Effect of Superoxide Dismutase on the Release of [$^3H$]-5-Hydroxytrytamine after Hypoxia from Rat Hippocampal Slices (흰쥐 해마 절편에서 저산소증에 의한 [$^3H$-5-Hydroxytrytamine의 유리 변동에 미치는 superoxide dismutase/catalase의 영향)

  • Many factors are known to be responsible for cerebral ischemic injury, such as excitatory neurotransmitters, increased intraneuronal calcium, or disturbance of cellular energy metabolism. Recently, oxygen free radicals, formed during ischemia/reperfusion, have been proposed as one of the main causes of ischemia/reperfusion injury. Therefore, to investigate the role of oxygen free radical during ischemia/reperfusion, in the present study the effect of endogenous oxygen free radical scavenger, superoxide dismutase / catalase(SOD / catalase) on the release of [$^3$H]-5-hydroxytryptamine([$^3$H]-5-HT) during hypoxia/reoxygenation in rat hippocampal slices was measured. The hippocampus was obtained from the rat brain and sliced 400 gm thickness with manual chopper. After 30 min's preincubation in the normal buffer, the slices were incubated for 20 min in a buffer containing [$^3$H]-5-HT(0.1 $\mu$M, 74 $\mu$Ci) for uptake, and washed. To measure the release of [$^3$H]-5-HT into the buffer, the incubation medium was drained off and refilled every ten minutes through a sequence of 14 tubes. Induction of hypoxia for 20 min (gassing it with 95% N$_2$/5% CO$_2$) was done in the 6th and 7th tube, and oxygen free radical scavenger, SOD / catalase was added 10 minutes prior to induction of hypoxia. The radioactivity in each buffer and the tissue were counted using liquid scintillation counter and the results were expressed as a percentage of the total activity. When slices were exposed to hypoxia for 20 min, [$^3$H]-5-HT release was markedly decreased and a rebound release of [$^3$H]-5-HT was observed on the post-hypoxic reoxygenation period. SOD / catalase did not changed the release of [$^3$H]-5-HT in control group, but inhibited the decrease of [$^3$H]-5-HT release in hypoxic period and rebound increase of [$^3$H]-5-HT in reoxygenation period. This result suggest that superoxide anion may play a role in the hypoxic-, and reoxygenation-induced change of [$^3$H]-5-HT release in rat hippocampal slices.

Transmission and Histochemical Detection of Mulberry Dwarf Mycoplasma in Several Herbaceous Plants (뽕나무 오갈병 마이코플라스마의 몇가지 초본식물에의 전염과 조직화학적 검정)

  • Transmission of mulberry dwarf mycoplasma (MDM) from diseased mulberry to 5 herbaceous plants (periwinkle, white clover, Ladino clover, red clover, and Chinese milk vetch) through insect vector, Hishimonus sellatus, was confirmed by symptom expression and microscopic evidences. The earliest symptom appearance was noticed on periwinkle in which incubation period was 25-30 days, while it ranged 35-40 days in the other plant species. The common symptoms of MDM infected plants were characterized by poor plant growth with accompanying discolorations of leaves (chlorosis with vein clearing in periwinkle, reddish in red clover, brownish in white and Ladino clovers, and yellowish in Chinese milk vetch). Mycoplasmal infections were diagnosed light microscopically by Dienes' and toluidine blue staining of hand-cut and Epon-embedded sections, respectively. In Dienes' stain, all the plants infected with MDM showed specific staining reaction in phloems. In toluidine blue stain, mycoplasmal existence was noted by granular appearance in sieve tubes which were confirmed to be mycoplasma-like organisms under an electron microscope.

Initial Subcellular Responses of Susceptible and Resistant Soybeans Infected with the Soybean Cyst Nematode

  • Initial subcellular responses in susceptible (PI 274420) and resistant (cv. Hartwig) soybeans infected with the soybean cyst nematode (SCN) were examined 2 and 4 days after inoculation (DAI). Subcellular features common to both soybeans at 2 DAI included hypertrophied initial syncytial cells (ISCs) and syncytium-component cells (SCs) with a dense cytoplasm containing proliferated rough and smooth endoplasmic reticulum (RER and SER), a hypertrophied nucleolus, and reduced vacuoles, suggesting that the nematode-infected cells were dedifferentiated. In the resistant soybean, a striking initial subcellular difference from the susceptible soybean was the dilation of the RER, indicating ER dysfunction and leading to cell death. This disturbed nematode feeding, as evidenced by disrupted feeding tubes. In PI 274420, the ISC cytoplasm was depleted, with the exception of ER membranes, at 4 DAI, while the SC cytoplasm was dense with proliferation of starch-containing plastids around multiple nuclei that might be derived from the congregation of nuclei in the neighboring SCs and in part by nuclear division without cytokinesis. In cv. Hartwig, syncytia were necrotized with secondary cell wall thickening outside the plasma membrane and an extremely dense cytoplasm containing a nucleus with an electron-lucent nucleolus, accompanied by the proliferation of closely stacked parallel RER and ribosomes. These results suggest that syncytia develop continuously in PI 274420 to produce and store nutritional substances in SCs, providing for the nematode through ISC until maturation, but in cv. Hartwig, syncytia degenerate early due to excessive metabolism, blocking nematode feeding and cytoplasmic connections with adjacent intact cells.

