Quinine Assay with Home-Built UV-LED Fluorometer: Quantitative Analysis, Photo-Bleaching, Fluorescence Quenching, and Urine Analysis

  • Quinine quantitative analysis, photo-bleaching, fluorescence quenching, and urine analysis have been performed by means of a UV-LED fluorometer, which can be easily built and used in a high-school laboratory. The quinine detection range is estimated to be 0.05-80 ppm, enough for many classroom luminescence experiments. The quinine content in commercial tonic water is determined from the calibration curve, and UV photo-bleaching of this anti-malarial drug is demonstrated with clear wavelength dependence. Halide quenching of quinine fluorescence is also observed and the increase in quenching efficiency in the order of $Cl^-$, $Br^-$, and $I^-$ is evident. Urine analyses for the student volunteers have been carried out and the results clearly reveal excretion of the ingested quinine. The student participants are exuberant throughout the course of this study and sense the practices resourceful.

  • In this paper, we suggested and made a classifier or qualitative and quantitative analysis in urine analysis system. Input variable number and fuzzy membership function was made from determination of standard sample, and the fuzzy rules were determined by the analysis of spectroscopic properties of pads in strip. Fuzzy classifier used in urine analysis system was evaluated or the standard samples in each items and degrees. Negative and positive response of urine test was classified in good property, but detail classification or quantitative analysis had 8% maximum error in each items. If fuzzy membership unction and generation of rule are supplemented, suggested fuzzy classifier can be applied to the clinical test.

Development of objective indicators for quantitative analysis of sodium intake: the sodium to potassium ratio of second-void urine is correlated with 24-hour urinary sodium excretion

  • BACKGROUND/OBJECTIVES: To date, sodium intake has been evaluated based on spot urine instead of 24-hour (hr) urine collection. Nevertheless, the optimal method for assessing daily sodium intake remains unclear. SUBJECTS/METHODS: Fifteen male (age 32.7 ± 6.5 years) participants were offered 3 meals with a total of 9-10 g salt over 24 hours, and 24-hr urine was collected from the second-void urine of the first day to the first-void urine of the second day. Twenty-four-hr urinary sodium (24UNa) was estimated using Tanaka's equation and the Korean formula, and spot urine Na, potassium (K), chloride (Cl), urea nitrogen (UN), creatinine (Cr), specific gravity (SG) and osmolality (Osm) were measured. The ratios of urinary Na to other parameters were calculated, and correlations with total measured 24UNa were identified. RESULTS: Average 24-hr urine volume was 1,403 ± 475 mL, and measured 24UNa was 143.9 ± 42.1 mEq (range, 87.1-239.4 mEq). Measured 24UNa was significantly correlated with urinary Na/UN (r = 0.560, P < 0.01), urinary Na/Osm (r = 0.510, P < 0.01), urinary Na/Cr (r = 0.392, P < 0.01), urinary Na/K (r = 0.290, P < 0.01), 24UNa estimated using Tanaka's equation (r = 0.452, P < 0.01) and the Korean formula (r = 0.414, P < 0.01), age (r = 0.548, P < 0.01), weight (r = 0.497, P < 0.01), and height (r = 0.393, P < 0.01) in all spot urine samples. Estimated 24UNa based on the second-void spot urine of the first day tended to be more closely correlated with measured 24UNa than were estimates from the other spot urine samples. The significant parameters correlated with the second-void urine of the first day were urinary Na/K (r = 0.647, P < 0.01), urinary Na/Cr (r = 0.558, P < 0.05), and estimated 24UNa using Tanaka's equation (r = 0.616, P < 0.05) and the Korean formula (r = 0.588, P < 0.05). CONCLUSIONS: Second-void urine is more reliable than first-void urine for estimating 24UNa. Urinary Na/K in the second-void urine on the first day is significantly correlated with 24UNa. Further studies are needed to establish the most reliable index and the optimal time of urine sampling for predicting 24UNa.

