Chelation of Tannin from Sorghum Extract using Fe(II) (수수 타닌의 Fe(II) 킬레이트)

  • This study investigated the chelation of a sorghum bran extract using iron (Fe) as a new natural colorant. The composition of the sorghum bran extract and chelation conditions were both examined. The thermal properties of the chelated colorants were analyzed using differential scanning calorimetry (DSC) and a thermal analyzer system(TGA). The sorghum bran extract solution showed a maximum absorbance at 281 nm based on UV/Vis spectrophotometry. According to the chelation pH conditions, pH 7.5 was the most effective. The chelation of the sorghum bran extract increased rapidly when increasing the iron concentration up to 2 mg/L, with no further chelation at a higher concentration. The particle size distribution curve for the chelated tannin revealed four groups: $4.5{\sim}17{\mu}m$, $20{\sim}42{\mu}m$, $45{\sim}80{\mu}m$, and $83{\sim}160{\mu}m$. In a DSC analysis, endothermic peaks attributed to the pyrolysis of the extract and chelated tannin were found at $318^{\circ}C$ and $415^{\circ}C$, respectively. In a TGA analysis, the chelation was shown to increase the final degradation temperature from $253^{\circ}C$ to $382^{\circ}C$, confirming that the chelation improved the thermal stability.

Improving the Chitinolytic Activity of Bacillus pumilus SG2 by Random Mutagenesis

  • Bacillus pumilus SG2, a halotolerant strain, expresses two major chitinases designated ChiS and ChiL that were induced by chitin and secreted into the supernatant. The present work aimed to obtain a mutant with higher chitinolytic activity through mutagenesis of Bacillus pumilus SG2 using a combination of UV irradiation and nitrous acid treatment. Following mutagenesis and screening on chitin agar and subsequent formation of halos, the mutated strains were examined for degradation of chitin under different conditions. A mutant designated AV2-9 was selected owing to its higher chitinase activity. To search for possible mutations in the whole operon including ChiS and ChiL, the entire chitinase operon, including the intergenic region, promoter, and two areas corresponding to the ChiS and ChiL ORF, was suquenced. Nucleotide sequence analysis of the complete chitinase operon from the SG2 and AV2-9 strains showed the presence of a mutation in the catalytic domain (GH18) of chitinase (ChiL). The results demonstrated that a single base change had occurred in the ChiL sequence in AV2-9. The wild-type chitinase, ChiL, and the mutant (designated ChiLm) were cloned, expressed, and purified in E. coli. Both enzymes showed similar profiles of activity at different ranges of pH, NaCl concentration, and temperature, but the mutant enzyme showed approximately 30% higher catalytic activity under all the conditions tested. The results obtained in this study showed that the thermal stability of chitinase increased in the mutant strain. Bioinformatics analysis was performed to predict changes in the stability of proteins caused by mutation.

Downregulation of $\textrm{NF}_{-{\kappa}}\textrm{B}$ activation in human keratinocytes by melanogenic inhibitors

