The Study of Age Determination Using Stepwise Dissolution Technique (단계적 용해에 의한 연대측정법 연구)

  • Recently developing method of age determination using stepwise dissolution technique to expand the applicability of absolute age determination significantly is evaluated whether it is applicable to the Korean samples. The materials selected for the study are uranium-bearing black slates from Changni Formation of Ogcheon metamorphic belt, tourmaline separated from Naedeongni granite of Yeongnam massif, garnet and ilmenite separated from ilmenite-bearing anorthositic rock of Yeongnam massif, scheelites from Ogbang mine, and magnetite separated from Gyemyeongsan Formation of Ogcheon metamorphic belt. For the stepwise dissolution, various acid steps with different normalities and different durations were applied to leach the samples. The leachate from each step was analyzed to determine the Pb isotopic composition and concentrations of Pb and U using thermal ionization mass spectrometer. The black slates from the Changni Formation and the tourmaline from the Naedeongni granite reveal significant variation of Pb isotopic composition, which reveals the potential of such stepwise dissolution technique as a dating method. The behaviors of uranium and lead during the each stage of step leaching are different, which seem to reflect the differences in positions within the crystal lattices depending upon mineral species.

  • Lead-acid battery is used widely as a power source at a automobile, industrial machines, folk lifts, U.P.S. etc. Lead-acid battery is cheaper than arty other secondary battery, but this battery has many disadvantages such as heavy, low energy density, environment problem etc. In this article, we introduce cyclic voltammetry methods to investigate corrosion behaviour of positive grids of Pb-Ca alleys.

  • The characteristics of heavy metal biosorption on the seaweeds were investigated to develop a biological treatment technology for wastewater polluted with heavy metals. The heavy metal biosorption on seaweeds ranked in the tallowing order: U. pinnatifida$\geq$E. stolonifera$\geq$Laminaria sp.>G. amansii. The Pb was biosorbed in the range of $93{\sim}99%$, and the Cu and Cd were biosorbed in the range of $70{\sim}80%$ at the concentration of the heavy metal of $100mg/{\ell}$ respectively. The seaweed which was pretreated with $CaCl_2$ solution improved the biosorption of the heavy metals. The temperature and pH didn't affect the biosorption of heavy metals. The Langmuir isotherm reasonably fit the data of heavy metal biosorption compared to the Freundlich isotherm. The affinity of metals on the biosorption ranked in the following order: Pb>Zn>Cu>Cd. The biosorption efficiency of the heavy metals on the U. pinnatifida decreased in the multi-component rather than the single component. The heavy metals adsorbed on the U. pinnatifida were recovered using 0.3%-NTA. U. pinnatifida among the seaweed used in this work showed the best performance for the biosorption of the heavy metals.

Isolation and Characterization of Aeromons hydrophila PBl6 and Properties of Synthetic Wastewater Degradation (Protease 생성균 Aeromonas hydrophila PB16의 분리 및 합성폐수처리능)

  • Protease producing bacterium, PB16 was isolated from food processing wastewater sludge and paddy field soil samples and selected by the clear zone and enzyme activity test. The isolate was gram negative, rod type and its protease productivity was 6.49 U/ml. As a result of API20NE kit test and 16S rDNA sequencying, the isolated PB16 was identified as Aeromonas hydrophila (99%). The growth rate ($h^{-1}$) was 0.21 in synthetic waste water only and 0.26 in synthetic waste water containing vitamin and mineral using a bioscreen C. Synthetic wastewater removal rate was 59 and 87%, respectively after 1 and 3 day reaction (intial CODcr was 2,472 mg/l).

Growth and Chlorophyiil Biosynthesis of Vigna angularis under Lead Stress

  • The effect of various supplies of lead singly and in combination with aluminium on growth and chlorophyll biosynthesis was investigated in 7-day-old Vigna angularis seedlings. Expose to 50 uM Pb or more drastically reduced root elongation rate. Significant depressions in root growth was observed within 1 day and no recovery of growth was seen over the duration of treatment period. Root elongation decreased depending on the Pb concentrations. Root growth inhibition was stronger than shoot growth inhibition. The initiation of lateral roots appeared to be more sensitive to Pb than the growth of main roots. Inhibition of root and shoot elongation by Pb was lessened by combined exposure of Pb and Al, suggesting that the presence of Al reverse the inhibitory effect of Pb alone. With the histochemical sodium rhodizonate method the rate of Pb uptake was dependent on the Pb concentration and exposure time of the roots to Pb salts. Pb was first deposited on the root surface and then translocated radially in the root cap cells. During a longer Pb administration (up to 72 h) Pb penetration was nonuniform, with accumulation within the cortex or endodermis. There was drastic reduction in chlorophyll content by Pb. The Pb inhibition of chlorophyll synthesis was concentration dependent. $\delta-Aminolevulinic$ acid dehydratase (ALAD) activity exhibited distinct inhibition from control. Reduction in chlorophyll content was accompanied by proportional changes in ALAD activity. Chlorophyll content and ALAD activity were less affected by combined exposure of Pb and Al, suggesting that Al has a protective effect against the inhibiting action of Pb on photosynthetic activity.