  • A simple and sensitive high performance liquid chromatography for analysis of Putrescine, Spermidine and Spermine in urine is described. The benzoyl chloride derivatives of Putrescine, Spermidine and Spermine are separated on a ${\mu}$ Bondapak $C_{18}$ reverse-phase column with methanol/$H_2O$(50/50v/v) as the mobile phase. The results show that the mean level of polyamines in uterine cancer patients urine is much higher than in normal human urine.

  • This study was undertaken to determined the sulfamethazine residues In urine, serum and muscle of slaughtered pigs(n: 230) by the method of TLC, EEC-4-plate and HPLC. 1. Inhibition diameter characteristics of reference bacterial substance by EEC-4-plate method and antibacterial inhibition zone was appeared sulfonamides in BS pH 7.2 2. Residual sulfamethazine was detceted from 7 serum(3.04%) and 10 urine(4.35%) by TLC test and concentration of residues was higher In urine than serum. 3. Residual sulfamethazine was detected from 9 muscle(3.91%) by EEC-4-plate method. 4. Positive samples by TLC test and EEC-4-plate method were exceed 0.1ppm quantitative analysis from 7 muscle(3.04%).

Analysis of metabolomic patterns in thoroughbreds before and after exercise

  • Objective: Evaluation of exercise effects in racehorses is important in horseracing industry and animal health care. In this study, we compared metabolic patterns between before and after exercise to screen metabolic biomarkers for exercise effects in thoroughbreds. Methods: The concentration of metabolites in muscle, plasma, and urine was measured by $^1H$ nuclear magnetic resonance (NMR) spectroscopy analysis and the relative metabolite levels in the three samples were compared between before and after exercise. Subsequently, multivariate data analysis based on the metabolic profiles was performed using orthogonal partial least square discriminant analysis (OPLS-DA) and variable important plots and t-test was used for basic statistical analysis. Results: From $^1H$ NMR spectroscopy analysis, 35, 25, and 34 metabolites were detected in the muscle, plasma, and urine. Aspartate, betaine, choline, cysteine, ethanol, and threonine were increased over 2-fold in the muscle; propionate and trimethylamine were increased over 2-fold in the plasma; and alanine, glycerol, inosine, lactate, and pyruvate were increased over 2-fold whereas acetoacetate, arginine, citrulline, creatine, glutamine, glutarate, hippurate, lysine, methionine, phenylacetylglycine, taurine, trigonelline, trimethylamine, and trimethylamine N-oxide were decreased below 0.5-fold in the urine. The OPLS-DA showed clear separation of the metabolic patterns before and after exercise in the muscle, plasma, and urine. Statistical analysis showed that after exercise, acetoacetate, arginine, glutamine, hippurate, phenylacetylglycine trimethylamine, trimethylamine N-oxide, and trigonelline were significantly decreased and alanine, glycerol, inosine, lactate, and pyruvate were significantly increased in the urine (p<0.05). Conclusion: In conclusion, we analyzed integrated metabolic patterns in the muscle, plasma, and urine before and after exercise in racehorses. We found changed patterns of metabolites in the muscle, plasma, and urine of racehorses before and after exercise.

Change of Laboratory Parameters during Treatment of Lead Poisoning (연중독치료시 혈중연, 뇨중연, 뇨중 Coproporphrin, 뇨중 ${\delta}$-Aminolevulinic acid의 변화)

