HCl, Surfactant (SDS, Tween 80) Enhanced Remediation of Contaminated Soil with Lead and Copper (계면활성제 (SDS, Tween 80)와 HCl을 활용한 중금속 오염 토양의 복원)

  • Soils collected from an agricultural field in Youngdong, Chungbuk province were intentionally: contaminated with lead and copper. The efficiencies of soil washing with HCl, SDS and Tween 80 were investigated through the column mode experiments. Washing with 0.1 N HCl obtained the best result for lead and copper removal (95.04 %, 95.94 %). In case of SDS, lead and copper removal rate was such poor as 7.1 % and copper was 24.04 %, respectively. Meanwhile, washing of contaminated soil with Tween 80, did not show any significant removal effect. It was found that the washing efficiency was dependent on pH of washing agent.

Effects of Surfactant Tween 80 on Enzymatic Accessibility and Degradation of Orchardgrass (Dactylis glomerata L.) at Different Growth Stages

  • The study evaluates the enzymatic dry matter (DM) degradability and water holding capacity of leaf and stem fractions of orchardgrass (Dactylis glomerata L.) at different growth stages with or without the presence of surfactant Tween 80. While Tween 80 significantly (p<0.05) increased water and enzyme holding capacities in the leaf blades fraction, less was observed in the fraction of leaf sheath and stem of orchardgrass. The enzyme holding capacity in the leaves was also altered more than that for water holding capacity. This resulted in the increased rate and extent of enzymatic hydrolysis of the leaf blade fractions at two growth stages, whereas little was with leaf sheath and stem fractions. It was also observed that at 0.005% concentrations of Tween 80 the enzymatic DM degradability of young leaf blades was higher (p<0.05) by 20-30% compared to that of the control, as well as for water and enzyme holding capacity. For matured leaf blades the DM degradability were increased with over 0.01% concentrations of the surfactant, but the increase was less than leaf blades of young orchardgrass. This result suggests the possibility of using the surfactant Tween 80 to improve forage digestibility in the rumen.

  • The effects of changing aqueous solution properties by nonionic surfactants on wetting behavior and water retention properies of hydrophobic PET (polyethylene terephthalate) fabric were reported. The aqueous solution properties were diversified by mixing and diluting two nonionic surfactants, i.e., sorbitan monolaurate (Span 20) and polyoxyethylene(20) sorbitan monolaurate (Tween 20). The surface wetting properties ($cos{\theta}$) of PET fabric were greatly improved by adding $10^{-1}g/dl$ Tween 20 and further improved by mixing Span 20 to the system. The water retention properties (W) of PET fabric were also greatly increased by addition of $10^{-1}g/dl$ Tween 20. In diluted surfactant systems, the $cos{\theta}'s$ were increased with decreasing surface tension of aqueous liquids. The ratios of aqueous liquid retained in the pore structure to liquid retention capacity (W/H) were also increased with decreasing surface tension, however, W/H values were dramatically increased right after critical micelle concentration (cmc). The existence of micelles was important for the retention of aqueous liquids in the fabric. The critical surface tension of PET fabric used was found to be 28.7dyne/cm.

Effects of Non-ionic Surfactant Tween 80 on the in vitro Gas Production, Dry Matter Digestibility, Enzyme Activity and Microbial Growth Rate by Rumen Mixed Microorganisms (비이온성 계면활성제 Tween 80의 첨가가 반추위 혼합 미생물에 의한 in vitro 가스발생량, 건물소화율, 효소활력 및 미생물 성장율에 미치는 영향)

