In order to understand molecular phylogenetics of silkworm (Bombyx mori), we analyzed the sequences of BmoMAR isolated from Bomhyx mori that is partial coding gene of transposase of mariner-like element(MLE). By pairwise comparing nucleotide sequences of BmoMAR with ten previously reported insect MLEs accessed in GeneBank, the average genetic distance was estimated to be 0.4840. The phylogenetics tree constructed from nine insect species except for human MLE(Hsmarl) by UPGMA method indicated that MLEs are divided into three clusters, and Drosophila mariutiana was independently subgrouped. Bombyx mori(BmoMAR) was subgrouped with microcaddishfly (Orthotrichia cristata), webworm(Atteva punctella), almond moth(Ephestia cautella), Hyalopora cecropia which we lepidoptera. Phylogenetics tree according to UPGMA principle, on the basis of informative nucleotide sequences of nine insect MLEs, indicated that Bombyx mori was more closely related to microcaddishfly(Orthotrichia cristata) and webworm (Atteva punctella) of lepidoptera. We suggest that insect MLEs are a useful key for studying molecular phylogenetics among intra species of insects.
The objectives of this study was to identify genetic variations of Tricholoma matsutake (S. Ito et Imai) Sing. growing in different geographic ranges in South Korea. Mushrooms were collected during fruiting seasons from 1994 to 1997 from 11 major sites which included four sites (Bonghwa, UIjin, Goryoung, and Chungdo) in Kyongbuk Province, three sites (Changnyung, Hadong, and Hamyang) in Kyongnam Province, two sites (Yangyang and Inje) in Kangwon Province, one site (Goisan) in Choongbuk Province, and one site (Namwon) in Chonbuk Province. Two mushrooms each from three to eight shiros in each sites were collected. Genetic characteristics were analyzed by Inter-Simple Sequence Repeat Polymerase Chain Reaction (I-SSR PCR) method using six primers. With a total of 131 DNA bands identified, Nei's genetic distance and UPGMA tree were constructed. It was estimated that genetic variations between sites amounted to 12.9%, while 87.1% of total variation was explained by variations among individuals within sites. The cluster analysis indicated that the eleven major sites were clustered into four groups, group I (Yangyang, Hamyang, Inje, Hadong and UIjin), group II (Changnyung, Namwon and Chungdo), group III (Goryoung), and group IV (Bonghwa and Goisan). It is concluded that matsutake mushrooms in South Korea have a considerable degree of genetic variations between major sites.
Inter-simple sequence repeat (I-SSR) markers were analyzed from diploid genomes of 95 nutmeg trees (Torreya nucifera Siev. et Zucc.) in 5 populations. A total of 62 I-SSR amplicons were observed and 7 of them were monomorphic in 95 individuals. DNA fingerprint of each tree was verified by pooling the observed I-SSR amplicons. Most of the genetic diversity was allocated within population (90.65%) and all the populations revealed similar level of I-SSR amplicon diversity within population. Degree of population differentiation (${\phi}_{ST}=9.35%$) was moderate on the basis of criteria obtained from isozyme analysis. Based on the results of the cluster analysis of UPGMA, genetic relationships among 5 populations were not coincided with the pattern of geographic distribution. Non-significant confidence interval at each node also suggests that all the nutmeg populations are genetically not much differentiated.

