바위돌꽃(Rhodiola rosea L.)의 촉성재배시 질소 시비에 따른 수량과 Salidroside 함량 (Comparison of Yield and Content of Salidroside with Application Rates of Nitrogenic Fertilizer under Forcing Culture of Rhodiola rosea L.)

  • 본 연구에서는 질소비료의 종류 및 처리량 차이가 바위돌꽃의 뿌리 수량 및 유효성분인 salidroside의 함량변화에 미치는 영향을 구명하여 고품질의 바위돌꽃을 생산할 수 있는 최적의 시비조건을 확립하고자 하였다. 요소비료와 유안비료 모두 48kg N/10 a까지는 처리량이 증가할수록 바위돌꽃 뿌리의 건물 생산량이 증가하는 경향이었으나, 그 이상의 처리량부터는 오히려 건물 생산량이 감소하였다. 요소비료와 유안비료 모두 화학비료 성분 처리량 48-8-20-10-10 kg/10a에서 가장 높은 salidroside 함량을 나타내었다. 곡선회귀 방정식에 기준한 요소와 유안비료의 적정 시비량은 45-8-20-10-10 kg/10 a로 나타났으나, 요소비료와 유안비료의 처리후 토양중 질소 축척과 토양 산성화, 식물체로의 질소 흡수이행량, 바위돌꽃 뿌리의 건물생산량 그리고 바위돌꽃 뿌리에 함유되어 있는 약리성분인 salidroside의 함량 등 제반사항을 고려하였을 때, 요소비료는 질소성분량으로 40 kg/10 a, 유안비료는 질소성분량으로 35 kg/10 a가 합리적일 것으로 판단된다.

  • 답토양(畓土壤)의 질소(室素)의 동태(動態)를 파악(把握)하기 위하여 성질(性質)이 다른 3개지역(個地域)의 토양(土壤)을 공시(供試)하여 지력질소(地力窒素)의 발현양식(發現樣式)과 토양(土壤)의 종류(種類), 온도(溫度), 토양(土壤)의 길이, 질소비료(窒素肥料)의 종류(種類), 담수일수(湛水日數)에 따른 $NH_4-N$, $NO_3-N$, 2가철(價鐵) pH의 변화(變化)와 이들 상호관계(相互關係)를 검토(檢討)하여 탈질량(脫窒量)을 추정(推定)하고 지역(地域)에 알맞는 질소시비법(窒素施肥法)을 확립(確立)하기 위한 기초적(基礎的) 지견(知見)을 얻을 목적(目的)으로 실험(實驗)하여 얻어진 결과(結果)를 요약(要約)하면 다음과 같다. 1. 온도상승효과는 화순(和順), 서창(西倉), 서산토양(瑞山土壤)의 순(順)이었으나 건토효과는 서산(瑞山), 화순(和順), 서창(西倉)의 순(順)이였고 $NH_4-N$의 생성율(生成率)은 서산(瑞山), 서창(西倉), 화순(和順)의 순(順)으로 토양(土壤)의 성질(性質)에 따라 상이(相異)했다. 2. 표면수중(表面水中) $NH_4-N$의 변화(變化)는 일반적(一般的)으로 토양중(土壤中)의 함량(含量)에 좌우(左右)되며 염안(鹽安), 유안구(硫安區)가 많고 담수(湛水) 3일(日)에 감소(減少)했다가 40일(日)에 다시 증가(增加)된 후(後) 50일(日)에 감소(減少)하였고 요소구(尿素區), SCU는 함량(含量)이 적었으며 담수기간(湛水期間) 비교적(比較的) 일정(一定)했다. 3. 토양중(土壤中) $NH_4-N$의 변화(變化)는 1층(層)은 염안(鹽安), 유안구(硫安區)가 집적(集積)이 많으며 SCU, 요소구(尿素區)는 적었다. 토양(土壤)의 종류(種類)에 따라서도 비교적(比較的) 같은 경향(傾向)을 나타냈으며 경시적(經時的)으로 함량(含量)은 감소(減少)했다. 2층(層)도 산화층(酸化層)과 같은 경향(傾向)을 나타냈다. 4. 표면수중(表面水中) $NO_3-N$의 변화(變化)는 토양(土壤)과 비료(肥料)의 종류(種類)에 따라 다르나 일반적(一般的)으로 요소(尿素), SCU구(區)가 함량(含量)이 많고 염안(鹽安), 유안구(硫安區)가 적었으며 담수(湛水) 30~40일(日)에 소실(消失)되었다. 5. 토양중(土壤中) $NO_3-N$의 변화(變化)는 1층(層)은 2층(層)에 비(比)하여 $NO_3-N$의 함량(含量)이 많고 토양(土壤)과 비료(肥料)에 따라 변화(變化)되는 양상(樣相)이 다르며 일반적(一般的)으로 담수(湛水) 30일(日)에 소실(消失)되었다가 그 후(後) 증가(增加)되었으며 요소(尿素), SCU구(區)가 질화(窒化)가 잘되는 편이며 2층(層)은 담수(湛水) 30일(日)까지는 함량(含量)이 적거나 거의 소실(消失)되었다가 40일(日) 이후(以後)는 야간증가(若干增加)했다. 6. 표면수(表面水)와 토양중(土壤中) $NH_4-N$, $NO_3-N$의 함량(含量)으로 보아 담수(湛水) 기간중(期間中) 시용(施用)한 질소(窒素)의 반량이상(半量以上)이 탈질(脫窒)되는 것으로 추정(推定)되며 요소(尿素) SCU 구(區)는 염안(鹽安) 유안구(硫安區)에 비(比)하여 탈질량(脫窒量)이 많으리라 추정(推定)되였다.

