Effects of enzymatically hydrolyzed fish by-products in diet of juvenile rainbow trout (Oncorhynchus mykiss)

  • Five experimental diets were formulated to evaluate the effects of dietary enzymatically hydrolyzed tuna by-product on growth, non-specific immune responses, and hematology of juvenile rainbow trout (Oncorhynchus mykiss). A basal diet with 50% of fishmeal was used as control (CON) and four other diets replaced 12.5% ($TBB_{12.5}$), 25% ($TBB_{25}$), 37.5% ($TBB_{37.5}$), and 50% ($TBB_{50}$) of fish meal in the CON diet. Juvenile rainbow trout ($4.87{\pm}0.05g$) were randomly distributed into 15 tanks (50 L) and fed 3-4% of wet body weight two times a day. At the end of 7 weeks of feeding trial, weight gain, specific growth rate, feed efficiency, and protein efficiency ratio of fish fed CON diet were significantly higher than those of fish fed $TB_{50}$ diet (P < 0.05). But there were no significant differences among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets (P > 0.05). There were no significant differences in GPT levels among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets. Also, there were no significant differences in lysozyme, superoxide dismutase, glucose, and total protein levels in all experimental diet (P > 0.05). The broken-line analysis indicated that the minimum dietary level of enzymatically hydrolyzed tuna by-product to replace fishmeal could be 29.7% in rainbow trout. These results indicated that the optimum level of dietary enzymatically hydrolyzed tuna by-product could replace greater than 29.7% but less than 37.5% of fishmeal in juvenile rainbow trout diet.

2021년 강원도 양식 무지개송어 및 은연어 비법정전염병 모니터링 (Disease monitoring of cultured rainbow trout and coho salmon in Gangwon province in 2021)

  • Disease including parasite, bacteria and virus cause serious mortality to salmonid fish in the aquaculture. In this study, we investigated the current disease status of the rainbow trout (Oncorhynchus mykiss) and coho salmon (Oncorhynchus kisutch) in Yanayang, Pyeongchang, Jeongseon and Yeongwol of Gangwon province in 2021 and performed molecular characterization of those pathogen. For parasites, Ichthyophthirius multifiliis was observed at 2 farms. For bacteria, we identified Aeromonas sobria from kidney of rainbow trout using phylogenetic analysis of gyrB gene. A. salmonicida were isolated from necrosis site of gill cover and fin in coho salmon and necrotic lesion of fin in rainbow trout. Phylogenetic analysis using vap gene indicated that A. salmonicida isolated in this study were clustered with previously reported A. salmonicida subsp. salmonicida isolates. For virus, JRt-Nagano type of infectious haematopoietic necrosis virus was detected in rainbow trout, but infectious pancreatic necrosis virus and Oncorhynchus masou virus were not detected. These results provide useful information for the prevention of disease spread and transmission when cultivating new species such as Atlantic salmon in Korea.

송어 보일드 통조림의 개발 및 식품학적 성분 특성 (Development and Food Component Characteristics of Canned Boiled Rainbow Trout)

  • 송어의 용도 확대에 의한 어민 소득 증대를 목적으로 송어를 이용한 통조림 제품의 개발을 시도하였다. 송어 통조림의 일반성분은 수분이 71.5%, 단백질이 18.2%, 지방이 3.1% 및 회분이 4.5%로, 시판 연어 통조림에 비하여 수분의 경우 약간 낮았고, 단백질의 경우 차이가 없었으며, 지방 빛 회분의 경우 높았다. 송어 통조림의 유리아미노산 및 총 아미노산의 총 함량은 각각 330.9 mg/100 g 및 18.2 g/l00 g이었고, 주요 아미노산은 유리 아미노산의 경우 glutamic acid(68.6 mg/100 g, 20.7%) 및 anserine(124.1 mg/100 g, 37.5%) 등이었으며, 총 아미노산의 경우 glutamic acid(18.0%), aspartic acid(8.6%), lysine(8.4%) 및 leucine(8.9%)등이었다. 송어 통조림의 무기질 함량은 칼륨이 123.3 mg/100 g, 칼슘이 271.3 mg/100 g, 마그네슘이 40.3 mg/100 g, 철이 2.4 mg/100 g 및 인이 244.3 mg/100 g으로, 송어 통조림을 100 g 섭취하는 경우 한국 성인 1일 영양 섭취량(칼슘 및 인: 모두 700 mg, 철: 12 mg)과 비교하여 보면 칼슘의 경우 38.8%를, 인의 경우 34.9%를, 그리고 철의 경우 20%를 섭취하는 효과가 있어 송어 통조림의 섭취에 의한 무기질 보강 효과는 기대할 수 있으리라 판단되었다. 송어 통조림의 지방산 조성은 폴리엔산이 43.7% 로 가장 높았고, 다음으로 모노엔산(28.8%) 및 포화산(27.5%)의 순이었으며, 주요 구성 지방산으로는 16:0(18.4%), 18:1n-9(20.6%), 18:2n-6(17.3%) 및 22:6n-3(12.7%) 등이었다.