Iatrogenic Large Esophageal Perforation Caused by Sengstaken-Blackmore Tube (식도 정맥류 출현 환자에서 Sengataken-Blackmore관에 의한 의인성 거대 흉부식도 파열)

  • This patient was an 53-year-old man who had undergone Sengstaken-Blackmore tube insertion for esophageal varix bleeding. Two days after Sengstaken-Blackmore tube insertion, he developed severe left hemothorax and was transferred to our hospital. The esophagoscopic findings revealed a large perforation lengthening 8-cm in the intrathoracic esophagus. A left thoracotomy was performed 33 days after the injury due to repeated varix bleedings and poor conditions. An 8-cm longitudinal perforation of the intrathoracic esophagus with gross suppurative empyema was found. Primary repair and esophageal exclusion was performed 2cm proximal and distal to the perforation, using rows of nonabsorbable staplers(TA stapler 60 $\times$ 4.8) and large bore thoracostomy tubes were placed for local drainage. Six days after intrathoracic esophageal exclusion, an esophagogram revealed a leakage at just above the proximal stapling site. A cervical esophageal exclusion was performed using the same method. One hundred thirty seven days after exclusion operation for the intra-thoracic esophageal perforation, the patient was able to eat per orally without any secondary esophageal reconstructive surgery.

Fabrication and Use of Corrosion Defect Specimens for Enhancement of ECT Reliability for Nuclear Steam Generator Tubing (증기발생기 전열관 와전류 검사의 신뢰성 향상을 위한 부식결함 시편의 제작 및 활용)

  • The development of an integrated technology including fabrication of corrosion defect specimens and their practical use is needed to enhance the reliability of eddy current test for nuclear steam generator tubing. In this paper, the necessity and importance are presented from the viewpoint of the structural integrity, simulation specimens for real defects, and experiences from the destructive examination of pulled tubes. The models for several corrosion defects we also briefly introduced, with the scheme for their practical use.

The Effects of Hot Corrosion on the Creep Rupture Properties of Boiler Tube Material (보일러 管材料의 크리프破斷特性에 미치는 고온부식의 影響)

  • In order to investigate the effects of hot corrosion on the creep rupture properties and creep life of 304 stainless steel being used as tube materials of heavy oil fired boiler, the creep rupture tests were carried out at temperature 630.deg.C, 690.deg.C and 750.deg.C in static air for the specimens with or without coating of double layer corrosives according to the new hot corrosion test method simulating the situation commonly observed on superheater tubes of the actual boiler. The double layer corrosives are 85% V$_{2}$O$_{5}$ + 10% Na$_{2}$So$_{4}$ + 5% Fe$_{2}$O$_{3}$ as the inner layer corrosive being once melted at 900.deg. C and crushed to powder, and 10% V$_{2}$O$_{5}$ + 85% Na$_{2}$SO$_{4}$ +5% Fe$_{2}$O$_{3}$ as the outer layer corrosive. As results, in the specimen coated with the double layer corrosives, the rupture strength was extremely lowered and showed a large difference each other. The rupture ductility also lowered remarkably as a result of the brittle fracture mode due to hot corrosion. These results indicate that hot corrosion could essentially alter the creep fracture mechanism. From the metallographic observation, it was clarified that the rupture life of 304 stainless steel subjected to hot corrosion was chiefly determined by the behavior of the aggressive intergranular penetration of sulfides.des.

Evaporation kent transfer characteristics of R-290 and R-600a in the horizontal tubes (수평관내 R-290과 R-600a의 증발 열전달 특성)

  • This paper presents the experimental results of evaporation heat transfer coefficients of HC(e.g. R290 and R600a), R-22 as a HCFCs refrigerant in horizontal double pipe heat exchangers, having four different inner diameters of 10.07 mm and 6.54 mm respectively. The experiments of the evaporation process were conducted at mass flux of $35.5{\sim}210.4\;kg/m^2s$ and cooling capacity of $0.95{\sim}10.1\;kW$. The main results were summarized as follows : The average evaporation heat transfer coefficient of R-290 and R-600a was higher value than that of R-22. In comparison with R-22, the evaporation heat transfer coefficient of R-290 and 600a is approximately $56.7{\sim}70.1$ and $46.6{\sim}59.7%$ higher, respectively. In comparison with experimental data and some correlations, the evaporation heat transfer coefficients are well matched with the Kandlikar's correlation regardless of a type of refrigerants and tube diameters.

