  • To develop a stalk detaching system the effect of shape factor of red pepper affecting the performance of unfolding, arranging and cutting was analysed. The obtained results are as follows : By cutting experiment, it was found that the bending of stalk affected the cutting rate of stalk, and that the bending of body increased the amount of peppers that were expelled from the cutting guide by conveying brush. The ratios, 'bending length of a body/body length'and 'bending length of a stalk/stalk length', could be used as criteria far abnormality of body and stalk of peppers, respectively. As a result of experiment, it was concluded that mechanical treatment would be difficult for the peppers with indexes greater than 0.4 and 0.3 fur body bending and stack bending. respectively. So, these indexes were used as criteria for distinguishing abnormality from normality of peppers. In the unfolding unit, conveyance of peppers was impossible for both of normal and abnormal ones at the inclination angle of 10°, especially, at the frequency of 8.3 Hz peppers maintained stationary state. At the inclination angle of 20°, both of normal and abnormal peppers showed similar tendencies, but abnormal ones showed an accumulation trend gradually with increased feeding speed. In the arranging unit, conveyance of peppers was almost impossible for both of normal and abnormal ones at the inclination angle of 20°, showing almost no difference between the conveyances of normal and abnormal ones. In the case of the inclination angle of 30°, at the condition of the feeding speeds and frequency corresponding 0.06 m/s, 0.08 m/s and 8.3 Hz, respectively, the passing time of the abnormal peppers on the arranging plate increased rapidly.

A Fusion Tag to Fold on: The S-Layer Protein SgsE Confers Improved Folding Kinetics to Translationally Fused Enhanced Green Fluorescent Protein

  • Genetic fusion of two proteins frequently induces beneficial effects to the proteins, such as increased solubility, besides the combination of two protein functions. Here, we study the effects of the bacterial surface layer protein SgsE from Geobacillus stearothermophilus NRS 2004/3a on the folding of a C-terminally fused enhanced green fluorescent protein (EGFP) moiety. Although GFPs are generally unable to adopt a functional confirmation in the bacterial periplasm of Escherichia coli cells, we observed periplasmic fluorescence from a chimera of a 150-amino-acid N-terminal truncation of SgsE and EGFP. Based on this finding, unfolding and refolding kinetics of different S-layer-EGFP chimeras, a maltose binding protein-EGFP chimera, and sole EGFP were monitored using green fluorescence as indicator for the folded protein state. Calculated apparent rate constants for unfolding and refolding indicated different folding pathways for EGFP depending on the fusion partner used, and a clearly stabilizing effect was observed for the SgsE_C fusion moiety. Thermal stability, as determined by differential scanning calorimetry, and unfolding equilibria were found to be independent of the fused partner. We conclude that the stabilizing effect SgsE_C exerts on EGFP is due to a reduction of degrees of freedom for folding of EGFP in the fused state.

Factors affecting the vertical distribution of Betula platyphylla var. japonica and Betula ermanii on Mt. Neko in Nagano Prefecture, Japan

  • Betula platyphylla var. japonica and Betula ermanii segregate vertically at an elevation of approximately 1,850 m on Mt. Neko in Nagano Prefecture, Japan. B. platyphylla var. japonica and B. ermanii were the dominant species below and above this altitude, at which the mean-annual and growing-season air temperatures were $4^{\circ}C$ and $14.1^{\circ}C$, respectively. Based on a modification of Kira's warmth index which employs cumulative temperature represented as $^{\circ}C$ day, leaf unfolding in both species was observed to be initiated at $58^{\circ}C$ day and $169^{\circ}C$ day, respectively. In 1996, leaf unfolding was initiated on 18 May in B. platyphylla var. japonica (+/-6 days) and on 5 June in B. ermanii (+/-8 days), shortly after the last frost which occurred on 5 May 1995 above 1,850 m; below this elevation there was no risk of frost at the time. At elevations above 1,850 m, the unfolded leaves of B. platyphylla were damaged by late frost, while B. ermanii escaped injury because the leaves were still protected by winter buds. The optimum temperature for seed germination in both B. platyphylla and B. ermanii was $30^{\circ}C$. Temperature alternation from 10 to $30^{\circ}C$ and moist storage of seeds at $4^{\circ}C$ (stratification) prior to incubation increased germination rates in both species. The seedlings of B. ermanii had a greater survival rates than those of B. platyphylla var. japonica when planted above 1,850 m. Comparisons of the timing of leaf unfolding and the latest frost at a site appeared to be the main factors affecting the vertical distribution of these species.

