Determinant of parasite clearance and density on uncomplicated falciparum malaria infections in malaria-endemic area of Lampung Province, Indonesia

  • Lampung is a malaria-endemic region in Indonesia with an annual parasite incidence of 0.06 per 1,000 population. The socio-demographic factors, clinical conditions, and artemisinin combination therapy (ACT) types might affect parasite clearance and parasite density. This study aims to investigate factors that influence parasite clearance and parasite density in malaria patients. A retrospective analytic observational and a cross-sectional approach was used to conduct this study. A total of 66 malaria patients were examined to investigate parasite density and clearance, socio-demographic profiles, clinical conditions, and ACT types. To analyze data, univariate, bivariate, and multivariate tests were used. Age (P=0.045; r=0.238) and ACT type (P=0.021; r=0.273) were the only variables that had a significant correlation with parasite clearance. Age (P=0.003; r=0.345) had a significant correlation with parasite density. The most influential factors related to parasite clearance were the ACT type (dihydroartemisinin piperaquine) (P=0.017; odds ratio (OR) 0.109; 95.0% confidence interval (CI), 0.018-0.675) and age (P=0.030; OR 0.132; 95.0% CI, 0.021-0.823). Age (P=0.046; OR 0.320; 0.105-0.978, 95.0% CI) was the most significant variable associated with parasite density.

  • Indoor thermal conditions greatly affect the health and comfort of humans who occupy the space in it. The purpose of this research is to analyze the influence of water and vegetation elements as a microclimate modifier in buildings to obtain thermal comfort through the study of thermal environment models. This research covers two objects, namely public buildings and housing in Makassar City, South Sulawesi Prov-ince - Indonesia. Quantitative methods through field surveys and measurements based on thermal and personal variables. Data analysis based on ASHRAE 55 2020 standard. The data was processed with a parametric statistical approach and then simulated with the Computational Fluid Dynamics (CFD) simulation method to find a thermal prediction model. The model was made by increasing the ventilation area by 2.0 m2, adding 10% vegetation with shade plant characteristics, moving water features in the form of fountains and increasing the pool area by 15% to obtain PMV + 0.23, PPD + 8%, TSV-1 - +0, Ta_25.7℃, and relative humidity 63.5 - 66%. The evaluation shows that the operating temperature can analyze the visitor's comfort temperature range of >80% and comply with the ASHRAE 55-2020 standard. It is concluded that water elements and indoor vegetation can be microclimate modifiers in buildings to create desired comfort conditions and adaptive con-trols in buildings such as the arrangement of water elements and vegetation and ventilation systems to provide passive cooling effects in buildings.

Effect of Improved Cooling System on Reproduction and Lactation in Dairy Cows under Tropical Conditions

  • The effects of utilizing evaporative cooling system equipped with tunnel ventilation on postpartum ovarian activities, energy balance and milk production of early lactating dairy cows under hot and humid climates were studied from parturition to 22 wk postpartum. Thirty-four crossbred Holstein-Friesian (93.75% HF$\times$.25% Bos indicus) primiparous cows were randomly assigned to one of two groups. Cooled cows (n = 17; treatment) were housed in the tunnel ventilated barn equipped with evaporative cooling system and uncooled (n = 17; control) were housed in the naturally ventilated barn without supplemental cooling system. Cooled cows had greater (p<0.05) dry matter intake and milk production than uncooled cows. Days to the energy balance (EB) nadir did not differ between groups. However, days to equilibrium EB for uncooled cows was longer (p<0.05) than for cooled cows. There was no significant difference in postpartum anovular condition between cooled and uncooled cows. The interval from parturition to first postpartum ovulation did not differ between groups ($31.4{\pm}4.3$ and $26.1{\pm}3.6$ day, respectively). These results suggest that the evaporative cooling and tunnel ventilation has the potential to decrease the severity of heat stress and improve both milk production and metabolic efficiency during early lactation without affecting reproductive function in dairy cows under hot and humid climates.