  • Exposure of skin cells, particularly keratinocytes to various nuclear factor-kappaB ($\textrm{NF}_{-{\kappa}}\textrm{B}$) activators [e.g. tumor necrosis factor-$\alpha$, interleukin-1, lipopolysaccharides, and ultraviolet light] leads to phosphorylation and degradation of the inhibitory protein, $\textrm{I}_{{\kappa}}\textrm{B}$. Liberated $\textrm{NF}_{-{\kappa}}\textrm{B}$ is translocated into the nucleus where it can change or alter expression of target genes, resulting in the secretion of extracellular signaling molecules including melanotrophic factors affecting melanocyte. In order to demonstrate the possible role of $\textrm{NF}_{-{\kappa}}\textrm{B}$ activation on the synthesis of melanotrophic factors from the keratinocytes, the activities of $\textrm{NF}_{-{\kappa}}\textrm{B}$ induced by melanogenic inhibitors (MIs) were determined in human HaCaT keratinocytes transfected with $\textrm{pNF}_{-{\kappa}}\textrm{B}$-SEAP-NPT plasmid. Transfectant cells released the secretory alkaline phosphatase (SEAP) as a transcription reporter in response to the $\textrm{NF}_{-{\kappa}}\textrm{B}$ activity and contain the neomycin phosphotransferase (NPT) gene for the dominant selection marker for geneticin resistance. MIs such as niacinamide, kojic acid, hydroquinone, resorcinol, arbutin, and glycolic acid were preincubated with transfectant HaCaT cells for 3 h and then ultraviolet B (UVB) was irradiated. $\textrm{NF}_{-{\kappa}}\textrm{B}$ activation was measured with the SEAP reporter gene assay using a fluorescence detection method. Of the Mis tested, kojic acid ($IC_{50}$/ = 60 $\mu$M) was found to be the most potent inhibitor of UVB-upregulating $\textrm{NF}_{-{\kappa}}\textrm{B}$ activation in transfectant HaCaT cells, which is followed by niacinamide ($IC_{50}$/= 540 $\mu$M). Pretreatment of the transfectant HaCaT cells with the Mis, especially kojic acid and niacinamide, effectively lowered $\textrm{NF}_{-{\kappa}}\textrm{B}$ binding measured by electrophoretic mobility shift assay. Furthermore, these two inhibitors remarkably reduced the secretion level of IL-6, one of melanotrophic factors, triggered by UV-radiation of the HaCaT cells. These observations suggest that Mis working at the in vivo level might act partially through the modulation of the synthesis of melanotrophic factors in keratinocyte.

Inhibitory Effects of Dunaliella salina Extracts on Thermally-Induced Skin Aging (두날리엘라 살리나 추출물의 피부 열노화 억제 효과)

  • Just like UV radiation, heat increases collagen degradation and accumulation of abnormal elastin fiber and this is termed thermal skin aging. Dunaliella salina (DS), a green alga, is known for its beta-carotene accumulation, having various applications in the health and nutritional products. However, the effects of DS on heat-induced skin aging remain unexplored. In this study, we performed anti-thermal aging tests of the ethanol extract of DS (DSE). We measured the cellular levels of type I procollagen and MMP-1 using ELISA in human dermal fibroblast cells after heat shock. DSE reduced the expression of MMP-1 protein and increased the expression of type I procollagen. In addition, DSE upregulated the mRNA expression of HSP47 reduced by heat shock, which is involved in collagen synthesis. Also, DSE reduced the expression of inflammation mediator (TGF-${\beta}$, IL-12, etc). We demonstrate that DSE regulates the heat-induced solar elastosis through the regulation of tropoelastin and fibrillin-1, two major proteins of elastic fibers, and MMP-12 expression. These results suggest that DSE may be effective for preventing thermally induced skin aging.

Synthesis of TiO2/active carbon composites via hydrothermal process and their photocatalytic performance (수열합성법에 의한 TiO2/active carbon 복합체의 제조 및 광촉매특성)

  • Granular bamboo-derived active carbons (AC) were impregnated (or coated) with $TiO_2$ nano crystalline powders. The photocatalytic activity of the $TiO_2$-impregnated active carbons ($TiO_2$/AC) were determined on the basis of the degradation rate of methylene-blue aqueous solution under UV irradiation. The active compounds of $TiO_2$ were impregnated onto the AC under moderate hydrothermal conditions (${\leq}200^{\circ}C$, pH 11). The mean size of $TiO_2$ particles calculated from BET surface area were found to be as 50 nm. The $TiO_2$ precipitates were coated on the cavities or pores on the surfaces of highly activated carbons. Since the hydrothermal process led to a lowering of the on-set temperature of the anatase-to-rutile transition of $TiO_2$ as low as $200^{\circ}C$, $TiO_2$ crystallites of a pure anatase or a mixed form with rutile were successfully coated on the AC depending on the synthesis temperatures.

