Biological Functions of N- and O-linked Oligosaccharides of Equine Chorionic Gonadotropin and Lutropin/Chorionic Gonadotropin Receptor

  • Members of the glycoprotein family, which includes CG, LH, FSH and TSH, comprise two noncovalently linked $\alpha$- and $\beta$-subunits. Equine chorionic gonadotropin (eCG), known as PMSG, has a number of interesting and unique characteristics since it appears to be a single molecule that possesses both LH- and FSH-like activities in other species than the horse. This dual activity of eCG in heterologous species is of fundamental interest to the study of the structure-function relationships of gonadotropins and their receptors. CG and LH $\beta$ genes are different in primates. In horse, however, a single gene encodes both eCG and eLH $\beta$ -subunits. The subunit mRNA levels seem to be independently regulated and their imbalance may account for differences in the quantities of $\alpha$ - and $\beta$-subunits in the placenta and pituitary. The dual activities of eCG could be separated by removal of the N-linked oligosaccharide on the $\alpha$-subunit Asn 56 or CTP-associated O-linked oligosaccharides. The tethered-eCG was efficiently secreted and showed similar LH-like activity to the dimeric eCG. Interestingly, the FSH-like activity of the tethered-eCG was increased markedly in comparison with the native and wild type eCG. These results also suggest that this molecular can implay particular models of FSH-like activity not LH-like activity in the eCG/indicate that the constructs of tethered molecule will be useful in the study of mutants that affect subunit association and/or secretion. A single-chain analog can also be constructed to include additional hormone-specific bioactive generating potentially efficacious compounds that have only FSH-like activity. The LH/CG receptor (LH/CGR), a membrane glycoprotein that is present on testicular Leydig cells and ovarian theca, granulosa, luteal, and interstitial cells, plays a pivotal role in the regulation of gonadal development and function in males as well as in nonpregnant and pregnant females. The LH/CGR is a member of the family of G protein-coupled receptors and its structure is predicted to of a large extracellular domain connected to a bundle of seven membrane-spanning a-helices. The LH/CGR phosphorylation can be induced with a phorbol ester, but not with a calcium ionophore. The truncated form of LHR also was down-regulated normally in response to hCG stimulation. In contrast, the cell lines expressing LHR-t631 or LHR-628, the two phosphorylation-negative receptor mutant, showed a delay in the early phase of hCG-induced desensitization, a complete loss of PMA-induced desensitization, and an increase in the rate of hCG-induced receptor down-regulation. These results clearly show that residues 632~653 in the C-terminal tail of the LHR are involved in PMA-induced desensitization, hCG-induced desensitization, and hCG-induced down-regulation. Recently, constitutively activating mutations of the receptor have been identified that are associated with familial male-precocious puberty. Cells expressing LHR-D556Y bind hCG with normal affinity, exhibit a 25-fold increase in basal cAMP and respond to hCG with a normal increase in cAMP accumulation. This mutation enhances the internalization of the free and agoinst-occupied receptors ~2- and ~17- fold, respectively. We conclude that the state of activation of the LHR can modulate its basal and/or agonist-stimulated internalization. Since the internalization of hCG is involved in the termination of hCG actions, we suggest that the lack of responsiveness detected in cells expressing LHR-L435R is due to the fast rate of internalization of the bound hCG. This statement is supported by the finding that hCG responsiveness is restored when the cells are lysed and signal transduction is measured in a subcellular fraction (membranes) that cannot internalize the bound hormone.

The Study on Chinese Comics Characteristics (중국 만화 <삼모 유랑기> 의 특성 연구)

  • Mainland China is under different conditions both socially and politically than other countries, so there is a lack of practical researches on cartoon characters. With a profound interest, the researcher has selected a character that is realistic and loved by many Chinese. The subject of the study is "Shan Mao Series." Out of 16 seasons, the most famous one is the Chronicle of Shan Mao's Wander. I analyzed the character thoroughly, dealing with the beginning of the character, the process of character development, and a reality shown in the chronicle, etc. on chapter II, and the character's image component on chapter III. Chinese modern cartoons were affected by a situational anxiety because of foreign powers and war. Thus satire cartoons went in fads among the public. The epitome of a typical cartoon was the Shan Mao Series. From 1935, when the character was first created, to now it has eaten into Chinese people's hearts. It's because the story happens during a war period and it deals with a playful, humorous main character's life and these facts show the hidden side of the unfair social system. Most of readers prefer a smart, playful, righteous, and brave character. Shan means three and mao, hair in Chinese, so Shan Mao is a child who has three hair. The character is not just a simple cartoon character; it has developed into a national figure among many Chinese. The reason why the researcher has chosen the series created by Zhang Leping, the author of the cartoon, is because a good cartoon, good animation, and even good movie stimulate the feelings that we get from our surroundings. The character which is created in China seems very unique and bizarre but there is a sense of friendliness. Also its character image and scenes make people laugh and it has become a typical symbol of a modern cartoon in China.