Study of pathogenicity and severity of Lactococcus garvieae isolated from rainbow trout (Oncorhynchus mykiss) farms in Kohkilooieh and Boyerahmad province

  • Background: Lactococcus garvieae is one of the most important risk factors in the rainbow trout culture. Therefore, the purpose of this study was to identify and detect strains isolated from rainbow trout suspected of having Lactococcus garvieae using biochemical characteristics and PCR and determination of the degree of severity of isolated strains. Methods: In this study, the cause of lactococcosis in selected rainbow trout farms in Kohkilooieh and Boyerahmad province was assayed. Gram-positive and catalase-negative bacterial isolates were first obtained from selected trout fish farms using conventional biochemical tests and PCR assay. The 10-day LD50 method (concentration causing 50% mortality in 10 days) was used to determine the severity of the isolated bacteria. Results: One bacterial isolate was detected from all sampled fish which confirmed as Lactococcus garvieae using a specific PCR assay based on the 16S rDNA gene by producing a single band of 1107 bp. Analysis of the rate of mortality showed that the 10-day LD50 was 4.6 × 105 CFU/fish. The results of this study showed that isolated bacteria had high severity for rainbow trout. The presence of bacteria in internal organs of suspected fish showed a severe systemic infection in challenged fish. Antibiogram assay also indicated that the isolated Lactococcus garvieae were resistant to some mostly used antibiotics in rainbow trout. Conclusions: According to current research, it can be concluded that the condition of lactococcosis in the studied area is not suitable, and despite the presence of disease, there is no proper action to control and prevent the disease. Unfortunately, isolated bacteria from the studied area have a very high severity compared to bacteria isolated from other regions of the country or other countries. Therefore, further investigation is needed to determine the cause of this difference and possibly in the design of the vaccine.

Effect of β-Mannanase and α-Galactosidase Supplementation to Soybean Meal Based Diets on Growth, Feed Efficiency and Nutrient Digestibility of Rainbow Trout, Oncorhynchus mykiss (Walbaum)

  • A 12-week feeding trial was conducted with 87 g rainbow trout to evaluate the effects on growth performances, feed efficiency and nutrient digestibility of adding ${\beta}$-mannanase and ${\alpha}$-galactosidase enzymes, solely or in combination. Seven diets were prepared by adding ${\beta}$-mannanase, ${\alpha}$-galactosidase and mixed enzyme at two different levels (1 g/kg and 2 g/kg) to control diet (without enzyme) including soybean meal. Mixed enzymes (1 g/kg, 2 g/kg) were prepared by adding ${\beta}$-mannanase and ${\alpha}$-galactosidase at the same doses (0.5+0.5 g/kg and 1+1 g/kg). At the end of the experiment, addition of ${\beta}$-mannanase, ${\alpha}$-galactosidase and mixed enzyme to diet containing 44% soybean meal had no significant effects on growth performance and gain:feed (p>0.05). In addition, adding ${\beta}$-mannanase, ${\alpha}$-galactosidase and mixed enzyme in different rations to trout diets had no affect on nutrient digestibility and body composition (p>0.05).

