Enzymatic Characteristics of an Extracellular Agarase of Cytophaga sp. KY-1 and Molecular Cloning of the Agarase gene

  • A bacterial strain KY-l isolated from sewage was able to produce an extracellular agarase(agarose 4-glycanohydrolase. EC 3.2.1.81). The strain KY-1 was identified as Cytophaga fermentans subsp. agarovorans based on its morphological and physiological characteristics. The agarase was purified by ammonium sulfate precipitation followed by DEAE-Sephadex A-50. Bio-Gel P-100. and CM-Cellulose column chromatography. The molecular weight of the purified enzyme was 24 kDa by SDS-polyacrylamide gel electrophoresis. The optimum temperature and pH for the enzyme activity were 30^{circ}C and 7.5, respectively. The enzyme activity was significantly inhibited in the presence of 0.1 mM $HgCl_2$. whereas it was elevated 3 times by $MnSO_4$ at 1 mM concentration. The Km value and Vmax were 16.67 mg/ml and 3.77 unit/ml.min. The agarase gene was cloned into Escherichia coli MC1061 using the plasmid vector pBR322. A 1.4 Kb DNA fragment of PstI-digested chromosomal DNA of C. fermentans KY-l was inserted into the PstI site of pBR322. expressed in the E. coli. and up to 60% of the total enzyme was extracellularly secreted. Enzymatic properties of the extracellular agarases produced by both the transformant and the donor were very similar in terms of optimal pH and temperature.

An Efficient Protocol for Causal Message Delivery in Distributed Mobile Systems (분산 이동 시스템에서 인과적 메시지 전달을 위한 효율적인 프로토콜)

  • There is a growing trend in developing system for distributed mobile system that supports services - job flow management, video conference, replicated data management and resource allocation. Supporting these services, applications have to use causally ordered message delivery. Previous proposals that provide causally ordered message delivery have problems such as communication overhead, message delaying, scalability, computing overload of mobile host. In this paper, we proposed efficient protocol for causally ordered message delivery using the methods that MSS maintains dependency information matrix between MSS and MH, Piggybacking dependency information about each immediate predecessor message. Proposed algorithm, when compared with previous proposals, provides a low message overhead, and low probability of unnecessary inhibition in delivering messages. Also, it consider resource restriction of MH and low bandwidth of wireless communication by computing most of algorithm at MSS, and reduce processing delay by executing causally ordered message delivery a unit of MH.

  • Traditional Medicinal System (TMS) is one of the centuries-old practices and long-serving companions to the human kind to fight against disease and to lead a healthy life. Every indigenous people have been using their unique approaches of TMS practice where among, the Chinese, Indian and African TMSs are world-wide renowned. India has a unique Indian System of Medicines (ISM) consisting of Ayurveda, Siddha, Unani, Naturopathy and Homoeopathy. Siddhars are the saints as well as the eminent scholars, who have attained Ashta-mahasiddhi [Tamil: (Ashta-Eight; Mahasiddhi-Power)] or enlightment. They have postulated, practiced, immensely contributed and have established the concept of the Tamil medicinal system called Siddha System of Medicine (SSM). From ancient time, SSM has flourished and has been widely practiced in the southern part of India particularly in Tamil Nadu. The induction of the modern medicinal system has immensely influenced the existence of SSM and has made the SSM principles and practices undervalued/extinct. However, at present, still a considerable group of people are using the SSM as a basic health-care modality. In this context, the present scrutiny deals with the TMS history, its significance with a special reference to SSM history, Siddhars, the basic concept of SSM, its diagnostic procedures, materia medica and treatment. Conclusively, Siddha is one of the most ancient indigenous health practices despite its several thorny challenges and issues, which needs to be flagged effectively and to be preserved and revitalized in the international arena in the near future.