A Study on Construction of Region-Based Cartoon Creation & Production Center (지역 중심의 만화 창· 제작센터 구축에 대한 연구)

  • This thesis aims to research ways for the regional balanced development of cartoon creation & production environment being centered in metropolitan area in Korean cartoon industry which has rapidly changed since 2013. As a cartoon can not only be produced with relative lower production cost comparing to those of other cultural contents industries, but also can be produced only if the minimal requirements for cartoon production is prepared, so the cartoon is a field that the decentralization can be accomplished very easily. Currently, most cartoon-relevant companies and cartoon promotion institutions are located in Seoul an Bucheon, etc. However, cartoon artists live nationwide, and even cartoon artists producing their works abroad are reached to a significant number. In some regions like Daejeon, Busan, Suncheon and Gyeongbuk (Gyeongsangbuk-do), there have been appeared full-scale movement to construct regional cartoon creation & production centers since 2015. This thesis aimed to investigate each region's movement to construct cartoon creation & production center with oversea cases, and to check how such movement could be balanced and harmonized with each region's unique features. First of all, this thesis analyzed the status quo of government's policy nurturing the cartoon industry. Korean government's cartoon-promotion policy around the axis of the Cartoon Industry's Mid.Long-Term Development Plan has been developed around the Korea Creative Content Agency and the Korea Manwha(cartoon) Contents Agency in Bucheon, but as the webtoon industry has rapidly grown up, the necessity for building a cartoon promotion institution in each region has been raised since 2015. With the establishment of 4th Cartoon Industry Mid.Long-Term Development Plan to be executed from 2019, it seems that full-scaled support framework for cartoon regional balanced development should be occupied. For the case of foreign countries, cartoon promotion institutions and relevant events have been developed around regions from early times like San-Diego, USA(Comicon), Angouleme, France(National Image Center), Kyoto (Cartoon Museum), Sakaiminato(Misuki Sigeru Road), Japan gave a lot of implications. In the section of conclusion, this study aimed to suggest the importance of and necessity for establishing a cartoon creation & production center in each region appropriately for the region's identity and characteristics with specific plans. Based on that, this thesis aimed to suggest a vision for cartoon & webtoon industry that regional creation & production system can be settled almost only in the cultural contents industry.

Research on Science, Technology & Society in Korea: A Critical Review (과학기술과 사회 연구의 현황과 과제)

  • The goal of the present study is reviewing the literature on the scientific community and also on science, technology & society to increase interactions between innovation studies and social studies of science and technology. Up until now, various empirical studies on Korean scientists and engineers have been concentrated on researchers at universities, while they have paid inadequate attention to researchers at state-funded research institutes and private companies. In addition, these studies have tended to use concepts in Western academia to elucidate Korean cases. On the other hand, recent empirical researches on the effects of the evaluation systems in universities, PBS system, and the network of school ties suggest that these topics may reveal the unique characteristics of Korean scientific community. Empirical studies on the scientific community have also shown that Korean research institutes and researchers who are in charge of innovation in Korea have demonstrated a tendency to conform to the government's guidance due to long experiences of state-led R&D and nationalism. Research on science, technology and society has viewed the participation of citizens in science and technology as a way toward science and technology democracy, and tended to have a strong practical orientation. However, there has been a relatively small amount of research on how citizen participation influences the direction and content of technological innovation. Also, although, from the viewpoint of technological innovation, how participation of citizens in science and technology can contribute to knowledge production and innovation is a critical issue, relatively small numbers of case studies on this subject have been conducted. Therefore, as the scholars who have emphasized the democracy of science and technology have actually experimented with various ways of citizen participation, innovation researchers may have to design and implement citizen participation through which citizens' local knowledge can contribute to technological innovation.

