Ca과 Vitamin D 보충이 폐경 이후 여성의 뼈대사에 미치는 영향 (The Effect of Ca and Vitamin D Supplementation on Bone Metabolism in Postmenopausal Women)

  • This study was designed to investigate the effects of Ca and/or vitamin D supplementation for 53 weeks on bone metabolism in postmenopausal women. The subjects were healthy 18 women aged from 59 to 69 years old. They were divided into three groups : placebo, Ca(1000mg/day) supplementation and Ca(1000mg/day) with vitamin D(12.5$\mu\textrm{g}$/day) supplementation. During the experimental periods except for metabolic studies, the subjects ate their usual diets and the use of drugs as well as excessive exercise was prohibited. Metabolic studies were conducted in the 1st week and in the 53rd week of the experimental periods. The subjects ate experimental diets which consisted of 1787.3kcal, 69.6g of protein, 561.5mg of Ca and 1078.6mg of P daily during both of the metabolic study periods. The results were summarized as follows; 1) Bone density of the second lumbar spine and trochanter measured after treatment decreased significantly in control group as compared with pre-experimental level(p<0.05). On the contrary, bone density of femoral neck and Ward's triangle in Ca group and the second lumbar spine in Ca.Vit D group increased significantly after treatment. 2) Serum PTH and calcitonin levels did not show any significant differences among groups before and after treatment. But serum PTH level increased significantly in all groups after treatment(P<0.05). 3) Serum Ca and P levels did not show any significant differences among groups before and after treatment. But serum Ca level increased significantly in all groups after treatment (P<0.05) and serum P level decreased significantly in Ca.Vit D group after treatment(P<0.05). 4) Mean 24-hours fecal Ca excretion of Ca group was the highest in the 1st week of treatment(P<0.01), and that of control group was the lowest in the 53rd week of treatment(P<0.01). Fecal Ca excretion increased significantly in control and Ca.Vit D group in the 53rd week of treatment(P<0.05). Urinary Ca excretion did not show any significant differences among groups in the 1st and 53rd week of treatment, but that of Ca.Vit D group was the highest the 1st week of treatment(P<0.01). In the 53rd week of treatment Ca and Ca.Vit D group showed positive Ca balance, but control group showed negative Ca balance. The above results showed that it will be difficult to prevent degenerative bone loss without Ca and/or vitamin D supplementation in postmenopausal women eating Korean usual diets.

Dietary maifanite supplementation did not affect the apparent total tract digestibility of calcium and phosphorus in growing pigs

  • Objective: This study was conducted to determine the effects of dietary maifanite supplementation and fecal collection method on the apparent total tract digestibility (ATTD) of calcium (Ca) and phosphorus (P) and blood parameters in growing pigs. Methods: Thirty-six growing barrows (Duroc${\times}$Landrace${\times}$Yorkshire; $27.0{\pm}2.6kg$) were allotted to six dietary treatments with 6 pigs per treatment according to body weight in a completely randomized design. The experimental treatments were: i) Low Ca+cornstarch (2.25%), ii) Low Ca+maifanite (2.25%), iii) Medium Ca+cornstarch (1.42%), iv) Medium Ca+maifanite (1.42%), v) High Ca+cornstarch (0.64%), and vi) High Ca+maifanite (0.64%). Feces were collected by the total collection (TC) and indicator method (IM). At the beginning and the end of the experiment, blood samples were collected from each pig. Results: For the TC method, there were no difference in Ca intake, fecal Ca output, Ca retention and the ATTD of Ca between cornstarch and maifanite diets at the same dietary Ca level. However, urinary Ca excretion was lower (p = 0.01) in pigs fed low Ca diets without maifanite supplementation compared with other dietary treatments. Dietary maifanite supplementation had no effect on the P metabolism in growing pigs. For the IM method, there was no difference in Ca digestibility between cornstarch and maifanite diets at the same dietary Ca level. The ATTD of P was greater (p<0.01) in pigs fed the high Ca diet with maifanite supplementation compared with the high Ca diet with cornstarch treatment. Dietary inclusion of maifanite had no effect on blood parameters in growing pigs. Conclusion: Dietary maifanite supplementation had no effect on the ATTD of Ca and P and serum parameters in growing pigs. The IM resulted in lower digestibility values than the TC method.

