Noise Cancellation using Microphone Array in Digital Hearing Aids (디지털 보청기에서 마이크로폰 어레이를 이용한 잡음제거)

  • In this paper, a noise cancellation-method using microphone array for digital hearing aids is proposed. The microphone array is located around the ear of a dummy. Speech sound is generated from the forward speaker positioned in the front of the dummy and noise sound is generated from the backward speaker. The speech and noise are mixed in the air space and entered into the microphones. VAD(voice activity detector) and ANC(adaptive noise cancellation) methods were used to eliminate noise in the sound of the microphones. 10 two-syllable words and 4 sentences were used for speech signals. Babble and car interior noise were used for noise signals. The performance of the proposed algorithm was evaluated by SNR(signal-to-noise ratio) and PESQ-MOS(perceptual evaluation of speech quality-mean opinion score). In babble noise condition, SNR was improved as much as $7.963{\pm}1.3620dB\;and\;3.968{\pm}0.6659dB$ for words and sentences respectively. In the case of car interior noise, SNR was improved as $10.512{\pm}2.0665dB\;and\;6.000{\pm}1.7642dB$ for words and sentences respectively. PESQ-MOS of the babble noise was improved as much as $0.1722{\pm}0.0861$ score for words and $0.083{\pm}0.0417$ score for sentences. And PESQ-MOS of the car interior noise was improved as $0.2661{\pm}0.0335$ score and $0.040{\pm}0.0201$ score for words and sentences respectively. It is verified that the proposed algorithm has a good performance in noise cancellation of microphone array for digital hearing aids.

Speech Segmentation using Weighted Cross-correlation in CASA System (계산적 청각 장면 분석 시스템에서 가중치 상호상관계수를 이용한 음성 분리)

  • The feature extraction mechanism of the CASA(Computational Auditory Scene Analysis) system uses time continuity and frequency channel similarity to compose a correlogram of auditory elements. In segmentation, we compose a binary mask by using cross-correlation function, mask 1(speech) has the same periodicity and synchronization. However, when there is delay between autocorrelation signals with the same periodicity, it is determined as a speech, which is considered to be a drawback. In this paper, we proposed an algorithm to improve discrimination of channel similarity using Weighted Cross-correlation in segmentation. We conducted experiments to evaluate the speech segregation performance of the CASA system in background noise(siren, machine, white, car, crowd) environments by changing SNR 5dB and 0dB. In this paper, we compared the proposed algorithm to the conventional algorithm. The performance of the proposed algorithm has been improved as following: improvement of 2.75dB at SNR 5dB and 4.84dB at SNR 0dB for background noise environment.

Artemisia capillaris Thunb. inhibits cell growth and induces apoptosis in human hepatic stellate cell line LX2

  • Artemisia capillaris (A. capillaries) is known to play roles in many cellular events, such as cell proliferation, differentiation, and apoptosis. We investigated the antifibrogenic efficacy of A. capillaris in the immortalized human hepatic stellate cell line LX2. Cell proliferation was determined by the MTT assay. Cell cycle was analyzed by the flow cytometry. Apoptotic cells were measured using a cell death detection ELISA. Caspase activity was detected by a colorimetric assay. The mRNA level of Bcl-2 and Bax mRNA were measured by real-time PCR. MEK and ERK protein were detected by Western blot analysis. We provide evidence that A. capillaris induces cell cycle arrest, apoptosis, and potently inhibits the mitogen-activated protein kinase pathway. A. capillaris inhibited cell proliferation of LX2 cells in a dose- and time-dependent manner, increased the apoptosis fraction at cell cycle analysis with an accompanying DNA fragmentation, and resulted in a significant decrease in Bcl-2 mRNA levels and an increase in Bax expression. Exposure of LX2 cells to A. capillaris induced caspase-3 activation, but co-treatment of A. capillaris with the pan-caspase inhibitor Z-VAD-FMK, and the caspase-3 inhibitor Z-DEVE-FMK, blocked apoptosis. A. capillaris down-regulated Mcl-1 protein levels and inhibited phosphorylation of MEK/ERK, suggesting that it mediates cell death in LX2 cells through the down-regulation of Mcl-1 protein via a MEK/ERK-independent pathway.

