Ultra-structural Observations of Colletotrichum orbiculare on Cucumber Leaves Pre-treated with Chlorella fusca (Chlorella fusca를 전처리한 오이 잎에서 오이탄저병균의 초미세 감염구조 관찰)

  • Chlorella is one of the microorganisms which can live autotrophically by their own photosynthesis. It was previously revealed that pre-treatment of Chlorella fusca caused a suppression of appressorium formation on the cucumber leaves after inoculation with Colletothrichum orbiculare. In this study, the ultrastructures of C. orbiculare on the cucumber leaves pretreated with C. fusca were observed using both scanning electron microscope (SEM) and transmission electron microscope (TEM). The SEM images revealed that most fungal conidia and hyphae were attached with lots of C. fusca cells. Also, the conidia could germinate but not form appressorium, which is necessary to penetrate into host tissue. These observations suggested that C. fusca adjoined to the fungus may play a role in suppression of the appressorium formation. On the other hand, the observations of TEM showed no remarkable cytological differences on the ultrastructures of the intracellular hyphae between in the pre-treated and untreated leaves. It seemed that the fungus could grow in the pre-treated plant tissues as in the untreated one. Based on these observations, it is suggested that the suppression of appressorium on the leaf surfaces by the C. fusca cells may be a main cause of the reduction of the anthracnose disease.

  • A scanning electron microscopic study was performed to observe the tegumental ultrastructures of Paragonimus iloktuenensis according to its developmental stages. The metacercariae were obtained from the liver of the brackish water crab, Sesarma dehaani. Juvenile and adult p. iloktsuenensis were recovered from the experimental rats on 2, 4 and 8 weeks after infection. The findings were summarized as follows: 1. The encysted metacercariae were charaeteristically gourd-shape, with their whole body surface beset with numerous spade-shape spines. The Barge, type II sensory papillae (non-ciliated round swellings) were arranged along the rim of the oral and ventral suckers, 11~12 and 6~8 in numbers respectively, 2. Two-week old juvenile worms, recovered chieay from the liver of the experimental rats, were slender in body shape: with their ventral sucker near the anterior one-third level. The distribution of tegumental spines was less dense than in the encysted metacercariae. The spines were with 1~2 pointed tips and 3~.4 longitudinal splits. Numerous ciliated knob-like, type I papillae were observed in both sides of the oral sucker, and 6 large, type If papillae were arranged along the rim of the ventral sucker. 3. Four week old worms, recovered from the thoracic cavity and/or lung parenchyme of the experimental rats, were thicker than wide in body configuration, and their ventral sucker was located near the anterior one.fourth level. The tegumental spines at ventral surface were grouped, each group with 3~5 aggregated ones. The type I and type II papillae (small-sired) were distributed chiefly around the rim of two suckers. 4. Adult (eight-week old) worms, recovered from the capsules in the lung parenchyme, were very stout, and covered densely with bearfoot like spines. At dorsal surface, cobblestone-like cytoplasmic processes were well-developed, with many tegumental spines embedded in them. It was observed in this study that the tegument of p. iloktsuenensis continued to change and differentiate as the worms grew to be adults.

  • Mast cells (MCs) are granulated cells that play a pivotal role in allergic reaction and inflammation. The granules of mast cells are known to be rich in zinc (Zn). Male Sprague-Dawley rats were used. We injected $200{\mu}L$ of complete Freund's adjuvant (CFA) subcutaneously in the dorsal aspect of one hindpaw Finally, zinc selenium autometallography(AMG) was done by Danscher's method. The present study showed the ultrastructures of zinc-containing mast cells found in inflammatory area following an complete freund's adjuvant (CFA) inoculation into the rat hindpaw. At light microscopic level, mast cells were round or oval, at average $12{\mu}m$ in diameter, with many filopodia extending from the cell surface. Because the rather small and spherical nucleus was centrally placed; it was frequently obscured by the cytoplasmic granules, it sometimes could not be seen. Mast cells were distributed chiefly in the vicinity of small blood vessels. In most preparation many mast cells were ruptured and their granules escaped into the surrounding tissue. In electron micrographs, The secretory granules were at average $0.5{\mu}m$ in diameter and were limited by a membrane. The cell surface contained numerous microvilli and folds. Their interior was heterogenous in appearance. The nucleus was surrounded by large numbers of prominent vesicels and a well developed Golgi apparatus, but scant endoplasmic reticulum.