Effects of Different Dietary Acidifier Sources of Calcium and Phosphorus on Ammonia, Methane and Odorant Emission from Growing-finishing Pigs

  • The objective of this study was to investigate the effects of different sources of Ca and P on urine and ileal digesta pH, and ammonia ($NH_{3}$), methane ($CH_{4}$), and odor emission. In experiment 1, eight pigs (commercial three-way cross; initial BW 67$\pm$3 kg) were arranged in a repeated 4$\times$4 Latin Square design. All pigs were equipped with a T-cannula in the distal ileum. Four corn-soybean meal based diets were formulated. Diet 1 was the control in which dicalcium phosphate (DCP) and limestone ($CaCO_{3}$) were used as the sources of inorganic P and Ca. In Diets 2 and 3, ${H_{3}}{PO_{4}}$, monocalcium phosphate (MCP), and $CaSO_{4}$replaced DCP and $CaCO_{3}$ as the inorganic sources of P and Ca. Diet 4 was similar to Diet 1 except that it was fortified with HCl to provide an acid load similar to that of diet 2. Urine and ileal digesta pH were determined in pigs fed each of these diets. In Exp. 1, urine pH decreased (p<0.05) in animals consuming diets containing ${H_{3}}{PO_{4}}$-$CaSO_{4}$ (5.85$\pm$0.38) and MCP-$CaSO_{4}$(5.73$\pm$0.30) compared with the DCP-$CaCO_{3}$ diet (6.89$\pm$0.24). In the pigs consuming ${H_{3}}{PO_{4}}$-$CaSO_{4}$, ileal digesta pH decreased compared with the control (5.52$\pm$0.28 vs. 6.66$\pm$0.17; p<0.05). Based on the results of Exp. 1, a total of four trials were performed in environmental chambers for determining how $NH_{3}$, $NH_{4}$, and odor were affected by the different dietary Ca and P sources (Exp. 2). In Exp. 2, pigs fed the ${H_{3}}{PO_{4}}$-$CaSO_{4}$ diet had decreased (30%) $NH_{3}$ emissions compared with the control (p<0.05). Also, a combination of MCP-$CaCO_{3}$-$CaCl_{12}$ decreased $NH_{3}$ emission by 15% (p<0.05). Emission of $CH_{4}$ was decreased only with the ${H_{3}}{PO_{4}}$-$CaSO_{4}$ diet with 14% (p<0.05). Odorant emission of phenolics and volatile fatty acids increased roughly three-fold with the DCP-$CaSO_{4}$ diet but was not affected by other test diets. In conclusion, acidogenic Ca and P sources in swine diets can decrease the urinary pH and reduce $NH_{3}$ and $CH_{4}$ emission from swine facilities.

성숙한 쥐의 대퇴골과 요추의 조성과 골격관련 인자에 대한 난소절제와 칼슘섭취 효과 (Effect of Ovariectomy and Calcium Intake on Femur and Lumbar Compositions and Bone Related Factors in Adult Rats)

