We have previously found that mycelia culture broth of eight kinds of traditional herbal extracts fermented with Phellinus linteus (previously named as 8-HsPLCB) not only inhibited melanin and tyrosinase activity, but also reduced the contents of melanogenesis-related proteins, including tyrosinase and microphthalmia-associated transcription factor, in 3-isobutyl-1-methylxanthine-stimulated B16F0 melanoma cells. For a further study, the effect of 8-HsPLCB against skin pigmentation in brown guinea pigs with ultraviolet B (UVB)-induced hyperpigmentation was investigated. 8-HsPLCB (3%) and arbutin (2%) as positive controls were applied topically twice daily for 4 weeks to the hyperpigmented areas. 8-HsPLCB showed skin-lightening effect as effective as arbutin, one of the most widely used in whitening cosmetics. Melanin index values as the degree of pigmentation showed a significant reduction week by week post 8-HsPLCB treatment and then substantially reduced by 4 weeks. The degree of depigmentation after 4 weeks of topical application with 8-HsPLCB was 32.2% as compared with before treatment (0 week). Moreover, using Fontana-Masson staining and hematoxylin-eosin staining, 8-HsPLCB reduced melanin pigmentation in the basal layer of the epidermis and epidermal thickness changes exposed to the UV-B irradiation as compared with non-treatment and vehicle treatment. The intensity of the skin-lightening effect of 8-HsPLCB was similar to arbutin. These results suggest that the skin-lightening effect of 8-HsPLCB might be resulted from inhibition of melanin synthesis by tyrosinase in melanocytes. To conclude, 8-HsPLCB treatment showed reduction of the melanin pigment and histological changes induced by UV irradiation in brown guinea pigs.
The effects of bamboo (Phyllostachys nigra var. henenis Strapf) leaf extract (BLE) on the changes of ultraviolet (UV) light B radiation-induced apoptotic sunburn cell (SBC) and epidermal ATPase-positive dendritic cell (DC) in SKH1-hr or ICR mouse were investigated. The mice were treated with UVB ($200mJ/cm^2$) and were sacrificed 24 hours later. BLE (50 mg/kg of body weight) or vehicle (saline) was given i.p. at 36 and 12 hours before irradiation, and 30 minutes after irradiation. BLE cream (0.2%) or cream base (vehicle) was also topically treated at 24 hours and 15 minutes before irradiation, and immediately after irradiation. The skin of SKH1-hr mouse prepared from the back of untreated mice exhibited about 0.3 SBC/cm length of epidermis, and 24 hours after UV irradiation, the applied areas show an increased number of SBCs. But the frequency of UVB-induced SBC formation was significantly reduced by intraperitoneal injection (59.0%) and topical application (31.8%) of BLE extract. The numbers of DC in normal ICR mouse were $628.00{\pm}51.56\;or\;663.20{\pm}62.58\;per\;mm^2$ of ear epidermis. By 1 day after UVB treatment, the number of ATPase-positive $cells/mm^2$ were decreased by 39.0% or 27.1% in i.p. or topical application group with vehicle. The frequency of UVB ($200mJ/cm^2$)-induced DC decrease was reduced by treatment of BLE as 25.7% in i.p. group and 3.2% in topical application group compared with the irradiation control group. The results presented herein that BLE administration could reduce the extent of skin damages produced by UVB.
Raf-1, a cytoplasmic serine/threonine protein kinase, serves as a central intermediate in many signaling pathways in cell proliferation, differentiation, and development. In this study, we investigated that B-raf, Drosophila homolog of the human c-raf-1, is involved in ultraviolet (UV) responsive events by using hypomorphic mutant $D-raf^{c110}$ and Draf-lacZ transgenic fly. At first, effect of UV damage on the survival of wild-type and $D-raf^{C110}$ strains was examined. In terms of $1/LD_{50}$ value, the relative ratio of UV sensitivities of wild-type versus $D-raf^{C110}$ strain was 1 : 2.2. By using quantitative $\beta$-galactosidase activity analysis, transcriptional activity of the D-raf gene promoter was also examined in UV-irradiated Draf-lacZ transgenic larvae. UV irradiation increased the expression of lacZ reporter gene in Draf-lacZ transgenic fly. However, in $D-raf^{C110}$ strain the transcriptional activity of D-raf gene promoter by UV irradiation was extensively reduced. Results obtained in this study suggest that D-raf plays a role in UV response, leading to better survival of Drosophila to UV damage.
