Preliminary Proteomic Analysis of Thiobacillus ferrooxidans Growing on Elemental Sulphur and Fe2+ Separately

  • Thiobacillus ferrooxidans is one of the most important bacterium used in bioleaching, and can utilize $Fe^{2+}$ or sulphide as energy source. Growth curves for Thiobacillus ferrooxidans have been tested, which show lag, logarithmic, stationary and aging phases as seen in other bacteria. The logarithmic phases were from 10 to 32 hours for Thiobacillus ferrooxidans cultivated with $Fe^{2+}$ and from 4 to 12 days for Thiobacillus ferrooxidans cultivated with elemental sulphur. Differences of protein patterns of Thiobacillus ferrooxidans growing on elemental sulphur and $Fe^{2+}$ separately were investigated after cultivation at $30^{\circ}C$ by the analysis of two-dimensional gel electrophoresis (2-DE), matrix-assisted laser desorption/ ionization (MALDI)-Mass spectrometry and ESI-MS/MS. From the 7 identified protein spots, 11 spots were found more abundant when growing on elemental sulphur. By contrast 6 protein spots were found decreased at elemental cultivation condition. Among the proteins identified, cytochrome C have been previously identified as necessary elements of electron-transfering pathway for Thiobacillus ferrooxidans to oxidize $Fe^{2+}$; ATP synthase alpha chain and beta are expressed increased when Thiobacillus ferrooxidans cultivated with $Fe^{2+}$ as energy source. ATP synthase Beta chain is the catalytic subunit, and ATP synthase alpha chain is a regulatory subunit. The function of ATPase produces ATP from ADP in the presence of a proton gradient across the membrane.

  • Escherichia coli 세포에서 intramolecular triplex가 형성될 수 있는지를 조사하기 위하여 genetic assay를 도입하였다. Two dimensional gel electrophoresis 방법을 사용하여 $(G)_9AATTC(G)_9$ 혹은 $(GAA)_9TTC(GAA)_8$ sequences가 in vitro에서 intramolecular triplex 구조를 형성하는 것을 확인하였다. Genetic assay를 위하여 temperature-sensitive EcoRI methylase 유전자를 포함한 플라스미드와 triplex를 형성할 수 있는 $(G)_9AATTC(G)_9$ 혹은 $(GAA)_9TTC(GAA)_8$ sequences를 포함한 플라스미드로 E. coli를 cotransform 하였다. EcoRI methylase가 발현되는 permissive temperature에서 pur pyr sequences를 포함하는 플라스미드는 EcoRI methylase 작용에 kinetically 저항성을 보여주었다. 이러한 결과는 pur pyr sequence들이 EcoRI methylase가 작용하기 어려운 triplex 구조를 E. coli 세포에서 형성한다는 것을 증거한다.

성장기 소의 등심에 발현되는 단백질들의 분리 및 동정 (Isolation and Identification of Proteins Increasingly Expressed in Beef Loin on Maturation)

  • 각각 다른 성장기의 한우 등심에서 추출한 단백질을 이차원 전기영동법으로 분리하여 젤 상의 단백질 전개 양상을 비교하였다. 성장 0, 6, 12, 24 개월령의 한우 등심 단백질들을 길이 16 cm 튜브젤에서 등전점에 따라 분리하고, 이차원적으로 $18{\times}20$ cm, 12% SDS-polyacrylamide gel 전기영동 하여 단백질을 분리하였다. 등전점 3.0에서 9.0 그리고 분자량 15,000에서 100,000 Da 사이의 단백질들이 분리되어 Silver 염색법으로 명확히 구분할 수 있었다. 흥미롭게, 성장과정에서 단백질 발현이 증가했거나 감소한 단백질들은 저분자 단백질들 이었다. 성장 과정 중 증가된 단백질들을 분리하기 위해 수용성 단백질들을 조직으로부터 1% Triton X-100 으로 추출하였다. 그리고 이를 30%와 50% 황산암모니아로 분획하였다. 이와 같이하여 각 단백질들의 분리조건을 결정하였다. 이들 조건을 이용하여 발현이 증가된 단백질들을 분리하고 PVDF membrane에 옮겨서 아미노산 서열을 결정하여 단백질을 규명하였다.