  • The non-ionic surfactant (NIS) Tween 80 was evaluated for its ability to influence invitro cumulative gas production, dry matter digestibility, cellulolytic enzyme activities, anaerobic microbial growth rates, and adhesion to substrates by mixed rumen microorganisms on rice straw, alfalfa hay, cellulose filter paper and tall fescue hay. The addition of NIS Tween 80 at a level of 0.05% increased significantly (P<0.05) in vitro DM digestibility, cumulative gas production, microbial growth rate and cellulolytic enzyme activity from all of substrates used in this study. In vitro cumulative gas production from the NIS-treated substrates; rice straw, alfalfa hay, filter paper and tall fescue hay was significantly (P<0.05) improved by 274.8, 235.2, 231.1 and 719.5% compared with the control, when substrates were incubated for 48 hr in vitro. The addition of 0.05% NIS Tween 80 to cultures growing on alfalfa hay resulted in a significant increase in CMCase (38.1%), xylanase (121.4%), Avicelase (not changed) and amylase (38.2%) activities after 36 h incubation. These results indicated that the addition of 0.05% Tween 80 could greatly stimulate the release of some kinds of cellulolytic enzymes without decreasing cell growth rate in contrast to trends reported with aerobic microorganism. Our SEM observation showed that NIS Tween. 80 did not influence the microbial adhesion to substrates used in the study. Present data clearly show that improved gas production, DM digestibility and cellulolytic enzyme activity by Tween 80 is not due to increased bacterial adhesion on the substrates.

  • This study is to investigate the effect of torrefaction on enzymatic hydrolysis of lignocellulosic biomass for bio-ethanol production. As a pretreatment, the torrefaction of lignocellulosic biomass was conducted in temperature of $250{\sim}350^{\circ}C$ in the absence of oxygen. Tween-80, nonionic surfactant, was tested to enhance saccharification efficiency by coping with hydrophobicity resulted from torrefaction. As a result, the glucose production from enzymatic hydrolysis of biomass pretreated by torrefaction was greater than that obtained from the non-pretreated biomass. Sugar conversion was higher when the biomass was saccharified with addition of tween-80. It was found that torrefaction can be applied as a preptreatment for lignocellulosic biomass and tween-80 is needed to enhance its enzyme saccharification.

Effects of Surfactant Tween 80 on Forage Degradability and Microbial Growth on the In vitro Rumen Mixed and Pure Cultures

  • Effect of a surfactant Tween 80 on the bacterial growth in the rumen was examined on the in vitro pure cultures of Streptococcus bovis, Selenomonas ruminantium, Butyrivibrio fibrisolvens, Prevotella ruminicola, Megasphaera elsidenni, Fibrobacta succinogenes, Ruminanococcus albus and Ruminococcus flavefaciens. Dry matter degradability (DMD), concentrations and compositions of volatile fatty acids (VFA), and the most probable number (MPN) of cellulolytic bacteria and total number of bacteria in the presence of Tween 80 were also examined on the in vitro rumen mixed culture either with barley grain or orchardgrass hay. The growth of S. bovis, S. ruminantium, B. fibrisolvens, P. ruminicola, M. elsidenni and F. succinogenes were significantly higher (p<0.05) at over 0.05% concentrations of Tween 80 than those of the control cultures, while was not changed with R. albus and R. flavefaciens. With rumen mixed culture the DMD of barley grain and orchardgrass hay was significantly higher (p<0.05) at a 0.2% concentration of Tween 80 than the control, being reflected in the significantly higher (p<0.05) VFA production (mmol $g^{-1}$DDM) with orchardgrass hay. The higher (p<0.05) ratio of propionate to acetate at a 0.2% concentration of Tween 80 was also observed with orchardgrass hay, showing a similar trend with barley grain. No changes in the total bacterial number and MPN of cellulolytic bacteria were observed.

Effects of Mixtures of Tween80 and Cellulolytic Enzymes on Nutrient Digestion and Cellulolytic Bacterial Adhesion

