Regulatory Viral and Cellular Elements Required for Potato Virus X Replication

  • Potato virus X (PVX) is a flexuous rod-shaped virus containing a single plus-strand RNA. Viral RNA synthesis is precisely regulated by regulatory viral sequences and by viral and/or host proteins. RNA sequence element as well as stable RNA stem-loop structure in the 5' end of the genome affect accumulation of genomic RNA and subgenomic RNA (sgRNA). The putative sgRNA promoter regions upstream of the PVX triple gene block (TB) and coat protein (CP) gene were critical for both TB and CP sgRNA accumulation. Mutations that disrupted complementarity between a region at the 5' end of the genomic RNA and the sequences located upstream of each sgRNA initiation site is important for PVX RNA accumulation. Compensatory mutations that restore complementarity restored sgRNA accumulation levels. However, the extent of reductions in RNA levels did not directly correlate with the degree of complementarity, suggesting that the sequences of these elements are also important. Gel-retardation assays showed that the 5' end of the positive-strand RNA formed an RNA-protein complex with cellular proteins, suggesting possible involvement of cellular proteins for PVX replication. Future studies on cellular protein binding to the PVX RNA and their role in virus replication will bring a fresh understanding of PVX RNA replication.

NMR study of the interaction of T4 Endonuclease V with DNA

  • T4 Endonuclease V (Mw 16,000) acts as a repair enzyme for UV induced pyrimidine dimers in DNA. Many researchers have studied the biochemical characteristics of the enzyme. However the precise action mechanism of T4 endo V has not fully elucidated yet. In our laboratory NMR spectroscopy technique is being used for the structural study of T4 endo V. Because of its low temperature stability and high content of ${\alpha}$-helix, the conventional $^1$H NMR technique was inapplicable. Therefore we utilized stable isotope labeling technique and so far prepared about 10 amino acid specific labeled proteins. The HSQC spectra of amino acid specific labeled proteins will help us to interpret the triple resonance 3D, 4D data which are under processing, We also studied the behaviors of specific amino acid residues whose roles might be critical. When the enzyme labeled by $\^$15/N-Thr was mixed with the substrate oligonucleotide (semispecific -TT- sequence), one crosspeak in its HSQC spectrum was completely desappeared, which means that one of seven Thr residues is in the binding site of the enzyme with DNA, This result is well consistent with previous report that implicated the Thr 2 residue in the activity of the enzyme. Similar studies were carried on the behaviors of Arg and Tyr residues.

Backbone NMR Assignments of an Uncharacterized Protein, SF1002 from Shigella flexneri 5a M90T

  • The causative agent of shigellosis, Shigella flexneri, is a Gram-negative anaerobic bacterial pathogen that causes one of the most infectious bacterial dysenteries in humans. It originates infection by invading cells of the colonic epithelium using a type III secretion system. Despite S. flexneri is closely linked with the human disease, structural study is very deficient. Here, we have initiated NMR study of SF1002 which is the uncharacterized protein from S. flexneri strain 5a M90T. Based on a series of triple resonance spectra, sequence-specific assignments of the backbone amide resonances of SF1002 could be completed. This NMR study would contribute to the structural genomics of S.flexneri.

Metallo-${\beta}$-lactamase를 생성하여 Imipenem에 내성인 Pseudomonas aeruginosa에 대한 항균제 병합요법의 효과 (Effect of Antibiotic Combination Therapy on Metallo-${\beta}$-Lactamase Producing Imipenem Resistant Pseudomonas aeruginosa)