Streptomyces coelicolor (Muller)의 과산화수소 대응 반응에 관련된 단백질 양상의 분석 (Profile Analysis of Proteins Related with Hydrogen Peroxide Response in Strep-tomyces coelicolor (Muller))

  • Streptomyces coelicolor (Muller) 의 세포에 $100 \mu$M 의 과산화수소를 1시간 동안 처리하여 과산화수소 스트레스중에 생성되는 단백질을 L-[$^{35}S$] methionine 을 이용하여 순간 표지하였다. 단백질을 추출하여 2차원 겔 전기영동으로 분석한 결과 지수 성장기의 세포가 가지는 총세포 단백질 중 약 100개의 단백질의 합성이 과산화수소에 의해 증가되는 것을 관찰하였다. 이들을 Pin(peroxide inducible) 단백질이라고 명명하고 과상화수소 처리 후 발현이 증가되는 시간에 따라 세 그룹으로 나누었다. 약 60 개의 Pin 단백질은 과산화수소 처리후 20분 이내에 발현이 증가하여 1시간동안 지속적으로 다량 합성되었다. 정체 성장기의 세포에서는 62개의 단백질의 합성이 과산화수소에 의해 증가되었으며, 이 중 21 개의 단백질은 지수성장기의 Pin 단백질과 일치하였다. 과산화수소에 대한 저항성이 증가한 세가지 돌연변이체의 단백질을 지수 성장기에서 추출하여 2 차원 겔 전기영동으로 분석한 결과, 각각의 경우 15, 17, 15개의 Pin 단백질을 야생형보다 항상적으로 다량합성하는 것을 관찰하였다. 이 pin 단백질 중 9개 (D74.7 a, E76.0c, E23.3, F50.7, F47.2a, F25.5, G39.6b, G24.0, H39.6a) 는 두가지 돌연변이체 모두에게 증가되었다. 따라서 이 단백질들은 S. coelicolor 가 과산화수소 스트레스에 대응하는데 있어 중요한 역할을 담당한다고 추정된다.

Iron Starvation-Induced Proteomic Changes in Anabaena (Nostoc) sp. PCC 7120: Exploring Survival Strategy

  • This study provides first-hand proteomic data on the survival strategy of Anabaena sp. PCC 7120 when subjected to long-term iron-starvation conditions. 2D-gel electrophoresis followed by MALDI-TOF/MS analysis of iron-deficient Anabaena revealed significant and reproducible alterations in ten proteins, of which six are associated with photosynthesis and respiration, three with the antioxidative defense system, and the last, hypothetical protein all1861, conceivably connected with iron homeostasis. Iron-starved Anabaena registered a reduction in growth, photosynthetic pigments, PSI, PSII, whole-chain electron transport, carbon and nitrogen fixation, and ATP and NADPH content. The kinetics of hypothetical protein all1861 expression, with no change in expression until day 3, maximum expression on the $7^{th}$ day, and a decline in expression from the $15^{th}$ day onward, coupled with in silico analysis, suggested its role in iron sequestration and homeostasis. Interestingly, the up-regulated FBP-aldolase, Mn/Fe-SOD, and all1861 all appear to assist the survival of Anabeana subjected to iron-starvation conditions. Furthermore, the $N_2$-fixation capabilities of the iron-starved Anabaena encourage us to recommend its application as a biofertilizer, particularly in iron-limited paddy soils.