  • A series of in vitro and in vivo experiments were conducted to investigate the effects of the mixture of Tween 80 and cellulolytic enzymes (xylanase and cellulase) on total tract nutrient digestibility and rumen cellulolytic bacterial adhesion rates in Holstein steers. Ground timothy hay sprayed with various levels of Tween 80 and cellulolytic enzymes was used as substrates in an in vitro experiment to find out the best combinations for DM degradation. The application level of 2.5% (v/w) Tween 80 and the combination of 5 U xylanase and 2.5 U cellulase per gram of ground timothy hay (DM basis) resulted in the highest in vitro dry matter degradation rate (p<0.05). Feeding the same timothy hay to Holstein steers also improved in vivo nutrient (DM, CP, CF, NDF and ADF) digesibilities compared to non-treated hay (p<0.05). Moreover, Tween 80 and enzyme combination treatment increased total ruminal VFA and concentrations of propionic acid and isovaleric acid with decreased acetate to propionate ratio (p<0.001). However, adhesion rates of Fibrobacter succinogenes and Ruminococcus flavefaciens determined by Real Time PCR were not influenced by the treatment while that of Ruminococcus albus was decreased (p<0.05). The present results indicate that a mixture of Tween 80 and cellulolytic enzymes can improve rumen environment and feed digestibility with variable influence on cellulolytic bacterial adhesion on feed.

  • C6-Benzylamino purine (BAP) 400 ppm with 500 ppm Spreader, 500 ppm Cover, 250 ppm Spreadersticker, 350 ppm Siloxane, or 1% Tween 20 was sprayed and the effect of surface-active agents on the feathering of maiden apple trees was evaluated. Only Tween 20 showed additional effect on the feathering in comparison with the single BAP treatment. In the second year different concentrations of Tween 20 were added to 400 ppm BAP. Each BAP treatment added with 0.2, 0.5, 0.8, or 1.0% Tween 20 was more effective in feathering than BAP 400ppm alone, but no significant differences among the concentrations.

Production of Tween 80-inducing Esterase by Acinetobacter sp. B1 Using Response Surface Methodology

  • Esterase produced by Acinetobacter sp. B1 (strain B1) was optimized by means of one-variable-at-a-time and response surface methodologies. Results of the one-variable-at-a-time experiment showed that Tween 80 significantly increased esterase production of strain B1. The addition of Tween 80 to the culture medium increased the biomass and esterase activity of strain B1, stimulated content of total extracellular protein, and enhanced the oleic acid (C18:1) composition in the cell membrane of strain B1. The influence of eight culture variables on esterase production was evaluated by Plackett-Burman design. Results showed that Tween 80, pH, and $K_2HPO_4$ significantly affected the esterase production of strain B1. Tween 80, pH, and $K_2HPO_4$ were further optimized by central composite design. Under the optimized conditions (w/v, soluble starch 2.5%, tryptone 1.5%, Tween 80 0.8%, $K_2HPO_4$ 0.5%, NaCl 0.5%, pH 8.0, inoculum size 1%, and inoculum age 24 h), the maximum esterase activity of strain B1 was 152.13 U/ml, which was 10-fold higher than that of non-optimization after 36 h cultivation.

Treatability of Heavy Metals in the Washing Technology of Marine Sediments Contaminated with Organic Matter (세척기반처리에 의한 해양오염퇴적물에 함유된 유기 오염물질 제거 공정 중 중금속 처리 가능성)

  • Treatability of heavy metals in marine sediments contaminated with mainly organic matter was investigated on the basis of washing technology using oxidizers and surfactants. Sediment samples were collected at N area which expected for remediation project of contaminated marine sediment. For additives, hydrogen peroxide ($H_2O_2$) and Tween-80 were used at oxidizer and nonionic surfactant, respectively. In experiments, sediments was mixed with sea water at the ratio of 1 : 3 than $H_2O_2$ (1 M, 3 M, 4 M, 5 M) and Tween-80 (0.05%) were added. Samples were gathered at following reaction time (10, 20, 30, 40, 50, 60, 70, 80 min and 24 h). Total Organic Carbon (TOC) was 55.2% at the conditions of 5 M $H_2O_2+0.05%$ Tween-80 24 h. Hence total heavy metals were Cu 29.5%, Zn 42.3%, Cd 73.0% and bioavailable heavy metals were Cu 60.0%, Zn 77.7%, Cd 90.2% at the conditions of 5 M $H_2O_2+0.05%$ Tween-80 10 min. The correlations for between bioavailable metals (Cu, Zn, Cd) and TOC were significant (Cu, Zn, Cd; $r^2=0.94$, 0.85, 0.69, respectively).
