Anti-inflammatory Effects and Mechanisms of Ulmus Davidiana Planch(Ulmaceae) in Collagen-induced Arthritis Rats (쥐의 콜라겐 유도 관절염에 대한 유근피의 효과 및 기전)

  • 목적 : 쥐의 콜라겐 유도 관절염에 대한 유근피 추출액의 면역 반응 효과 및 그 기전을 살펴보고자 하였다. 방법 : 유근피 추출액의 면역 반응을 관찰하기 위하여 콜라겐 유도 관절염 쥐가 사용되었다. 실험에 쓰인 쥐 뒷다리의 부종 용적은 volume meter로 측정하였고, lymphocyte 증식, 표1, 표2 및 TNF-${\alpha}$ 레벨은 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolirun bromide(MTT) assay에의해 측정하였다. 활막세포의 cAMP 레벨은 경쟁적 단백 결합검사 (CPBA)를 통하여 측정하였다. 2형 콜라겐에 대한 항체는 효소면역 협착검사법(ELISA)을 반복 사용하여 측정하였다. 결과 : 실험에서 유근피 추출액( 20, 80, 150mg/kg, ig ${\times}$ 7days)의 시술은 면역 반응을 억제하고 콜라겐 유도관절염 쥐의 체중과 면역 기관의 무게를 유지하였다. 콜라겐 유도 관절염 쥐에서 림프구의 증식과 IL-2의 생산은 복막의 대식세포 및 활막세포의 IL-1 , TNF-${\alpha}$와 함께 증가하였고, 유근피 추출액 (20, 80, 150mg/kg, ig ${\times}$ 7days)의 시술은 이러한 변화를 유의성 있게 감소시켰다. 0.5, 2.5, 12.5, 62.5, 125mg/l 농도에서의 유근피 추출액은 활막세포의 cAMP 레벨을 증가시킨데 반해 콜라겐 유도관절염 쥐에서의 시험관 실험결과에서는 감소시켰다. 유근피 추출액은 2형 콜라겐 항체의 농도에 대하여는 효과가 없었다. 결론 : 유근피 추출액은 항염증 작용과 면역조절 작용을 갖고 있고, 활막세포의 G protein-AC-cAMP transmembrane signal transduction 형질 도입 신호에의한 콜라겐 유도관절염 쥐의 치료 효과를 가지고 있는 것으로 여겨진다.

Effect of Ulmus davidiana Planch (Ulmaceae) on T-lymphocyte-producing cytokines such as IL-2, IL-6, and $IFN-{\gamma}$ production in collagen-induced arthritis of rats

  • Objective : The effect of Ulmus davidiana Planch(UD), which has long been known to have anti-inflammation and protective effects on damaged tissue, inflammation and bone among other functions, on the development of type II collagen (CII)-induced arthritis (CIA) in rats was studied. Methods : Male rats were immunized with an emulsion of $200\;{\mu}g$ of CII and complete Freund's adjuvant (CFA). The rats were then given intraperitoneal stimulation of Ulmus davidiana Planch herbal acupuncture(UDHA)or saline during the experiment. When compared with rats treated with saline as control, UDHA at doses of more than $20{\mu}g/100\;g$ rat once a day for 7 days inhibited the ability of inguinal lymph node cells to produce T cell cytokines interleukin-2, interleukin-6, $IFN-{\gamma}$ when the cells were obtained from rats 14 days after immunization and cultured in vitro with CII. Results : When rats were injected intraperitoneally, UD -treated group and control group rats did not differ significantly when low doses of UD was given to rats. Conclusion : The recommended dose of UD in the management and treatment of rat CIA will be more than $20{\mu}g/100\;g$, which is two-firth of human therapeutic dose. From the results, it was concluded that the effect of UDHA is dependent of dosage.

Inhibitory effect of Ulmus davidiana Planch extracts on bone resorption mediated by processing of cathepsin K in cultured mouse osteoclasts

  • Objective: Ulmus davidiana Planch (Ulmaceae) has long been known to have anti-inflammnatory in the traditional Korean medicine. UD has been reported as a good enhancer for bone healing. Methods : In this experiment, we investigate the Inhibitory effects of UD on bone resorption using the bone cells culture. Different concentrations of crude extract of UD were added to mouse bone cells culture. The mitochondria activity of the bone cells after exposure was determined by colorimetric MIT assay. It was demonstrated that UD has potential effects on bone cells culture without any cytotoxicity. The most effective concentration of UD on bone cells were $100\;{\mu}g/ml$. Cathepsin K (Cat K) is the major cysteine protease expressed in osteoclasts and is thought to play a key role in matrix degradation during bone resorption. Results : When mouse long bone cells including osteoclasts and osteoblast were treated with the PI3-Kinase inhibitor, wortmannin (WT), WT prevented the osteoclast-mediated intracellular processing of Cat K. Similarly, treatment of osteoclasts-containing long bone cells with UD extracts prevented the intracellular maturation of Cat K, suggesting that UD may disrupt the intracellular trafficking of pro Cat K. This is similar to that of WT. Since secreted proenzymes have the potential to reenter the cell via mannose-6-phosphate (M6P) receptor, to prevent this possibility, we tested WT and UD in the absence or presence of M6P. Inhibition of Cat K processing by WT or UD was observed in a dose-dependent manner. Furthermore, the addition of M6P resulted in enhanced potency of WT and UD. Conclusion : UD dose-dependently inhibited in vitro bone resorption with a potency similar to that observed for inhibition of Cat K processing.

