Action Spectra of Apoptosis Induction and Reproductive Cell Death in L5178Y cells in UV-B Region

  • It is important to determine the action spectrum of UV-B radiation contained in the sunlight to estimate the risk of skin cancer. We have investigated action spectra for induction of apoptosis and reproductive cell death in L5178Y cells using the Okazaki Large Spectrograph at NIBB. L5178Y cells were exposed to light at different wavelengths in UV-B or UV-A region. Frequencies of apoptosis induction and reproductive cell death were determined by counting cells with chromatin condensation, and by the colony formation assay, respectively. The measured sensitivity spectra for the two end-points were in very good agreement. Sensitivity decreased steeply with increase of wavelength in UV-B region and remains nearly constant in UV-A region. The action spectra were also slightly steeper than that for the minimum erythematic dose (MED), but very similar to the light absorption spectrum of DNA in UV-B region. On the other hand, the spectra for both endpoints were similar to MED spectrum but not DNA spectrum in the UV-A region. Also different time-course and morphological difference of apoptosis were found between UV-B (long time, fragmentation) and UV-A (short time, shrinkage) region. These results suggest that DNA damage induced by UV-B light triggers apoptosis and reproductive cell death, but other damaged targets (membrane, protein and so on) trigger these effects in UV-A region.

Impacts of Ultraviolet-B Radiation on Rice-Field Cyanobacteria

  • Cyanobacteria are the dominant micro flora in rice-fields, contributing significantly to fertility as a natural biofertilizer. Recent studies show a continuous depletion of the stratospheric ozone layer, and the consequent increase in solar UV-B (280-315 nm) radiation reaching the Earth's surface. UV-B radiation causes reduction in growth, survival, protein content, heterocyst frequency and fixation of carbon and nitrogen in many cyanobacteria. UV -B induced bleaching of pigments, disassembly of phycobilisomal complexes, thymine dimer formation and alterations in membrane permeability have also been encounterd in a number of cyanobacteria. However, certain cyanobacteria produce photoprotective compounds such as water soluble colorless mycosporine-like amino acids (MAAs) and the lipid soluble yellow-brown colored sheath pigment, scytonemin, to counteract the damaging effects of UV-B. Cyanobacteria, such as Anabaena sp., Nostoc commune, Scytonema sp. and Lyngbya sp. were isolated from rice fields and other habitats in India and screened for the presence of photoprotective compounds. A circadian induction of the synthesis of MAAs by UV -B was noted in a number of cyanobacteria. Polychromatic action spectra for the induction of MAAs in Anabaena sp. and Nostoc commune also show the induction to be UV-B dependent peaking at 290 nm. Another photoprotective compound, scytonemin, with an absorption maximum at 386 nm (also absorbs at 300, 278, 252 and 212 nm), was detected in many cyanobacteria. In conclusion, a particular cyanobacterium having photoprotective compounds may be a potent candidate as biofertilizer for crop plants.

Compositional changes in mycosporine-like amino acids induced by UV radiation: marine dinoflagellate Scrippsiella sweeneyae

  • The compositional changes in mycosporine-like amino acids (MAAs) were investigated in the marine dinoflagellate Scrippsiella sweeneyae exposed to four different spectral compositions and five relative intensities of UV-B (280-320 nm) to UV-A (320-400 nm) + photosynthetically available radiation (PAR: 400-700 nm). Neither dose nor wavelengths of UVR significantly affected the growth rates. UVR caused a significantly increase in cell volume. Cell volume in the >280nm treatment was more than two times greater at 6.8 % of UVR intensity. Production of UVR induced MAAs was dependent on the dose of UVR. However. the induction of MAAs was related to the cell growth. Greater induction of MAAs was observed at shorter wavelengths. The composition of MAAs varied with increasing light intensity of UVR.

