A triplex real-time PCR assay for simultaneous and differential detection of Bordetella bronchiseptica, Mycoplasma cynos, and Mycoplasma canis in respiratory diseased dogs

  • Bordetella (B.) bronchiseptica, Mycoplasma (M.) cynos, and M. canis are the major bacterial pathogens that cause canine infectious respiratory disease complex (CIRDC). In this study, we developed a triplex real-time polymerase chain reaction (tqPCR) assay for the differential detection of these bacteria in a single reaction. The assay specifically amplified three bacterial genes with a detection limit of below 10 copies/reaction. The assay showed high repeatability and reproducibility, with coefficients of intra- and inter-assay variations of less than 1%. The diagnostic results of the assay using 94 clinical samples from household dogs with CIRDC clinical signs, the prevalence of B. bronchiseptica, M. cynos, and M. canis was 22.3%, 18.1%, and 20.2%, respectively, indicating that the diagnostic sensitivity was comparable to those of previously reported qPCR assays. The dual infection rate of B. bronchiseptica and M. cynos, B. bronchiseptica and M. canis, and M. cynos and M. canis was 5.3%, 7.4%, and 3.1%, respectively. Moreover, the triple infection rate of B. bronchiseptica, M. cynos, and M. canis was 2.1%. These results indicate that coinfections with B. bronchiseptica, M. cynos, and M. canis have frequently occurred in the Korean dog population. The newly developed tqPCR assay in the present study will be a useful tool for etiological and epidemiological studies on these three CIRDC-associated bacterial pathogens. The prevalence and coinfection data revealed through this study will contribute to expanding knowledge on the epidemiology of CIRDC in the recent Korean dog population.

Development and evaluation of a triplex real-time quantitative reverse transcription-polymerase chain reaction for rapid and differential detection of three feline respiratory viral pathogens

  • In this study, a new triplex real-time quantitative reverse transcription polymerase chain reaction (tqRT-PCR) assay was developed for the rapid and differential detection of three feline viral pathogens including feline calicivirus (FCV), feline herpesvirus 1 (FHV-1), and influenza A virus (IAV) in a single reaction. The assay specifically amplified three targeted viral genes with a detection limit of below 10 copies/reaction. The assay showed high repeatability and reproducibility, with intra- and inter-assay coefficients of variation of less than 1%. Based on the diagnostic results of the assay using 120 clinical samples obtained from cats with feline respiratory disease complex (FRDC)-suspected signs, the prevalence of FCV, FHV-1, or IAV was 43.3%, 22.5%, or 0%, respectively, indicating that the diagnostic sensitivity was comparable or superior to those of previously reported monoplex qRT-PCR/qPCR assays. The dual infection rate for FCV and FHV-1 was 8.3%. These results indicate that FCV and FHV-1 are widespread and that co-infection with FCV and FHV-1 frequently occur in the Korean cat population. The developed tqRT-PCR assay will serve as a promising tool for etiological and epidemiological studies of these three bacterial pathogens, and the prevalence data for three feline viruses obtained in this study will contribute to expanding knowledge about the epidemiology of FRDC in the current Korean cat population.

연마 과정에 따른 열중합 의치상 레진의 표면 거칠기 평가 (Evaluation of surface roughness of heat-polymerized denture base resin according to the polishing step)

  • Purpose: The objective of this study was to compare the surface roughness according to polishing process in conventional laboratory techniques used for polishing three different acrylic denture base resins. Materials and methods: Specimen preparation and surface polishing procedures were conducted to manufacturer's recommendation with three heat-polymerized denture base resins. Surface roughness and gloss were measured by a contact type tester and a LED gloss checker using thickness 2 mm and diameter 10 mm. There were five specimens for each acrylic resin material and polishing procedures. Mean average surface roughness (Ra) values of each specimen group were analyzed using a one-way ANOVA analysis of variance and Scheffe's post hoc test. Surfaces after surface roughness and gloss testing according to each polishing process were evaluated under a stereoscopic microscope. Results: The highest mean average surface roughness was measured($Ra=2.43{\pm}0.47$) for surfaces finished with a denture tungsten carbide bur in Triplex. The lowest surface roughness values ($Ra=0.11{\pm}0.07$) were determined in Vertex polished with a lathe. In addition, all materials revealed that surface roughness determined highly in HP1 and HP2 than other procedures. All correlation between surface roughness and gloss showed highly with three heat-polymerized resins. Specially, topmost correlation revealed than other material in Triplex. Significant differences in mean average surface roughness were found between polishing process used high speed lathe and low speed hand-piece. Conclusion: Laboratory polishing used to high speed was found to produce the smoothest surface of heat-polymerized denture base acrylic resin. Therefore, we recommended that high polishing process need to get smooth surface.