Effects of TNFalpha, NOS3, MDR1 Gene Polymorphisms on Clinical Parameters, Prognosis and Survival of Multiple Myeloma Cases

  • It is not clear how gene polymorphisms affecting drugs can contributes totheir efficacy in multiple myeloma (MM). We here aimed to explore associations among gene polymorphisms of tumor necrosis factor alpha (TNFalpha), nitric oxide synthesis 3 (NOS3) and multi-drug resistance 1 (MDR1), clinical parameters, prognosis and survival in MM patients treated with VAD (vincristine-adriamycine-dexamethasone), MP (mephalane-prednisolone), autolougus stem cell transplantation (ASCT), BODEC (bortezomib-dexamethasone-cyclophosphamide) and TD (thalidomide-dexamethasone). We analyzed TNFalpha, NOS 3 and MDR1 in 77 patients with MM and 77 healthy controls. The genotyping was performed with PCR and/or PCR-RFLP. There was no clinically significant difference between MM and control groups when TNFalpha (-238) and (-857) and MDR1 gene polymorphisms were studied. However, the TNFalpha gene polymorphism (-308) GG genotype (p=0.012) and NOS3 (+894) TT genotype (p=0.008) were more common in the MM group compared to healthy controls. NOS3 (VNTR) AA (p=0.007) and NOS3 (+894) GG genotypes (p=0.004) were decreased in the MM group in contrast. In conclusion, the NOS3 (+894) TT and TNFalpha (-308) GG genotypes may have roles in myeloma pathogenesis.

20(S)-Protopanaxadiol Induces Human Breast Cancer MCF-7 Apoptosis through a Caspase-Mediated Pathway

  • 20(S)-Protopanaxadiol (PPD), a ginsenoside isolated from Pananx quinquefolium L., has been shown to inhibit growth and proliferation in several cancer cell lines. The aim of this study was to evaluate its anticancer activity in human breast cancer cells. MCF-7 cells were incubated with different concentrations of 20(S)-PPD and cytotoxicity was evaluated by MTT assay. Occurrence of apoptosis was detected by DAPI and Annexin V-FITC/PI double staining. Mitochondrial membrane potential was measured with Rhodamine 123. The Bcl-2 and Bax expression were determined by Western blot analysis. Caspase activity was measured by colorimetric assay. 20(S)-PPD dose-dependently inhibited cell proliferation in MCF-7 cells, with an $IC_{50}$ value of $33.3{\mu}M$ at 24h. MCF-7 cells treated with 20(S)-PPD presented typical apoptosis, as observed by morphological analysis in cell stained with DAPI. The percentages of annexin V-FITC positive cells were 8.92%, 17.8%, 24.5% and 30.5% in MCF-7 cells treated with 0, 15, 30 and $60{\mu}M$ of 20(S)-PPD, respectively. Moreover, 20(S)-PPD could induce mitochondrial membrane potential loss, up-regulate Bax expression and down-regulate Bcl-2 expression. These events paralleled activation of caspase-9, -3 and PARP cleavage. Apoptosis induced by 20(S)-PPD was blocked by z-VAD-fmk, a pan-caspase inhibitor, suggesting induction of caspase-mediated apoptotic cell death. In conclusion, the 20(S)-PPD investigated is able to inhibit cell proliferation and to induce cancer cell death by a caspase-mediated apoptosis pathway.

Robust Distributed Speech Recognition under noise environment using MESS and EH-VAD (멀티밴드 스펙트럼 차감법과 엔트로피 하모닉을 이용한 잡음환경에 강인한 분산음성인식)

  • The background noises and distortions by channel are major factors that disturb the practical use of speech recognition. Usually, noise reduce the performance of speech recognition system DSR(Distributed Speech Recognition) based speech recognition also bas difficulty of improving performance for this reason. Therefore, to improve DSR-based speech recognition under noisy environment, this paper proposes a method which detects accurate speech region to extract accurate features. The proposed method distinguish speech and noise by using entropy and detection of spectral energy of speech. The speech detection by the spectral energy of speech shows good performance under relatively high SNR(SNR 15dB). But when the noise environment varies, the threshold between speech and noise also varies, and speech detection performance reduces under low SNR(SNR 0dB) environment. The proposed method uses the spectral entropy and harmonics of speech for better speech detection. Also, the performance of AFE is increased by precise speech detections. According to the result of experiment, the proposed method shows better recognition performance under noise environment.

