The Effects of Prunus on Diabetic Nephropathy Rats Induced by Unilateral Nephrectomy and Streptozotocin (도인(挑仁)이 일측 신절제와 streptozotocin으로 유발된 당뇨병성 신증 Rat에 미치는 영향)

  • Objectives: Diabetic nephropathy is the most common cause of end stage renal disease. Transforming growth factor (TGF)-${\beta}1$, type IV collagen, advanced glycation end-products (AGEs), and angiotensin II type 1 receptor (AT1) are the main factors of diabetic nephropathy. We investigated the effects of Prunus on renal function and histopathological changes of diabetic nephropathy rat model induced by unilateral nephrectomy and streptozotocin. Methods: Diabetes was induced in male Sprague-Dawley rats ($290{\pm}10g$) by injecting streptozotocin (55 mg/kg) into the tail vein after unilateral nephrectomy. Rats were divided into 3 groups (n=6): normal, control, and Prunus. After 8 weeks of oral administration of Prunus extract on the Prunus group from 3 days after streptozotocin injection, we checked weight, 24 hrs urine, blood biochemistry and renal tissue to evaluate renal function and histopathological changes by examining parameters including albuminuria, BUN, creatinine, cholesterol, low density lipoprotein (LDL), triglyceride, TGF-${\beta}1$, type IV collagen, AGEs, and AT1. We also measured mRNA expression of TGF-${\beta}1$, type IV collagen, AGEs, and AT1 by Real Time polymerase chain reaction (RT-PCR). Results: Prunus decreased the amount of 24 hrs proteinuria, and inhibited histopathological changes of diabetic nephropathy including the expression and accumulation of TGF-${\beta}1$, type IV collagen and AGEs which could promote development of diabetic nephropathy. Prunus also inhibited mRNA expression of TGF-${\beta}1$, type IV collagen. Conclusions: These findings suggest that Prunus might protect the renal function and inhibit the development of renal injury by regulating factors including TGF-${\beta}1$, type IV collagen, AGEs, except AT1, so Prunus can be used for diabetic patients to prevent the progression of diabetic nephropathy.

Radiographic and Ultrasonographic Diagnosis of Unilateral Ovarian Cysts (개에서 편측성 난소낭종의 방사선학적 및 초음파학적 진단 2례)

  • A female, 14-year-old mixed dog weighing 4 kg and a female,8-year-old schnauzer dog weighing 6 kg showing vaginal discharges and vomiting for a few weeks were examined for the preoperative check of anterior cruciate ligament rupture and the recheck after the surgery of nephrectomy. There were no distinct abnormal signs except for the increased CBC at schnauzer dog on blood chemistry analysis. Radiographic and ultrasonographic findings were a large abdominal mass and a cyst-like hypoechogenic or anechogenic cavitary lesion on the ovary region. These findings were consistent with an ovarian cyst. Thereafter highly suspected ovarian cysts by diagnostic imaging were supported by high estrogen concentration on hormonal assay and the results of Pathologic examination as the unilateral ovarian cysts with secondary pyometra.

A Rare Case of Canine Huge Renal Cell Carcinoma with Ovarian Metastasis

  • A 10-year-old female Pomeranian weighing 2.25 kg with a history of gradual weight loss and an intraabdominal mass confirmed with ultrasonography at a local hospital was referred to the Veterinary Medical Teaching Hospital, Jeju National University. Physical examination revealed abdominal distention. Blood analysis revealed hypoglycemia (57 mg/dL; reference range, 60-110 mg/dL). On abdominal ultrasonography, a heterogeneously hyperechoic mass measuring 7.51 × 10.6 cm was found at the level of the left kidney. Computed tomographic findings showed a unilateral left kidney mass with a 10-cm diameter and vessel contrast enhancement in the corticomedullary phase. Unilateral nephrectomy and ovariohysterectomy were performed because left ovary enlargement was observed during surgery. The renal mass was adjacent to the aorta and vena cava and attached to a portion of the small intestine, greater omentum, and left ovary. The mass was huge (12.5 × 10 × 7 cm) and was 16.5% of the body weight. Histopathological examination revealed renal cell carcinoma (RCC) and ovarian metastasis. After surgery, clinical signs improved remarkably, and serum glucose level returned to normal. As RCC is resistant to radiation and chemotherapy, the owner decided not to proceed with postoperative adjunctive therapies. To the best of our knowledge, this is the first case report of ovarian metastasis with huge RCC in a dog.