  • 본 논문에서는 제어 시스템의 신뢰도를 높이고 중단없는 제어 동작을 실현하기 위하여 제어기를 삼중화한 내고장성 디지털 조속기를 설계하였으며, 이 시스템을 Markov process로 모델링하여 신뢰도를 해석하였다. 임무시간에서 삼중화 시스템은 단일 제어기 시스템에 비해 신뢰도 86[%]수준에서 약 1.8 배, 95[%]수준에서 약 2.8배, 99[%]수준에서 약 6배 증가함을 확인하였다. 또한 북제주 화력 발전소의 터빈 속도제어 시스템을 대상으로 디지털 속도제어 시스템을 설계하였으며, 설계된 디지털 속도제어 시스템의 유효성을 입증하기 위해 모의실험을 행하였다.

  • In this study, turbine speed control algorithm is studied for Buk-Jeju steam turbine power plant and also digital governor system is designed for speed control of steam turbine in power plant. By using duplex I/O module, triplex CPU module, 2 out of 3 voting algorithm and adding self diagnostic ability, the reliability of the designed digital governor system can be acquired satisfactorily. Designed and manufactured digital governor system is implemented in a pilot steam turbine plant of 0.3kw output power Installed in Korea Maritime University. After a series of experiment the reliability and availability is confirmed and also stable operation is achieved.

  • 전동기의 고정자 권선법에는 통상적인 3상 권선법과 multiplex 3상 권선법이 있다. 본 논문에서는 multiplex 3상 권선 전동기의 장점인 토크와 토크 리플 성능 향상에 대해 연구하였고 그 원인을 분석하였다. 8극 48슬롯 권선계자형 동기전동기(WFSM)에 대해 기존 3상 권선법과 duplex 3상 권선법을 비교 분석하였으며, 32극 144슬롯 표면 부착형 영구자석 동기전동기(SPMSM)에 대해 유한요소해석법을 이용하여 기존 3상과 triplex 3상 권선법을 비교 분석하였다. 마지막으로 두 종류의 전동기에서 multiplex 권선법의 장점을 알아보고자 평균 토크와 토크 리플을 비교하였다.

  • Programmable electronic interlocking system plays key role in railway operation and is closely related to railway accidents, in which the programmable electronic controller of interlocking system may become sources. Redundant digital controllers are adopted as the interlocking controllers to prevent the accidents from the controllers being out of order. The redundant digital controllers being fault tolerant are realized through dual or triplex controllers. In this paper, we calculated safety and availability of the redundant digital controllers using Markov models, demonstrated key part to determine the availability ana the safety.

SiO2/ZnS:Cu/ZnS Triplex Layer Coatings for Phosphorescence Enhancement

  • The objective of this study is to coat the $SiO_2$ layer uniformly on the ZnS:Cu phosphors by using Sol-Gel method. From results of SEM micrographs observation, XRD and XPS analysis, it could be confirmed that $SiO_2$ layer was relatively well coated on ZnS:Cu particles. $Ag_2S$ was used as a decoding chemical to analyze the dense and uniform coating performance of $SiO_2$ layer on phosphor particles. It could be concluded that phosphors synthesized from our two step replacement method showed strong blue peak comparing to other method and rather weak green peak also. Obtained particle size of phosphors were about 20m diameter. Luminescence properties of the phosphors were examined by photoluminescence spectra at the excitation wavelength of 270 nm.

  • This thesis presents a development of intelligent digital governing system and it's application on Sumjingang Hydro-Power plant. The developed system consists of hardware, software and governing algorithm. The feature of hardware is triplex-modular fail safe redundant system for a safe turbine running. The software consists of operating system and application program. The operating system has real-time and multi-tasking features. And also, application algorithm is composed to run francis type hydro-turbine. The developed digital governing system is applied to Sumjingang hydro-power plant, Korea Hydro Nuclear Power Corporation.
