Effect of temperature on pharmacokinetics of nalidixic acid, piromidic acid and oxolinic acid in olive flounder Paralichthys olivaceus following oral administration (넙치, Paralichthys olivaceus에 nalidixic acid, piromidic acid, oxolinic acid의 경구투여 약물동태에 미치는 수온의 영향)

  • Effects of temperature ($13{\pm}1.5^{\circ}C$, $23{\pm}1.5^{\circ}C$) on the pharmacokinetic properties of nalidixic acid (NA), piromidic acid (PA) and oxolinic acid (OA) were studied after oral administration to cultured olive flounder, Paralichthys olivaceus. Serum concentrations of these antimicrobials were determined after oral administration of a single dosage of 60 mg/kg body weight (average 700 g). At $23{\pm}1.5^{\circ}C$, the peak serum concentrations of NA, PA and OA, which attained at 10 h, 24 h and 30 h post-dose, were 11.55, 3.79 and $1.12{\mu}g/m\ell$, respectively. At $13{\pm}1.5^{\circ}C$, the peak serum concentrations of NA, PA and OA, which attained at 10 h, 15 h and 30 h post-dose, were 6.36, 1.4 and $1.01{\mu}g/m\ell$, respectively. Better absorption of NA and PA was noted at $23{\pm}1.5^{\circ}C$ compared to $23{\pm}13^{\circ}C$. The elimination of NA from serum of olive flounder was considerably faster at $23{\pm}1.5^{\circ}C$ than at $13{\pm}1.5^{\circ}C$. However, both absorption and elimination of OA were not affected significantly by temperature. The kinetic profile of absorption, distribution and elimination of these antimicrobials in serum were analyzed by fitting to a one- and two compartment model, with WinNonlin program. In the one compartment model for NA, AUC, Tmax and Cmax at $23{\pm}1.5^{\circ}C$ were $258.26{\mu}g{\cdot}h/m\ell$, 10.67 h and $8.91{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $C_{max}$ at $13{\pm}1.5^{\circ}C$ were $341.45 {\mu}g{\cdot}h/m\ell$, 7.72 h and $6.23{\mu}g/m\ell$, respectively. In the one compartment model for PA, AUC, $T_{max}$ and $C_{max}$ at $23{\pm}1.5^{\circ}C$ were $248.12{\mu}g{\cdot}h/m\ell$, 21.15 h and $3.09{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $C_{max}$ at $13{\pm}1.5^{\circ}C$ were $103.89{\mu}g{\cdot}h/m\ell$, 12.89 h and $1.22{\mu}g/m\ell$, respectively. In the two compartment model for OA, AUC, $T_{max}$ and $C_{max}$ at $23{\pm}1.5^{\circ}C$ were $138.20{\mu}g{\cdot}h/m\ell$, 23.95 h and $1.06{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $T_{max}$ at $13{\pm}1.5^{\circ}C$ were $159.10{\mu}g{\cdot}h/m\ell$, 28.03 h and $1.02{\mu}g/m\ell$, respectively.

Antimicrobial Activity of Florfenicol Against Fish Pathogenic Bacteria and Pharmacokinetics in Blood of Cultured Olive Flounder by Oral Administration (Florfenicol의 어류 병원성 세균에 대한 항균 효과와 경구 투여후 넙치 혈액에서의 약물동태학적 특성)

  • The pharmacokinetics of florfenicol (FF) after oral administration was studied in the cultured olive flounder, Paralichthys olivaceus, using high performance liquid chromatography (HPLC). After single administration of FF (20 mg/kg body weight) by oral route in olive flounder ($700{\pm}50$ g, $23{\pm}1.5^{\circ}C$), the concentration in the serum was determined at 1, 5, 10, 15, 24, 30, 50 and 168 h post-dose. The kinetic profile of absorption, distribution and elimination of FF in serum were analyzed fitting to a two-compartment model by WinNonlin program. The area under the concentration-time curve (AUC), maximum concentration ($C_{max}$), time for maximum concentration ($T_{max}$) and elimination time were 22.51 ${\mu}g{\cdot}h/mL$, 0.84 ${\mu}g/mL$, 8.62 h and 447 h, respectively. The results of this study related to dosage and withdrawal times could be used for prescription of FF in field for the treatment of bacterial diseases in olive flounder.

