In vitro Evaluation of Cytotoxic Activities of Essential Oil from Moringa oleifera Seeds on HeLa, HepG2, MCF-7, CACO-2 and L929 Cell Lines

  • Moringa oleifera Lam. (Moringaceae) is widely consumed in tropical and subtropical regions for their valuable nutritional and medicinal characteristics. Recently, extensive research has been conducted on leaf extracts of M. oleifera to evaluate their potential cytotoxic effects. However, with the exception of antimicrobial and antioxidant activities, little information is present on the cytotoxic activity of the essential oil obtained from M. oleifera seeds. Therefore, the present investigation was designed to investigate the potential cytotoxic activity of seed essential oil obtained from M. oleifera on HeLa, HepG2, MCF-7, CACO-2 and L929 cell lines. The different cell lines were subjected to increasing oil concentrations ranging from 0.15 to 1 mg/mL for 24h, and the cytotoxicity was assessed using MTT assay. All treated cell lines showed a significant reduction in cell viability in response to the increasing oil concentration. Moreover, the reduction depended on the cell line as well as the oil concentration applied. Additionally, HeLa cells were the most affected cells followed by HepG2, MCF-7, L929 and CACO-2, where the percentages of cell toxicity recorded were 76.1, 65.1, 59.5, 57.0 and 49.7%, respectively. Furthermore, the $IC_{50}$ values obtained for MCF-7, HeLa and HepG2 cells were 226.1, 422.8 and $751.9{\mu}g/mL$, respectively. Conclusively, the present investigation provides preliminary results which suggest that seed essential oil from M. oleifera has potent cytotoxic activities against cancer cell lines.

In vitro antimicrobial activity of aqueous and ethanol extracts of Euphorbia hirta

  • Euphorbia hirta, locally called 'ara tanah' or 'susun nabi' in Malaysia is a small annual herb common to the tropical countries and belongs to the same family as the tic and tapioca. E. hirta has had a long history of usage in the treatment of various ailments. In this current study, in vitro sensitivity test of crude aqueous and ethanol extracts of leaves and barks of E. hirta was carried out against bacteria (Escherichia coli, Salmonella enteritidis, Staphylocccus aureus and Bacillus subtilis) and fungi (Microsporum canis, Aspergillus fumigatus, Candida albicans and Candida tropicalis) using the discs diffusion method. The extract-impregnated discs (20, 40 and $80\;{\mu}g/{\mu}l$), the E. hirta extracts inhibited the growth of all the bacteria tested. The growth of C. albicans was inhibited in a concentration dependent manner by the aqueous leaves and barks extracts. C. tropicalis was found to be sensitive to the aqueous leaves extracts. The results were compared to antibacterial drugs of chloramphenicol, ampicilin, penicillin G, and enrofloxacine; and to antifungal drug of ketoconazole, itraconazole and miconazole. In this current study, it can be concluded that this plant has antimicrobial activity that is as potent as the standard antimicrobial drugs against certain microorganisms.

중합효소연쇄반응을 이용한 Theileria sergenti의 신속한 검출 (Rapid detection of Theileria sergenti by polymerase chain reaction)

  • Theileria sergeni의 진단방법으로 Giemsa 염색에 의한 광학현미경적 관찰이 가장 통상 적으로 이용되고 있으나 감염이 아주 적거나 내과성인 경우 검출하기가 매우 곤란하다. 이에 PCR 진단을 위한 대강유전자로서 p33의 염기서열을 이용하여 4개의 oligonucleotide primers. TS1, TS2 ,TS3,, TS4를 작성하였다. 작성된 primer의 각 조합에 따라 PCR한 결과 TS1과 TS4 조합에서는 499 bps, TS1과 TS3 조합에서는 381 bps, TS2와 TS4 조합에서는 365 bps, TS2 와 TS3 조합에서는 247 bps 크기의 산물을 획득하였다 이 PCR산물은 p33 유전자 염기서열 분석을 통한 제한효소처리 및 Southern blot hybridization 방법을 통하여 그 특이성을 확인하였다. Primer의 특이성을 조사한 결과 미감염 백혈구 및 다른 주혈기생충인 Babesic ouota, Anaplosmn marginate에 대해서는 교차 반응을 나타내지 않았다. 또한 야외시료에 PCR 기법을 적용한 결과 Giemsa 염색에 의한 광학현미경적 관찰에서는 64.8%의 양성률을 보인 반면, PCR 진단에서는 본 실험에서 작성된 TS1과 TS4, TS2와 TS3 조합이 공희 88.7%의 양성률을 나타내었다.