폴더 여닫이에 따른 휴대폰 디자인 이미지 비교 연구 (A Study on the Mobile Phone Design Image Comparison in the State of Folding and Unfolding)

  • 본 연구는 소비자의 감성에 민감할 것으로 예상되는 휴대폰 제품에 관하여 감성 어휘를 추출하고 어휘 관계 구조를 분석하여 휴대폰의 디자인 이미지를 파악하기 위해 수행되었다. 휴대폰 폴더가 열린 경우와 닫힌 경우로 나누어 각각의 경우 감성 이미지가 다르게 나타날 것으로 예상하였다. 어휘 수집을 통해 휴대폰 관련 감성 대표 어휘 25개가 추출되었다. 추출된 감성 어휘로 20개의 휴대폰 샘플을 감성 평가하여 요인 분석한 결과, 휴대폰감성 이미지는 열린 경우와 닫힌 경우가 다르게 나타났다. 열린 경우는 제1요인에서 '첨단적이다, 세련되다, 좋다, 돋보이다. 고급스럽다. 아름답다. 이쁘다, 정교하다'가 제2요인에서 '곡선적이다, 부드럽다, 여성적이다, 자연적이다' 가 제3요인에서 '가볍다. 슬림하다, 작다' 가 추출되어 각기 '쿨하다', '소프트하다', '핸디하다' 로 명명하였다. 닫힌 경우는 제1요인에서 '세련되다, 좋다, 정교하다. 아름답다, 이쁘다. 균형적이다, 고급스럽다, 뚜렷하다'가, 제2요인에서 '화려하다, 개성있다' 가, 제3요인에서 '기계적이다, 남성적이다, 딱딱하다, 현대적이다' 가 추출되었으며 이는 각각 '미려하다', 다채롭다', '하드하다' 로 명명하였다. 이를 통해 휴대폰은 폴더 여닫이에 따라 추구해야 할 이미지가 다르게 디자인되는 것이 고려되어야함을 알 수 있다.

Improving the Surface Functionality of Curdlan by Conjugation with Unfolding Protein through Naturally Occurring Maillard Reaction

  • Protein conjugation of curdlan belonging to $\beta$-1, 3-glucan was carried out to improve it surface functionalities. The glucan was mixed with phosvitin, {TEX}$$\alpha$_{s}${/TEX}-casein, lysozyme or ovalbumin, respectively. The mixture was freeze-dried, and he resulting powder was incubated at 6$0^{\circ}C$ and 79% relative humidity for 12 days in order to generate a controlled Maillard reaction between curdlan and proteins. conjugation with unfolding proteins, i.e., phosvitin and {TEX}$$\alpha$_{s}${/TEX}-casein, drastically increased the solubility of the glucan, whereas lysozyme and ovalbmin did not. The solubility in water of curdlan was 3.44% for the phosvitin conjugate and 1.09% for the {TEX}$$\alpha$_{s}${/TEX}-casein conjugate. SDS-slab polyacrylamide gel electrophoresis showed that curdlan was solubilized due to covalent binding with phosvitin. Emulsifying properties of curdlan were substantially improved by the conjugation with phosvitin and {TEX}$$\alpha$_{s}${/TEX}-casein. Emulsion stability of the curdlan-phosvitin conjugate was about 2.9 times greater than that of the curdlan-phosvitin mixture.

영양소분지에 분포하는 경상누층군에 대한 고지자기 연구 (Paleomagnetism of the cretaceous sedimentary rocks in the Yongyang Sub-Basin)

  • Paleomagnetic and rock magnetic investigations have been carried out for the Cretaceous Hanyang Group, exposed in the Yongyang Sub-Basins within the Kyeongsang Basin, eastern South Korea. A total of 452 oriented core samples was drilled from 31 sits for the study. The in-situ site mean direction is more dispersed than the mean direction after bedding correction, indicating that the fold test is positive at 95% confidence level. In addition, the stepwise unfolding of the characteristic remanent magfold test is positive at 95% confidence level. In addition, the stepwise unfolding of the characteristic remanent magnetization reveals that a maximum value of k is observed at 90% unfolding. Furthermore, the rock magnetic investigations and electron microscope observations of the representative samples show that the main magnetic carrier of the Hayang Group is the detrital specular hematite of single and pseudo-single domain sizes with negligible contribution of pigmentary hematite grains. These results collectively imply that the ChRM direction is the primary component acquired at the time of the formation of the strata. Provided the primary nature of the ChRM, a magnetostratigraphic correlation between polarities of the studied formation and the Geomagnetic Time Scale indicates that the Hayang Group in the Yongyang Sub-Basin can be correlated to the Cretaceous Long Normal superchron. The paleomagnetic pole position from this study is significantly different from those of the Hayang group in the Euiseong the Milyang sub-Basins. Rather the paleomagnetic pole position of the Hayang Group of the study area is closer to that of the Quaternary period or present time of the Korean Peninsula. It is hypothesized that the study area might be rotated about 25$^{\circ}$ aticlockwise with respect to the Euiseong and Milyang Sub-Basins after the formation of the strata and aquisition of the ChRM, although there is not enough geologic evidence supporting the rotation hypothesis.