LS-SVM Based Modeling of Winter Time Apartment Hot Water Supply Load in District Heating System (지역난방 동절기 공동주택 온수급탕부하의 LS-SVM 기반 모델링)

  • Continuing to the modeling of heating load, this paper, as the second part of consecutive works, presents LS-SVM (least square support vector machine) based model of winter time apartment hot water supply load in a district heating system, so as to be used in prediction of heating energy usage. Similar, but more severely, to heating load, hot water supply load varies in highly nonlinear manner. Such nonlinearity makes analytical model of it hardly exist in the literatures. LS-SVM is known as a good modeling tool for the system, especially for the nonlinear system depended by many independent factors. We collect 26,208 data of hot water supply load over a 13-week period in winter time, from 12 heat exchangers in seven different apartments. Then part of the collected data were used to construct LS-SVM based model and the rest of those were used to test the formed model accuracy. In modeling, we first constructed the model of district heating system's hot water supply load, using the unit heating area's hot water supply load of seven apartments. Such model will be used to estimate the total hot water supply load of which the district heating system needs to provide. Then the individual apartment hot water supply load model is also formed, which can be used to predict and to control the energy consumption of the individual apartment. The results obtained show that the total hot water supply load, which will be provided by the district heating system in winter time, can be predicted within 10% in MAPE (mean absolute percentage error). Also the individual apartment models can predict the individual apartment energy consumption for hot water supply load within 10% ~ 20% in MAPE.

  • This study was conducted to obtain the transgenic Brnssica napus plants with tobacco Apase gene using the binary vector system of Agrobacteriurn fumefociens. The results obtained were summarized as follows: A repressible acid phosphatase gene of Saccharon~yces cerevisiae, pho105 was used for screening of tobacco Apase cDNA. In order to identify Apase gene in tobacco genome, Southern blot analysis was pcrformed and the Apase gcnc may be present as a single copy, or at most two or three copies, in tobacco genome. To isolate the tobacco Apase gene, tobacco cDNA library was constructed using purifed mRNA from -Pi treated tobacco root and the plaque forming unit of the library was 2.8 x $10^5$ pfu/m${\ell}$, therefore the library might cover all expressed mRNAs. Using pho5 as a probe. tobacco Apase cDNA was cloned, and restriction mapping and Southern blot analysis of cDNA insert were revealed that the 3.6 kb cDNA contained tobacco acid phosphatase cDNA. Plasmid pGA695 -tcAPl was constructed by subcloning tobacco Apase cDNA into the Hind site of pGA695 with 35s promoter which can be expressed constitutively in plants. The Brassica napus cotyledonary petioles were cocultivated with the ,4 grobacteriunz and transferred to the selection medium. The transformed and regenerated plants were transplanted to soil medium. Southern blot analysis was done on the transformed plants, and it was confirmed that a foregin gene was stably integrated into the genonies of B. nnpus plants.

Rapid Ecoassessment Technique about Anthropogenic Disturbance Potentiality of Land Use (토지의 훼손 잠재성에 대한 신속한 생태평가기법)

  • In order to determine the degree of anthropogenic disturbance potentiality (ADP) of the area a rapid ecoassessment technique was developed on the basis of actual vegetation map. ADP degree of relevant unit cell was computed by using four criteria of land use patterns such as forested area, open water and stream, agricultural area, and urbanized area. Ultimate ADP degree of each cell was obtained by means of direct and indirect computation process. Finally the map of ADP was drawn and analyzed. Vulnerable cell and disturbance nuclei were determined according to disturbance vector which is a kind of potential disturbance pressure of relevant cell influenced by surrounding cell. A case study was accomplished in the Gijang area of Pusan metropolitan city. 973 meshes (500m×500m) were analyzed and a total of 79 meshes were currently threatened. Present technique of rapid ecoassessment was practically useful for diagnosing and planning land use.

