The Anti-tumor Effect of Bojungikkeehapdaechilkitang with Doxorubicin in Colon-26 (보중익기합대칠기탕과 Doxorubicin의 병용이 Colon-26의 항암효과에 미치는 영향)

  • Objectives : To evaluate the anti-tumor and synergic effect of Bojungikkeehapdaechilki-tang (BJDC) with doxorubicin. Methods : The inhibitory concentration (IC), $IC_{50}{\;}and{\;}IC_{90}$ of single use of doxorubicin and BIDC with their concomitant treatment against Colon-26 (murine rectum carcinoma) cell line were observed using MTT (microculture tetrazolium test) assay. In addition, their anti-tumor effects were also observed in xenograft nude mice models against Colon-26 cell line. Results : BJDC had only minimal direct anti-tumor effect against Colon-26 cell line but it reduced general depressed signs induced by implantation of the tumor cell lines and increased the total WBC and lymphocyte numbers. Conclusions : It is considered or expected that BJDC extract is reducing the critical toxicity of doxorubicin and has favorable synergic anti-tumor effect when administered conconitently with doxorubicin.

Growth Factor Receptor Expression on Brain Tumor Cell Lines : Preliminary Study for in vitro and in vivo Experiments of Immunotoxin Therapy (뇌종양세포주에서의 성장인자수용체의 발현 : 면역독소 치료의 연구를 위한 예비실험)

  • Objective : Growth factor receptors on the tumor cells are known to be expressed highly allowing the tumor cells to bind growth factors to stimulate cellular division. Immunotoxin therapy is one of the novel approaches to the primary malignant brain tumor, and expression of cell-surface receptor is essential for the immunotoxin to have specific anti-tumor activity. Despite promising cytotoxic activity of immunotoxin, tumor responses are not curative on clinical trials, and additional studies are needed regarding various factors influencing the efficacy of the immunotoxin. The purpose of this study is to detect the expression of various growth factor receptors on brain tumor cell lines which are going to be used in these studies. Materials and Methods : The authors detected transferrin receptor(TR), insulin-like growth factor-1 receptor(IGF-1R), and interleukin-4 receptor(IL-4R) on medulloblastoma cell line(Daoy) and glioblastoma cell lines(U373 MG and T98 G) by flow cytometric analysis. Results : TR was expressed on Daoy, U373 MG, and T98 G. IGF-1R was expressed on Daoy and U373 MG, but not on T98 G. IL-4R was expressed on all cell lines tested. Conclusion : The transferrin and interleukin-4 receptors might be good targets for immunotoxin therapy. The results should be considered in additional in vitro and in vivo studies regarding immunotoxin and in establishing the proper treatment model of the immunotoxin therapy including selection of the adequate immunotoxin.

  • Isoguanosine and berberine 1:1 mixture[l:[1(mole:mole)] has been prepared and evaluated by measuring antitumor effects against various tumor cell lines in culture and in mice. We reported that the synergistic effect of isoguanosine and berberine mixture has been revealed compared with each of isoguanosine and berberine increased by $3{\sim}8$ times than that of each components in various tumor cell lines in vitro. The most effective dose of isoguanosine and berberine mixture was 60 mg/kg/day in mice bearing S-180 solid tumor, the %(1-T/C) values were 70%. Against the P-388 leukemia, isoguanosine-berberine mixture was the most effective at the dose of 60 mg/kg/day, the %T/C values were 163%.

Current status and clinical application of patient-derived tumor organoid model in kidney and prostate cancers

  • Urological cancers such as kidney, bladder, prostate, and testicular cancers are the most common types of cancers worldwide with high mortality and morbidity. To date, traditional cell lines and animal models have been broadly used to study pre-clinical applications and underlying molecular mechanisms of urological cancers. However, they cannot reflect biological phenotypes of real tissues and clinical diversities of urological cancers in vitro system. In vitro models cannot be utilized to reflect the tumor microenvironment or heterogeneity. Cancer organoids in three-dimensional culture have emerged as a promising platform for simulating tumor microenvironment and revealing heterogeneity. In this review, we summarize recent advances in prostate and kidney cancer organoids regarding culture conditions, advantages, and applications of these cancer organoids.

