Uptake and Recovery of Urea-15N Blended with Different Rates of Composted Manure (퇴비의 혼합 시비율에 따른 Urea-15N의 이용율 및 회수율)

  • To utilize composts more efficiently, combining composts with fertilizer to meet crop requirements is an appealing alternative. A pot experiment was conducted to study the effect of application rate of composted pig manure blended with fertilizer on the availability and loss of fertilizer-N. Chinese cabbage (Brassica campestris L. cv. Samjin) plants were cultivated for 30 and 60 days. 15N-Labeled urea ($5.24\;^{15}N\;atom\;%$) was added to soil at $450mg\;N\;kg^{-1}$, and unlabeled compost ($0.37\;^{15}N\;atom\;%$) was added at 0, 200, 400, and $600mg\;N\;kg^{-1}$. The amount of plant-N derived from urea was not affected by compost application at rate of $200mg\;N\;kg^{-1}$. However, compost application at 400 and $600mg\;N\;kg^{-1}$ significantly (P<0.05) increased plant assimilation of N from urea irrespective of sampling time, probably because of physicochemical changes in the soil properties allowing urea-N to be assimilated more efficiently. The amount of immobilized urea-N increased with increasing rate of compost application at both growth periods, as the results of increased microbial activities using organic C in the compost. Total recovery of urea-N (as percentage of added N) by Chinese cabbage and soil also increased with increasing rate of compost from 71.5 to 95.6% and from 67.0 to 88.2% at the 30- and 60-days of growth, respectively. These results suggest that increasing rate of compost blending increases plant uptake of fertilizer-N and enhances immobilization of fertilizer-N, which leads to decrease in loss of fertilizer-N. However, information about the fate of immobilized N during future crop cultivation is necessary to verify long-term effect of compost blending.

Safety Assessment of Oriental Medicines and Their Preparations (한약 및 한약제제의 안전성 평가)

  • This study was carried out to evaluate the safety of circulated oriental medicines and their related preparations in Gyeonggi-do. Total 366 samples (165 species) were analyzed about heavy metals, residual contents of sulfur dioxide ($SO_2$), and 68 samples were analyzed about preservatives. 17 samples (13 species, 4.6%) were exceeded the legal limit of heavy metal. The concentrations over the legal limit for Pb, Cd, As and Hg were 6.1~19.2 mg/kg, 0.4~0.7 mg/kg, 6.9 mg/kg and 0.7 mg/kg, respectively. In particular, the exceeding ratio (5.9%) of the legal limit of heavy metals in foreign products was 3.3 times more than domestic products (1.8%). 13 samples (10 species, 3.6%) exceeded the limit of residual sulfur dioxide and the concentration ranges were 105 to 428 mg/kg in domestic products, on the other hand foreign products were from 114 to 2,468 mg/kg. The mean concentration over the limit of residual sulfur dioxide of foreign products (804 mg/kg) was 2.4 times more than domestic products (338 mg/kg). In studying of the preservatives in oriental preparation, the contents of dehydroacetic acid (48.9~64.1%) in 3 samples of labeled solutions were under the labeled preservative contents and the contents of benzoic acid (139.9%) in 1 sample of labeled pill product was exceeded the labeled preservative contents. The numbers of the detected preservatives in unlabeled solutions, pills and granules were 4, 11 and 7, respectively.

Serial MR Imaging of Magnetically Labeled Humen Umbilical Vein Endothelial Cells in Acute Renal Failure Rat Model (급성 신부전 쥐 모델에서 자기 표지된 인간 제대정맥 내피세포의 연속 자기공명영상)