Dissolution Profile Analysis of Hydroxypropyl Methylcellulose-based Vitamin C Tablets (Hydroxypropyl methylcellulose를 활용한 비타민 C 지속성 정제의 용출 특성 분석)

  • The objective of this study was to develop oral matrix tablets for the sustained release of vitamin C. In this study hydroxypropyl methylcellulose (HPMC) has been utilized as an excipient, as it is one of the most widely used polymers, for use during long periods of time in formations. The vitamin C tablet formulation depends on the molecular weight and concentration of sustained-delivery in HPMC. Anti-oxidants have been added as a dissolution medium in order to prevent vitamin C degradation in water. The dissolution test was carried out in a distilled water medium, and the release model equation was applied to analyze the vitamin C release pattern. The results demonstrated that the release and lasting power of vitamin C tablets, containing HPMC, lasted for more than 12 h.

A Study on the Classifying Quality Standard by Comparison with Physicochemical Characteristics of Virgin, Pure, Pomace Olive Oil (버진, 퓨어, 포마스 올리브유의 이화학적인 특성 비교를 통한 품질등급 구분에 관한 연구)

  • This study investigated the classification of olive oils that are mainly distributed in Korea via imports. The fatty acid contents, degree of color, pigments, anti-oxidants, and sterol contents are analyzed on the different types of olive oil as follows: 10 kinds of extra virgin olive oil, 5 kinds of pure olive oil, and 5 kinds of refined olive-pomace oil. As a result of fatty acid analysis, the majority of oleic acid ($C_{18:1}$) and palmitic acid ($C_{16:0}$), and minority of linoleic acid ($C_{18:2}$) and stearic acid ($C_{18:0}$) were detected without any significant differences between the grades of olive oils. The UV spectrum is related to the ${\Delta}K$, and it is a part of the analysis factor for the purity and degree of degradation of the oil. Extra virgin olive oil had ${\Delta}K$ of almost 0, pure olive oil had 0.07~0.12, and refined olive-pomace oil had 0.1~0.13. These differed from extra virgin oil, and the pure or pomace oil ${\Delta}K$ had a confirmed distinct difference. The color degrees of chlorophyll with a low $L^*$ value and $(-)a^*$ (green) and carotenoid with $(+)b^*$ (yellow) were confirmed to have correlation between extra virgin and other olive oils. To compare chlorophyll and carotenoid as natural pigment in olive oils, 417 nm and the ratio of the absorbance at 480 nm (417/480) was calculated at 1.62 of extra virgin, 1.85 of pure olive oil, and 3.32 of refined olive-pomace oil. Therefore, it will be possible to distinguish when the extra virgin or pure olive oil are mixed with olive-pomace oil. The total amount of tocopherol, an anti-oxidant, were 19.06 in extra virgin, 10.91 in pure olive oil, and 27.88 in refined olive-pomace oil. The high content of tocopherol in pomace oil caused recovery of solvent extraction from olive pulp. Thus, extra virgin oil and pure olive oil were distinguished by olive-pomace oil. Polyphenol compounds in extra virgin olive oil measured high only in ferulic acid with 0.543 mg/kg, caffeic acid with 0.393 mg/kg, and other vanillic acid, vanillin, and p-coumaric acid had similar amount of 0.3 mg/kg. All grade of olive oils had the highest ${\beta}$-sitosterol content. Af (Authenticity factor) value were estimated with campesterol and stigmasterol content ratio (%). Af value was 19.2 in extra virgin olive oil, 17.1 in pure olive oil, 16.9 in refined olive-pomace oil, which were distinctive from sunflower oil with 3.7, corn oil with 2.4, and soybean oil with 2.0. It can provide important indicator of olive oil adulteration with other cheap vegetable oils. The results of this study can be used as a database for the classification of olive oil grade and distinguishing between the different types of oils.

