Candida albicans를 탐식(貪食)한 다핵백혈구(多核白血球)의 전자현미경적(電子顯微鏡的) 관찰(觀察) (Phagocytosis of Candida albicans by human polymorpho-nuclear leukocytes: An ultrastructural study)

  • These studies were carried out the observation of polymorphonuclear leukocyte phagocytosis of Candida albicans in vitro and also detected to the cytoplasmic changes of polymorphonuclear leukocyte during phagocytosis by the method of electron microscopy. The results were summarized as follows: 1. In normal polymorphonuclear leukocyte, nuclear lobes showed a preponderance of dense, granular chromatin located peripherally. The cytoplasm of polymorphonuclear leukocyte was not extensive; the cytoplasmic matrix was moderate dense and of a granular appearance. Golgi complex and rough endoplasmic reticulum system was poorly developed. But a various type of granules were seen abundantly. 2. After 30 minutes of incubation, Candida albicans was completely engulfed. These had come to lie in the vacuole which was limited by the membrane. 3. After 90 minutes of incubation, the phagocytic vacuoles were larger, and many granules devoid of membranes were seen within them. Though the granules has lost their membrane after entering the vacuoles. 4. After 2 hours of incubation, the cytoplamsic components of polymorphonuclear leuko cytes were changed their original morphology.

  • Aspirin이 위점막(胃粘膜)에 손상(損傷)을 일으키는 기전(機轉)을 해명(解明)하고 침해(侵害)받는 세포(細胞)의 종류(種類) 및 형태(形態)를 구명(究明)하고자 rat에 체중(體重) 1kg당(當) 60mg을 경구(經口) 및 피하(皮下)로 투여(投與)한 후(後) 경시적(經時的)으로 임상소견(臨床所見)을 관찰(觀察)함과 동시(同時)에 위점막세포(胃粘膜細胞)의 형태적변화(形態的變化)를 광학(光學) 및 전자현미경적(電子顯微鏡的)으로 관찰(觀察)한 결과(結果)를 요약(要約)하면 다음과 같다. aspirin의 경구(經口) 및 피하투여(皮下投與)에 의(衣)하여 특별(特別)한 임상소견(臨床所見)이나 위점막(胃粘膜)의 육안적(肉眼的) 및 광학현미경적(光學顯微鏡的) 소견(所見)은 인정(認定)되지 않았으나 미세형태학적(微細形態學的)으로는 투여경로(投與經路)에 관계(關係)없이 명백(明白)한 변화(變化)를 인정(認定)할 수 있었다. 즉(卽) 위점막세포중(胃粘膜細胞中) 가장 심하게 손상(損傷)받는 세포(細胞)는 벽세포(壁細胞)와 주세포(主細胞)로서 벽세포(壁細胞)에서는 SER과 intracellular canaliculi의 증식(增殖)이, 그리고 주세포(主細胞)에서는 RER의 단열(斷裂), 확장(擴張) 및 효소원과립(酵素原顆粒)의 수적감소(數的減少)와 구조적이상(構造的異常)과 같은 가역적(可逆的) 변성변화(變性變化)가 인정(認定)되었으며 점액세포(粘液細胞), 점액경세포(粘液頸細胞), 기저과립세포(基底顆粒細胞)는 비교적(比較的) 경(經)한 변화(變化)를 보였다. 경구투여군(經口投與群)보다는 경미(輕微)하지만 피하주입군(皮下注入群)에서도 경구투여군(經口投與群)에 상응(相應)하는 변화(變化)를 나타낸 점(点)은 aspirin에 의한 위점막(胃粘膜) 손상(損傷)이 aspirin의 직접적영향외(直接的影響外)에 혈중(血中) aspirin 농도(濃度)에 의(依)한 간접적손상(間接的損傷)도 가미(加味)된다는 것을 시사(示唆)하는 형태적소견(形態的所見)이라고 생각(生覺)되었다.

배양한 사구체 상피세포에서 고농도 당과 후기 당화합물에 의한 P-cadherin의 변화 (High Glucose and Advanced Glycosylation Endproducts(AGE) Modulate the P-cadherin Expression in Glomerular Epithelial Cells(GEpC))