  • Purpose : To evaluate the usefulness of in vivo magnetic resonance (MR) imaging for tracking intravenously injected superparamagnetic iron oxide (SPIO)-labeled human umbilical vein endothelial cells (HUVECs) in an acute renal failure (ARF) rat model. Materials and Methods: HUVECs were labeled with SPIO and poly-L-lysine (PLL) complex. Relaxation rates at 1.5-T MR, cell viability, and labeling stability were assessed. HUVECs were injected into the tail vein of ARF rats (labeled cells in 10 rats, unlabeled cells in 2 rats). Follow-up serial $T2^*$-weighted gradient-echo MR imaging was performed at 1, 3, 5 and 7 days after injection, and the MR findings were compared with histologic findings. Results: There was an average of $98.4{\pm}2.4%$ Prussian blue stain-positive cells after labeling with SPIOPLL complex. Relaxation rates ($R2^*$) of all cultured HUVECs at day 3 and 5 were not markedly decreased compared with that at day 1. The stability of SPIO in HUVECs was maintained during the proliferation of HUVECs in culture media. In the presence of left unilateral renal artery ischemia, $T2^*$-weighted MR imaging performed 1 day after the intravenous injection of labeled HUVECs revealed a significant signal intensity (SI) loss exclusively in the left renal outer medulla regions, but not in the right kidney. The MR imaging findings at days 3, 5 and 7 after intravenous injection of HUVECs showed a SI loss in the outer medulla regions of the ischemically injured kidney, but the SI progressively recovered with time and the right kidney did not have a significant change in SI in the same period. Upon histologic analysis, the SI loss on MR images was correspondent to the presence of Prussian blue stained cells, primarily in the renal outer medulla. Conclusion: MR imaging appears to be useful for in vivo monitoring of intravenously injected SPIO-labeled HUVECs in an ischemically injured rat kidney.

Synaptic Organization of Vibrissa Afferent Terminals in the Trigeminal Interpolar Nucleus (삼차신경중간핵에서 저역치기계자극수용기 유래 들신경섬유 종말의 연접양상)

  • In order to evaluate the mechanism of transmission as well as processing of sensory information originating from low-threshold mechanoreceptor in oral and maxillofacial region at primary synaptic region of trigeminal nervous system, vibrissa afferent fibers of adult cat were labeled with intra-axonal HRP injection. Serial sections containing labeled boutons were obtained from the piece of trigeminal interpolar nucleus. Under electron microscope, total 30 labeled boutons were observed, and ultrastructural characteristics, frequency of occurence, synaptic organizations of vibrissa afferent terminals were analysed. The results were as follows: 1. Labeled boutons contained clear, spherical synaptic vesicles with diameter of 45$\sim$55nm. They formed asymmetrical synapse with dendrites showing definite postsynaptic density, larger synaptic cleft, multiple synaptic structures at various regions. With unlabeled axon terminals(p-ending) containing polymorphic synaptic vesicles, they formed symmetrical synapse showing indefinite postsynaptic density and narrower synaptic area. 2. Each labeled bouton formed 1 to 15 synapses, the average of 4.77$\pm$3.37 contacts per labeled bouton, with adjacent neuronal profiles. Relatively complex synaptic organization, which formed synapses with more than 5 neuronal profiles, was observed in a large number(46.7%, n=14) of labeled boutons. 3. Axo-somatic synapse was not observed. The number of axo-dendritic synapse was 1.83$\pm$1.37 per labeled bouton. Majority(85.0%) of axo-dendritic synapses were formed with dendritic shafts, nonprimary dendrites(n=47, 1.57$\pm$1.38/1 bouton), however, synapses formed with primary dendrites(n=6, 0.20$\pm$0.41/1 bouton) or dendritic spines(n=2, 0.07$\pm$0.25/1 bouton) were rare. 4. 76.7%(n=23) of labeled boutons formed axo-axonic synapse (2.93$\pm$2.36/1 bouton) with p-endings containing pleomorphic vesicles. Synaptic triad, in which p-endings formed synapses with labeled boutons and dendrites adjacent to the labeled boutons simultaneoulsy, were also observed in 60.0%(n=18) of labeled boutons. From the above results, vibrissa afferent terminals of adult cat showed distinctive synaptic organization in the trigeminal interpolar nucleus, thus, suggests their correlation with the function of the trigeminal interpolaris nucleus, which participates in processing of complex sensory information such as two-point discrimination and motivational-affective action. Further studies on physiologic functions such as quantitative analysis on ultrastructures of afferent terminals and nerve transmitters participating in presynaptic inhibition are required.