  • Inoculation with the compatible strain Ds 1 of Xanthomonas campestris pv. vesicatoria caused brownish ad water-soaked lesions, but incompatible strain Bv5-4a produced hypersensitive symptoms with local necrosis on tomato (cv. Kwangyang) leaves. Bacterial populations of the compatible strains Ds 1 propagated more greatly than the incompatible strain Bv5-4a at the frist onset, but no differences were observed 5 days after inoculation. The bacterial infection induced the synthesis and accumulation of soluble proteins in tomato leaves, especially in the incompatible interaction. Native-polyacrylamide gel electrophoresis distinguished the soluble proteins in the tomato leaves infected by the compatible or incompatible strains. A protein of low molecular weight occurred only in the incompatible interaction. Some pathogenesis-related (PR) proteins, especially the 15, 18, 23, 26 and 54 kDa proteins, were detected only in the infected tomato leaves. In the two-dimensional electrophoresis, some proteins with different molecular weights (Mr. 21∼29 kDa) and the pI 8∼9 appeared more distinctly only in the incompatible interaction. These data suggest that the de novo synthesis of some PR proteins in tomato may be significant in defense against X. c. pv. vesicatoria.

Protein profiling in disomic addition lines of wheat carrying Leymus racemosus chromosomes

  • Wheat wild relatives that have never been domesticated contained useful genetic resources such as the resistance to abiotic and biotic stresses. Leymus racemosus is one of the wild species. It can grow in a harsh environment like seaside and distribute by healthy rhizomes. Also, it has a useful genetic resource such as salt tolerance and different diseases resistance. Wheat (Triticum aestivum L. cv. Chinese Spring; CS) was crossed with L. racemosus. Wheat-L. racemosus disomic addition lines were produced. The purpose of this study is to identify protein expression in each disomic addition line compared to CS. We performed two-dimensional electrophoresis. Two-dimensional gels stained with coomassie brilliant blue (CBB), a total of 1566 differentially expressed proteins were identified by Progenesis Same Spots software from the cultivars. However, a total of 90 protein spots were identified to be either present or absent or showing significantly differential expression when the difference threshold was set to more than 1.5 fold. However, out of the 90 differentially protein spots, a total of 74 spots were sorted for mass spectrometry analysis. The identified proteins may provide important clues for better understanding the molecular changes in the chromosomes carrying Leymus racemosus.

한우육의 육질에 중요 영향을 미치는 근내지방축적 최적기에 발현되는 단백질 동정 (Identification of the Proteins Expressed at Optimum Marbling in Hanwoo Loin)

  • Proteins from the loin tissues age ranged from 0 to 24 months of ten Korean cattle were extracted, separated and compared on two dimensional(2 D) gels to identify the proteins whose expression is highly correlated to marbling. We also compared the difference of loin proteins between castrated and non castrated bull cows on two dimensional gels. As the marbling in the loin of the cattle is optimized at 18 to 24 months, eight proteins expressed significantly higher level in 24 month than in 0 or 6 month were selected in terms of isoelectric points(pIs) and molecular weights. Using these values, we searched the Swiss Prot database via the ExPASy molecular biology server with TagIdent program. The proteins with the nearest molecular weights and isoelectric points were selected from the lists. These possible candidates were confirmed by N terminal microsequencing of the eight selected proteins. Three proteins, myoglobin, hemoglobin and ATPase, whose N termini were not blocked could be microsequenced and found to be exactly matched to the selected candidates. It is suggested that the proteins increasingly expressed in marbling periods can be involved in meat color, lipid transport and flavor improvement.

Two-Dimensional Reference Map of Schizosaccharomyces pombe Proteins (Update)

  • Based on the first 2D reference map of the fission yeast Schizosaccharomyces pombe protein reported previously, we expanded and updated the map using narrower pI ranges. In this paper, 240 protein spots were identified on our reference map. In the pI 4-7 range, 144 spots corresponding to 86 different proteins were identified. In the pI 6-9 range, 43 spots corresponding to 35 different proteins were identified. Fifty-three new spots corresponding to 39 different proteins were further identified in the pI 5-6 range.