외국의 교육제도와 교육시설의 특징 - 독일, 싱가포르, 미국의 사례를 중심으로 - (The Study on the foreign education system and educational facilities - Cases of the Germany, Singapore and the United States -)

  • Nowadays, the knowledge of modern society through education has become more and more complicated. The education system and the educational philosophy have created various education systems and methods for each country due to the difference of history and culture. In Korea, there are various teaching methods in recent years. However, if the traditional teaching method in Korea is based on educating teachers with a good understanding of the contents taught, some foreign countries such as USA and Germany can express their thoughts of the total population. In this situation, this study researches cases of other countries that have different educational systems from ours. It compares the differences of social background, educational program, and educational space in Germany, Singapore and the United States. As a result of this study, the following contents can be derived. The educational program is based on the social background of each country and plans an educational space suitable for the education program. In Germany, the emphasis is on the students' experience and community consciousness rather than on the amount of knowledge, so a home base space is planned between the classrooms to solve the problem of lack of solidarity and belonging among the students who are lacking in the curriculum. Some practical courses are run in the community. In Singapore, educational programs are elite-oriented compared to the other country, and because of the typical tropical climate, educational facilities are characterized by a lot of common space in semi-exterior space. Many schools in the United States have IMC in the central part of their schools that adopt self-directed learning and discussion.

Methanol extract of Elsholtzia fruticosa promotes 3T3-L1 preadipocyte differentiation

  • Elsholtzia fruticosa (EF) is present in tropical regions throughout South Asian countries as well as the Himalayas. Although it has been used as a traditional medicine to treat digestive, respiratory, and inflammatory issues, its effect on preadipocyte differentiation is unknown. In this study, we examined the effects of a methanol extract prepared from EF on the differentiation of 3T3-L1 preadipocytes. Cell differentiation was assessed by microscopic observation and oil-red O staining. The expression of adipogenic and lipogenic genes, including PPARγ and C/EBPα, was measured by western blot analysis and quantitative real-time polymerase chain reaction (qRT-PCR), to provide insight into adipogenesis and lipogenesis mechanisms. The results indicated that EF promotes the differentiation of 3T3-L1 preadipocytes, with elevated lipid accumulation occurring in a concentration-dependent manner without apparent cytotoxicity. EF enhances the expression of adipogenic and lipogenic genes, including PPARγ, FABP4, adiponectin, and FAS, at the mRNA and protein levels. The effect of EF was more pronounced during the early and middle stages of 3T3-L1 cell differentiation. Treatment with EF decreased C/EBP homologous protein (CHOP) mRNA and protein levels, while increasing C/EBPα and PPARγ expression. Treatment with EF resulted in the upregulation of cyclin E and CDK2 gene expression within 24 h, followed by a decrease at 48 h, demonstrating the early-stage impact of EF. A concomitant increase in cyclin-D1 levels was observed compared with untreated cells, indicating that EF modulates lipogenic and adipogenic genes through intricate mechanisms involving CHOP and cell cycle pathways. In summary, EF induces the differentiation of 3T3-L1 preadipocytes by increasing the expression of adipogenic and lipogenic genes, possibly through CHOP and cell cycle-dependent mechanisms.

Psychobiotic Effects of Multi-Strain Probiotics Originated from Thai Fermented Foods in a Rat Model

  • This work aimed to investigate the psychobiotic effects of six bacterial strains on the mind and behavior of male Wistar rats. The probiotic (PRO) group (n=7) were rats pre-treated with antibiotics for 7 days followed by 14-day probiotic administration, antibiotics (ANT) group (n=7) were rats treated with antibiotics for 21 days without probiotics. The control (CON) group (n=7) were rats that received sham treatment for 21 days. The six bacterial strains with probiotic properties were mostly isolated from Thai fermented foods; Pedicoccus pentosaceus WS11, Lactobacillus plantarum SK321, L. fermentum SK324, L. brevis TRBC 3003, Bifidobacterium adolescentis TBRC 7154 and Lactococcus lactis subsp. lactis TBRC 375. The probiotics were freeze-dried into powder (6×109 CFU/5 g) and administered to the PRO group via oral gavage. Behavioral tests were performed. The PRO group displayed significantly reduced anxiety level and increased locomotor function using a marble burying test and open field test, respectively and significantly improved short-term memory performance using a novel object recognition test. Antibiotics significantly reduced microbial counts in rat feces in the ANT group by 100 fold compared to the PRO group. Probiotics significantly enhanced antioxidant enzymatic and non-enzymatic defenses in rat brains as assessed using catalase activity and ferric reducing antioxidant power assay, respectively. Probiotics also showed neuroprotective effects with less pyknotic cells and lower frequency of vacuolization in cerebral cortex. This multi-strain probiotic formulation from Thai fermented foods may offer a potential to develop psychobiotic-rich functional foods to modulate human mind and behaviors.