  • Emphysematous pyelonephritis is a necrotizing renal infections characterized by intrarenal and occasional perirenal gas production. Although uncommon (89 cases in the literature), it occurs almost exclusively in diabetic patients (87% of the cases). Prompt and aggressive management is required to salvage these patients. We describe a recent case of a diabetic woman with emphysematous pyelonephritis due to E. Coli, successfully managed with unilateral nephrectomy.

A Case of Unilateral Renal Lymphosarcoma in a Dog (개에서의 편측성 신장 림프육종 1예)

  • A 10-year-old, female spayed mixed-breed dog with a history of vomiting and anorexia was examined. Abnormal findings of comlete blood count and serum chemistry included polycythemia, thrombocytopenia, hyper-globulinemia and hypoalbuminemia. Abdominal radiographs revealed severe unilateral renomegaly, and ultrasonography showed a left-sided renal mass. During the operation, left kidney was resected. Cytologial and histopathological examinations revealed neoplastic lymphoid proliferation with high mitotic figures in renal mass. Immunohistochemistry revealed tumor cells were CD3-positive and CD79${\alpha}$-negative, consistent with T-cell lineage. The renal mass of this case was diagnosed as renal T cell lymphosarcoma.

Antioxidant Effects of Ascorbic Acid on Renal-Ischemia Reperfusion Injury in Rabbit Model

  • Renal ischemia-reperfusion (I/R) injury is great clinical important because viability of the organ depends on the tolerance to ischemia-reperfusion injury, an inevitable processing during surgery. The purpose of this study was to investigate the effects of premedicated ascorbic acid alone in I/R injury model induced by cross-clamping of renal vessels. In the rabbit models, 2-4 kg New Zealand white rabbits were subjected to 30 minutes of warm unilateral renal ischemia followed by removal of contralateral kidney and then divided into five groups, control (2) arid treatment groups (3). In control group 1, the rabbits only received right nephrectomy. In control group 2, the rabbits received I/R on left kidney after the right nephrectomy. In treatment group 1, the rabbits received ascorbic acid 50 mg/kg IV before the operation. In treatment group 2, the rabbits received ascorbic acid 100 mg/kg IV before the operation. In treatment group 3, the rabbits received ascorbic acid 200 mg/kg IV before the operation. Blood samples were collected from these rabbits for measurement of kidney function tests at the 0, 1 st, 3rd and 7th day and antioxidant enzyme( SOD, GSHPx, CAT) at 24 hours. Kidney function tests (serum creatinine and BUN) showed a significant difference between group 2 and group 4, 5. Activity of antioxidant enzymes in plasma were significant decrease in group 4, 5 compare to group 2. The result of this study suggested that the exogenous ascorbic acid had a role of attenuation of renal I/R injury in rabbit model.

Clinicopathological Studies on the Experimentally Induced Nephrosis of Korea Black Goats (한국흑염소에서의 실험적 신증의 임상병리학적 연구)

  • In order to study the effects of administration of neomycin on the kidney of Korean black goats, 4.5(group A), 9(group B) or 14(group C) mg/kg, body weight of neomycin was injected intramuscularly turice a day for 84 days. On day 31, unilateral nephreotomy was performed in one animal of each group. The results were as follows: Serum neomycin concentrations were 8.7, 13.8, and 20.2 ${\mu}$g/ml 12 hours after injection. Serum BUN and creatinine concentration in nephreotomized goats, regardless of dosage, rapidly increased and came to peak on 3 days after, and then began to decrease to normal range, gradually, In non-nephrectomized goats, regardless of dosage, there was no notable change. In nephreotomized goats, PSP Tl/2 was delayed to the utmost 1 day after nephrectomy, regard-less of dosage, and then began to decrease to normal range, gradually in non-nephrectomized goats, there was not notable change in PSP T$\frac{1}{2}$. The intact kidney was markedly enlarged compared with the kidney of non-nephrectomized goats, but there was no microscopic change. It is concluded that administration of neomycin up to 14mg/kg body weight twice a day for 84 days was not nephrotoxic to the Korean black goats.