  • 목 적 : 단백뇨 질환에서 볼 수 있는 사구체 상피세포(glomerular epithelial cells, GEpC) 족돌기 사이에 위치한 세극막(slit diaphragm)의 P-cadherin의 당뇨조건에 따른 병리학적 변화를 알아보고자 하였다. 방 법 : 백서 GEpC을 배양하고 고농도의 당과 후기당화합물(advanced glycosylation endproducts, AGE)을 적용하여 당뇨병 환경에 가까운 조건을 설정한 후, p-cadherin 단백양은 Western 분석으로, 유전자 표현의 변화는 RT-PCR로 관찰하였다. 실험군은 당의 농도를 5 또는 30mM로, AGE와 BSA를 첨가하고 osmotic control로서 당 5 mM에 mannitol 25 mM을 섞은 것을 조합하여 A5, A30, B5, B30, Aosm로 하였다. 결 과 : P-cadherin 단백양은 B5 결과를 대조군으로 비교하여 당을 첨가한 B30에서 50.4$\%$의 감소, AGE를 추가한 조건인 A5와 A30에서 각각 7.4$\%$와 30.4$\%$의 의미 있는 감소를 보였다. 또한 P-cadherin mRNA의 표현은 B30에서 40.3$\%$의 감소, A30에서 27.2$\%$의 의의 있는 감소를 보였다. 이러한 감소 소견은 osmotic control(Aosm)에서는 관찰할 수 없었다. 결 론 : 고농도의 당과 AGE에 의한 GEpC의 P-cadherin을 유전자 수준에서의 억제로 단백의 생성 감소를 초래함으로써, 당뇨환경에서 세극막 성분의 변화를 설명할 수 있으며, 추후 이의 변화 기전에 대한 연구가 필요할 것으로 사료된다.

방사선조사에 따른 백서 악하선내 혈관 내피세포 미세구조의 조기변화 (EARLY IRRADIATION EFFECTS ON THE ULTRASTRUCTURE OF THE CAPILLARY ENDOTHELIAL CELL IN THE RAT SUBMANDIBULAR GLANDS)

  • The purpose of this study was to investigate the early irradiation changes on the ultrastructure of the capillary endothelial cell in the rat submandibular glands. For the study, 110 Sprague-Dawley strain male rats were singly irradiated to their neck regions with the doses of 2Gy, 5Gy, and 10Gy by 6MV X -irradiation, and sacrificed on the 3 hours, 6 hours, 12 hours, 1 day, 3 days, 7 days, and 14 days after irradiation. The authors observed the histologic and ultrastructural changes of the capillary endothelial cell using the light and electron microscopes. The results were as follows: I. In the light microscopic examination, the capillary dilation was observed on the 6 hours group and the capillary density was slightly increased on the 12 hours group after 2Gy and 5Gy irradiation. And luminal size and capillary density were decreased on the 3 days and the 7 days groups after irradiation, after then, they were recovered. But capillary density was still decreased on the 14 days group after 10Gy irradiation. 2. In the transmission electron microscopic examination, the mild proliferation of cytoplasmic process of the endothelial cell and reduction in luminal size were observed on the 3 hours group after irradiation. After then, endothelial swelling, marked proliferation of cytoplasmic process, thickened basal lamina, and numerous pinocytotic vesicles were observed after the 1 day group after irradiation. Thickened basal lamina and numerous pinocytotic vesicles were still observed until the 7 days group after irradiation. These changes were recovered to normal on the 14 days group after 2Gy and 5Gy irradiation, but not after 10Gy irradiation. 3. In the scanning electron microscopic examination, the dilation of conduits and constriction, and meandering were observed on the 1 day group after 10Gy irradiation. These changes were observed with increased coarseness of the surface of the vascular resin casting on the 3 days group after irradiation. 4. From the above results, endothelial swelling, proliferation of cytoplasmic process, and thickening of the basal lamina appeared before the 6 hours group after irradiation. And these changes may also induce the increase of the capillary number and luminal size, after then, capillary permeability was increased via the increase of the number of pinocytotic vesicles. The changes were observed earlier and more apparent with the increase of the irradiation doses under the dose of 10Gy irradiation.

재관류시 외인성 칼슘 자극이 허혈 심근에 미치는 영향에 대한 전자현미경적 연구 (An Ultrastructural Study on the Effect of Exogenous $Ca^{2+}$ Stimulation to Ischemic Myocardium during Post-ischemic Reperfusion)

