fermentation characteristics, nutrient disappearance, and ruminal gas production. AC was added at the levels of 0.00, 0.25, and 0.50 % to each of the four types of diets (roughage/concentrate ratio : 8/2, 6/4, 4/6 and 2/8), respectively. Although not significant, ruminal pH tended to increase by adding AC, and as the concentrate level increased, ruminal pH decreased (P<0.05). Acetate concentration and acetate/propionate molar ratio tended to decrease in AC diets. but molar % of propionate tended to increase by the addition of AC. Ruminal degradation of dry matter, crude protein, NDF, ADF, and hemicellulose in AC diets tended to increase than in non-AC diet, however, no tendency in ruminal degradation of crude fat was observed. As the concentrate level increased, rumunal degradation of dry matter and nutrients in AC diets increased significantly(P<0.05). Ruminal gas production tended to decrease in the 0.50 % AC diets, however, it tended to increase in high roughage diets. Although there appeared some beneficial effects in adding AC to ruminant diets in this study, more works should be done with AC before we can make clear conclusion on the use of AC in the ruminant diets.

  • Anaerobic digestion (AD) is one of the most widely used process that can convert the organic fraction of waste activated sludge (WAS) into biogas. However, most researched actual methane yields of anaerobic digester (AD) on lab scale is lower than theoretical ones. Bioelectrochemical, anaerobic digester was used to increase methane yield from waste activated sludge. The influence of anaerobic digestion sludge and raw sludge mixing ratio (3:7, 5:5) on methane yield and organic matter removal efficiency were explored. As a result, when the mixing ratio of bioelectrochemical anaerobic sludge was 5:5 compared with 3:7, the highest methane yields were 294.2 mL $CH_4/L$ (0.63 times increase) and 52.5% (7.5% increase), the bioelectrochemical anaerobic digester(5:5) was more stable in the pH, t otal alkalinity and VFAs, respectively. These results showed that the increase in the mixing ratio of anaerobic digestion sludge was found to be effective for maintaining the stable performance of bioelectrochemical anaerobic digester.

  • The present study was conducted to examine the fermentation characteristics and effective degradability (ED) in the rumen, and whole tract digestibility of whole crop silage based TMR in comparison with conventional separate feeding of concentrate and roughage. Three ruminally fistulated non-lactating Holstein cattle were used in a 3 $\times$ 3 Latin square design. The cattle were fed 8kg of whole crop barley silage based TMR (BS-TMR) or 8kg (DM basis) of whole crop rye silage based TMR (RS-TMR) twice (08:00 and 18:00) daily in an equal amount. The cattle were also fed concentrate (5.6kg) and rice straw (1.4kg) seperately (DM basis, Control) twice daily in an equal amount. The both silages were included in TMR at 20% level (as fed basis). pH in the rumen fluid was not influenced by the diets but was slightly higher from TMR than from control. No difference was found in ammonia-N concentration between diets. Total VFA concentration was relatively increased in the cattle fed RS-TMR to the other diets up to 6h post feeding. The proportion of acetate was increased in the TMR feeding at right before feeding (0 h, p<0.005) and 9 h (p<0.048) post feeding compared with control. Propionate proportion was increased (p<0.046) in both TMRs while butyrate proportion was increased (p<0.029) at 1h post feeding compared to other diets. Effective degradability (ED) of DM and CP of RS-TMR was relatively increased to other diets, and EDNDF of both TMRs was higher than that of control diet due to the increased parameters b (p<0.039) and c (p<0.006) in TMR treatments. Whole track digestibility of most components in the TMRs was slightly increased compared to that in control diet, and RS-TMR had a tendency to be increased whole track digestibility except for NDF compared to BS-TMR. Based on the results observed from the present study, nutrient availability of whole crop silage based TMR looked slightly better than conventional separate feeding of concentrate and rice straw, mainly due to the improved stabilization of fermentation in the rumen and increased NDF digestibility of whole crop silage in TMR.