중국 영남지방 여음산방 원림의 특징에 관한 연구 (A Study on the Characteristics of Yuyin ShanFang in China Lǐngnán Region)

  • 본 연구는 중국 영남지방의 4대 명원 가운데 하나인 여음산방에 대해 현지조사와 시문 분석 등을 수행한 결과 다음과 같은 결론을 도출하였다. 첫째, 여음산방의 조영자는 우빈(?彬 1824~1897)이며, 원림 면적은 $1,598m^2$이다. 우빈은 원림의 이름을 '여음(餘蔭)'이라고 하였는데 그 뜻은 조상의 음덕을 의미하였다. 둘째, 조영자가 쓴 시문을 분석해 보면, 미래 이상세계를 표현한 시문, 가문에 대한 오복사상을 표현한 시문, 그리고 풍류사상에 대한 시문으로 분류할 수 있었다. 셋째, 여음산방의 건축물의 배치와 공간구성의 특징을 분석해 본 결과, 부지 한가운데 사당건물(善言?公)을 중심으로 남쪽에 펼쳐진 공간은 주로 거주하는 공간이며, 북쪽에 펼쳐진 공간은 주로 제사와 관련된 공간이었다. 넷째, 여음산방의 공간구성요소를 분석해 본 결과이다. 정자부분은 원림에 지정(池亭)인 한취정(閒趣亭), 후화원(後花園)에 유상곡수연을 표현한 계상정(?賞亭), 독특한 반정(半亭) 형태인 내훈정(來熹亭) 등을 조영한 것으로 나타났다. 다섯째, 여음산방에는 형태가 매우 다양한 연못을 5개소 조성하였으며 기학적인 경계선을 가지는 것은 특징이다. 여섯째, 여음산방의 석가산은 괘방청산(掛榜靑山)이라고 하였는데, 과거시험에 합격한 사람의 이름을 적어서 붙이는(괘방) 곳이 곧, 이 석가산(청산)이라는 의미를 갖고 있다. 일곱째, 여음산방의 화창은 나무로 만든 문격창(紋格?)과 격단창(隔斷?) 등이 특징이며, 색채유리창(色彩?璃?)은 쑤저우 원림과 또 다른 차이점을 보이고 있다. 여덟째, 동문 양식은 쑤저우 고전원림의 동문 양식에 비해 다양하지 않은 편이지만, 동문 전면(前面)에 적혀있는 시문 장식물의 다양한 형태와 화려한 색채가 특징이다. 아홉째, 여음산방 원림에 설치된 건물장식용 조각예술품들이 표현된 주제를 분류해 보면 길상, 문자, 도교 팔선, 동식물, 설화와 기타 등으로 분류할 수 있다. 마지막으로 여음산방에 식재된 수목들은 주로 아열대식물이고 이중에 상징적 의미를 가진 수목도 있으며, 날씨가 좋기 때문에 과일나무를 많이 심었다.

An International Collaborative Program To Discover New Drugs from Tropical Biodiversity of Vietnam and Laos

  • An International Cooperative Biodiversity Group (ICBG) program based at the University of Illinois at Chicago initiated its activities in 1998, with the following specific objectives: (a) inventory and conservation of of plants of Cuc Phuong National Park in Vietnam and of medicinal plants of Laos; (b) drug discovery (and development) based on plants of Vietnam and Laos; and (c) economic development of communities participating in the ICBG project both in Vietnam and Laos. Member-institutions and an industrial partner of this ICBG are bound by a Memorandum of Agreement that recognizes property and intellectual property rights, prior informed consent for access to genetic resources and to indigenous knowledge, the sharing of benefits that may arise from the drug discovery effort, and the provision of short-term and long-term benefits to host country institutions and communities. The drug discovery effort is targeted to the search for agents for therapies against malaria (antimalarial assay of plant extracts, using Plasmodium falciparum clones), AIDS (anti-HIV-l activity using HOG.R5 reporter cell line (through transactivation of the green fluorescent protein/GFP gene), cancer (screening of plant extracts in 6 human tumor cell lines - KB, Col-2, LU-l, LNCaP, HUVEC, hTert-RPEl), tuberculosis (screening of extracts in the microplate Alamar Blue assay against Mycobacterium tuberculosis $H_{37}Ra\;and\;H_{37}Rv),$ all performed at UIC, and CNS-related diseases (with special focus on Alzheimer's disease, pain and rheumatoid arthritis, and asthma), peformed at Glaxo Smith Kline (UK). Source plants were selected based on two approaches: biodiversity-based (plants of Cuc Phuong National Park) and ethnobotany-based (medicinal plants of Cuc Phuong National Park in Vietnam and medicinal plants of Laos). At mc, as of July, 2001, active leads had been identified in the anti-HIV, anticancer, antimalarial, and anti- TB assay, after the screening of more than 800 extracts. At least 25 biologically active compounds have been isolated, 13 of which are new with anti-HIV activity, and 3 also new with antimalarial activity. At GSK of 21 plant samples with a history of use to treat CNS-related diseases tested to date, a number showed activity against one or more of the CNS assay targets used, but no new compounds have been isolated. The results of the drug discovery effort to date indicate that tropical plant diversity of Vietnam and Laos unquestionably harbors biologically active chemical entities, which, through further research, may eventually yield candidates for drug development. Although the substantial monetary benefit of the drug discovery process (royalties) is a long way off, the UIC ICBG program provides direct and real-term benefits to host country institutions and communities.

