Atypical Granular Cell Tumor of the Sellar Region

  • We report a very rare case of atypical granular cell tumor arising in the neurohypophysis of a 56-year-old woman. The tumor was seen on radiology to be lobulated, soft and diffusely enhanced, the same as pituitary macroadenoma, but it was anatomically localized within the posterior part of the hypophysis. We partially removed the tumor via the transsphenoidal approach. The pathology showed nuclear pleomorphism, spindling features, and lymphoplasmacytic infiltration. Ki-67 and S-100 protein were focally positive in tumor cells. Histological diagnosis confirmed an atypical granular cell tumor in the sellar region, which is a rare tumor that often has the clinical appearance of a pituitary adenoma. Ophthalmologic symptoms are the most common, followed by endocrinologic manifestations. Here we describe its symptoms and radiological and pathological features.

  • We experienced a case of giant cell tumor arising from the right 2nd rib in the diagnostic and therapeutic thoracotomy at our department of Thoracic and Cardiovascular Surgery, Chungnam Univ. Hospital in May 3rd, 1991. The reports of this tumor are very rare and are known to have the incidence of 1 percent or less of all giant cell bone tumor. The tumor of the right 2nd rib, infant head-sized, covered with parietal pleura was invaded into the vertebra on operation finding. The tumor was resected partially[three fourths] and revealed to be the giant cell tumor on pathologic finding. At now, the patient is taking the radiotherapy to achieve the therapeutic goal to the remnant tumor.

  • Diffuse-type giant cell tumor is relatively rare than localized giant cell tumor. Moreover, diffuse type giant cell tumor is common in intraarticular area, rarely occurs at intramuscular or subcutaneous layer. We experienced 1 case of giant cell tumor within the deltoid muscle. So we report this case with review of the literatures.

  • It is not uncommon for sarcomatous transformation of giant cell tumor of bone to occur after radiation, but osteosarcoma arising from giant cell tumor after surgical treatment is very rare and remains an aggressive form of sarcoma of bone with high mortality rate. We experienced 2 cases in whom a osteosarcoma developed long after benign giant cell tumor of bone was removed surgically from the same site. Malignant transformation was presented at 2 years 1 month and 9 years 8 months each after initial surgery. We describe our experience concerning clinical features, methods of treatment and outcomes of osteosarcoma arising from giant cell tumor.

Anti-Cancer Effects of Salvia Miltiorrhiza, Carydalis Turtschaminovii, Reynoutria Elliptica Herbal Acupuncture on Solid Tumor of Rats induced by Injection of RK3E-ras Cells (RK3E-ras cells로 유발된 흰쥐의 고형종양에 대한 단삼, 현호색, 호장근 약침의 항종양 효과)

  • Objectives : The present study was carried out investigate the anti-cancer effect of Salvia miltiorrhiza, Carydalis turtschaminovii and Reynoutria elliptica herbal acupuncture on solid tumor of rats induced by injection of RK3E-ras cells. Methods : RK3E-ras cells were injected on the right lumbar region of rats. After 1 weeks, the experimental rats were divided into four groups : Control group, Salvia miltiorrhiza herbal acupuncture group(SM), Carydalis turtschaminovii herbal acupuncture group(CT), Reynoutria elliptica herbal acupuncture group(RE). And we investigated the weight and size of tumor tissue, gross anatomy, histological and PCNA immunohistochemical study, hepatic and renal metastasis for tumor of each group. Results : 1. In the weight of tumor tissue assessment, SM and CT's weight of tumor tissue was decreased. 2. In the size of tumor tissue assessment, SM was smaller than any other group. 3. In the histological observation, SM's formation of tunica fibrosa that surround the tumor cell was obvious and vasculature that developes circumference of tumor cell was not observed, and density of tumor cell was very low. 4. In the PCNA immunohistochemical study, Control group, SM, RE showed strong immune response in the central site of tumor tissue. 5. In observation of liver and kidney tissue, we were not able to observe tumor cell in the SM. Conclusions : Consequently, SM and CT showed a inhibition of growth and metastasis.

  • Giant cell tumor is the second most common benign tumor of the bone. But, the incidence in the ribs is rare. Clinical features of giant cell tumor may vary markedly from latent to very aggressive. Recently, we experienced clinically aggressive form of giant cell tumor in the right 2nd rib and report this case with reviewing the articles of giant cell tumor.