Japanese Black cattle of Hyogo prefecture (Tajima strain) are famous for its ability to produce high-quality meat and have been maintained as a closed system for more than 80 years. In order to assess the usefulness of microsatellite markers in closed cattle populations, and evaluate the genetic structure of the Tajima strain, we analyzed representative dams of the Tajima strain comprised of the substrains Nakadoi and Kinosaki. Genetic variability analyses indicated low genetic diversity in the Tajima strain. In addition, a recent genetic bottleneck, which could be accounted for by the high level of inbreeding, was detected in both substrains. In phylogenetic analyses, relationship coefficients and genetic distances between individuals were calculated using pedigree and microsatellite information. Two phylogenetic trees were constructed from microsatellite and pedigree information using the UPGMA method. Both trees illustrated that most individuals were distinguished clearly on the basis of the two substrains, although in the microsatellite tree some individuals appeared in clusters of different substrains. Comparing the two phylogenetic trees revealed good consistency between the microsatellite analysis tree and the pedigree information. The correlation coefficient between genetic distances derived from microsatellite and pedigree information was 0.686 with a high significance level (p<0.001). These results indicated that microsatellite information may provide data substantially equivalent to pedigree information even in unusually inbred herds of cattle, and suggested that microsatellite markers may be useful in revealing genetic structure without accurate or complete pedigree nformation. RAPD analysis was performed to discuss the taxonomic status and phylogenetic relationships among the tribe Forsythieae and related groups. Two hundred and eighteen scorable polymorphic bands were detected from fourteen oligonucleotide primers. From the results of RAPD analysis by Nei and Li's genetic distance, each individuals of Abeliophyllum distichum showed high genetic relationships with ranging from 0.085 to 0.301, also the genus Forsythia showed from 0.042 to 0.655 among the species and populations. But, Abeliophyllum and Forsythia showed distinct dissimilarity, ranging from 0.610 to 1.258. And genetic differences among the population of Forsythia were 0.042 in F. koreana, 0.275 in F. saxatilis, 0.275 in F. ovata, 0.279 in F. nakaii, and 0.249 in F. viridissima. The UPGMA phenogram of tribe Forsythieae based on the results of RAPD analysis were presented that Abeliophyllum is distinct genus different from Forsythia. NJ tree which applied as the outgroups Fontanesia and Jasminum was derived, and it showed that tribe Forsythieae might be a monophyletic group. The genus Fontanesia was showed as sister group of tribe Forsythieae. Among the populations of taxa in Forsythia, F. koreana and F. saxatilis were more closely related, and F. ovata and F. nakaii were very closely related to F.japonica. And Fontanesia was the sister group of tribe Forsythieae.
Genomic DNA (gDNA) extracted from two populations of natural and cultured river pufferfish (Takifugu obscurus) was amplified by polymerase chain reaction (PCR). The complexity of the fragments derived from the two locations varied dramatically. The genetic distances (GDs) between individuals numbered 15 and 12 in the cultured population was 0.053, which was the lowest acknowledged. The oligonucleotide primer OPC-11 identified 88 unique loci shared within each population reflecting the natural population. The OPC-05 primer identified 44 loci shared by the two populations. The average band-sharing (BS) values of individuals in the natural population (0.683±0.014) were lower than in those derived from the cultured population (0.759±0.009) (p<0.05). The shortest GD demonstrating a significant molecular difference was found between the cultured individuals # 15 and # 12 (GD=0.053). Individual # 02 of the natural population was most distantly related to cultured individual # 22 (GD=0.827). A cluster tree was built using the unweighted pair group method with arithmetic mean (UPGMA) Euclidean GD analysis based on a total of 578 various fragments derived from five primers in the two populations. Obvious markers identified in this study represent the genetic structure, species security, and proliferation of river pufferfish in the rivers of the Korean peninsula.
Chitin synthases(UDP-N-acetyl-D-glucosamine: chitin 4-$\beta$-N-acetyl-D-glucosaminyl transferase, EC 2.4.1.16) catalyze the synthesis of chitin from UDP-N-acetyl-D-glucosamine. Two zymogenic type of chitin synthase gene(TmCHS1 and TmCHS2) were amplified and its nucleotide sequences were determined. By the amino acid comparison and UPGMA tree grouping, TmChs1 and TmChs2 were classified as class II and class IV chitin synthases respectively. The class II type TmChs1 was grouped with others of Agaricales ectomycorrhizal mushroom. Additionally the phylogenetic tree was well adapted to Hymenomycete previously classified by morphological and physiological characteristics.
In order to presume the relationships between two species of P. ternata and P. tripartia, and their populations of the Korean Pinellia, RAPD analysis was performed. The length of the amplified DNA fragments ranged from 300 to 2,500bp. Seventy scorable RAPD makers were found from the PCR reactions with 7 random oligoprimers and were analyzed by Nei-Li's genetic coefficient. Also, some regional groups instead of same taxa were clustered from the phenogram of UPGMA analysis and NJ tree. Populations within each species were clustered at low genetic distance, there had the closed relationship. According to the regional individuals, Pinellia ternata was showed the variation pattern of morphological (leaf shape and flower color) and cytological characters(somatic chromosome numbers). So we suggested to difference of characteristic variety based on variety of habitat. According to this study, new species (Pinellia sp.) was affiliated with Pinellia and had the closest relationship with Hallasan and Japan population. The RAPD data was very useful to define the genetic variation and to discuss the relationships among the intraspecific taxa and their populations of the Korean Pinellia.

Vaccinium oldhami Miq. is a Korean native tree, which is deciduous and shrub tree with broad leaf. It grows 1~4m in height generally. Ecologically, this tree grows well in shady place even in barren soil. Also, the tree has resistance to cold and dry, which tend to form a little community. This research investigates quantitative morphological characteristics of leaf and fruit among the V. oldhami in South Korea and then considers its relationship on the basis of raw data among the 10 populations. This study will give us invaluable information about growing conditions, reasonable management and breeding by selection of V. oldhami in South Korea. The main results obtained from this study are summarized as follows; Leaf size of Mudeung population was larger than other populations. Naebyeon population was smaller in size of the leaf than other populations. Anmyeondo population was larger in fruit characteristics compared with other populations and Deogyu population was the smallest among populations. According to cluster analysis based on the leaf and fruit morphological characteristics, the natural V. oldhami populations were classified into four groups such as the first group of Kumo population, the second group of Mudeung population, the third group of Anmyundo, Daedun, Doolyun population and the fourth group of the other five populations.

For the variant identification of Korean dogwood, Cornus kousa Buerg., we investigated useful genetic marker using the ISSR primer. The overall amplicons was 58 in six primers and the mean number of amplicons per primer was 9.67. The UPGMA dendrogram based on the genetic distance showed two major clusters composed of pink-bracted and white-bracted groups respectively. And white-bracted group was divided to two groups, island and inland groups. This result showed that ISSR analysis provides useful markers for the variant identification even in the seedling or young tree of Cornus kousa, especially pink-bracted individuals of Isl. Oenaro.