The Relationship between Korea Agricultural Productions and Greenhouse Gas Emissions Using Environmental Kuznets Curve (환경쿠즈네츠곡선을 이용한 한국의 농업 생산과 온실가스 배출의 관계 분석)

  • Purpose - The purpose of this study was to investigate the relationship between Korea agricultural productions and Greenhouse Gas (GHG) emissions based on Environmental Kuznets Curve (EKC) hypothesis. Design/methodology/approach - This study utilized time series data of economic growth, greenhouse gas, agricultural productions, trade dependency, and energy usages. In order to econometric procedure of EKC hypothesis, this study utilized unit root test and cointegration test to check staionarity of each variable and also adopted Vector Error Correction Model (VECM) and Ordinary Least Square (OLS) to analyze the short and long run relationships. Findings - In the short run, greenhouse gas emissions resulting from economic growth show an inverse U-shape relationship, and an increase in agricultural production and energy consumption led to increase in greenhouse gas emission. In the long run, total GHG emissions and CO2 emissions show an N-shaped relationship with economic growth, and an increase in agricultural production has resulted in a decrease in total GHG and CO2 emissions. However, methane (CH4) and nitrous oxide (N2O) emissions showed an inverse U-shape relationship with economic growth, which indicated the environment and production process of agricultural production. Research implications or Originality - Korea agricultural production has different effects on the GHG emission sources, and in particular, methane (CH4) and nitrous oxide (N2O) emissions show to increase as the agricultural production expansions, so policy or technological development in related sector is required. Especially, in the context of the 2030 GHG reduction road-map, if GHG-related reduction technologies or policies are spread, national GHG emission reduction targets can be achieved and this is possible to predict the decline in production in the sector and damage to the related industries.

Constructing for Korean Traditional culture Corpus and Development of Named Entity Recognition Model using Bi-LSTM-CNN-CRFs (한국 전통문화 말뭉치구축 및 Bi-LSTM-CNN-CRF를 활용한 전통문화 개체명 인식 모델 개발)

  • Named Entity Recognition is a system that extracts entity names such as Persons(PS), Locations(LC), and Organizations(OG) that can have a unique meaning from a document and determines the categories of extracted entity names. Recently, Bi-LSTM-CRF, which is a combination of CRF using the transition probability between output data from LSTM-based Bi-LSTM model considering forward and backward directions of input data, showed excellent performance in the study of object name recognition using deep-learning, and it has a good performance on the efficient embedding vector creation by character and word unit and the model using CNN and LSTM. In this research, we describe the Bi-LSTM-CNN-CRF model that enhances the features of the Korean named entity recognition system and propose a method for constructing the traditional culture corpus. We also present the results of learning the constructed corpus with the feature augmentation model for the recognition of Korean object names.

The Analysis of the Effect of Fiscal Decentralization on Economic Growth: Centering The U. S. (재정분권화가 경제성장에 미치는 영향에 관한 실증연구: 미국의 경우를 중심으로)

  • Estimated coefficients has serious problems including inconsistency, biasness, etc. because many researches about the effect of fiscal decentralization on a country's economic growth use the traditional OLS method. Researches use the data intactly so that so called "spurious regression" phenomenon exists. This causes fundamental fallacy. This research tries unit root test, cointegration test, and then estimates the United States' economic time series by using VECM. The analysis of the effect of the state level-fiscal decentralization on economic growth shows two long term-equilibriums. During short term-dynamic adjustment, fiscal decentralization and economic growth move the same or different directions. In case of prediction GDP increases steeply and then from 2015 gently; and fiscal decentralization index shows a general reduction trend and then decreases slowly. At local level it shows two long term-equilibriums. During short term-dynamic adjustment, fiscal decentralization and economic growth also move the same or different directions. Impulse response analysis shows the very negative effect of fiscal decentralization on economic growth.

The Long-Run Relationship between House Prices and Economic Fundamentals: Evidence from Korean Panel Data (주택가격과 기초경제여건의 장기 관계: 우리나라의 패널 자료를 이용하여)

  • This paper adopts recently developed panel unit root test that is cross-sectionally robust. Cointegration test is also used to find whether regional house prices are in line with gross regional domestic production (GRDP) in the long run in Korea during 1989-2009. Based on the panel VECM and the panel ARDL models, we examine causal relationships among the variables and estimate the long-run elasticity. We find evidence of cointegration and bidirectional causal relationships between regional house prices and GRDP. The results of long-run estimates, using both fixed effect and ARDL models, show that house prices positively and significantly influence on the GRDP and vice versa. Together with these results, the findings of ARDL-ECM imply that there exists a long-run equilibrium relationship between house prices and regional economic variables even if there is a possibility of short-run deviation from its long-run path.