The Effect of Information Service Quality on Customer Loyalty: A Customer Relationship Management Perspective (정보서비스품질이 고객로열티에 미치는 영향에 관한 연구: 고객관계관리 관점)

  • As managing customer relationship gets more important, companies are strengthening information service using multi-channels to their customers as a part of their customer relationship management (CRM) initiatives. It means companies are now accepting such information services not as simple information -delivering tools, but as strategic initiatives for acquiring and maintaining customer loyalty. In this paper, we attempt to validate whether or not such various information services would impact on organizational performance in terms of CRM strategy. More specifically, our research objective is to answer the next three questions: first, how to construct the instruments to measure not information quality but information service quality?; second, which attributes of information service quality can influence corporate image and customer loyalty?; finally, does each information service type have unique characteristics compared with others in terms of influencing corporate image and customer loyalty? With respect to providing answers to those questions, the previous studies had been limited in that those studies failed to consider the variety of types of information service or restricted the quality of information service to information quality. An appropriate research model answering the above questions should consider the fact that most companies are utilizing multi channels for their information services, and include the recent strategic information service such as customer online community. Moreover, since corporate information service could be regarded as a type of products or services delivered to customer, it is necessary to adopt the criteria for assessing customer's perceived value when to measure the quality of information service. Therefore, considering both multi-channels and multi-traits may enable us to tell the detailed causal routes showing which quality attributes of which information service would affect corporate image and customer loyalty. As information service channels, we include not only homepage and DM (direct mail), which are the most frequently applied information service channels, but also online community, which is getting more strategic importance in recent years. With respect to information service quality, we abstract information quality, convenience of information service, and timeliness of information service through a wide range of relevant literature reviews. As our dependant variables, we consider corporate image and customer loyalty that both of them are the critical determinants of organizational performance, and also attempt to grasp the relationship between the two constructs. We conducted a huge online survey at the homepage of one of representative dairy companies in Korea, and gathered 367 valid samples from 407 customers. The reliability and validity of our measurements were tested by using Cronbach's alpha coefficient and principal factor analysis respectively, and seven hypotheses were tested through performing correlation test and multiple regression analysis. The results from data analysis demonstrated that timeliness and convenience of homepage have positive effects on both corporate image and customer loyalty. In terms of DM, its' information quality was represented to influence both corporate image and customer loyalty, but we found its' convenience have a positive effect only on corporate image. With respect to online community, we found its timeliness contribute significantly both to corporate image and customer loyalty. Finally, as we expected, corporate image was revealed to provide a great influence to customer loyalty. This paper provides several academic and practical implications. Firstly, we think our research reinforces CRM literatures by developing the instruments for measuring information service quality. The previous relevant studies have mainly depended on the measurements of information quality or service quality which were developed independently. Secondly, the fact that we conducted our research in a real situation may enable academics and practitioners to understand the effects of information services more clearly. Finally, since our study involved three different types of information service which are most frequently applied in recent years, the results from our study might provide operational guidelines to the companies that are delivering their customers information by multi-channel. In other words, since we found that, in terms of customer loyalty, the key areas would be different from each other according to the types of information services, our analysis would help to make decisions such as selecting strengthening points or allocating resources by information service channels.

The Relationship between attribution styles and attitude toward mathematics of mathematically gifted students and those of regular students at elementary schools (초등학교 수학영재와 일반학생의 귀인성향과 수학에 대한 태도와의 관계)

  • The purpose of this study is to provide information that will help understand unique characteristics of mathematically gifted students and that can be utilized for special programs for mathematically gifted students, by investigating difference and relationship between attribution styles and attitude toward mathematics of mathematically gifted students and those of regular students. For that purpose, 202 mathematically gifted students and 415 regular students in 5th and 6th grades at elementary schools were surveyed in terms of attribution styles and attitude toward mathematics, and the result of the study is as follows. First, as for attribution styles, there was no difference between gifted students and regular students in terms of grade and gender, but there was significant difference in sub factors because of giftedness. Second, there was not significant difference between grades. but there was significant difference in sub factors between genders. Mathematically gifted students were more positive than regular students in every sub factor excepting gender role conformity, and especially they showed higher confidence and motivation. Third, according to the result of correlation analysis, there was significant static correlation between inner tendencies and attitude toward mathematics with both groups. The gifted group showed higher correlation between attribution of effort and attitude toward mathematics and inner tendencies and confidence than the regular group. The gifted group showed higher correlation in sub factors, and especially there was high static correlation between attribution of talent and confidence, and attribution of effort and motivation. Fourth, according to the result of multiple regression analysis, inner tendencies showed significant relation to attitude toward mathematics with both groups, and especially the influence of attribution of effort was high. Both attribution of effort and attribution of talent were higher in the gifted group than the regular group, and attribution of effort had a major influence on practicality and attribution of talent had a major influence on confidence.