UV Actions in Plant Photomorphogenesis -Induction and Amplification of Anthocyanin Synthesis in Broom Sorghum-

  • In broom sorghum, Sorghum bicolor Moench, UV causes anthocyanin synthesis having action peaks in UVA and UVB regions. We previously reported that UV induces anthocyanin synthesis through UVB photoreceptor and phytochrome activated by UV. Furthermore, UVA and UVB amplify phytochrome-induced anthocyanin synthesis (PIAS). Our action- spectroscopic research indicated that a UV -receptor for amplification of PIAS is likely to be the same or same type of UVB photoreceptor for induction of anthocyanin synthesis. UVA-amplification of PIAS can be explained by the action of a cryptic red light signal (CRS), an amplification factor for PIAS produced by a distinct phytochrome-species activated by UVA. We suggest that UVA photoreceptors are not involved in anthocyanin synthesis in the broom sorghum.

살리실산이 오이 잎의 산화적 스트레스와 UV-B 내성에 미치는 영향 (Effects of Salicylic Acid on Oxidative Stress and UV-B Tolerance in Cucumber Leaves)

  • The effect of salicylic acid(SA) on antioxidant system and protective mechanisms against UV-B induced oxidative stress was investigated in cucumber(Cucumis sativus L.) leaves. UV-B radiation and SA were applied separately or in combination to first leaves of cucumber seedlings, and dry matter accumulation, lipid peroxidation and activities of antioxidant enzymes were measured in both dose and time-dependant manner. UV-B exposure showed reduced levels of fresh weight and dry matter production, whereas SA treatment significantly increased them. SA noticeably recovered the UV-B induced inhibition of biomass production. UV-B stress also affected lipid peroxidation and antioxidant enzyme defense system. Malondialdehyde(MDA), a product of lipid peroxidation, was greatly increased under UV-B stress, showing a significant enhancement of a secondary metabolites, which may have antioxidative properties in cucumber leaves exposed to UV-B radiation. Combined application of UV-B and SA caused a moderate increase in lipid peroxidation. These results suggest that SA may mediate protection against oxidative stress. UV-B exposure significantly increased SOD, APX, and GR activity compared with untreated control plants. Those plants treated with 1.0 mM SA showed a similar pattern of changes in activities of antioxidant enzymes. SA-mediated induction of antioxidant enzyme activity may involve a protective accumulation of $H_2O_2$ against UV-B stress. Moreover, their activities were stimulated with a greater increase by UV-B+SA treatment. The UV-B+SA plants always presented higher values than UV-B and SA plants, considering the adverse effects of UV-B on the antioxidant cell system. ABA and JA, second messengers in signaling in response to stresses, showed similar mode of action in UV-B stress, supporting that they may be important in acquired stress tolerance. Based on these results, it can be suggested that SA may participates in the induction of protective mechanisms involved in tolerance to UV-B induced oxidative stress.

Lambs Infected with UV-Attenuated Sporocysts of Sarcocystis ovicanis Produced Abnormal Sarcocysts and Induced Protective Immunity against a Challenge Infection

  • The present study surveyed the prevalence of natural infection of the sheep esphagus muscle with sarcocysts of Sarcocystis ovicanis and examined induction of protective immunity using UV-attenuated sporocysts. The overall prevalence of natural infection of the sheep was 95%. Infectivity of the collected sarcocysts was confirmed by shedding of sporulated oocysts after feeding infected esophageal tissues to dogs. To induce protective immunity, lambs were immunized 3 times (once a week) with $1.5{\times}10^4$ sporocysts exposed to UV-light for 30min (UV-30 group) or 60 (UV-60 group) min and then challenged with $1.5{\times}10^4$ normal sporocysts at the 3rd week post the 1st vaccination. These lambs showed high survival and less clinical signs of sarcocystosis than normal infected lambs. The attenuated sporocysts produced abnormal cysts; small in size and detached from the muscle fiber. These abnormalities were more obvious in UV-60 group than UV-30 group. Also, the $IFN-{\gamma}$ level and lymphocyte percentage were increased while the total leukocyte count was decreased in the UV-60 group compared with other groups. The high level of $IFN-{\gamma}$ may be an evidence for the induction of Th1 responses which may have protective effect against a challenge infection.

