Isolation and Characterization of Cryptococcus sp. CS-2 Secreting Polygalacturonase from Soil (토양으로부터 Cryptococcus sp. CS-2의 분리 및 균주가 분비하는 Polygalacturonase의 특성에 관한 연구)

  • A ploygalacturonase-produchg yeast was isolated from Cheju soil by selective eivichment media. One strain which has the highesl activity of polygalacturonase was selected. The characle~ishcs of the strain CS-2 were as follows: CS-2 utilized xylose. sucrose, maltose, u.ehalose, cellobiose. melibiose, lactose, raffinose, inosiiol, dulicilol, and dextrose, but did not utilized galactose, nitrate. nit~te, and lysine. Growth of CS-2 was inhibited by cyclohexamide, 1% acetic acid, and high concenaation (over 50%) of glucose. It grew at $30^{\circ}C$ but did 'IIOL $35^{\circ}C$. The cell size ofthe strain CS-2 was 2.9 p ~ n in length and 1.3 $\mu$ in diameter. Vegetable reproductmn was multiple budding and ascospre was present I to 4. Pseudomycelia or true myceliua formation were not observed In any of the cullureq. These results suggest that strain CS-2 is most likely a strain related Cryptococcus spp. (Cryptococcu spp. CS-2). When polygalacturonase or ihe yeast was induced by addition of polygalactoronic acid, polygalacturonase activity was detected in culture supernatent. There was a peak of specific activity a1 he mid-stationary phase(3 days culture) of growth. Polygalacturonase specific activity of Crylmcoccus sp. CS-2 was 2.96 unitsling. The molecular weighl ol'polygalacturonase was showed to be 46 KDa by both SDS-PAGE and activity stailling.

  • Stress-strain curves based on Ramberg-Osgood and Hollomon relations are strongly dependent upon the regressed range of strain. This work investigates mathematical expressions of true stress-strain curves of metallic materials. We first observe the variation of yield strength, strain hardening exponent and stress-strain curve with regressed range of stain. Based on sectional regression and expression using one or two parameters, we propose an optimal strain range for which yield strength and nonlinear material behavior are quite appropriate.

On Relevant Ramberg-Osgood Fit to Engineering Non-Linear Fracture Mechanics Analysis (정확한 비선형 파괴역학 해석을 위한 새로운 Ramberg-Osgood 상수 결정법)

  • This paper proposes a robust method for the Ramberg-Osgood (R-O) fit to accurately estimate elastic-plastic J from engineering fracture mechanics analysis based on deformation plasticity. The proposal is based on engineering stress-strain data to determine the R-O parameters, instead of true stress-strain data. Moreover, for practical applications, the method is given not only for the case when full stress-strain data are available but also for the case when only yield and tensile strengths are available. Reliability of the proposed method for the R-O fit is validated against detailed 3-D Finite Element (FE) analyses for circumferential through-wall cracked pipes under global bending using five different materials, three stainless steels and two ferritic steels. Taking the FE J results based on incremental plasticity using actual stress-strain data as reference, the FE J results based on deformation plasticity using various R-O fits are compared with reference J values. Comparisons show that the proposed R-O fit provides more accurate J values for all cases, compared to existing methods for the R-O fit. Advantages of the proposed R-O fit in practical applications are discussed, together with its accuracy.

The Hot Deformation Behaviors of Intermediate Thermo-Mechanical Treated Al-Li Based Alloy (중간가공열처리한 AI-Li계 합금의 고온변형거동)