Species Diversity, Composition and Stand Structure of Tropical Deciduous Forests in Myanmar

  • The characterization of tree species and forest stand conditions is useful in the planning of activities aimed to conserve biodiversity. The main objective of this study was to describe tree species diversity, species composition and stand structure of tropical deciduous forests distributed in three regions in Myanmar. Forest inventory was conducted in the Oktwin teak bearing forest, the Letpanpin community forest and Alaungdaw Kathapa National Park. According to the Jackknife estimator of species richness, 85 species (${\pm}18.16$), 70 species (${\pm}5.88$) and 186 species (${\pm}17.10$) belonging to 31 families were found in the Oktwin teak bearing forest, 33 families in Letpanpin community forest and 53 families in Alaungdaw Kathapa national park, respectively. Shannon's diversity indices were significantly different among the forests (p<0.05). It ranged from 3.36 to 4.36. Mean tree density (n/ha) of the Oktwin teak bearing forest, Letpanpin community forest and Alaungdaw Kathapa National Park were 488 (${\pm}18.6$), 535 (${\pm}15.6$) and 412 (${\pm}14.1$), while basal areas per hectare were $46.96m^2({\pm}3.23),\;49.01m^2({\pm}5.08)\;and\;60.03m^2({\pm}3.88)$, respectively. At the family level, Verbenaceae, Myrtaceae and Combretaceae occupied the highest importance value index, while at the species level it was Tectona grandis, Lagerstoremia speciosa and Xylia xylocarpa.

열대리슈마니아 핵형에 대한 열쇽, 약제 및 감마선 조사의 영향 (Influence of heat shock, drugs, and radiation on karyotype of Leishmania major)

  • 세계적으로 중요한 열대풍토병 병원체의 하나인 열대리슈마니아의 핵형에 영향을 줄 것으로 기대되는 인자와 그 효과를 관찰하고자 하였다. 토기의 혈액을 포함한 N.N.N. 배지에서 유지하고 있는 열대리슈마니아(Leishmania major)의 promastigote를 열쇽. 약제 첨가 자외선 조사 및 감마선을 이용한 방사선 조사를 여러 가지 방법으로 시행하고 주기변동전기영동(pulsed field gradient gel electrophoresis)을 이용하여 핵형의 변동 여부를 관찰하였다 그 결과 여러 방식에 의한 열쇽과 약제 처리 및 자외선 조사에 따른 핵형의 변화가 없었다 그러나 방사선 조사군에서는 50 Gy 이상 조사한 군에서 1 mesa base pair(Hb) 크기에 있는 염색체부터 소실되기 시작하여 방사선 조사량이 증가함에 따라서 250-500 Kb의 작은 염색체도 파괴되어 500 Gy 이상 군에서는 뚜렷한 염색체 분획이 없이 젤 하단 200 Kb 크기 아래 부분에 몰려 있었다. 이러한 소견은 방사선에 의하여 염색체가 불규칙하게 파손된 것을 의미한다고 하겠다. 방사선을 300 Gy까지 조사한 충체는 계대 배양이 가능하였고. 이들은 원래의 핵형을 유지하였다. 방사선조사 후에 배양된 충체는 염색체가 파괴되지 않았거나 부분적인 손상 후에 DNA 재결합에 의해 원상회복된 것으로 판단된다. 열대리슈마니아의 핵형은 일시적인 자극에 의하여 쉽게 변형되지 않는 안정된 것임을 확인하였다.

인도산 및 아프리카산 식물체 추출물의 장내세균에 대한 생육억제 반응 (Growth-inhibitory Responses of Human Intestinal Bacteria to Extracts from Indian and African Plants)

  • 31과 50종 인도산 식물체 84 시료와 12파 20종 아프리카산 식물체 27 시료의 메탄을 조추출물의 5종 장내세균(Bifidobacterium bifidum, Bifidobacterium longum, Lactobacillus acidophilus, Clostridium perfringens, Escherichia coli)에 대한 생육억제활성을 in vitro 혐기조건하에서 여지확산법으로 검정한 결과 억제활성은 공시세균과 식물체의 종류 및 부위에 따라 달리 나타났다. 인도산 식물체 시료중, Cymbopogon citratus 전부위, Ocimum basilicum 전부위, Madhuca indica 꽃 및 Aegle marmelos 잎 추출물이 Cl. perfringens에 대하여 강한 생육저해활성을 나타낸 반면, O. basilicum 전부위 추출물은 E. coli에 대하여 중간정도의 생육저해활성을 보였다. 그러나, 이들 식물체들은 공시 유산균의 생육에는 영향을 미치지 않았다. 이러한 결과로 부터 이들 열대식물들의 약리학적인 작용을 일부 설명할 수 있을 것으로 사료된다.