  • This study explored the effect of calcium levels and/or ovariectomy on bone composition and its related factors using the female Sprague-Dawley rats which achieved peak bone mass in normal(0.5%) calcium intake during growth period. The rats were randomly divided into six groups and fed 0.1%, 0.5% and 1.5% calcium diets for 8 weeks after ovariectomized and sham operation. The results indicated that body weight gain was higher in ovariectomized groups than sham groups, regardless of dietary calcium levels and food intakes. Serum Ca and P concentrations were normal level regardless of dietary calcium levels and operation. Estrogen concentration was decreased in ovariectomized rat groups. Serum alkaline phosphatase activity and urinary hydroxyproline were increased in ovariectomized groups. When rats were fed normal Ca diet during growing period, weight, length and breaking force of femur were not significantly different in all groups but increased the same level. Generally, lipid contents in lumbar and femur were lower in low calcium groups and this effect was more pronounced in femur. In case of femur, the significant decrease in Ca contents of bone was observed in a relatively short period of feeding low Ca diet, even though it appeared th desirable peak bone mass had achieved through the growing period by supplementing the adequate amount of calcium. The marked decrease of estrogen levels after ovariectomy did not seem to influence greatly bone parameters measured except bone wet weight. Therefore, this study indicated that lower intake of Ca might be more important determinating factor against osteoporosis than postmenopausal state stimulated by ovariectomy in female rats. With normal or high intake of Ca it appears possible to prevent bone loss in postmenopausal period, and this might apply only in case of achieving peak bone mass in the growing with the adequate intake of calcium.   • The effects of mulberry-leaf powder Tofu(MPT) on anthropometric measurements, including bone mineral density(BMD) in the right carpus, biochemical bone turnover markers, serum levels of lipids and macrominerals, were investigated in 30 smoking male adults who lived in Choongnam were given MPT(100 g/day) for 4 weeks. The average ages, number of smoked cigarettes and packyear were 22.38 years, 15.12/day and 3.54 years, respectively. The nutrient contents per 100 g MPT were 86.10 kcal energy, 8.98 g protein, 0.53 mg fiber, 211.33 mg Ca and 1.59 g fat. Anthropometric measurements, including dietary intake using the 24-hours recall method, carpal BMD using DEXA, serum levels of protein, albumin and glucose, lipid profiles (cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol) with Al(atherosclerosis index), HTR, CRF, LHBt, some biomarkers of BMD(serum alkaline phosphatase activity, osteocalcin, urinary DPD), and serum macrominerals(Ca, Ca/P ratio, Mg) and Pb were analyzed before and after consumption of MPT. After MPT consumption, dietary intakes of plant protein, total Ca and plant Ca increased significantly, but there were no significant differences in anthropometric measurements, BMD with bone metabolism markers, serum levels of protein, albumin or glucose, lipid profiles with AI, HTR, LHR and CRF.

한국 성인 여자에 있어서, 나트륨 섭취 수준이 체내 칼슘대사에 미치는 영향 (The Effect of Dietary Sodium on Calcium Metabolism in Healthy Young Adult Women)

  • The present study was conducted to examine the effect of an increased level of dietary sodium on calcium excretion in 8 health young adult Korean women on a controlled diet. After adaptation period of 2 days, each subject received 2811.8$\pm$68.1 mgNa(day during the initial period of 5 days (low sodium period) and 6417.1$\pm$248.6mgNa(day during the following period of 5 days (high sodium period). Calcium intake was 593.7$\pm$15.7 mg Ca/day during the low sodium period of 596.1$\pm$25.1 Ca/day duing the high sodium period. When the low sodium period is compared with the high soidum period, the results were as following. 1) Mean urinary sodium excretion was significantly higher during the high sodium period (5760.1$\pm$156.5mg0 than during the low sodium period (2272.2$\pm$108.6mg)(P<0.001). Fecal sodium excretion of the high sodium period was also significantly higher than that of the low sodium period(P<0.001). Mean value of sodium balance during the high sodium period was higher than that of the low sodium period . However, the difference was not significant. 2) Mean urinary calcium excretion was significantly higher during the high sodium period than during the low sodium period ; mean value of the low sodium period was 124.7$\pm$11.3mg and that of the high sodium period was 202.6$\pm$17.2mg)P<0.001). Fecal calcium excretion was higher during the high sodium period (284.9$\pm$31.0mg) than during the low sodium period (253.9$\pm$15.3mg0, but there was no significance. Mean value of calcium balance during the high sodium period was significantly lower than that of the low sodium period(P<0.001). The above results show that high sodium intake increases calcium excretion as well as sodium excretion.

식이내 단백질 수준이 성장기 흰 쥐의 신장 기능에 미치는 영향 (The Effect of Level of Dietary Protein on Kidney Development and Function in Growing Rats)

  • 본 연구에서는 식이단백질 수준이 성장기 흰쥐의 신장 발달과 기능에 미치는 영향을 조사하기 위해 평균체중이 97.5$\pm$1.9g인 숫컷 흰쥐를 5%, 15% 그리고 50% casein 식이로 6주간 사육하였다. 체중증가량은 단백질 수준이 높을수록 높았으며 특히 단백질수준이 신장무게에 미친영향이 간, 비장, 흉선 등이 다른 장기에 비해 컸다. 신장내 총 DNA와 RNA 함량에서는 유의적인 차이가 없었다. 총 protein과 단위 g당 protein 함량은 식이내 단백질수준이 높을 수록 증가했다. 사구체여과율(GFR)은 식이단백질 수준이 높을수록 높았으나 신혈류(RBF)는 각 군 사이에 유의적인 차이를 보이지 않았다. 뇨질소배설량은 단백질 수준에 비례하여 유의적인 차이를 보였고 노칼슘배설량은 5%군이 낮은 경향을 보였다. 본 연구 결과로 성장기의 흰쥐에서 고단백식이는 신장의 무게증가를 촉진하며 사구체여과율을 증가시킴을 알 수 있다.