  • In this study, intermediate thermo-mechanical treated Al-2.0 wt%Li, and Al-2.0 wt%Li-1.2 wt%Cu-1.0 wt%Mg-0.12 wt%Zr alloys were tested in tension at $10^{\circ}C$ and elevated temperature(100, 200 and $300^{\circ}C$). The results are follows : The tensile strength of Al-Li-Cu-Mg-Zr alloy is the highest but the elongation of Al-Li alloy is the highest(106%) among the all alloys in tension at $300^{\circ}C$. The Portervin-LeChartlier effect is showed in AI-Li-Cu-Mg-Zr alloy at 10 and $100^{\circ}C$, because of tangled dislocation by Mg and Cu. In the true stress-strain curves of all alloy, the peaks of stress at $300^{\circ}C$ are showed at the strain less than 0.1. In the binary alloy, the dynamic restoration process at 200 and $300^{\circ}C$ is nearly similar to dynamic recovery type. The hot deformation stress is decreased with increase of dynamic recovery degree, but the elongation is increased. When the strain the strain rate are constant, the temperature dependence of hot deformation stress is increased with increase of deformation temperature. The elongation and degree of dynamic recovery are decreased with increase of hot deformation activation energy, but the deformation stresses slightly increased.

Study on Mechanical Behavior of a Pre-Heat Treated Steel of ESW95 (선조질강 ESW95의 기계적 성질에 관한 연구)

  • We investigate the mechanical behavior of a pre-heat treated steel of ESW95, which is being used for automotive parts including tie-rods to save manufacturing cost and enhance product quality. SCM435 is also investigated to reveal the characteristics of the pre-heat treated steel tested. AFDEX/MAT is used to extract the true stress-strain curve over the large strain with higher accuracy. It has been found that ESW95 has very week strain-hardening behavior which can be negligible compared with SCM435 and that the initial yield strength is quite high and the toughness of ESW95 reaches nearly 75% of SCM435. ESW95 is characterized by the weak strain-hardening behavior and high yield strength that can be lead to minimization of post-processing including heat treatment and straightening. ESW95 and SCM435 are also compared by applying them to ball-stud forging by computer simulation. It is expected that a great deal of change may take place in production as well as in service if the pre-heated steels are adopted.

  • Creep behavior of Ti had been studied in a stress from 9.8 to 29.4 MPa and temperature rang from 873K to 973K with a special reference to tertiary creep. It was found that stress exponent of Ti was larger than that of the general pure metal and the compound metal. The relationship between true strain and strain rate in tertiary creep was appeared as the equation, $ln{\dot{e}}$ = $ln{\dot{e}}_{0}$ + ${\Omega}$ e Also, Apparent activation energy of was appeared as 274.92kJ/mol by using the equation ${\dot{\varepsilon}}_{0}$ = A ${\sigma }_{0}^{\ast_0}$ exp$(-Q_{0}/RT)$

  • When a pipe bend is modelled with straight beam elements, its stiffness, particularly in bending behavior, is overestimated than its true value. In this paper, we propose a simple and practical beam-modelling technique to estimate its stiffness properly. When this technique, based on the strain energy concept, is employed to modify the beam sectional properties of the bend, quite satisfactory results can be obtained. To verify the validity of this method, we apply the present technique to the free vibration analysis of a center pipe with 2 bends, one of the three components of the automobile exhaust system.

  • The constraint effect $A_2$ has to be evaluated within plastic region near crack tip front using opening displacement. Plastic boundary and stress or strain conditions in the vicinity of the crack tip using recrystallization heat treatment was represented. It was found that the plastic deformation boundary by recrystallization heat treatment method was the true strain of ${\epsilon}t$ = 0.05mm/mm. With the estimation of constraint effects $A_2$, the region of proper displacement measurement point near crack tip was between 0mm and 1mm distance toward direction of crack propagation, and was between 1mm to 3mm distance toward direction of load line.

Structural damage identification using incomplete static displacement measurement

  • A local damage identification method using measured structural static displacement is proposed in this study. Based on the residual force vector deduced from the static equilibrium equation, residual strain energy (RSE) is introduced, which can localize the damage in the element level. In the case of all the nodal displacements are used, the RSE can localize the true location of damage, while incomplete displacement measurements are used, some suspicious damaged elements can be found. A model updating method based on static displacement response sensitivity analysis is further utilized for accurate identification of damage location and extent. The proposed method is verified by two numerical examples. The results indicate that the proposed method is efficient for damage identification. The advantage of the proposed method is that only limited static displacement measurements are needed in the identification, thus it is easy for engineering application.
