Characterization of Arthrospira platensis Mutants Generated by UV-B Irradiation (자외선 조사에 의해 유도된 미세조류 Arthrospira platensis 변이주의 특성)

  • Arthrospira platensis (A. platensis) is an economically important filamentous microalgae. The isolation of mutants by chemical or physical mutagen is a clue for the strain improvement. In this study, effects of ultraviolet-B (UV-B) radiation on A. platensis were investigated. Cells (or microalgae) were exposed to UV-B (15 Watt, 254 nm) for 1, 3, 5, and 10 min, and resulting mutants were designated UM1, UM3, UM5, and UM10, respectively. Especially, the lipid content of UM5 mutant was considerably increased by 8~11 fold compared to that of wild types. Moreover, the carotenoid content and antioxidant enzyme (peroxidase and superoxide dismutase) activity were increased. It was shown that UV-induced mutants can accumulate bioactive compounds, which will be useful for the industrial production of valuable products.

Removal of Sulfamethoxazole using Ozonation or UV Radiation; Kinetic Study and Effect of pH (오존 처리 및 UV 조사를 이용한 Sulfamethoxazole 제거; 동역학적 고찰 및 pH 영향)

  • This study was performed to assess the potential use of ozone or UV radiation for the treatment of water contaminated with sulfamethoxazole (SMX), which is frequently used antibiotic in human and veterinary medicines, especially focusing on the kinetic study and effect of pH. In a study using ozone alone, kinetic study was performed to determine second-order rate constant ($k_{O3,SMX}$) for the reactions of SMX with ozone, which was found to be $1.9{\times}10^6M^{-1}s^{-1}$ at pH 7. The removal efficiencies of SMX by ozone were decreased with increase of pH due to rapid decomposition of ozone under the condition of various pH (2.5, 5.3, 7, 8, 10). In a UV irradiation study at 254 nm, a kinetic model for direct photolysis of SMX was developed with determination of quantum yield ($0.08mol\;Einstein^{-1}$) and molar extinction coefficient ($15,872M^{-1}cm^{-1}$) values under the condition of quantum shielding due to the presence of reaction by-products formed during photolysis. For effect of pH on photolysis of SMX, SMX in the anionic state ($S^-$, pH > 5.6), most prevalent form at environmentally relevant pH values, degraded more slowly than in the neutral state (SH, 1.85 < pH < 5.6) by UV radiation at 254 nm.

  • To do breath alcohol measurement, a sensor is necessary that it can detect low alcohol gas concentration of 0.01% at least. In this work, a capacitance-type alcohol gas sensor using porous silicon layer is developed to measure low alcohol gas concentration. The sensor using porous silicon layer has some sensitivity at room temperature by very large effective surface area, but there is still much room for improvement. In this experiment, we measured the capacitance of the sensor under 254 nm UV light on the porous silicon layer, in which alcohol solution was kept in a flask at 25, 35, and $45^{\circ}C$ by a heater. As the result, the improvement of sensitivity by illuminating UV light was observed. The increasing rate of the capacitance was shown to be double more than those measured under UV-off state. It is supposed that UV light activates response of the oriental and interfacial polarizations which have slow relaxation time for AC field.

  • In order to removal turbidity at high turbidity, this study was carried to evaluate the coagulants(Alum, PACl, PACS) that was suited the characteristics of raw water in water treatment plants and to determinate the optimum method of lime feed. When the optimum coagulant was selected the organic matter removal was also investigated as $UV_{254}$. PACl, lime first feed had the highest turbidity removal efficiency rate as above 99.1% and then $UV_{254}$ removal rate was obtained over than 88.0%. If you had the necessary of the lime feed, among the method of lime feed time interval feed largely was improved than simultaneous feed. Also, lime feed dose had about 1/5 of coagulants dose in case of Alum and PACl, but always PACS should be considerated lime dose.

  • We have been proposed model equation which is able to predict the trihalomethane producing concentration formation, that is one of byproduct, in the water treatment processes. In proposed model, the effects of trihalomethane factors like chlorine contact time, pH, temperature, TOC and UV-254 are considered. The concentration of the trihalomethane produced is proportion to the contact with chlorine, pH of water, temperature of water TOC and UV-254, respectively. This proposed model could be predicted the formed concentration of trihalomethanes by trihalomethane factors.