Metal-Urea-Montmorillonite Hybrid Incorporated with Citric Acid

  • Massive intercalation of urea into montmorillonite (MUCH) was recently proposed to enhance urea use efficiency through smart suppression of emission of $NH_3$ and NOx. This study was to synthesize citrate-incorporated MUCH (Cit-MUCH) which can enhance suppression of $NH_3$ volatilization. The XRD pattern of Cit-MUCH was very similar to that of MUCH to indicate successful incorporation of citric acid into MUCH. Incorporation of citric acid was confirmed by the existence of $COO^-$ symmetric stretching vibration. During the initial 4 days after application, $NH_3$ volatilization from both bare and perilla-planted soils was much more suppressed by application of Cit-MUCH than MUCH. A peak volatilization rate decreased from 28.3 N mg $m^{-2}\;h^{-1}$ of MUCH-broadcasted soil to 22.2 N mg $m^{-2}\;h^{-1}$ of Cit-MUCH-broadcast soil. $NH_3$ volatilization was less in planted soil than bare soil for 72 hrs after application. These results showed that incorporation of citric acid led to increase in suppression of ammonia volatilization from urea-applied soils.

Performance of Growing Lambs Fed Urea Ammoniated and Urea Supplemented Wheat Straw Based Diets

  • Twelve growing male lambs ($10.05{\pm}0.41kg$, 5 months age) were assigned to three equal groups in a completely randomized design and fed respectively wheat straw (WS), ammoniated wheat straw (AWS) or urea supplemented wheat straw (USWS) along with concentrate mixtures of varying protein and energy contents to meet their requirements as per NRC (1985). Despite comparable nutrient intake and digestibility, the lambs fed A WS based diet digested lower (p<0.05) crude protein. The digestibility of NDF and hemicellulose were lower while the digestibility of cellulose was higher in lambs on AWS as compared to lambs on USWS based diet. The lambs of all the three groups were in positive and comparable N, Ca and P balance except higher Ca balance in lambs fed WS based diet. The body weight change, average daily gain and feed conversion efficiency were similar among the dietary groups. All the parameters of rumen fermentation pattern were comparable between lambs of AWS and USWS based diets except ammonia-N concentration that was higher in the latter however it was lower in WS fed group. Though, the feeding cost per unit gain was comparable in lambs fed all the three diets, the cost incurred towards roughage during 120 days of experimental feeding was significantly less in lambs fed AWS than UTWS. Thus, feeding of USWS can be practiced to minimize labour cost and environmental pollution involved in the process of urea treatment without affecting the performance of growing lambs.

EFFECT OF SUPPLEMENTING UREA MOLASSES MINERAL BLOCK LICKS ON BACTERIAL PRODUCTION RATE IN THE RUMEN OF CROSSBRED CALVES

  • Sixteen crossbred (Sahiwal $\times$ Holstein) male rumen fistulated calves of 18 to 24 months of age were randomly divided into four groups of four animal, each. Animals in all the groups were fed wheat straw ad lib as basal roughage. However, the animals in group I were fed concentrate mixture at maintenance level, whereas, the animals in groups II, III and IV had free access to existing, modified (A) and modified (B) urea molasses mineral block licks respectively. Daily wheat straw intake (kg) was significantly (p<0.01) higher in groups II ($4.20{\pm}0.13$), III ($4.07{\pm}0.16$) and IV ($4.22{\pm}0.20$) as compared to group I ($3.21{\pm}0.14$). Total N and TCA precistrained rumen liquor) was significantly higher in groups II ($22.36{\pm}0.25$), III ($21.63{\pm}0.25$) and IV ($21.77{\pm}0.55$) as compared to group I ($18.31{\pm}0.41$). Bacterial production rate (g/day and g/kg digestible organic matter intake) were non-significantly different amongst groups I ($214.4{\pm}13.28;\;85.38{\pm}3.69$); II ($198.7{\pm}5.70;\;86.17{\pm}3.53$); III ($214.4{\pm}8.19;\;96.15{\pm}2.16$) and IV ($218.2{\pm}10.62;\;94.44{\pm}5.52$). Similarly, percent efficiency of N incorporation into bacterial protein was not found significantly different amongst groups I, II, III and IV. These studies indicate that when concentrate mixture (upto maintenance level) in the diet of ruminants was replaced with UMMB licks, various N fraction in SRL and efficiency of bacterial production rates in the rumen were not affected.