CdSe-Mn nanocomposites were synthesized using a microwave method with sodium sulfite (Na2SO3), selenium (Se), cadmium sulfate octahydrate (3CdSO4·8H2O), ammonia solution (NH3·H2O), and manganese (II) sulfate monohydrate (MnSO4·H2O). We obtained CdSe-Mn-C60 nanocomposites by calcining CdSe-Mn nanocomposites and fullerene (C60) in an electric furnace at 700 ℃ for 2 h. X-ray diffraction, Raman spectroscopy, and scanning electron microscopy were used to characterize the crystal structures, lattice vibrations, and surface morphologies of the products, respectively. The photocatalytic activities of the CdSe-Mn-C60 nanocomposites were investigated based on the photocatalytic degradations of organic dyes such as BG, MB, MO, and RhB under ultraviolet (UV) irradiation at 254 nm. UV-visible spectrophotometry was used to confirm the degradation process.

Background: Naturally occurring antioxidants were used to regulate the skin damage caused by ultraviolet (UV) radiation because several antioxidants have demonstrated that they can inhibit wrinkle formation through prevention of matrix metalloproteinases (MMPs) and/or increase of collagen synthesis. We examined the effect of oral administration of the antioxidant mixture ("L-Skin Care") on UVB-induced wrinkle formation. In addition, we investigated the possible molecular mechanisms of photoprotection against UVB through inhibition of collagen-degrading MMP activity or through enhancing of pro collagen synthesis in mouse dorsal skin. Methods: Female SKH-l hairless mice were orally administrated "L-Skin Care" (test group) or vehicle (control group) for 10 weeks with UVB irradiation by three times a week. The intensity of irradiation was gradually increased from 30 to $180mJ/cm^2$. Microtopographic and histological assessments of the dorsal skins were carried out at the end of 10 weeks to evaluate wrinkle formation. Western blot analysis and EMSA were also carried out to investigate the changes in the balance of collagen synthesis and collagen degradation. Results: Our "L-Skin Care" significantly reduced UVB-induced wrinkle formation, accompanied by significant reduction of epidermal thickness, and UVB-induced hyperplasia, acanthosis and hyperkeratosis. Oral administration of "L-Skin Care" significantly prevented UVB-induced expressions of MMPs, mitogen-activated protein (MAP) kinases and activation of activator protein (AP)-1 transcriptional factor in addition to enhanced type I procollagen and transforming growth factor-$\beta$ (TGF-$\beta$) expression. Conclusion: Oral administration of "L-Skin Care" significantly inhibited wrinkle formation caused by chronic UVB irradiation through significant inhibition of UVB-induced MMP activity accompanied with enhancement of collagen synthesis.

UV-C and -B types of lamps, and $H_2O_2$ as a photocatalytic material were applied to optimize processing conditions for pesticides removal in Mandarine orange peel. Factors to affect the removal of pestrcides were arranged as a function of irradiation temperature, irradiation time, and $H_2O_2$ spray concentration. The optimum processing conditions for the chloropyrifos and the EPN removals in Mandarine orange peel were irradiation time of 60 min, irradiation temperature of $45^{\circ}C$ and $H_2O_2$ spray concentration of 1000 ppm. However, the optimum processing conditions for methidathion removal were 60 min of irradiation time, $40^{\circ}C$ of irradiation temperature and 1000 ppm of $H_2O_2$ spray concentration. The residual percentages of chloropyrifos, EPN and methidathion were 46, 49 and 28% after above irradiation processing, respectively.