  • Ozone alone and catalytic ozone processes were introduced for treatment of humic acid, which is representative refractory organic compound. The treatment efficiencies of humic acid in each process were analyzed in pH variation, DOC removal, and $UV_{254}$ decrease. Mn loaded GAC catalyst was prepared by loading potassium permanganate onto the granular activated carbon surface. BCM-GAC and BCM-Silica gel catalyst were prepared by BCM. $UV_{254}$ decrease in all processes was comparatively high with efficiency over 87%. DOC removal in ozone/GAC process was the highest with 78%, and removal rates for other processes followed the order ozone/BCM-GAC(62%) > ozone/BCM-silica gel(45%) > ozone/silica gel(43%) > ozone/Mn Loaded GAC(42%) > ozone alone(37%).

A Study on Ozone Oxidation of Algae (조류의 오존산화에 관한 연구)

  • The objectives of this research were to estimate variation characteristics of TOC, UV-254 and NH4+-N, etc, if odorous generated algaes flowed into water treatment plant and they contacted with ozone known as typical advanced treatment. It was estimated that pH decreased from initial 7.4 to 2.1 after ozone contact 100min. pH declined to early 10min. suddenly and then pH drop did almost occur. TOC and UV-254 continued to decrease with passed time of ozone contact. NO3--N concentration was much higher than NH4+-N and NO2--N for whole test period. Because ozone oxidized organic matters as various kinds of ion material and ion intensity of sample was extended, conductivity showed high as passed time of ozone contact. Owing to ozone oxidation of algae, color did almost disappear after ozone contact 20min.

  • Ozone alone, Ozone/GAC, Ozone/$H_2O_2$ and Ozone/GAC/$H_2O_2$ processes were introduced for treatment of humic acid, which is a representative refractory organic compound. $H_2O_2$ and GAC used as catalysts for experiment. The treatment efficiencies of humic acid in each process were analyzed for pH variation, DOC removal, and $UV_{254}$ decrease. $UV_{254}$ decrease in Ozone/GAC and Ozone/GAC/$H_2O_2$ processes were the highest with about 93%, and Ozone alone and Ozone/$H_2O_2$ processes were 88%. DOC removal in Ozone/GAC/$H_2O_2$ process was the highest with 71%. Removal by Ozone/GAC, Ozone alone, and Ozone/$H_2O_2$ processes were 66%, 39%, and 47%, respectively.

Improving Coagulation Performance with pH Preadjustment in Drinking Water Treatment (정수처리에서 pH 저감에 의한 응집효율향상에 관한 연구)

  • This paper reports on a pilot scale comparison of PACS coagulation with and without pH preadjustment. The pH of the water was adjusted with carbon dioxide and sulfuric acid. Process performance was assessed on the basis of total organic carbon(TOC), UV absorbance, turbidity and disinfection by-product(DBP) precursors. Coagulation pH appeared to be a determining factor for maximum NOM removal. The optimum coagulation pH in order to decrease TOC and turbidity were pH 7. Preadjustment of pH 7 increased TOC removal to as much as 43, 47 percent with sulfuric acid and carbon dioxide. Moreover, coagulation at pH 7 caused a reduction in UV$_{254}$, THMFP and HAAFP compared to the baseline coagulation. For preadjustment of pH 7 with carbon dioxide, the percentage of TOC, UV$_{254}$, THMFP and HAAFP shows the reduction rate of 3.8, 0.5, 4.8, 9.4% comparing to the coagulation rendition using sulfuric acid. Acid addition to depress pH during coagulation decrease Langelier Saturation Index(LSI), potentially causing increase corrosion in water distribution systems. LSI for carbon dioxide and sulfuric acid at pH 6 was -2.3, -3.3. Therefore, carbon dioxide was more effective at controlling corrosion than sulfuric acid.

A Study on the Removal of LAS using TiO2 Photocatalyst (TiO2 광촉매를 이용한 LAS의 제거에 관한 연구)

  • The objective of this study is to delineate removal efficiency of the Linear alkylbenzene sulfonates(LAS) in solution by $TiO_2$ photocatalytic oxidation as a function of the following different experimental conditions : initial concentration of LAS, $TiO_2$ concentration, UV wavelength and pH of the solution. It was increased with decreasing initial concentration of LAS and with decreasing pH of the solution. Removal efficiency increased with increasing $TiO_2$ concentration but was almost the same at $TiO_2$ concentration of 2 g/L and 3 g/L, i.e., for initial LAS concentration of 50 mg/L. It was removal efficiency was 85% at 150 min in the case of $TiO_2$ concentration of 0.5 g/L but 100% after 150 min in the case of $TiO_2$ concentration of 1 g/L, 100% after 110 min in the case of $TiO_2$ concentration of 2 g/L and 3 g/L. UV wavelength affection on the removal efficiency of LAS decreased in the order of 254, 312 and 365 nm as increasing wavelength. But the removal efficiency of LAS was nearly the same at UV wavelength of 254 nm and 312 nm.