콩단백질과 이소플라본이 성장기 암컷 쥐의 골밀도에 미치는 영향 (Effects of Soy Protein and Isoflavones on Bone Mineral Density in Crowing Female Rats)

  • The purpose of this study was to examine the impact of soy protein and soy isoflavones on bone and mineral density in young female Sprague-Dawley rats. Fifty eight rats (body Weight 75 $\pm$ 5 g) were randomly assigned to one of four groups, consuming casein, soy protein concentrate, soy protein isolate (57 mg isoflavones/100 g diet) or casein added isoflavones (57 mg isoflavones /100 g diet). All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin, USA) in spine and femur on 3, 6, 9 weeks after feeding. The serum and urine concentrations of Ca and P were determined. Diet did not affect weight gain and mean food intake. Food efficiency ratio was lower In soy protein groups. The serum concentration of Ca and P were not changed by soy protein and isoflavones. Urinary Ca and P excretion were not significantly different. Spine BMD was significantly increased by soy protein isolate on 3 and 6 weeks after feeding. Femur BMD was significantly increased in the groups of soy protein isolate and isoflavones adding on after 9 weeks. Therefore, soy protein with rich isoflavones may be beneficial on spine and femur BMD increasement in growing female rats.

Mode of Inhibitory Action of Amitriptyline on Carbachol-Induced Contraction of Isolated Rabbit Detrusor Muscle

  • The present study was aimed at elucidating the mode of inhibitory action of tricyclic antidepressants on the smooth muscle. Effects of amitriptyline on the isolated detrusor muscle strips of the urinary bladder of the rabbit were examined. The spontaneous rhythmic movement of the muscle preparation was frequently observed, which was decreased or abolished by addition of amitriptyline $(10^{-5}{\sim}10^{-3}\;M)$. The muscle preparation responded with contraction dose dependently to carbachol, of which dose response curve shifted to the right in the presence of either amitriptyline or atropine. However, amitriptyline produced a nonparallel shift, whereas atropine caused a parallel one. In calcium free medium, the contraction response to carbachol was markedly decreased, which was resumed by the addition of $CaCl_2$ (2.5mM), but not in the presence of either amitriptyline or nifedipine. KCI (60 mM) produced a potent contraction, which was abolished in the presence of amitriptyline or nifedipine. These results suggest that amitriptyline, unlike atropine, not only acts as a noncompetitive antagonist at cholinergic muscarine receptors but also inhibits Ca-influx through the muscle cell membrane.

성장기 암컷 쥐에서 Arginine 첨가가 골밀도에 미치는 영향 (Effects of Arginine Supplementation on Bone Mineral Density in Growing Female Rats)

  • The aim of this study was to define an arginine effect when added to a diet. The influence of arginine supplements on bone mineral density and content were studied in young female Sprague-Dawley rats fed either an arginine supplemented diet or control diet. Twenty four rats (body weight $83{\pm}5g$) were randomly assigned to one of two groups, consuming casein or casein with supplemented arginine diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin, USA) in spine and femur 3, 6, and 9 weeks after feeding. The serum and urine concentrations of Ca and P were determined. Diet did not affect weight gain and mean food intake. The serum concentration of Ca and P were not changed by arginine supplementation. Urinary Ca excretion was significantly decreased by arginine supplementation. Spine BMD was significantly increased by arginine supplementation on 3 and 6 weeks after feeding. Femur BMD was significantly increased in the group of arginine supplementation on 3, 6, and 9 weeks. Rats fed the arginine-supplemented diet had better bone mineral content than did control diet rats in the experimental period. Therefore, arginine supplementation may be beneficial on spine and femur BMD increment in growing female rats. These are thought to be associated with an arginine-induced growth hormone release. The exact mechanism of this effect remains to be elucidated.