  • The bark of Ulmus davidiana var. japonica(Ulmaceae) has been described in traditional books to have diuretic and laxative actions, to ameliorate edematous disease, and to be used in insomnia and gastointestinal disorders. Thus this study was carried out to elucidate some of the pharmacological activities of the plant extracts. The results obtained in tills experiment indicated that its methanol extract elicited remarkable inhibition of HCI.ethanol induced gastric lesion, Shay ulceration and gastric secretion. However, it showed no anti-inflammatory action. The acute toxicity of the extract was low, that is, the minimum lethal dose was more than 2000 mg/kg by oral administration in mice. The systematic fractionation of the methanol extract by hexane, ether, ethylacetate and butanol resulted in potent prevention of gastric erosion in butanol and water fractions.

  • Four lignan xylosides and two neolignan glycosides were isolated from the stem and root barks of Ulmus davidiana var. japonica. Their structures were identified as lyoniside, nudiposide, 5'-methoxyisolarciresinol-9'-O-$\beta$- D-xylopyranoside, isolariciresiol-9'-O-$\beta$-D-xylopyranoside, rel-trans-dihydrodehydroconiferyl alcohol 4'-O-$\alpha$-L-rhamnopyranoside and icariside E3 by comparison of their spectral data with those reported in the literatures, respectively.

  • From the $CHCl_3$ and BuOH soluble fractions of the heartwood of Hemiptelea davidii, eleven compounds have been isolated. On the basis of spectral data, they were identified as ${\beta}-sitosterol$ (1), scopoletin (2), kaempferol (3), 4-hydroxybenzoic acid (4), 2-(4-hydroxyphenyl) ethanol (5), aromadendrin (6), scopolin (7), kaempferol 6-C-glucoside (8), aromadendrin 6-C-glucoside (9), taxifolin 6-C-glucoside (10) and quercetin 6-C-glucoside (11), respectively. Among these compounds, compounds 3, 8, 10, and 11 showed potent DPPH radical scavenging activity with $IC_{50}$ values of 11.9, 14.7, 10.3 and $6.2\;{\mu}g/ml$, respectively.

  • The Barks of Ulmus davidiana (Ulmaceae) have been used for the treatment of insecticide, anti-boil and anti-fungi in Korean traditional medicine. Four phenolic compounds were isolated from 80% Acetone extracts. The structures of these compounds were elucidated as (+)-catechin, (+)-catechin 7-O-${\beta}$-D-glucopyranoside, (+)-catechin 7-O-${\beta}$-D-xylopyranoside and procyanidin B-1. These phenolic compounds showed significant nitrogen monoxide(NO) production inhibitory activity in IFN-${\gamma}$, LPS stimulated RAW 264.7 cell and also showed significant antioxidative activity on DPPH radical.   • In this study 2 cadalene homologues - 3,7-dimethoxycadalene and 7-hydroxycadalene-5,8-quinone (keyakinone A)-were further identified from ethanol extracts of Zelkova serrata wood, except 7-hydroxy-3-methoxycadalene. Two biological activities-scavenging activity of hydroxy radical and cell toxicity by MTT assay-were measured with 7-hydroxy-3-methoxycadalene. The scavenging activity of hydroxyl radical of the compound was excellent and increased with its concentration. At 100 ppm hydroxyl radicals were removed completely. However, MTT assay revealed that 7-hydroxy-3-methoxycadalene showed critical toxicity to the cells. When 1 ppm of the compound was treated to the cells, cell viability was reached up to 90%, while it was reduced to 22% after treatment of 9 ppm. In 4 different Ulmaceae species, such as Ulmus davidiana, Ulmus parvifolia, Ulmus macrocarpa, Ulmus macrophylla, 7-hydroxy-3-methoxycadalene was not found at all. Instead, 7-hydroxycadalene (Mw 214), in which methoxyl group is omitted from 7-hydroxy-3-methoxycadalene, was distributed in the heartwood of 4 Ulmaceae species as major cadalene compound.

Effect of Ulmus Davidiana Planch Herbal Acupuncture Solution on Inhibition of Experimental Bone Resorption in Mouse Calvarial Bone Cells (유근피 약침액이 생쥐의 두개골 파골세포에서 골재흡수의 저해에 미치는 영향)

  • 목적 : 골관절염치료에 빈용되는 유근피 약침액의 처치가 생쥐의 파골세포에 있어서 골재흡수의 저해에 미치는 영향을 알아보고자 하였다 방법 : 생쥐에게 유근피 약침액을 두개골 세포에 전, 후 처치하여 골재흡수에 대한 유근피 약침액의 억제 활성능을 검토하였다. 결론 : 염증성 cytokine 중 $IL-1{\beta}$ 유도인자인 PGE와 LPS처리로 $IL-1{\beta}$생성이 증가되었으나, 유근피 약침액 처치군은 이를 억제하였다. 유근피 약침액 전처리군에서도 $IL-1{\beta}$ 생성이 억제되었다. 유근피 약침액 처치군은 PGE2유발 $IL-1{\beta}$ 전사를 억제하였으며, PGE2 유발 $IL-1{\beta}$ 유도는 cAMP antagonist인 Rp-cAMP와 protein kinase A(PKA)저해제에 의해서도 억제되어 $IL-1{\beta}$ 발현에 cAMP, PKA 신호전달경로가 관여함을 시사하였다. 본 연구에서 유근피 약침액은 강한 항 관절염효과와 골재흡수 저해 활성이 있으며, 관절염 치료, 예방에 유의함을 밝힌 것으로 사료된다.