  • Protein unfolding is a key step in several cellular processes, including protein translocation across some membranes and protein degradation by ATP-dependent proteases. C1pAP protease and the proteasome can actively unfold proteins in a process that hydrolyzes ATP, These proteases catalyze unfolding by processively unraveling their substrates from the attachment point of the degradation signal. As a consequence, the ability of a protein to be degraded depends on its structure as well as its stability. An ${\alpha}$-helix is easier to unravel than a ${\beta}$-strand. In multidomain proteins, independently stable domains are unfolded sequentially. The steric constraints imposed on substrate proteins during their degradation by the proteasome were investigated by constructing a model protein in which specific parts of the polypeptide chain were covalently connected through disulfide bridges. The cross-linked model proteins were fully degraded by the proteasome, but two or more cross-links retarded the degradation slightly. Our results suggest that the pore of the proteasome allows the concurrent passage of at least three stretches of a polypeptide chain, and also explain the limited degradation by the proteasome that occurs in the processing of the transcription factor NF-KB, and also implicate difficulty in degradation of amyloidal aggregates by the proteasome

물질투과율 측정정보 기반 의료용 선형가속기의 에너지스펙트럼 유도기술 개발 (Development of Unfolding Energy Spectrum with Clinical Linear Accelerator based on Transmission Data)

  • 연구배경: 세기조절방사선치료와 같이 고선량을 암조직에 정밀하게 전달할 수 있는 방사선 치료기술이 개발됨에 따라, 보다 정확한 선량평가 기술의 개발이 요구되고 있다. 본 연구에서는 선형가속기에서 발생된 광자선의 물질투과율 정보를 통해 간접유도방식으로 에너지스펙트럼을 예측할 수 있는 기술의 개발을 목적으로 한다. 재료 및 방법: 의료용 선형가속기를 사용한 측정과 몬테칼로 전산모사를 통해 감쇠물질의 두께에 따른 X-선의 투과율이 평가되었으며, 이와 더불어 단일에너지에 의한 이온함의 반응함수가 결정되었다. 최종적으로 Unfolding 방법을 사용하는 HEOROW 프로그램을 통해 고선량률의 치료용 선형가속기에서 발생된 광자선의 에너지스펙트럼이 유도되었다. 본 연구에서 실험을 위해 Elekta Synergy Flatform 선형가속기가 사용되었으며, 몬테칼로 방법을 사용하는 Geant4 전산모사 프로그램이 사용되었다. 결과 및 논의: 감쇠물질의 두께에 따른 X-선 투과율과 몬테칼로 전산모사를 통해 계산된 X-선 투과율의 비교 결과 0.43%의 평균제곱근오차가 확인된다. 물질투과정보를 통해 간접적으로 유도된 에너지스펙트럼은 몬테칼로 전산모사를 통해 직접적으로 계산된 에너지스펙트럼과 비교분석 되었으며, 피크위치 및 평균에너지의 오차가 각각 0.066 MeV와 0.03 MeV로 평가되었다. 하지만 보다 정확한 에너지스펙트럼을 예측하기 위해서는 다양한 감쇠물질의 사용과 에너지스펙트럼 유도프로그램의 정밀도 향상을 위한 추가적인 연구가 필요할 것으로 판단된다. 결론: 본 연구를 통해 확인된 물질투과율 기반의 에너지스펙트럼 측정 기술은 에너지가 높고 선량률이 높기 때문에 광자선을 직접적으로 측정하는 것이 제한적인 의료용 선형가속기에 적용될 수 있을 것으로 판단된다.

기온과 봄 식물계절지수와의 관계 (The Relationship Between Temperature and Spring Phytophenological Index)

  • 본 연구는 봄 식물계절지수와 기온지표와의 관계를 알아보기 위해 지난 9년간(2010년-2018년) 대구수목원, 팔공산, 주왕산, 가야산에 공통적으로 생육하는 소나무, 일본잎갈나무, 신갈나무, 진달래, 생강나무, 당단풍나무 6종을 대상으로 발아, 개화, 개엽의 봄 식물계절 시기의 변화 경향과 기온간의 관계를 파악하였다. 기온의 변화는 9년동안2월 보다 3~4월의 월평균 기온이 증가하였으며, 지역별로 수목원과 팔공산의 평균기온이 높았다. 발아, 개화, 개엽은 수종별로 생강나무가 가장 빠르며 소나무가 가장 느렸고, 지역별로 수목원에서 가장 빨리 식물계절시기가 도래하였다. 봄 식물계절시기는 -1.267~-6.151일/9년 정도 앞당겨지고 있는 경향을 보이는데 소나무(-6.151일/9년)가 가장 크며, 진달래(-1.267일/9년)의 변화율이 가장 낮았다. 발아, 개화, 개엽에서 모두 유의한 값을 보이는 수종은 진달래와 소나무로서 이들은 1월~3월 평균기온과 상관관계를 보였다. 봄 식물계절지수(SPI)의 시계열 변화를 확인한 결과 4개 지역의 변화율이 모두 음의 값으로 식물계절 시기가 모두 빨라지고 있었다. 그 중 수목원, 팔공산 및 가야산과 같은 내륙지역일수록 변화율이 컸으며, 다소 거리가 떨어진 주왕산의 경우 변화율이 조금 낮게 나타났다.