A Study on a Basic Model for GIS Audit, Based on Various Types of GIS Projects (GIS 사업유형을 고려한 GIS 감리의 기반 모델 연구)

  • Since 1995, national and local governments have competitively initiated many and large GIS projects and audit for the projects becomes an important issue. So far, the audit in the Information Technology(IT) area has tried to deal with the issue but ineffectiveness has been found for the successful GIS project management. Effective auditing is a critical element for the project management. In order to establish a proper audit model for the GIS projects and to promote auditing activities in the projects, this study constructs two hypotheses and tries to prove them. The hypotheses are as follows : 1) For a good audits model for GIS, unique characteristics of a GIS project audit items and the scope of the audit need to be identified. 2) The scope of audit needs to be classified according to the requests from tasks in the projects. To prove the hypotheses, this study analyzes positive aspects of audit in IT and construction projects, clarifies the audit items in GIS projects by comparing with them, and classifies the scope of the GIS audit based on various types of GIS projects. As a results, 5 types of the GIS audit are identified : (1) audit for project management, (2) audit focused on IT, (3) audit characterized by GIS technologies, (4) GIS database audit and (5) consulting services for critical problems in the projects. In addition, 4 criteria in classifying the GIS projects are suggested for the GIS audit. The 4 criteria are domain, scope, duration, and GIS applications technologies. Especially, GIS technology considered in this study includes GIS software, methodologies for GIS development, GIS database and quality control of GIS data, which are not usually reflected in the existing studies about in GIS audit. Because the GIS audit depends on a type of GIS projects, scopes of the audit can be flexibly reconstructed in accordance with the types of GIS projects. This is a key to effective and realistic audit for the future GIS projects. Strategies for effective GIS audit are also proposed in terms of the following: GIS project management, goal establishment in each audit stage, documentation from GIS audit, timing strategies for intensive GIS audit, and designing team structure.

Flowering Patterns of Miscanthus Germplasms in Korea (국내 수집 억새 유전자원의 출수 특성)

  • Miscanthus has been considered as the most promising bioenergy crop for lignocellulosic biomass production. In Korea, M. sacchariflorus and M. sinensis can be found easily in all regions. It is a great advantage to utilize as important species with respect to genetic and cross-breeding programs materials for creation of novel hybrids. For successful breeding programs, it is important to precisely understand the variability of flowering traits among Miscanthus species as breeding parents materials. In this study, flowering traits were observed daily in 960 germplasms of two Miscanthus species (M. sacchariflorus and M. sinensis) for growing seasons over 2 years. The flowering process was divided into three stages. ST (sprouting time) was recorded when first leaf of the plant emerged on soil. FS1 (flowering stage 1) and FS2 (flowering stage 2) were recorded when flag leaf was firstly observed, and 1 cm of panicle was showing on at least one stem, respectively. For 2013 and 2014, the latest germplasms exerted flag leaf, i.e. September 30 (DOY of FS1 164.1) and September 4 (DOY of FS1 141.0) occurred M. sacchariflorus cv. Geodae 1 and M. sacchariflorus cv. Uram collected from Southern Korea (Jeollanam-do), while Miscanthus germplasms collected from northern Korea (Gyeonggi-do) which emerged the earliest flag leaf in July and August, significantly decreased DOY. For DOY from ST to FS2, M. sacchriflorus germplasms ranged from 140 to 190 days, and 110 to 170 days for 2013 and 2014. The highest frequency showed to 160 days for 2013, and 150 days for 2014. In M. sinensis germplasms, the highest frequency showed to 180 days for 2013, and 170 days for 2014. In the results of correlation between the day of years from ST to FS2 for 2013 and 2014, M. sacchriflorus and M. sinensis showed high coefficient of correlation (0.70 and 0.89). It can be supposed that flowering characteristics of Miscanthus are largely affected by the unique phenotypic characteristic of native habitat than environmental factors of the current planted site. This study for flowering traits of Miscanthus may provides an important information in order to expedite the introduction as breeding materials for creation of new hybrid.