Kinetics of Removing Nitrogenous and Phosphorus Compounds from Swine Waste by Growth of Microalga, Spirulina platensis

  • Abstract Spirulina platensis was grown in SWlUe waste to reduce inorganic compowlds and simultaneously produce feed resources. Spirulina platensis prefers nitrogenous compounds in Ibe order: $NH_4^{+}-N>NO_3^{-}-N>simple-N$ such as urea and simple amino acids. It even consumes $NH_4^{+}-N$ first when urea or nitrate are present. Therefore, the content of residual $NH_4^{+}-N$ in Spimlina platensis cultures can be determined by the relative extent of the following processes: (i) algal uptake and assimilation; (ii) ammonia stripping; and (iii) decomposition of urea to NH;-N by urease-positive bacteria. The removal rates of total nitrogen ffild total phosphorus were estimated as an indicator of the treatment effIciency. It was found that Spirulina platensis was able to reduce 70-93% of $P_4^{3-}-P$, 67-93% of inorganic nitrogen, 80-90% of COD, and 37-56% of organic nitrogen in various concentrations of swine waste over 12 days of batch cultivation. The removal of inorganic compounds from swine waste was mainly used for cell growth, however, the organic nitrogen removal was not related to cell growlb. A maximum cell density of 1.52 dry-g/l was maintained with a dilution rate of 0.2l/day in continuous cultivation by adding 30% swine waste. The nitrogen and phosphorus removal rates were correlated to the dilution rates. Based on the amino acid profile, the quality of the proteins in the Spirulina platensis grown in the waste was the same as that in a clean culture.ulture.

Improving Nutritional Quality of Cocoa Pod (Theobroma cacao) through Chemical and Biological Treatments for Ruminant Feeding: In vitro and In vivo Evaluation

  • Cocoa pod is among the by-products of cocoa (Theobroma cacao) plantations. The aim of this study was to apply a number of treatments in order to improve nutritional quality of cocoa pod for feeding of ruminants. Cocoa pod was subjected to different treatments, i.e. C (cocoa pod without any treatment or control), CAm (cocoa pod+1.5% urea), CMo (cocoa pod+3% molasses), CRu (cocoa pod+3% rumen content) and CPh (cocoa pod+3% molasses+Phanerochaete chrysosporium inoculum). Analysis of proximate and Van Soest's fiber fraction were performed on the respective treatments. The pods were then subjected to an in vitro digestibility evaluation by incubation in rumen fluid-buffer medium, employing a randomized complete block design (n = 3 replicates). Further, an in vivo evaluation of the pods (35% inclusion level in total mixed ration) was conducted by feeding to young Holstein steers (average body weight of $145{\pm}3.6kg$) with a $5{\times}5$ latin square design arrangement (n = 5 replicates). Each experimental period lasted for 30 d; the first 20 d was for feed adaptation, the next 3 d was for sampling of rumen liquid, and the last 7 d was for measurements of digestibility and N balance. Results revealed that lignin content was reduced significantly when cocoa pod was treated with urea, molasses, rumen content or P. chrysosporium (p<0.01) with the following order of effectiveness: CPh>CAm>CRu>CMo. Among all treatments, CAm and CPh treatments significantly improved the in vitro dry matter and organic matter digestibility (p<0.05) of cocoa pod. Average daily gain of steers receiving CAm or CPh treatment was significantly higher than that of control (p<0.01) with an increase of 105% and 92%, respectively. Such higher daily gain was concomitant with higher N retention and proportion of N retention to N intake in CAm and CPh treatments than those of control (p<0.05). It can be concluded from this study that treatment with either urea or P. chrysosporium is effective in improving the nutritive value of cocoa pod.