  • The tegumental ultrastructures oi Echinostoma hortense adults were observed by scanning electron microscopy. The worms of 4 weeks of age were harvested from albino rats experimentally infected with the metacercariae obtained from the loach. The results were as follows: 1. The worms were leaf-like and their anterior end portion, including oral sucker and head crown, ventrally curved to face posteriorly. The tegument of whole body was wrinkled transversely and covered with cobblestone-like cytoplasmic processes. 2. The oral sucker had roundly swollen (type I ) sensory papillae on the ventral half of its lip and mini-ciliated knob-like (type I ) sensory papillae, arranged in 2∼3 rows, on the dorsal outer surface. 3. Aspinous ventral sucker had many of type I papillae arranged in a circular band on its outer surface. The tegument around the genital opening was of similar feature to the ventral sucker, but sensory papillae were hardly found around the former. 4. Scale-like spines with broad base and round tip were distributed densely on the tegument anterior to the ventral sucker but they became sparse in posterior half of the ventral surface, finally to disappear at posterior extremity. A few number of type I papillae were observed on the ventral surface. The results suggest that the tegument of 5. hortense is similar to that of other echinostomes etapecially 5. revolutum. But the number and arrangement of collar spines, and/or the type and distribution of sensory papillae seem characteristic features of E. hortense differed from other echinostomes.

  • Ultrastructures of the large and elongated cells (idioblasts) in the haustorium of a parasitic angiosperm, Cuscuta australis R. Brown growing on the host plant, Trifolium repens L. were investigated by the electron microscopy. The idioblasts were characterized by the presence of a large nucleus, small vacuoles, and dense cytoplasm including a number of various cell organelles such as ribosome, rough endoplasmic reticulum(r-ER), mitochondrion, dictyosome, proplastid, multilamellar structure(MLS), microfilament bundle(MFB), and cytosegresome. Therefore, it is suggested that the idioblasts are metabolical1y very active. Particularly, MLS, MFB, and cytosegresome observed in this study did not appear in the haustorial cells of the other parasitic angiosperms. MLS was transformed into vacuole and also incorporated with cell wall. MFB composed of microfilaments, about each 7.5 nm in diameter, was observed in nucleus and also cytoplasm. Some types of MFB were distinguished on the basis of arrangement of microfilaments. A part of cytoplasm sequestered by stacked cisternae of smooth ER(s-ER), cytosegresome, was altered into a vacuole which was formed by digestion of the sequestered cytoplasm and of cisternae of s-ER. Cell organelles such as MLS, MFB, and cytosegresome were discussed in relation to the metabolic control of the idioblasts.

  • The ultrastructures of the hemal node and the hemolymph node in the Korean native goat were observed by transmission and scanning elelctron microscopies for their morphological features. The sinus of hemal node was lined by endothelial-like reticular cells that had an euchromatin-rich nucleus and many cytoplasmic processes by which reticular fibers were surrounded. In the hemolymph node, the mast cell and the plasma cell were closely contact each other by the cytoplasmic process. The hemal node had venous sinusal-like vessels which were different from the deep sinus, and the hemolymph node had lymph capillaries. The lymph vessels with valves were observed in the capsule of the hemolymph node.