Hydrophobic Polydimethylsiloxane Thin Films Prepared by Chemical Vapor Deposition: Application in Water Purification (화학적 증기 증착 방법을 통해 제조한 소수성 폴리디메틸실록산 박막: 수처리로의 응용)

  • Polydimethylsiloxane (PDMS) can be deposited on various substrates using chemical vapor deposition process, which results in the formation of PDMS thin films with thickness below 5 nm. PDMS layers can be evenly deposited on surfaces of nanoparticles composed of various chemical compositions such as $SiO_2$, $TiO_2$, ZnO, C, Ni, and NiO, and the PDMS-coated surface becomes completely hydrophobic. These hydrophobic layers are highly resistant towards degradation under acidic and basic environments and UV-exposures. Nanoparticles coated with PDMS can be used in various environmental applications: hydrophobic silica nanoparticles can selectively interact with oil from oil/water mixture, suppressing fast diffusion of spill-oil on water and allowing more facile physical separation of spill-oil from the water. Upon heat-treatments of PDMS-coated $TiO_2$ under vacuum conditions, $TiO_2$ surface becomes completely hydrophilic, accompanying formation oxygen vacancies responsible for visible-light absorption. The post-annealed $PDMS-TiO_2$ shows enhanced photocatalytic activity with respect to the bare $TiO_2$ for decomposition of organic dyes in water under visible light illumination. We show that the simple PDMS-coating process presented here can be useful in a variety of field of environmental science and technology.

Imaging Performance Analysis of an EO/IR Dual Band Airborne Camera

  • An airborne sensor is developed for remote sensing on an aerial vehicle (UV). The sensor is an optical payload for an eletro-optical/infrared (EO/IR) dual band camera that combines visible and IR imaging capabilities in a compact and lightweight package. It adopts a Ritchey-Chr$\'{e}$tien telescope for the common front end optics with several relay optics that divide and deliver EO and IR bands to a charge-coupled-device (CCD) and an IR detector, respectively. The EO/IR camera for dual bands is mounted on a two-axis gimbal that provides stabilized imaging and precision pointing in both the along and cross-track directions. We first investigate the mechanical deformations, displacements and stress of the EO/IR camera through finite element analysis (FEA) for five cases: three gravitational effects and two thermal conditions. For investigating gravitational effects, one gravitational acceleration (1 g) is given along each of the +x, +y and +z directions. The two thermal conditions are the overall temperature change to $30^{\circ}C$ from $20^{\circ}C$ and the temperature gradient across the primary mirror pupil from $-5^{\circ}C$ to $+5^{\circ}C$. Optical performance, represented by the modulation transfer function (MTF), is then predicted by integrating the FEA results into optics design/analysis software. This analysis shows the IR channel can sustain imaging performance as good as designed, i.e., MTF 38% at 13 line-pairs-per-mm (lpm), with refocus capability. Similarly, the EO channel can keep the designed performance (MTF 73% at 27.3 lpm) except in the case of the overall temperature change, in which the EO channel experiences slight performance degradation (MTF 16% drop) for $20^{\circ}C$ overall temperate change.

Effects of the Spatholobi calulis extract on Antioxidation and Inhibition of Matrix Metalloproteinase in Human Skin Fibroblasts (계혈등 추출물의 항산화와 사람 피부 섬유아세포에서의 Matrix Metalloproteinase-1 발현저해 효과)

  • The production of matrix metalloproteinases (MMPs) by the UV irradiated skin fibroblast and the degradation of exracellular matrix (ECM) by these enzymes is known as one of the main reasons of photoaging. In this study, to investigate the relationship between aging and Spatholobi caulis extract, we examined the effects of antioxidant, in vitro MMP inhibition and expression of UVA-induced MMP-1 in human dermal fibroblasts. Spathoiobi caulis extract was found to show scavenging activities of radicals and reactive oxygen species (ROS) with the $IC_{50}$ values of $45.81{\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and $3.11{\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. Spatholobi caulis extract inhibited the activities of MMP-1 in a does-dependent manner and the IC50 value calculated from semi-log plots was $31.96{\mu}g/ml$. Also, UVA induced MMP expression was reduced $74.66\%$ by treatment with Spatholobi caulis extract, and MMP-1 mRNA expression was reduced in a dose-dependent manner. Therefore Spatholobi caulis extract was able to significantly inhibit MMP expression in protein and mRNA level. All these results suggested that Spatholobi caulis extract may act as an anti-aging agent by antioxidation and reducing UVA-induced MMP-1 production.