The Effect of Nitrogen Fertilization to the Sward on Guineagrass (Panicum maximum Jacq cv. Gatton) Silage Fermentation

  • To investigate the effect of nitrogen fertilization on the quality of tropical grass silage, guinea grass grown with 3 types of nitrogen fertilizers, namely, urea, ammonium sulfate, and compound fertilizer 804, at 2 fertilization levels, 0.5 and 2.5 kg $Na^{-1}$ (0.5 N and 2.5 N, respectively), was subjected to silage fermentation. Silage fertilized with 0.5 N showed butyrate-dominant fermentation, irrespective of the type of fertilizer used. On the other hand, fermentation of silage fertilized with 2.5 N was significantly affected by the type of fertilizer used; fertilization with ammonium sulfate and compound fertilizer 804 resulted in silage that contained a large amount of butyrate and no lactate; this silage was considered to be of a significantly low quality as compared with silage fertilized with 0.5 N. Among silage fertilized with 2.5 N, the desirable butyrate-free fermentation was found only in urea-fertilized silage, which had the best quality. Grass material fertilized with a high level of urea accumulated a relatively high concentration of nitrate nitrogen (0.22% dry matter). Our results presented here suggest that nitrogen fertilizer management could affect the quality of tropical grass silage and that a relatively high concentration of nitrate in silage may promote butyrate-free fermentation even in tropical grass silage.

A Comparison of Ammonia and Preformed Protein as a Source of Nitrogen for Microbial Growth in the Rumen of Sheep Given Oaten Chaff

  • Microbial growth efficiency in the rumen was studied in sheep given hourly, 31.25 g oaten chaff with either 0.31 and 0.88 g urea or 1.88 and 5.63 g casein (exp. 1) and 33.33 g oaten chaff with 1.04 casein or 0.3, 0.6 and 0.9 g urea or the mixture of the casein and urea (exp. 2). Concentrations of ruminal fluid ammonia increased with increasing nitrogenous supplements. Organic matter digestibility in sacco in the rumen was not different irrespective of N sources. Isoacids and valeric acid increased with increasing ingested casein but decreased with increasing urea intake. Peptide and amino acid pools in ruminal fluid increased with increasing ammonia concentrations (exp. 2) suggesting that proteolytic activity and transportation of peptides and amino acids across microbial membrane of rumen microbes may be regulated by the metabolite mechanism (intracellular amino acids and $NH_4{^+}$, respectively). Densities of total viable and cellulolytic bacteria in ruminal fluid increased with increasing ammonia levels but that of small Entodinia decreased. The density of fungal sporangia growth on oat leaf blades decreased with increasing ammonia concentrations but appeared to remain constant in the presence of casein. Efficiency of net microbial cell synthesis was 15-28% higher when ammonia concentrations increased from 100 to above 200 mg N/l regardless of N sources. In conclusion, supplementation of preformed protein had no effect on rumen digestion and microbial growth efficiency. This could not be accounted for its effect on ruminal fluid ammonia. Increased microbial growth efficiency with increasing ammonia levels may be due to a reduction in the turnover of microbial cells within the rumen.

Functional Characterization of Mammary Gland of Holstein Cows under Humid Tropical Summer Climates

  • Physiological parameters were measured on six primiparous, non-pregnant Holstein cows prior to peak lactation over a 3-month summer season in southwestern Taiwan. The objectives were to characterize heat stress-induced change in functionality of mammary gland under natural climates of tropical summer and to establish physiological indices applicable to this environment in referring to this change. Environmental and physiological readings, milk and blood samples were taken at 15:00 h biweekly for totally five time points during the study. Climate readings showed that the afternoon humidex value reached the highest (53.5) around mid summer. Rectal temperature of cows taken simultaneously varied between $38.26^{\circ}C$ and $40.02^{\circ}C$ in parallel to humidex. Milk production declined drastically from 29.2 to 22.2 kg/d the first month entering summer but leveled up at end of the summer season suggesting effects exerted by heat stress rather than stages of lactation. Lactose content decreased linearly (p<0.05) with times in summer, from 4.69 to 4.38%. On the other hand, activity of N-acetylglucosaminidase (NAGase) in milk increased linearly to over two folds (p<0.05) during the same intervals. Elevations of fractional constituent of BSA in whey protein and serum cortisol level were also noticed in the course. Measurement of arteriovenous concentration (A-V) difference across the mammary gland demonstrated net uptake of glucose and net release of urea throughout the study period. The amount of urea released from mammary gland increased (p<0.05) progressively from 1.54 to 7.76 mg/dl during summer. It is concluded that gradual regression of mammary gland occurred along the humid tropical summer season. This regression is likely initiated through elevation of body temperature, which is irreversible above certain point. The increased release of urea from mammary gland during heat stress suggests its potential role as an early indicator of suboptimal mammary function.