Effects of Interrupted Wetness Periods on Conidial Germination, Germ Tube Elongation and Infection Periods of Botryosphaeria dothidea Causing Apple White Rot

  • Responses of Botryosphaeria dothidea to interrupted wetness periods were investigated under in vivo and in vitro conditions. Conidia of B. dothidea were allowed to germinate on apple fruits under wetting condition at $25^{\circ}C$ for 5 hr. They were air-dried for 0, 1, 2 or 4 hr, and then rewetted at $25^{\circ}C$ for 5 hr. Following an initial wetness period of 5 hr, 83% of the conidia germinated. The percent conidial germination increased to 96% when wetting was extended continuously another 5 hr. However, no further conidial germination was observed when wetting was interrupted by dry periods of 1, 2 and 4 hr, resulting in 83, 81 and 82%, respectively. The mean length of the germ tubes was $37{\mu}m$ after 5 hr of wetting and elongated to $157{\mu}m$ after 10 hr of continuous wetting. On the other hand, interruption of wetting by a dry period of 1 hr or longer after the 5 hr of initial wetting arrested the germ tube elongation at approximately $42{\mu}m$ long. Prolonged rewetting up to 40 hr did not restore germ tube elongation on slide glasses under substrate treatments. Model simulation using weather data sets revealed that ending infection periods by a dry period of at least 1 hr decreased the daily infection periods, avoiding the overestimation of infection warning. This information can be incorporated into infection models for scheduling fungicide sprays to control apple white rot with fewer fungicide applications.

Biocontrol Characteristics of Bacillus Species in Suppressing Stem Rot of Grafted Cactus Caused by Bipolaris cactivora

  • One of the most important limiting factors for the production of the grafted cactus in Korea is the qualitative and quantitative yield loss derived from stem rots especially caused by Bipolaris cactivora. This study is aimed to develop microbial control agents useful for the control of the bipolaris stem rot. Two bacteria (GA1-23 and GA4-4) selected out of 943 microbial isolates because of their strong antibiotic activity against B. cactivora were identified as Bacillus subtilis and B. amyloliquefaciens, respectively, by the cultural characteristics, Biolog program and 16S rRNA sequencing analyses. Both bacterial isolates significantly inhibited the conidial germination and mycelial growth of the pathogen with no significant difference between the two, of which the inhibitory efficacies varied depending on the cultural conditions such as temperature, nutritional compositions and concentrations. Light and electron microscopy of the pathogen treated with the bacterial isolates showed the inhibition of spore germination with initial malformation of germ tubes and later formation of circle-like vesicles with no hyphal growth and hyphal disruption sometimes accompanied by hyphal swellings and shrinkages adjacent to the bacteria, suggesting their antibiotic mode of antagonistic activity. Control efficacy of B. subtilis GA1-23 and B. amyloliquefaciens GA4-4 on the cactus stem rot were not as high as but comparable to that of fungicide difenoconazole when they were treated simultaneously at the time of pathogen inoculation. All of these results suggest the two bacterial isolates have a good potential to be developed as biocontrol agents for the bipolaris stem rot of the grafted cactus.

Rapid and Sensitive Detection of Lettuce Necrotic Yellows Virus and Cucumber Mosaic Virus Infecting Lettuce (Lactuca sativa L.) by Reverse Transcription Loop-Mediated Isothermal Amplification

  • Cucumber mosaic virus (CMV) is damaging to the growth and quality of lettuce crops in Lanzhou, China. Recently, however, for the first time an isolate of lettuce necrotic yellows virus (LNYV) has been detected in lettuce crops in China, and there is concern that this virus may also pose a threat to lettuce production in China. Consequently, there is a need to develop a rapid and efficient detection method to accurately identify LNYV and CMV infections and help limit their spread. Reverse transcription loop-mediated isothermal amplification (RT-LAMP) assays were developed to detect the nucleoprotein (N) and coat protein (CP) genes of LNYV and CMV, respectively. RT-LAMP amplification products were visually assessed in reaction tubes separately using green fluorescence and gel electrophoresis. The assays successfully detected both viruses in infected plants without cross reactivity recorded from either CMV or LNYV or four other related plant viruses. Optimum LAMP reactions were conducted in betaine-free media with 6 mM Mg2+ at 65℃ for LNYV and 60℃ for 60 min for CMV, respectively. The detection limit was 3.5 pg/ml and 20 fg/ml using RT-LAMP for LNYV and CMV plasmids, respectively. Detection sensitivity for both RT-LAMP assays was greater by a factor of 100 compared to the conventional reverse transcription polymerase chain reaction assays. This rapid, specific, and sensitive technique should be more widely applied due to its low cost and minimal equipment requirements.