The Study on Geology and Volcanism in Jeju Island (II): Petrochemistry and $^{40}Ar/^{39}Ar$ Absolute Ages of the Volcanic Rocks in Gapado-Marado, Jeju Island (제주도의 지질과 화산활동에 관한 연구 (II): 가파도와 마라도 화산암류의 암석화학 및 $^{40}Ar/^{39}Ar$ 절대연대)

  • We report petrologic characteristics including $^{40}Ar-^{39}Ar$ absolute ages of the subsurface lavas recovered from borehole cores in two islets, Marado and Gapado, off the southwestern coast of Jeju in southernmost Korea and discuss on the volcanism in the region. The lavas in Gapado are apparently divided into one unit with bright colored, aphanitic texture and sheet jointed, and another unit with dark colored and massive. The outcrops often show differentially weathered pattern due to textural difference. While, the lavas in Marado have vesicular and glomerporphyric texture, even though each lava flow unit in Marado has slight unique texture with variation of vesicularity and phenocrysts. The chemical composition of rock core samples from Gapa borehole and Mara borehole shows that the lavas from Gapado and Marado are classified into basaltic trachyandesite($SiO_2$ 52.6-53.6 wt%, $Na_2O+K_2O$ 7.3-7.5 wt%) and tholeiitic andesite($SiO_2$ 51.7-52.8 wt%, $Na_2O+K_2O$ 3.6-4.1 wt%), respectively. The measured $^{40}Ar-^{39}Ar$ plateau ages range from $824{\pm}32\;Ka$(MSL -69 m) to $758{\pm}\;Ka$(MSL 19 m) for core samples of Gapa borehole and $259{\pm}168\;Ka$(MSL -26 m) for a core sample of Mara borehole, respectively. The absolute age of Gapado basaltic trachyandesite is well correlated with that of Sanbangsan trachyte(Won et al., 1986). Meanwhile, the age of a sample in Marado has $259{\pm}168\;Ka$(MSL -26 m) with poor plateau age formation and high error range. We report the data in caution but the rock composition and absolute age of Marado tholeiitic andesite are relatively correlated with those of lava units from Duksu and Sangmo-2 boreholes, indicating the volcanism during 260-150 Ka. On the basis of interpretation of occurrences of exposed and subsurface volcanic rocks of the study area, stratigraphic relationship with adjacent borehole cores and the bathymetry chart of surrounding area, it indicates that the lavas in Gapado were formed around 800 Ka during relatively early stage of volcanic activity in Jeju Island. Meanwhile, Marado may have originated around 260-150 Ka during relatively young stage of volcanism in Jeju Island. It is inferred that the volcanisms have originated in land and these islets were individual ancient volcanoes. The apparent topography has been re-shaped by tidal erosion due to transgression.

Analysis of Genetic Characteristics and Probability of Individual Discrimination in Korean Indigenous Chicken Brands by Microsatellite Marker (MS 마커를 이용한 토종닭 브랜드의 유전적 특성 및 개체 식별력 분석)

  • Microsatellite markers have been a useful genetic tool in determining diversity, relationships and individual discrimination studies of livestock. The level of genetic diversity, relationships among two Korean indigenous chicken brand populations (Woorimatdag: WR, Hanhyup3: HH) as well as two pure populations (White Leghorn: WL, Rhode Island Red: RIR) were analyzed, based on 26 MS markers. A total of 191 distinct alleles were observed across the four chicken populations, and 47 (24.6%) of these alleles were unique to only one population. The mean $H_{Exp}$ and PIC were estimated as 0.667 and 0.630. Nei's $D_A$ genetic distance and factorial correspondence analysis (FCA) showed that the four populations represented four distinct groups. However, the genetic distance between each Korean indigenous chicken brand (WR, HH) and the pure population (WL, RIR) were threefold that among the WR and HH. For the STRUCTURE analyses, the most appropriate number of clusters for modeling the data was determined to be three. The expected probabilities of identity among genotypes of random individuals (PI) were calculated as $1.17{\times}10^{-49}$ (All 26 markers) and $1.14{\times}10^{-15}$, $7.33{\times}10^{-20}$ (9, 12 with the highest PI value, respectively). The results indicated that the brand chicken breed traceability system employing the own highest PI value 9 to 12 markers, and might be applicable to individual identification of Korean indigenous chicken brand.