  • This study investigates a simple mathematical model for the quantitative estimation of regional myocardial blood flow in experimental canine coronary artery thrombosis using Rb-82 dynamic myocardial positron emission tomography. The coronary thrombosis was induced using the new catheter technique by narrowing the lumen of coronary vessel gradually, which finally led to partial obstruction of coronary artery. Ten Rb-82 dynamic myocardial PET scans were performed sequentially for each experiment using our 5, 10 and 20 second acquisition protocol, respectively, and three regions of interest were drawn on the transaxial slices, one on left ventricular chamber for input function and the other two on normal and decreased perfusion segments for the flow estimation in those regions. Single compartment model has been applied to the measured sets of regional PET data, and the rate constants of influx to myocardial tissue were calculated for regional myocardial flow estimates with the three parameter fits of raw data by the Levenberg-Marquardt method. The results showed that, (1) single compartment model suggested by Kety-Schmidt could be used for the simple estimation of regional myocardial blood flow, (2) the calculated regional myocardial blood flow estimates were dependent on the selection of input function, which reflected partial volume effect and left ventricular wall motion, and (3) mathematically fitted input and tissue time activity curves were more suitable than the direct application of the measured data in terms of convergence.

  • We used rats as the experimental animal for the elucidation of metabolic patterns and pharmacokinetic profiles of SMZ in the rat, by use of the urine and plasma from predetermined intervals, respectively. Information herefrom would give some insight into species differences and sex differences in the metabolism and pharamcokinetics of drugs, at least SMZ in particular. Results would be summarized as follows: 1. There were two hydorxy metabolites(5-hydroxysulfamethazine and 6-hydroxyethylsulfamethazine) and an acetyl derivative($N_4$-acetyl sulfamethazine) in the 24h-collected urine, on confirmation with each standard materials. There were also two unknown metabolites therein. 2. In the viewpoint of quantitative aspect, $N_4$-acetylsulfamethazine was the largest, hence it is assumed that the acetyl pathway is the major one in the metabolism of SMZ in the rat. 3. As regards sex difference in the rat, the male had more metabolic capacity than the female in metabolism of SMZ. 4. The concenteration-time curves of sulfamethazine(20mg/kg, po) in the plasma compartment were fitted to a one-compartment open model by use of a computer program(NONLIN). 5. There were significant differences(P<0.05) in the pharmacokinetics of sulfamethazine between two sexes in the rat, with higher disposition rate in the male. 6. The emergence of $N_4AcSMZ$ metabolized from SMZ was fast in the plasma of the rat. Half-life of $N_4AcSMZ$ was also. significantly different(P<0.05) between two sexes, suggesting differences in the eliminatory capacity of $N_4AcSMZ$.

Prediction Performance of FDS on the Carbon Monoxide Production in the Under-Ventilated Fires (환기부족 화재에서 일산화탄소 발생에 대한 FDS의 예측성능)

  • In the present study, a numerical simulation was conducted to estimate the prediction performance of FDS on the carbon monoxide production in the under-ventilated compartment fires. Methane and heptane fires located in the a 2/5 scale compartment based on the ISO-9705 standard room was simulated using FDS Ver. 5.5. Through the comparison between the computed results and the earlier published experimental data, the performance of FDS was estimated on the predictions of the combustion gases concentration in the hot upper layer of the compartment and the effects of CO yield rate on the estimation of CO production at local points were analyzed. From the results, it was known that FDS Ver. 5.5, in which the two-step reaction mixture fraction model implemented, was more effective on the prediction of CO concentration compared to the previous FDS version. In addition, controlling CO yield rate made the predicted CO concentration get closer to the experimental data for the fires of the under-ventilated condition.