  • The effects of exogenous $Ca^{2+}$ stimulation on the post-ischemic myocardial cells were studied using isolated Langendorff-perfused guinea pig hearts. At the starting point of reperfusion, Tyrode solutions, each containing 2.0mM, 4.0mM and 8.0mM $CaCl_2$ respectively, were administered for 2 minutes apart by descending, ascending, or by combined sequences followed by standard Tyrode solution containing 1.0mM $CaCl_2$. The minutes of global ischemia produced reversible but moderate to severe degree of myocardial ultrastructrual changes including focal destruction of sarcolemma, loss of nuclear matrix, clumping and margination of chromatins, mitochondrial swelling, destruction of mitochondrial cristae, shortening of sarcomeres, focal loss of myofibrils, and separation of cell junctions. In spite of reperfusion, the ultrastructure was more severely damaged and irreversible changes such as intracellular fluid accumulation, contracted sarcomeres, mitochondrial destruction, disruption of sarcolemma, loss of nuclear matrix, and separation of cell junction were observed in a large number of cells. In contrast, Tyrode-perfused $Ca^{2+}$-stimulated myocardial cells showed relatively well preserved ultrastucture, except slight changes including focal mitochondrial swelling, widening of T-tubule, and widening of cell junctions, especially at fasciae adherentes. The post-ischemic $Ca^{2+}$-stimulated reperfused myocardial cells produced focal changes such as mitochondrial destruction, disintegration of sarcolemma, widening of T-tubule, and intracellular fluid accumulation with slight variation in degree of changes by the method of $Ca^{2+}$ administration sequence. However, in a large number of the myocardial cells, chromatins were redistributed relatively evenly in the nuclear matrix, mitochondrial cristae were tightly packed, and a considerable number of intramitochondrial granules and glycogen granules reap-pealed. These results indicate that exogenous $Ca^{2+}$ stimulation in the initial period of reperfusion may be beneficial to salvage or to reduce the post-ischemic myocardium from further deleterious changes, and that the beneficial effects may be derived from the reserves of the function of the intracellular $Ca^{2+}$ regulating organelles and/or from the responsiveness of contractile apparatus to $Ca^{2+}$ stimulation.

고사리삼 지하경의 유관속 분열조직 미세구조 (Ultrastructure of Vascular Meristems in the Rhizome of Botrychium ternatum)

  • 고사리삼의 지하경에서 2기생장의 기원을 구명하기 위하여 전형성층으로부터 유관속 형성층의 분화과정을 미세구조적인 측면에서 관찰하였다. 유관속 형성층은 종자식물에서와 같이 전형성층으로부터 점진적인 분화과정을 거쳐서 기원되지만, 그 활성은 짧아서 화석 은화식물에서와 같은 유한 형성층인 것으로 관찰되었다. 유관속 형성층의 점진적인 기원은 세포질이 아주 농후한 전형성층으로부터 세포질이 점차 희박해지며, 수가 많고 소형인 액포가 점차로 크기가 커지면서 수가 적어지는 액포로 되는 점 등으로 뒷받침될 수 있다. 한편 유관속 형성층이 유한성을 보이는 점은 후기 유관속 형성층이 지질과립 등의 저장물 축적이 심하고 세포소기관은 물론 세포질이 매우 희박해지면서 세포벽이 일반 분열조직세포와는 달리 상당히 비후되어 있는 특징으로도 알 수 있다. 그러므로 고사리삼의 유관속 형성층은 종자식물의 유관속 형성층의 특징과 화석은화식물의 특징을 모두 공유하고 있는 것으로 생각된다.

수용성조영제의 주입이 가토악하선조직에 미치는 영향에 관한 전자현미경적 연구 (ULTRASTRUCTURAL STUDY ON THE EFFECTS OF RETROGRADE INFUSION OF WATER-SOLUBLE CONTRAST MEDIA ON THE RABBIT SUBMANDIBULAR GLAND)

  • The author observed the effects of retrograde infusion of water soluble contrast media (Tele- brix 30) on the rabbit submandibular glands and compared the effects of different degrees of filling. 26 rabbits were divided into 2 groups of 12 each as experimentals and I group of 2 as normal controls. One experimental group was filled with 0.2㎖ and the other with 0.4㎖. Right submandibular gland of each rabbit was infused with contrast media and left one with physiologic saline as a experimental control, at a constant rate of 0.12㎖/min. using an infusion pump via the main excretory duct. Immediately after the infusion of contrast media, oblique lateral radiographs of the glands were made with occlusal film in order to confirm the glandular filling. The rabbits were sacrificed after varying periods (1, 8, 24 hours and 3, 6, 10 days) and the tissues were prepared for light and electron microscopic examination. The results were as follows: 1. In glands filled with 0.2㎖ contrast media, the initial changes were a few vacuole formation in the acini and slight dilation of the intralobular duct. The moderately severe changes such as vacuole formation in the acini, the abnormal substructure within the secretory granule, dilation of acinar and intercalated duct lumen, scalloping of striated duct lumen and inflammatory cell infiltrate were observed at 3 days. The general appearance was successively recovered, so the tissue had a normal appearance at 10 days. 2. In glands filled with 0.4㎖ contrast media, the most prominent alterations such as severe acinar atrophy, decreased number of secretory granules, proliferation of connective tissue stroma and pronounced inflammatory cell infiltrates appeared at 6 days. Although the general appearance returned to be almost normal at 10 days, acinar cells showed some atrophy and decreased secretory granules. 3. In glands subjected to 0.4㎖ infusion, the alterations were more severe and the recovery was slower than those seen in the glands to 0.2㎖ infusion.