Isolation and Characterization of UV-inducible gene in Eukaryotic cells

  • The present study intends to characterize the DNA damage-inducible responses in eukaryotic cells. The fission yeast, S. pombe, which displays efficient DNA repair systems, was used in this study as a model system for higher eukaryotes. To study UV-inducible responses in S. pombe, five UV-inducible cDNA clones were isolated from S. pombe by using subtration hybridization method. To investigate the expression of isolated genes, the cellular levels of the transcripts of these genes were determined by Northern blot analysis after UV-irradiation. The transcripts of isolated gene (UV130) increased rapidly and reached maximum accumulation after UV-irradiation. Compared to the message levels of control, the levels of maximal increase were approximately 5 fold to UV-irradiation. In order to investigation whether the increase of UV130 transcripts was a specific results of UV-irradiation, UV130 transcript levels were examined after treating the cells to Methylmethane sulfonate (MMS). The transcripts of UV130 were not induced by treatment of 0.25% MMS. These results implied that the effects of damaging agents are complex and different regulatory pathways exist for the induction of these genes. To characterize the structure of UV130 gene, nucleotide sequences were analyzed. The nucleotide sequence of 1,340 nucleotide excluding poly(A) tail contains one open reading frame, which encodes a protein of 270 amino acids. The predicted amino acid sequences of UV130 do not exhibit any significant similarity to ther known sequences in the database.

Caenorhabditis elegans에서 분리한 자외선 유도유전자 (UV100과 UV150)의 발현 및 특성에 관한 연구 (Characterization of Expression of UV-Inducible Gene (UV100 and UV150) in Caenorhabditis elegans)

  • 본 연구는 DNA 상해유도기작을 규명하기 위하여 꼬마선충 (Caenorhabditis elegans) 으로 부터 subtraction hybridization 방법을 이용하여 자외선 유도 유전자인 UV100과 UV150을 분리하고 그 유전자 구조와 발현양상을 조사하였다. 분리한 유전자의 발현양상을 Northern hybridization 방법으로 살펴본 결과 자외선 조사 후에 최대 2배 이상의 발현 증가를 나타내었다. 이 결과 이미 밝혀진 다른 UV-inducible 유전자와 유사하게 UV100과 UV150 유전자는 자외선에 의해서만 발현이 증가됨을 알 수 있었다. 또한 분리한 유전자의 기능을 알기 위하여 RNAi 실험을 한 결과 분리한 자외선 유도유전자는 발생단계에 따라 다양한 DNA 회복기작을 나타냄을 알 수 있었다.

Expression of Poly (ADP-ribose) Polymerase During Apoptosis Induced by Ultraviolet Radiation in HeLa $S_3$ Cells

  • Induction of apoptosis allows the organism to get rid of abnormal cells and also of tumor cells. Understanding the mechanism involved in Ultraviolet radiation (UV) induced apoptosis may improve its therapeutic efficacy. In this study, we present expression of poly (ADP-ribose) polymerase (PARP) during apoptosis induced by UV in HeLa $S_3$ cells. Four different assays were performed in this study: morphological assessment of apoptotic cells and cell viability, DNA fragmentation analysis by agarose gel electrophoresis, quantitative assay of fragmented DNA, and expression of PARP by the western blot analysis. The percentages of apoptotic HeLa $S_3$ cells irradiated with $75J/m^2$ UV was increased continuously from 3 hrs incubation. DNA ladder pattern was appeared at 6 hrs. The amount of nucleosomal DNA fragments in cells treated UV increased from 3 to 12 hrs incubation and gradually decreased. The cleavage of PARP in HeLa $S_3$ cells irradiated with UV was induced, and the cleavage of PARP was more delayed in the cells pretreated with $5J/m^2$ UV and subsequently irradiated with $75J/m^2$ UV. than that in the cells only irradiated with $75J/m^2$ UV. Thus these data suggest that the cleavage of PARP relates with DNA fragmentation associated with apoptosis.