  • In order to study the change of laboratory parameters of lead poisoning, 8 persona who had not been treated previously for lead poisoning (Group 1 and 6 persons who had been inadequately treated for few months for chronic lead poisoning at local clinic (Group 2) were examined. They had occupational exposure to lead for 3 to 18 years (mean, 7.6). In group 1 blood lead, urine lead, urine coproporphyrin and ${\delta}$-aminolevulinic acid levels before our treatment exceeded the critical levels of lead poisoning. In group 2 urine lead level exceeded but blood lead, urine coproporphyrin and ${\delta}$-aminolevulinic acid levels were within normal limits. All of them were treated with D-penicillamine for 4 months as inpatients at Industrial Accident Hospital. The dose of D-penicillamine was the same in all patients; 600 mg per day p.o. and the chelating agent was administer every other week. For laboratory analysis, 24 hour urine and 10 gm of whole blood were collected every 1 month on last day of non-administration period. The results were as follows: 1. It was found that urine lead level was decreased below the cirtical level of lead poisoning after 4 month's treatment with D-penicillamine and blood lead level was decreased more progressively below the critical level after 1 month treatment. 2. Urine coproporphyrin and ${\delta}$-aminolevulinic acid levels were decreased progressively to normal range after 1 month treatment. 3. Two months after treatment, blood lead, urine lead, urine coproporphyrin and ${\delta}$-aminolevulinic acid levels showed some increasing trends. 4. Urine lead level should be checked in a person who had been inadequately treated with chelating agents because blood lead, coproporphyrin and ${\delta}$-aminolevulinic acid might be in normal range.

  • In order to establish a new biochemical parameter to evaluate nuitrtitional status. the present paper presents a paper chromatographic procedure. which enables clear separation of the essential and nonessential free amino acids in the urine without desalting the sample prior to chromatographic analysis The children of school age. showing difference in their body physiques. from the rural and urban areas in Korea were selected in collecting the 24 hour urine specimens. with the following conclusions. (1) A new procedure utilyzing paper chromatographic analysis. without desalting the urine. was proposed to establish a new biochemical parameter for nutritional status by analyzing the ratio of the nonessential to the essential free amino acids in urine. (2) According to the pesent procedure. the ratio obtained with the urban children was proved to be $1.33{\pm}0.18$, whereas that with the rural children was $1.59{\pm}0.24$; suggesting the lower the ratio. the superior the body physiques and nutritional status. (3) The ratio had a statistically significant correlation with the body and sitting heights as well as the body weight. but not with the girth.

  • Central diabetes insipidus(CDI) results from deficient vasopressin(antidiuretic hormone) secretion and causes polydipsia and polyuria. Its etiologic diagnosis is confirmed with an increase of urine osmolality by administering desmopressin(DDAVP) after water restriction. Because cm is caused by deficiency of vasopressin, up to now, desmopressin, a synthetic analog of vasopressin, has been the drug of choice in the treatment of CDI. However, under such treatment, CDI patients suffer from the continual administration of DDAVP throughout one's life and high cost of the treatment We administrated oriental herb medicine on a cm patient in a state of discontinuance of DDAVP. Prior to the study, brain sella MRI was scanned to exclude germinoma. In addition, urine analysis, serum and urinary osmolality, daily urinary volume, serum electrolyte levels were measured. Chungsimyunjatang was administered for 15 days, and urine analysis, urine osmolality, daily urinary volume, serum Na were measured several times again during the therapy, As a result, urinary frequency increased, serum Na slightly elevated, but specific gravity of urine, urinary osmolality severely decreased and daily urinary volume substantially increased. However, the frequency of DDAVP treatment was reduced from four times per day to once or twice a day with the continual administration of the Chungsimyunja-tang for two months after the discharge.

  • The purpose of this study was to investigate the metallothionein of acute cadmium poisoning mice as a Cadmium index. Forty male ICR mice were injected with cadmium chloride solution from 1/8LD50 to 1/2LD50 dose. At 24 hours after exposed Cd, I examined Cd and metallothionein (MT) intissues(liver and kidney) and fluids(whole blood and urine) and also measured low molecular proteins, N-acetyl-D-glucosaminidase(NAG) and 2- microglobuline (2-MG) in urine. The concentration of Cd and MT of liver kidney whole blood and urine were increased with dose dependent manner. Urinary Cd and urinary MT and very good significance (p<0.01) and urinary MT had good significance with kidney Cd and NAG but not 2-MG Conclusionally MT in urine was very correlated with kidney Cd and urine Cd, So MT maybe useful as a Cd poisoning index.