Cytokine Induction of Intercellular Adhesion Molecule-1(ICAM-1) Expression on Human Glioblastoma Cell Line, U-251 MG, U-373 MG (교모세포종 U-251MG, U-373MG세포주의 Cytokines처리에 의한 세포내 ICAM-1 발현)

  • Objective : Despite advances in the understanding of tumor biology and the tumor immunology, there has been no effective treatment. The Intercellular adhesion molecule-1(ICAM-1) has been shown to be important in interaction involving cells of the immune system and to be upregulated in a number of cell culture systems by cytokines, including immune interferon($IFN-{\gamma}$) and tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$). ICAM-1 has been identified as one of the ligands for lymphocyte function-associated antigen-1(LFA-1). The effectiveness of various cytokines to ICAM-1 induction on cultured human glioblastoma cell lines and potential efficacy of immunotherapy were studied. Method : Human glioblastoma cell lines, U-251 MG, U-373 MG were trypsinized and suspended at $1{\times}10^5cells/ml$ and grown on 8 well chamber slide, the cells were incubated in 0.3ml medium alone or medium containing $IFN-{\gamma}$(1000U/ml) or $TNF-{\alpha}$(250U/ml) or $IFN-{\gamma}$ plus $TNF-{\alpha}$ for 6, 12, 24, 48 and 72 hours. The coverslip were then removed and stained with a 1/30 dilution of anti-ICAM-1 antibody. Result : Surface antigen expression of ICAM-1 was increased by incubating glioblastoma cell lines with $IFN-{\gamma}$ and $TNF-{\alpha}$. Combined effect of $IFN-{\gamma}$ and $TNF-{\alpha}$ has induced more ICAM-1 expression on glioblastoma cell lines. Upregulation of ICAM-1 expression in an established glioblastoma cell line was of greater magnitude and more rapid following incubation with $IFN-{\gamma}$ plus $TNF-{\alpha}$. Surface antigen expression of ICAM-1 was increased for up to 48 hours after cytokine treatment on both cell lines(p<0.05). There was no difference on both cell lines(p>0.05). Conclusion : The results of the present study indicate that ICAM-1 expression in glioblastoma cell lines, U-251 MG and U-373 MG, are induced and enhanced after treatment with $IFN-{\gamma}$ and $TNF-{\alpha}$. Combined effect of $IFN-{\alpha}$ and $TNF-{\gamma}$ is stronger and more rapid than $IFN-{\gamma}$ or $TNF-{\alpha}$ alone.

  • Methanolic and aqueous extracts of 26 red algae species collected from Jeju Island coast were prepared at a high $(70^{\circ}C)$ and a room temperature $(20^{\circ}C)$ and were examined for their cytotoxic activity against 4 tumor cell lines: U-937 (human monoblastoid leukemia cell line), HL-60 (human promyelocytic leukemia cell line), B-16 (murine melanoma cell line) and HeLa (woman cervical carcinoma cell line). $20^{\circ}C$ methanolic extract of Polysiphonia japonica showed cytotoxic activity of over 50% against U-937, HL-60 and B-16 cells. On the other hand, the $20^{\circ}C$ aqueous extract of Scinaia okamurae and $70^{\circ}C$ aqueous extract of Chondrus crispus showed cell growth inhibition activity of more than 50% against HL-60 and B-16 cells. The highest cytotoxic activity was observed in the $20^{\circ}C$ aqueous extract of Scinaia okamurae against B-16 cells (80.55%).

  • Naesosan(NSS) has been used in Oriental Medicine as a drug that treated carbuncle and cellulitis. So, the purpose of this Study was to investigate effects of NSS on the cytotoxicity of cancer cell lines and lymphocytes in vitro, proliferation of Ll210 cells and lymphocytes in L1210 cells transplanted mice, improvement of blood count in Ll210 cells transplanted mice, tumor weight and body weight in sarcoma-180 cells transplanted mice, survival prolongation in sarcoma-180 cells transplanted mice. We used NSS extract with freeze-dried, 8wks-old male mice(balb/c and ICR mouse $18{\pm}2g$). Ll210 cell lines, and sarcoma-180 cell lines for this Study, The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results of this Study were obtained as follows ; 1. NSS showed significantly cytotoxicitic effects of cancer cell lines, did not show cytotoxicitic effects of lymphocytes. 2, Proliferation of lymphocytes in L1210 cells transplanted mice did not effects by NSS. 3. NSS inhibited significantly the proliferation of L1210 cells in L1210 cells transplanted mice. 4. NSS improved significantly the blood count in Ll210 cells transplanted mice. 5. NSS increased significantly th body weight in sarcoma-180 cells transplanted mice. 6. NSS dereased significantly the tumor weight in sarcoma-180 cells transplanted mice. 7. NSS prolonged significantly the survival time in sarcoma-180 cells transplanted mice.