Studies on Effects of Ea4-Peptide of Rainbow Trout Pro-IGF-I on Heart Development, Vasculogenesis and Hematopoiesis in Fish Embryos by Transgenesis

  • Insulin-like growth factors (IGFs) are mitogenic peptide hormones that regulate embryonic development, postnatal growth and cellular differentiation in vertebrates IGFs are initially translated as pre-pro-peptides and then proteolytically processed to yield the mature IGFs and E-peptides. Like the C-peptide of pro-insulin, the E-peptides of pro-IGFs are generally believed to possess little or no biological activity other than their potential roles in the biosynthesis of the mature IGFs. Like human IGF-1, previous studies in our laboratory showed that the recombinant trout Ea4-peptide of pro-IGF-1 exhibited a dose-dependent mitegenic activity in cultured BALB/3T3 fibroblasts and other non-oncogenic transformed cells (Tian et al., 1999) We have also shown by in vitro and in vivo studies that Ea4-peptide possessed novel anti-tumor activities (Chen et al., 2002, Kuo and Chen, 2002; Kuo and Chen 2003). Recent results of studies conducted in chorionicallantoic membrane of developing chicken embryos revealed that Ea4-peptide of trout pro-IGF-1 also possesses a dose-dependent antiangiogenic activity. Together these results raised the question whether Ea4-peptide of trout pro-IGF-1 may affect heart and blood vessel development and hematopoiesis in fish embryos. (중략)

PERFORMANCE OF RAINBOW TROUT (Oncorhynchus mykiss) FED ON DIFFERENT DIETARY PROTEIN WITH FIXED ENERGY RATIO

  • An experiment was conducted on rainbow trout(Oncorhynclus mykiss) for eight weeks to investigate the growth performance of the fish fed with different dietary protein with constant diet energy of $20kJg^{-1}$. Four diets containing 25, 30, 35 and 40% crude protein were used. The highest mean final weight was obtained for the fish fed with diet having 35% protein. Growth performance in terms of Specific Growth Rate (SGR), Food Conversion Ratio (FCR) and Protein Efficiency Ratio (PER) were calculated for each diet. There were no significant differences in SGR but the highest value was exhibited by fish fed with 35% protein diet. Significant differences were found among FCR of different diets. Diets with 35 and 40% crude protein gave better FCR value than that of 25 and 30% crude protein. Although significant differences were not found between PER of different diets but PER of diet with 35% protein was found to be better than PER of both high and low protein diets (diets of 40 and 30% crude protein). It is concluded that diet having 35% protein with protein energy ratio of $17.53mgkJ^{-1}$ was suitable for rainbow trout (O. mykiss) among the protein spectrum used.

Effect of Extracellular Calcium on Vitellogenin Production in the Culture of Hepatocytes in the Rainbow Trout, Oncorhynchus mykiss

  • Effect of extracellular calcium in vitellogenin (VTG) production in response to estradiol-17 $\beta$ $(E_2,\;2\times10^{-6}M)$ was examined in primary hepatocyte culture of rainbow trout, Onchorhynchus mykiss. Total calcium in estrogenized sera significantly increased, compared with the control, while diffusible calcium was insignificant. However, diffusible calcium in the incubation medium with $E_2$ was significantly reduced, compared with the control. The uptake of extracellular calcium by cultured hepatocytes signifIcantly increased 90 min after $E_2$ addition. Moreover, the accumulation of intracellular calcium increased in the cultures with $E_2$, regardless of the calcium concentrations in the incubation media. In addition, $E_2-primed $ VTG production was significantly decreased by withdrawal of E_2$ from the incubation medium. Moreover, VTG production by $E_2-primed$ hepatocytes was reduced by removing calcium from the incubation medium with or without $E_2$. These results suggest that the entry of extracellular calcium into the cytoplasm is an important step for VTG production in primary hepatocyte cultures in rainbow trout.

무지개송어의 성숙에 따른 식품성분 및 지질과산화물의 변화 (Changes of Food Components and Lipid Peroxides in Rainbow Trout with Growth)

  • 양어장에서 양식된 무지개송어의 성장에 따른 식품성분과 지질과산화물의 변화를 측정하여 다음의 결과를 얻었다. 치어와 성어의 조지방 함량과 수분 함량은 3.54%, 9.59% 및 77.40%, 69.02%로 성장에 따라 조지방의 함량은 증가되고 수분은 감소하였다. 지방산 조성은 치어에서 saturates 30.18%, monoenes 37.81%, polyenes 25.09%이고, 성어에서 30.77%, 40.41%, 23.45%로 성장에 따라 monoenes의 비율은 증가하고 polyenes 은 감소하였다. 구성 아미노산조성은 성장에 따라 큰 차이를 보이지 않았으며, 모두 glutamic acid, aspartic acid, histidine이 전체 아미노산의 약 50%를 차지하였다. 지질과산화의 지표로 측정한 maiondialdehyde, lipofuscin은 성장에 따른 유의적인 증가를 보이지 않았다.