Seedlings of spinach (Spinacia oleracea L. "King of Denmark" and "Minsterland") were grown with or without supplemental UV-B under field conditions for five or nine days when their fifth or sixth leaves were expanding. Except two leaves which were just expanded and expanding, all other leaves of each seedling were removed before tested. One of these two leaves was exposed to UV-B irradiation and the other leaf was covered with lumiror film (no transmission below 320 nm) to prevent it from UV-B exposure. l,l-diphenyl-2-picrylhydrazyl scavenging activities (antioxidative activity) in leaves without covers were increased according to UV-B doses they were exposed to. And removal of UV-B exposure with lumiror decreased foliage antioxidative activities. This increase or decrease of activities in leaves did not dependent on the UV-B doses their paired leaves were exposed to in both two cultivars. The results demonstrated that spinach foliage antioxidants induced by UV-B exposure were not transported into another leaf.

To investigate the effects of different UV-B levels on plant growth, cucumber plants were subjected to three levels of biologically effective ultraviolet-B(UV--$B^{BE}$ radiation [daily dose : 0.03(No UV-B), 6.40(Low UV-B) and 11.30 (High UV-B) kJ $m^{-2}$, UV--$B^{BE}$] in the growth chambers for 3 weeks during the early growth period. High and low levels of UV-B irradiation drastically decreased both dry weight and leaf area, but increased specific leaf weight of cucumber. Plants subjected to UV-B resulted in 30% and 20% reduction of photosynthesis rate by high and low UV-B, respectively. However, respiration rate was not affected by the UV-B. With increasing UV-B intensity, total chlorophyll contents were decreased linearly, while the contents of flavonoid were increased linearly. These results suggest that the present levels of UV-B may affect the growth of cucumber plant compared with a UV-B-free condition. It is likely that the growth of cucumber will be affected by enhanced UV-B due to ozone depletion in the near future.
The present study assessed the effects of an aqueous extract of Acanthopanax koreanum root (AE) and of AE following fermentation by lactic acid bacteria (Lactobacillus plantarum and Bifidobacterium bifidum) (AEF) on human skin fibroblast HS68 cells exposed to ultraviolet B (UVB) irradiation and oxidative stress. AEF effectively antagonized the senescence-associated β-galactosidase staining and upregulation of p53 and p21Cip1/WAF1 induced by UVB or H2O2 treatment in HS68 cells. It also exhibited excellent antioxidant activities in radical scavenging assays and reduced the intracellular level of reactive oxygen species induced by UVB or H2O2 treatment. The antioxidant and antisenescent activities of AEF were greater than those of nonfermented A. koreanum extract. AEF significantly repressed the UVB- or H2O2-induced activities of matrix metalloproteinase (MMP)-1 and -3, overexpression of MMP-1, and nuclear factor κB (NF-κB) activation. This repression of NF-κB activation and MMP-1 overexpression was attenuated by a mitogen-activated protein kinase activator, suggesting that this AEF activity was dependent on this signaling pathway. Taken together, these data indicated that AEF-mediated antioxidant and anti-photoaging activities may produce anti-wrinkle effects on human skin.

It is well-documented that decreased antioxidant defense system by ultraviolet(UV) irradiation is the most important reason to induce the skin aging, especially photoaging. Chlorogenic acid(CA), a nonflavonoid catecholic compound, is present in the diet as part of fruits, tea, coffee and wine and has been reported to have anti-inflammatory, antimutagenic and anticarcinogenic activities. In this study, we examined the effects of CA on the UV -induced photoaging. Firstly, we investigated the protective effect of CA on antioxidant defense system in HaCaT human keratinocytes after UV irradiation treatment. UV irradiation decreased antioxidant defence enzyme activities of superoxide dismutase, catalase and GSH contents, which were restored by CA. To elucidate the effect of CA, 1% of CA and vehicle were applied to human buttock skin before and after UV irradiation (2MED). CA prevented UV -induced matrix metalloproteinase-1 mRNA expression and procollagen mRNA depression. And CA also increased CD1a(Langerhans cell) expression significantly. Our results suggest that CA has protective effects on UV -induced photoaging by increasing cellular antioxidant defense system. Therefore, CA may be a useful anti-aging agent for cosmetic purpose.