  • The ultrastructures of germ cells and the functions of Leydig cells and Sertoli cells during spermatogenesis in male Kareius bicoloratus (Pleuronectidae) were investigated by electron microscope observation. Each of the well-developed Leydig cells during active maturation division and before spermiation contained an ovoid vesicular nucleus, a number of smooth endoplasmic reticula, well-developed tubular or vesicular mitochondrial cristae, and several lipid droplets in the cytoplasm. It is assumed that Leydig cells are typical steroidogenic cells showing cytological characteristics associated with male steroidogenesis. No cyclic structural changes in the Leydig cells were observed through the year. However, although no clear evidence of steroidogenesis or of any transfer of nutrients from the Sertoli cells to spermatogenic cells was observed, cyclic structural changes in the Sertoli cells were observed over the year. During the period of undischarged germ cell degeneration after spermiation, the Sertoli cells evidenced a lysosomal system associated with phagocytic function in the seminiferous lobules. In this study, the Sertoli cells function in phagocytosis and the resorption of products originating from degenerating spermatids and spermatozoa after spermiation. The spermatozoon lacks an acrosome, as have been shown in all teleost fish spermatozoa. The flagellum or sperm tail of this species evidences the typical 9+2 array of microtubules.

  • We investigate the molecular and cellular etiologies that underlie the deletion of the six amino acid residues (${\Delta}F323-Y328$; ${\Delta}FY$) in human torsin A (HtorA). The most common and severe mutation involved with early-onset torsion dystonia is a glutamic acid deletion (${\Delta}E$ 302/303; ${\Delta}E$) in HtorA which induces protein aggregates in neurons and cells. Even though ${\Delta}FY$ HtorA forms no protein clusters, flies expressing ${\Delta}FY$ HtorA in neurons or muscles manifested a similar but delayed onset of adult locomotor disability compared with flies expressing ${\Delta}E$ in HtorA. In addition, flies expressing ${\Delta}FY$ HtorA had fewer aberrant ultrastructures at synapses compared with flies expressing ${\Delta}E$ HtorA. Taken together, the ${\Delta}FY$ mutation in HtorA may be responsible for behavioral and anatomical aberrations in Drosophila.

Ultrastructural Aspects of the Mixed Infections with Turnip mosaic virus and Ribgrass mosaic virus in Oriental Cabbage

  • Ultrastructural observation was conducted for the cells of oriental cabbage, Brassica campestris ssp. pekinensis 'Chungawang', inoculated simultaneously with Turnip mosaic virus (TuMV-ACT2-4vq) and Ribgrass mosaic virus (RMV-Ca1dn2) which were known as major destructive viruses of oriental cabbage in Korea. In cells infected with RMV alone, the virus particles were located as bundle or scattering in cytosols and vacuoles, which were typical ultrastructures of tobamovirus. Vessels of xylem were compacted with RMV particles. The cells infected only with TuMV had the cluster of virus particles scarcely and the typical potyvirus inclusions of scrolls, pinwheels, tubes and laminated aggregates in cytosols. The TuMV particles were jammed lineally between tonoplasts. In double infection, the two unrelated viruses of TuMV-ACT2-4vq and RMV-CA1dn2 were located together in a cell, and typical properties of each virus were also observed. The potyvirus inclusions and the tobamovirus particles were mixed entirely in cytoplasm. The virus particles of RMV wre presented strikingly near and in the center of potyvirus inclusions. In vascular cells, the tobamovirus particles were located abundantly than those in single infection. The potyvirus inclusions were embedded in the cluster of RMV particles in phloem parenchyma cells and the vascular elements were degenerated severely.

The Effect of Acrylamide on the Ultrastructures of Nervous System of the Mouse (생쥐 신경계의 미세구조에 미치는 Acrylamide의 영향)

  • The effed of acrylamide on the nervous system has heen morphologically studied using light and electron microscopes. The light micrographs on central and pedpheral nervous tissues of mouse treated with acrylamide monomer showed total vacuolation of spinal cord, cell degradation containing neuron and neuroglia, and distal nerve fiber degeneration. The electron micrographs showed ultrastrudural changes. Abnormal mitochondria in neuron, splitting of myelin sheath in lumbar ventral root nerve, partial disintergration of myelin sheath and axoplasmic degeneration in sciatic nerve, and overafl polyneuropathies in nervous system were observed. These results suggest that acrylarnide intoxicated mouse shows distal behavioral neuropathy as an earlist clinical sign, but the initial effect of acrylamide on the nervous system seems to appear at nearly the same time in both central and peripheral nervous systems.