Establishment of an Allo-Transplantable Hamster Cholangiocarcinoma Cell Line and Its Application for In Vivo Screening of Anti-cancer Drugs

  • Opisthorchis viverrini (O. viverrini) is a well-known causative agent of cholangiocarcinoma (CCA) in humans. CCA is very resistant to chemotherapy and is frequently fatal. To understand the pathogenesis of CCA in humans, a rodent model was developed. However, the development of CCA in rodents is time-consuming and the xenograft-transplantation model of human CCA in immunodeficient mice is costly. Therefore, the establishment of an in vivo screening model for O. viverrini-associated CCA treatment was of interest. We developed a hamster CCA cell line, Ham-1, derived from the CCA tissue of O. viverrini-infected and N-nitrosodimethylamine-treated Syrian golden hamsters. Ham-1 has been maintained in Dulbecco's Modified Essential Medium supplemented with 10% fetal bovine serum for more than 30 subcultures. These cells are mostly diploid (2n=44) with some being polyploid. Tumorigenic properties of Ham-1 were demonstrated by allograft transplantation in hamsters. The transplanted tissues were highly proliferative and exhibited a glandular-like structure retaining a bile duct marker, cytokeratin 19. The usefulness of this for in vivo model was demonstrated by berberine treatment, a traditional medicine that is active against various cancers. Growth inhibitory effects of berberine, mainly by an induction of G1 cell cycle arrest, were observed in vitro and in vivo. In summary, we developed the allo-transplantable hamster CCA cell line, which can be used for chemotherapeutic drug testing in vitro and in vivo.

Black ginseng extract ameliorates hypercholesterolemia in rats

  • Background: Ginseng (Panax ginseng Meyer) is a well-characterized medicinal herb listed in the classic oriental herbal dictionary as "Shin-nong-bon-cho-kyung." Ginseng has diverse pharmacologic and therapeutic properties. Black ginseng (BG, Ginseng Radix nigra) is produced by repeatedly steaming fresh ginseng nine times. Studies of BG have shown that prolonged heat treatment enhances the antioxidant activity with increased radical scavenging activity. Several recent studies have showed the effects of BG on increased lipid profiles in mice. In this study report the effects of water and ethanol extracts of BG on hypercholesterolemia in rats. To our knowledge, this is the first time such an effect has been reported. Methods: Experiments were conducted on male Sprague Dawley rats fed with a high-cholesterol diet supplemented with the water and ethanol extracts of BG (200 mg/kg). Their blood cholesterol levels, serum white blood cell levels, and cholesterol-metabolizing marker genes messenger RNA (mRNA) expression were determined. Liver and adipose tissues were histologically analyzed. Results: We found that BG extracts efficiently reduced the total serum cholesterol levels, low-density lipoprotein (LDL) levels with increased food efficiency ratio and increased number of neutrophil cells. It also attenuated the key genes responsible for lipogenesis, that is, acetyl-coenzyme A (CoA) acetyltransferase 2, 3-hydroxy-3-methyl-glutaryl-CoA reductase, and sterol regulatory element-binding protein 2, at the mRNA level inside liver cells. Furthermore, the BG extract also reduced the accumulation of fat in adipose tissues, and inhibited the neutral fat content in liver cells stained with hematoxylin and eosin and oil red O. Conclusion: Administration of BG extracts to Sprague Dawley rats fed with high-cholesterol diet ameliorated hypercholesterolemia, which was mediated via modulation of cholesterol-metabolizing marker genes. This data throw a light on BG's cardioprotective effects.