Treatment with a Small Synthetic Compound, KMU-193, induces Apoptosis in A549 Human Lung Carcinoma Cells through p53 Up-Regulation

  • Despite recent advances in therapeutic strategies for lung cancer, mortality still is increasing. In the present study, we investigated the anti-cancer effects of KMU-193, 2-(4-Ethoxy-phenyl)-N-{5-[2-fluoro-4-(4-methylpiperazine-1-carbonyl)-phenylamino]-1H-indazol-3-yl}-acetamide in a human non-small cell lung cancer cell line A549. KMU-193 strongly inhibited the proliferation of A549 cells, but it did not have anti-proliferative effect in other types of cancer cell lines. KMU-193 further induced apoptosis in association with activation of caspase-3 and cleavage of PLC-${\gamma}1$. However, KMU-193 had no apoptotic effect in untransformed cells such as TMCK-1 and BEAS-2B. Interestingly, pretreatment with z-VAD-fmk, a pan-caspase inhibitor, strongly abrogated KMU-193-induced apoptosis. KMU-193 treatment enhanced the expression levels of p53 and PUMA. Importantly, p53 siRNA transfection attenuated KMU-193-induced apoptosis. Collectively, these results for the first time demonstrate that KMU-193 has strong apoptotic effects on A549 cells and these are largely mediated through caspase-3- and p53-dependent pathways.

Over Expression of BCL2 and Low Expression of Caspase 8 Related to TRAIL Resistance in Brain Cancer Stem Cells

  • Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) has been investigated as an effective agent to treat various cancers. Cancer stem cells are resistant to TRAIL treatment, but the mechanism of TRAIL resistance remains unknown. In this study, brain cancer stem cells were isolated by CD133 magnetic sorting, and the number of CD133 positive cells detected by flow cytometry. The self-renewing capacity of brain cancer stem cells was examined by a neurosphere formation assay, and the percentage of cell death after TRAIL treatment was examined by an MTS assay. Expression of DR5, FADD, caspase 8 and BCL2 proteins was detected by western blot. The amount of CD133 positive cells was enriched to 71% after CD133 magnetic sorting. Brain cancer stem cell neurosphere formation was significantly increased after TRAIL treatment. TRAIL treatment also reduced the amount of viable cells and this decrease was inhibited by a caspase 8 inhibitor or by the pan-caspase inhibitor z-VAD (P<0.05). Brain cancer stem cells expressed lower levels caspase 8 protein and higher levels of BCL2 protein when compared with CD133 negative cells (P<0.05). Our data suggest that TRAIL resistance is related to overexpression of BCL2 and low expression of caspase 8 which limit activation of caspase 8 in brain cancer stem cells.

Voice Activity Detection Based on Entropy in Noisy Car Environment (차량 잡음 환경에서 엔트로피 기반의 음성 구간 검출)

  • Accurate voice activity detection have a great impact on performance of speech applications including speech recognition, speech coding, and speech communication. In this paper, we propose methods for voice activity detection that can adapt to various car noise situations during driving. Existing voice activity detection used various method such as time energy, frequency energy, zero crossing rate, and spectral entropy that have a weak point of rapid. decline performance in noisy environments. In this paper, the approach is based on existing spectral entropy for VAD that we propose voice activity detection method using MFB(Met-frequency filter banks) spectral entropy, gradient FFT(Fast Fourier Transform) spectral entropy. and gradient MFB spectral entropy. FFT multiplied by Mel-scale is MFB and Mel-scale is non linear scale when human sound perception reflects characteristic of speech. Proposed MFB spectral entropy method clearly improve the ability to discriminate between speech and non-speech for various in noisy car environments that achieves 93.21% accuracy as a result of experiments. Compared to the spectral entropy method, the proposed voice activity detection gives an average improvement in the correct detection rate of more than 3.2%.

  • The remote monitoring system including hemodynamic information and pump status of the implanted animal could be helpful during the in vivo experiment or clinical trial for an artificial heart Implantation. In order to monitor the course of the in vivo experiment continuously and anywhere, web-based remote monitoring system was developed, which can monitor pressures(AoP, LAP, RAP, PAP) and flow information as well as the pump operating conditions. The system consists of data sending, storing viewer part. The data sending part was constructed using component object model and the viewer part was constructed using the Java applet. In addition, the dialog box was introduced to communicate earth other instantly and the alarming function was also introduced when the hemodynamic values were out of the desired ranges. The developed remote monitoring system was applied during the in vivo experiment of the BVAD (Bi-ventricular Assist Device) implantation for 1 month and showed designed work without failure.