Granular Cell Tumor in the Lower Lip : A Case Report (하구순의 과립세포종 증례보고)

  • Purpose : Granular cell tumour(GCT) is a rare benign tumour that can arise in anywhere throughout the body. Histopathological diagnosis of malignancy is difficult. We report a case of a granular cell tumor which developed on lower lip. Methods : A 58-year-old male had a palpable mass and whitish plaque in the oral mucosa for three months. Results : Under local anasthesia, the tumor was removed through wide excision including normal skin margin. Immediate reconstruction using local flap was done. Pathologic observation showed nests of polygonal cells with abundant granular, eosinophilic cytoplasm and round nuclei. Immunohistopathologic staining showed positive reaction on S-100 protein Conclusion : The results support the hypothesis that granular cell tumor is derived from Schwann cells. We report here on a case of granular cell tumor of lower lip that was successfully treated with radical resection.

Exosomal Communication Between the Tumor Microenvironment and Innate Immunity and Its Therapeutic Application

  • Exosomes, which are well-known nanoscale extracellular vesicles, are multifunctional biomaterials derived from endosomes and perform various functions. The exosome is a critical material in cell-cell communication. In addition, it regulates the pathophysiological conditions of the tumor microenvironment in particular. In the tumor microenvironment, exosomes play a controversial role in supporting or killing cancer by conveying biomaterials derived from parent cells. Innate immunity is a crucial component of the host defense mechanism, as it prevents foreign substances, such as viruses and other microbes and tumorigenesis from invading the body. Early in the tumorigenesis process, the innate immunity explicitly recognizes the tumor via Ags and educates the adaptive immunity to eliminate it. Recent studies have revealed that exosomes regulate immunity in the tumor microenvironment. Tumor-derived exosomes regulate immunity against tumor progression and metastasis. Furthermore, tumor-derived exosomes regulate polarization, differentiation, proliferation, and activation of innate immune cells. Exosomes produced from innate immune cells can inhibit or support tumor progression and metastasis via immune cell activation and direct cancer inhibition. In this study, we investigated current knowledge regarding the communication between tumor-derived exosomes and innate immune cell-derived exosomes (from macrophages, dendritic cells, NK cells, and neutrophils) in the tumor microenvironment. In addition, we discussed the potential development of exosomal immunotherapy using native or engineered exosomes against cancer.

The Characteristics of Antitumor Agent Isolated from Streptomyces sp.409 (Streptomyces sp.409 에서 분리한 항암활성 물질의 특징)

  • This study was carried out to find new anti-tumor agent producing microbe and to characterize the anti-tumor agent produced from the microbe. Purified compound that has a high cytotoxicity against tumor cell-lines could be obtained from the broth culture filtrates of Streptomyces sp.409 strain isolated from soil in Korea. The in vitro cytotoxicity the in vivo evaluation of acute toxicity the safety assessment of the anti-tumor compounds and the taxonomic characteristics of the anti-tumor agent were measured. The antitumor compound 1 and 2 were obtained from the broth culture filtrates of Streptomyces sp.409 strain. The cytotoxicity of the compound 1 against tumor cell-line P388D$_1$ showed almost 4.5 times higher than that of adriamycin. However in the cytotoxicity against normal cell line Vero E6, adriamycin showed adversely 4 times higher than the compound 1 ($IC_{50}$/ value: 228.7 $\mu\textrm{g}$/$m\ell$). In comparison study with compound 1 and compound 2 in the in vitro cytotoxin productivity against tumor cell lines, $IC_{50}$/ value of the compound 1 was 0.25 $\mu\textrm{g}$/$m\ell$ in tumor cell line P388D$_1$and 0.53 $\mu\textrm{g}$/$m\ell$ in tumor cell-line L1210, and that of the compound 2 was 7.18 $\mu\textrm{g}$/$m\ell$ and 35.71 $\mu\textrm{g}$/$m\ell$, respectively; LD$_{50}$ value of the compound 1 in the in vivo acute toxicity in mice was 22.62 $\mu\textrm{g}$/kg body weight. These results suggest that compound 1 purified from Streptomyces sp. 409 has anti-tumor activity and will be developed as an anti-tumor drug.g.