Ellagic Acid Inhibits Migration and Invasion by Prostate Cancer Cell Lines

  • Polyphenolic compounds from pomegranate fruit extracts (PFEs) have been reported to possess antiproliferative, pro-apoptotic, anti-inflammatory and anti-invasion effects in prostate and other cancers. However, the mechanisms responsible for the inhibition of cancer invasion remain to be clarified. In the present study, we investigated anti-invasive effects of ellagic acid (EA) in androgen-independent human (PC-3) and rat (PLS10) prostate cancer cell lines in vitro. The results indicated that non-toxic concentrations of EA significantly inhibited the motility and invasion of cells examined in migration and invasion assays. The EA treatment slightly decreased secretion of matrix metalloproteinase (MMP)-2 but not MMP-9 from both cell lines. We further found that EA significantly reduced proteolytic activity of collagenase/gelatinase secreted from the PLS-10 cell line. Collagenase IV activity was also concentration-dependently inhibited by EA. These results demonstrated that EA has an ability to inhibit invasive potential of prostate cancer cells through action on protease activity.

Expression of $O^6$-methylguanine-DNA methyltransferase and Sensitivity to Anticancer Alkylating Agents in Human Cancer Cells (사람 암세포에서의 $O^6$-methylguanine-DNA methyltransferase의 발현과 알킬화 항암제에 대한 감수성)

  • Five human cancer cell lines (HeLa S3, Hep 3B, KATO III, Hs 683, HeLa MR) and one human normal cell line (WI-38) were examined cell viability, northern blot analysis, western blot analysis, and in situ hybridization for the expression $O_{6}$ -methylguanine-DNAmethyltransferase (MGMT), which can repair $O_{6}$ -methylguanine produced in DNA by alkylating agents. In cell viability test, the lethal sensitivities of each strain against anti-tumor drug N,N-bis(2-chloroethyl)- N-nitrosourea (BCNU) were counted, and both BCNU treated and untreated cell extracts were examined for their MGMT inducibility by RNA dot blot analysis. Cell lines did not show MGMT induction by BCNU pretreatment. Tlle MGMT activity was assayed by measuring the $^3$H radioactivity transferred from the substrate DNA containing [methyl-$^3$H)-O$_{6}$ -methylguanine to acceptor molecules in the cell extracts. Extracts from the majority of tumor strains and normal cells contained substantial MGMT activity of varying degree, while the known Mer$^{[-10]}$ cell (lacked or severely depleted in MGMT activity) Hela MR, and Hs 683 (proved to be Mer$^{[-10]}$ ) were much more sensitive to BCNU than the rest of tumor strains, as measured by cell viability test. Overall results above, KATO III showed the highest expression level of MGMT among the strains examined. Furthermore, with all the tumor and normal strains tested, a good correlation was observed between MGMT expression and cellular resistance to BCNU. The varying levels of expression of MGMT in human cancer cells found in this study should provide a molecular basis for MGMT expression among tumor strains from different tissue origin, the information of antitumor agents selection for chemotherapy of cancers.

Scientific Analysis of Formulation Theory of Chungpesagan-tang; In vitro Cytotoxicity of Cisplatin Combined with Chungpesagan-tang

  • In vitro cytotoxic activities of cisplatin combined with Chungpesagan-tang or puerarin, which were treated with or without human intestinal bacteria, were measured. When cisplatin was combined with Chungpesagan-tang and its ingredient treated without intestinal bacteria, they did not affect the in vitro cytotoxicity of cisplatin against tumor cell lines. However, when cisplatin was combined with intestinal bacteria-treated Chungpesagan-tang and its ingredients, the cytotoxicities against SNU C4, L1210, A549 and P388 tumor cell lines were synergistically increased. Puerarin, which was isolated from Puerariae Radix, did not show in vitro cytotoxicity. However, its metabolite, daidzein, showed potent cytotoxicity against tumor cell lines and was synergistic by the combined usage of cisplatin. These results suggest that natural glycosides are not only prodrugs which can be transformed to active compounds by intestinal microflora, but the combined usage of cisplatin with natural components, such as daidzein, and herbal medicinal polyprescriptions, such as Chungpesagan-tang, may be a new method for prevention and minimization of the toxicity of cisplatin.