Echinostoma revolutum: Freshwater Snails as the Second Intermediate Hosts in Chiang Mai, Thailand

  • The occurrence of 37-collar spined echinostome metacercariae in freshwater snails was investigated in 6 districts of Chiang Mai Province, Thailand, from October 2011 to April 2012. A total of 2,914 snails that belong to 12 species were examined, and 7 snail species (Clea helena, Eyriesia eyriesi, Bithynia funiculata, Bithynia siamensis siamensis, Filopaludina doliaris, Filopaludina sumatrensis polygramma, and Filopaludina martensi martensi) were found infected with echinostome metacercariae. The prevalence of metacercariae was the highest in Filopaludina spp. (38.5-58.7%) followed by B. funiculata (44.0%), E. eyriesi (12.5%), B. siamensis siamensis (8.2%), and C. helena (5.1%). Metacercariae were experimentally fed to hamsters and domestic chicks, and adult flukes were recovered from both hosts at days 15 and 20 post-infection. The adult flukes were identified based on morphological features, morphometrics, host-parasite relationships, and geographical distribution. They were compatible to Echinostoma revolutum or Echinostoma jurini, with only minor differences. As the adults were recovered from both hamsters and chicks, our specimens were more compatible to E. revolutum rather than E. jurini (reported only from mammals). This is the first report for metacercariae of E. revolutum in the snail host, C. helena, and also confirmed that Filopaludina spp., E. eryresi, and Bithynia spp. act as the second intermediate hosts of E. revolutum under natural conditions, which are indigenously distributed in Chiang Mai province.

Production of Virus Free Seeds using Meristem Culture in Tomato Plant under Tropical Conditions

  • Protocol was established for production of virus free healthy seeds using meristem ($0.3-0.5\;\cal{mm}$ in size) culture and field management under net house condition in tomato. The isolated meristem was found well established in MS liquid medium containing $0.1\;\cal{mg}\;1^{-1}\;of\;GA_3$. For shoot and root development either from primary meristem or from nodal segment of meristem derived plants, semisolid MS medium having $0.5\;\cal{mg}\;1^{-1}$ of IBA was found most effective. The elimination of the studied viruses (ToMV, CMV, ToLCV) in meristem-derived plants was confirmed by DAS-ELISA test. For field management of the virus eradicated meristem-derived plants, use of net house was found very effective measures to check viral vector visit and eventually infection. The meristem-derived plants were vigor and high yielder than the native seed derived plants and produced healthy seeds. Due to stop vector visit, no viral symptoms were observed in both $R_1\;and\;R_2$ plants cultivated in net house condition. Starting of viral infestation was observed in $R_2$ generation when they were planted in open house condition without control of vector visit. Therefore, for management of viral diseases, use of virus free meristem derived plantlets and their subsequent cultivation in soil under net house condition without using any vector killing insecticide can be recommended for producing healthy seeds in tomato. The developed protocol for environmentally healthy tomato seed production in Bangladesh may be used in the countries having similar tropical like environment conducive for viral vector visit.

Production, Nutritional Quality and In vitro Methane Production from Andropogon gayanus Grass Harvested at Different Maturities and Preserved as Hay or Silage

  • Andropogon gayanus is an important grass due to its high biomass production, drought tolerance and favorable growth on low fertility acidic soils. Currently, there is little research on the impact of growth stage on the nutritional quality or the degree of $CH_4$ production that may arise from this forage during ruminal fermentation. The objectives of this study were to determine the effects of regrowth stage of A. gayanus on its chemical composition, in vitro production of gas and CH4, as well as in vitro dry matter (DM) digestibility when grown under tropical Brazilian conditions and conserved as hay or as silage. The nutritional value of A. gayanus grass declined with increasing maturity; however digestible DM yield linearly increased. After 112 d of regrowth, A. gayanus produced higher quality silage (higher lactate and lower pH and butyrate content) and higher DM yield. However, the low levels of crude protein at this time would make protein supplementation a necessity for proper rumen fermentation. No differences in $CH_4$ kinetic parameters were found with advancing maturity or preservation method (hay or silage).

  • The basic and main nutritive ingredients of two temperature (Anguilla japonica and A. rostrata) and two tropical (A. bicolor pacifica and A. marmorata) fresh water eel species that are farmed domestically were evaluated. With exception of A. rostrata, eels cultured at the same farm were used for analysis. The contents of crude protein were in the order A. marmorata (17.7%)>A. rostrata (17.5%)>A. bicolor pacifica (17.4%)>A. japonica (15.8%) and the contents of crude lipids were A. japonica (21.5%)>A. rostrata (15.4%)>A. bicolor pacifica (10.5%)>A. marmorata (8.9%). These values differed significantly even among the three species of eel farmed under identical culture conditions. In comparison, all four species of eel showed similar pattern in overall amino acid composition, although slight differences in the compositions of some amino acids were observed. The fatty acid compositions of muscle tissues were notably different among four species of eel, especially between the tropical and temperature eels. In a taste-test of the meat of the four eel species, which considered taste, flavor and texture, the overall preference was in the order A. japonica, A. marmorata, A. bicolor pacifica and A. rostrata.