Comparison of Estimation Methods in NONMEM 7.2: Application to a Real Clinical Trial Dataset (실제 임상 데이터를 이용한 NONMEM 7.2에 도입된 추정법 비교 연구)

  • Purpose: This study compared the performance of new NONMEM estimation methods using a population analysis dataset collected from a clinical study that consisted of 40 individuals and 567 observations after a single oral dose of glimepiride. Method: The NONMEM 7.2 estimation methods tested were first-order conditional estimation with interaction (FOCEI), importance sampling (IMP), importance sampling assisted by mode a posteriori (IMPMAP), iterative two stage (ITS), stochastic approximation expectation-maximization (SAEM), and Markov chain Monte Carlo Bayesian (BAYES) using a two-compartment open model. Results: The parameters estimated by IMP, IMPMAP, ITS, SAEM, and BAYES were similar to those estimated using FOCEI, and the objective function value (OFV) for diagnosing the model criteria was significantly decreased in FOCEI, IMPMAP, SAEM, and BAYES in comparison with IMP. Parameter precision in terms of the estimated standard error was estimated precisely with FOCEI, IMP, IMPMAP, and BAYES. The run time for the model analysis was shortest with BAYES. Conclusion: In conclusion, the new estimation methods in NONMEM 7.2 performed similarly in terms of parameter estimation, but the results in terms of parameter precision and model run times using BAYES were most suitable for analyzing this dataset.

Risk Assessment of Indoor Pollution by BTEX Released from Groundwater (지하수내 BTEX에 의한 실내오염시 위해도 평가)

  • Benzene, Ethyl-benzene, Toluene and Xylene (BTEX) can be released to a groundwater in case of the oil leakage from underground storage tank of a gas station. These chemicals are found to contribute to the total inhalation risk from contaminated indoor air. This study presents the assessment of a human exposure to such chemicals released from the groundwater into indoor air. At first, a 2-compartment model is developed to describe the transfer and distribution of the chemicals released from groundwater in a house through showering, washing clothes, and flushing toilets. The model is used to estimate a daily human exposure through inhalation of such BTEX for adults based on two sets of exposure scenarios. Finally, a sensitivity analysis is used to identify important parameters. The results obtained from the study would help to increase the understanding of risk assessment issues associated with the indoor pollution by BTEX released from contaminated groundwater.

  • A report by the National Research Council in the United States suggested that many lung cancer deaths each year are associated with breathing radon in indoor air. Most of the indoor radon comes directly from soil beneath the basement of foundation. Recently, radon released from groundwater is found to contribute to the total inhalation risk from indoor air. This study presents the assessment of a exposure to radon released from the groundwater into indoor air. At first, a 3-compartment model is describe the transfer and distribution if radon released from groundwater in a house through showering, washing clothes, and flushing toilets. The model is used to estimate a daily human exposure through inhalation of such radon for adults based on two sets of exposure scenarios, Finally, a sensitivity analysis is used to identify important parameters. The results obtained from the study would help to increase the understanding of risk assessment issues associated with the indoor radon released from groundwater.

NOISE REDUCTION OF AN ENCLOSED CAVITY BY MEANS OF AIR-GAP SYSTEMS

  • The objective of this paper is to introduce the noise reduction characteristics of a double gap system, which is composed of two air-gaps and two partition sheets. The resonance of acoustic modes of an enclosed cavity can be effectively suppressed by installing the double gap system in the cavity. It is revealed from a simple, one-dimensional model that the double gap system is more effective than the single gap system that consists of one air-gap and one partition sheet, in that the former requires a smaller space than the latter. Finally, these theoretical conclusions are verified by comparison experiments using an actually manufactured enclosed cavity, of which the boundary surfaces are made of thick panels that can be assumed as rigid walls.

  • Removal of urea and vitamin B12 was simulated using two pool models with closed loop dialysis, open loop dialysis, and hemodiafiltrations with predilution and postdilulion. It was found that urea removal was limited by dialyzer clearance, however B12 removal was limited both by the resistance between ECF and ICF and by dialyzer clearance. Open loop dialysis was better than closed loop dialysis in terms of dialysate requirement and removal efficiency. Residual renal function plays an important role in removing vitamin B12. Dialysis frequency more than twice/wide does not have great effect on removal efficiency, but has the effect of reducing the difference between maximum and minimum cancentrations during dialysis period.