Effect of Dietary Protein Level on Renal Function : Lipid and Eicosanoids Metabolism in Uninephrectomized Aging Model in Rats (식이 단백질 수준이 한쪽 신장을 절제한 흰쥐에서 혈청 지질 및 Eicosanoid 대사에 미치는 영향)

  • This study was performed to elucidate the mechanism of dietary protein level on renal function through lipid and eicosanoids metabolism. Male rats of 337.8$\pm$5.7g body weight were undergone unilateral nephrectomy or sham-operation. The rats were divided into high protein(40% casein), normal protein(15% casein) and low protein(8% casein) diet groups and fed experimental diets ad libitum for 24 weeks. The results are summarized as follows. Serum total lipid, cholesterol and HDL-cholesterol of rats in 15% and 40% casein groups were higher than those of 8% casein group. But serum triglyceride was affected neither by uninephrectomy nor by dietary protein level. Serum thromboxane(TX) B2 and 6-keto prostaglandin F1$\alpha$ increased with increasing dietary protein level. Serum prostaglandin(PG) E2 was not affected by uninephrectomy nor by dietary protein level. Urinary PGE2 and TXB2 excretion tended to be lower in uninephrectomized groups. Renal tissue concentration of TXB2 was lower in uninephrectomized groups and in high protein group. These results suggest the possibility that the effects of dietary protein level on renal function could be due to changes in lipid and eicosanoids metabolism.

Transarterial embolization of renal artery in dogs with experimental hydronephrosis (개의 실험적 수신증에 실시한 신동맥 색전술)

  • This study was performed to validate the procedure of transarterial embolization of the renal artery (TAE-RA) using iohexol-ethanol solution in dogs with unilateral experimental hydronephrosis and to evaluate the embolized kidney using B-mode ultrasonography and selective angiography. Experimental hydronephrosis was induced by ligation of unilateral ureter in 12 dogs. Ultrasonographic findings revealed that size of the kidney was significantly increased at 9 days and 17 days and the length of renal cortex was significantly decreased at 17 days after ligation of the unilateral ureter and it was in accordance with dilation of ipsilateral renal pelvis. No significant change of BUN, creatinine, ALT, calcium, and phosphorus was found immediately after unilateral experimental hydronephrosis. Therefore, it was concluded that unilateral hydronephrosis was established in 12 dogs at 17 days after ligation of ureter. Renal artery embolization was performed using selective catheterization in the hydronephrotic kidney of seven dogs and EKG, $SpO_2$, body temperature, pulse, and respiratory rate were within normal ranges during procedures. Iohexol-ethanol solution was used as embolic material. Average ethanol dose for renal artery embolization was $1.94{\pm}1.24ml/kg$. There were no dogs expired after TAE-RA and no side effects associated with regurgitation of iohexol-ethanol solution. Revascularization of renal artery was not found in angiography in dogs treated by TAE-RA at immediately after TAE-RA and 14 days after TAE-RA. Ultrasonographically, the mean longitudinal length of the embolized kidney decreased significantly at 2 and 3 months after TAE-RA compared to that of contralateral normal kidney. In summary, marked shrinkage of the embolized kidney was observed in dogs with unilateral experimental hydronephrosis treated by TAE-RA with iohexol-ethanol and no adverse effects were observed throughout the observation period. It is concluded that TAE-RA with iohexol-ethanol solution is a viable alternative to nephrectomy in dogs with unilateral hydronephrosis.

Renal Tubular Carcinoma in a Cocker Spaniel Dog (코커 스파니엘에서 신장세포암종)

  • A 7-year-old neutered male Cocker spaniel was referred to Veterinary Medical Teaching Hospital, Kyungpook National University with a left abdominal mass, anorexia and weight loss. Hematologic test and biochemical panel showed severe leukocytosis, mild anemia, mild hypoalbuminemia and a marked elevation in gamma glutamyl transferase (GGT). Radiography demonstrated a large mass on the left kidney and there was no evidence of pulmonary metastases. Histopathologic features of the mass were consistent with renal tubular carcinoma. The dog was treated with unilateral nephrectomy. During the follow-up, the dog died 6 months after surgery.