Reversible histoarchitecture study of testis and cauda epididymis and changes in cauda epididymal epithelial cell types on treatment with benzene extract of Ocimum sanctum leaves in albino rats

  • In the present study, an attempt has been made to assess whether the effect of benzene extract of Ocimum sanctum leaves on the ultrastructural changes in the epithelial cells of the cauda epididymis, its subsequent recovery in the seminiferous epithelium and fertility of male albino rats. Wistar strain male albino rats were orally administered benzene extract of 250 mg/kg body weight of O. sanctum leaves followed by subsequent recovery maintaining suitable controls for 48 days. Results indicate decrease in the weights of testis, epididymis and seminal vesicles. Other accessory organs were not affected. Total count, cell and nuclei diameters of germ cells and Leydig cells were reduced. Cauda epididymis exhibited significant reduction in epithelial height and nuclei diameter of epithelial cells. Cells showed vacuolization with exhibit of signs of degeneration. Ultra study revealed that, in general, the cauda epididymis was affected and in particular, the principal, clear and basal cells were highly disturbed. Further, there was decrease in the size of lipid droplets, mitochondria, Golgi complex, endoplasmic reticulum and accumulation of lysosomal bodies. Fertility performance test showed no implantation in female rats mated with O. sanctum treated rats. Moreover, their recovery after withdrawal of treatment was observed suggesting that the effect of the treatment is transient and reversible. A recovery period resulted in normal spermatogenesis and fertility, suggesting reversible antispermatogenic and antifertility effects of the plant.

침자(鍼刺)한 경혈(經穴) 부위(部位)의 조직학적 변화(組織學的 變化) (Histological Changes of the Acupoint by Acupuncture Stimulation)

  • 침자시 해당부위에서 발현되는 특수한 수용체에 대한 반응을 형태학적으로 연구하고자 흰쥐의 족삼리(足三里) 부위에 침자를 한 다음 경혈부위 조직내의 비만세포의 변화와 침자주변 세포의 미세구조 변화를 관찰하였다. 침자시 형성된 진피층의 동공주변에서는 비만세포들이 정상군에 비하여 유의성있는 종가(p<0.01)를 하였다. 비만세포의 세포질에는 전자밀도와 형태가 다른 과립들이 충만되어 있었을 뿐만 아니라 세포질 밖으로 분비된 과립들도 관찰할 수 있었다. 침자 결과 형성된 골격근육층의 근섬유들은 근원섬유 마디의 밝은띠가 정상군에 비하여 않게 관찰되는 것으로 보아 침자시 자극에 의하여 근육섬유들이 수축하였음이 밝혀졌다. 이상의 결과로 보아 침자에 의하여 유도되는 유효한 생체반응은 진피층에서 관찰되는 비만세포의 수와 미세구적인 변화는 침자에 의한 경혈기능과 밀접한 상관관계를 가지고 있을 것으로 사료되며, 또한 근섬유를 수축시키는 형태학적 변화가 유도되는 것으로 보아, 경혈 부위는 기계적 자극에 반응하는 특수한 수용체와 밀접한 관련이 있는 것으로 사료된다.

Cyclophosphamide가 흰쥐의 부정소에 미치는 영향 III. 미 부 (Effects of Cyclophosphamide in the Epididymis of the Rat III. Cauda)

  • This research was undertaken to determine the effects of the anticancer and immunosuppressive drug cyclophosphamide (CP) on the epididymis of the male rat in terms of ultrastructural alteration and protein analysis by SDS-PAGE at different groups; control group, 1 week group, 3 weeks group, 5 weeks group were treated with saline (control group) or CP at doses of 20mg/Kg/week, 1 time a week, respectively. In the cytoplasm of the principal cells on the epididymis, the mitochondrial outer and inner membranes were significantly swollen or disrupted. The cisterns of rough endoplasmic reticulum (rER) were also swollen, and a number of Golgi vesicles were increased, respectively. It is suggested that treatment with CP alters the specific cell organelles in all segments of the epididymis. CP caused changes in protein concentrations in cauda of epididymis after CP treatment. Total proteins of 30 to 39 species such as lactate dehydrogenase, carnitine acetyltransferase and acid phosphatase were expressed in the cauda fluid. Then the more CP was increased, the more concentration of proteins caused to decrease, synthesize or increase in epididymal cauda. In contrast to the control group, in particular 29KD and the other 10 proteins in the cauda fluid were decreased or disappeared, respectively, whereas 89KD and the other 6 proteins in the cauda, were increased or synthesized, respectively. The other proteins are not showed distinctive difference. Therefore, it is possible that CP at a high dose accumulation alters epididymal function with dose-related increase or decrease in specific activity of marked proteins for all regions of the epididymis (particularly, specific segment of cauda). These alterations could be mediated by direct, toxic effects of the drug on the epithelium or be secondary to changes in the spermatozoa as a result of the CP treatment.