  • 국내 대학병원에서 분리되어 imipenem 에 대한 최소억제농도가 $8{\mu}g/ml$ 이상인 51개의 포도당비발효 그람음성 간균들 중 metallo-${\beta}$-lactamase (MBL)을 생성하는 균주들을 분리하고, 그들 중에서 내성이 강한 Pseudomonas aeruginosa에 대한 항균제 병합요법의 효과를 알아보기 위하여 상승효과를 보이는 항균제 조합을 찾아보았다. 9개의 균주(Pseudomonas aeruginosa 2주 및 Achromobacter xylosoxidans subsp. xylosoxidans 7주)가 MBL 양성을 나타냈으며, PCR 결과 9주 모두에서 $bla_{VIM-2}$ 유전자가 관찰되었다. 이들 중에서 P. aeruginosa DK569는 aztreonam (MIC; $8{\mu}g/ml$)을 제외하고 실험한 모든 ${\beta}$-lactam 항균제, aminoglycoside, ciprofloxacin에 내성을 보여 aztreonam 함유 배지를 이용하여 상승효과률 보이는 항균제를 찾고자 하였다. One disk synergy test 에서 선별된 항균제 조합을 이용하여 생존률 검사 실험을 한 결과, aztreonam (AZT)와 piperacillin-tazobactam (TZP)의 병합은 항균제 노출 6시간 후에 AZT 또는 TZP의 단독 항균제 노출시 보다 균수가 1/18.7로 감소하였다. 그리고 AZT와 amikacin (AN)의 병합에서도 항균제 노출 6 시간 후에 AZT 또는 AN의 단독항균제의 투여보다 균수가 1/17.1 로 감소하였다. 결국 위 두 조합은 의미있는 상승효과를 보이지 못하여 위 세 항균제를 조합하여 실험하였다. 위의 세 항균제를 병합하였을 때 항균제 노출 8시간 후에 AZT, TZP 및 AN의 단독 투여에 비하여 병합요법에 의해 균수가 1/183.3 로 감소하여 의미있는 상승효과를 보였다. 이 결과는 치료가 쉽지 앓은 MBL 생성균에 의한 감염에 대한 치료에 AZT, TZP 및 AN의 세 가지 항균제 병합요법이 유용할 것이라는 것을 의미한다.

Big Data and Knowledge Generation in Tertiary Education in the Philippines

  • This exploratory study investigates the use of a computational knowledge engine (WolframAlpha) and social networking sites (Gmail, Yahoo and Facebook) by 200 students at De La Salle-College of Saint Benilde, their "friends" and their "friends of friends" during the 2009 through 2013 school years, and how this appears to have added value in knowledge generation. The primary aim is to identify what enhances productiveness in knowledge generation in Philippine Tertiary Education. The phenomenological approach is used, therefore there are no specific research questions or hypotheses proposed in this paper. Considering that knowledge generation is a complex phenomenon, a stochastic modelling approach is also used for the investigation that was developed specifically to study un-deterministic complex systems. A list of salient features for knowledge generation is presented as a result. In addition to these features, various problem types are identified from literature. These are then integrated to provide a proposed framework of inclusive (friendly) and innovative social networks, for knowledge generation in Philippine tertiary education. Such a framework is necessarily multidisciplinary and useful for problem-solving in a globalized and pluralist reality. The implementation of this framework is illustrated in the three parts of the study: Part 1: Online lessons, discussions, and examinations in General Psychology, Introduction to Sociology, and Life and Works of Jose Rizal, for the author's students in De La Salle-College of Saint Benilde; Part 2: Facebook Report analytics of students and teachers, their friends and their friends of friends via WolframAlpha; and Part 3: Social Network Analysis of the people and groups influencing the courses' scope-and-sequence in the new General Education Curriculum for Tertiary Schools and Institutions in the Philippines.

비트 플레인 정합에 의한 움직임 추정기의 VLSI 설계 (VLSI Design for Motion Estimation Based on Bit-plane Matching)

  • 전역탐색알고리즘(full-search algorithm, FSA)은 탐색영역의 범위가 커짐에 따라 방대한 양의 계산을 필요로 하기 때문에 이에 따른 알고리듬의 처리시간이 커지고, 하드웨어로 구현했을 때 회로가 복잡해진다는 문제점을 안고 있다. 본 논문에서는 이러한 문제점을 개선하기 위한 방안으로 비트플레인 정합에 의한 움직임 추정기의 VLSI 구조를 제안한다. 제안된 움직임 추정기에서는 비트 플레인 정합기준을 이용하여 기존의 전역 탐색 알고리즘을 하나의 이진영상으로 적용함으로써 움직임 추정에 소요되는 연산의 양을 크게 줄이면 서도 전역탐색 알고리듬과 유사한 움직임 추정 성능을 갖도록 하였으며, 제안된 VLSI 구조에서는 두 개의 프로세싱 코어를 채택하여 데이터 흐름을 시스톨릭 (systolic) 어레이의 형태로 제어하여, 시스템 내부의 SRAM을 제거하여 동작 속도 상의 이득뿐만 아니라, 메모리 공정을 필요로 하지 않는 저가의 공정을 사용 가능하게 함으로써 제작상의 비용을 절감할 수 있는 해결책을 제시하였다. 구현된 하드웨어는 VHDL을 이용하여 설계하고, 기능 검증을 수행한 후 0.6-μm three-metal CMOS 공정을 이용하여 8.15 X 10.84㎟의 크기로 집적하였다.

Gene Mutations of 23S rRNA Associated with Clarithromycin Resistance in Helicobacter pylori Strains Isolated from Korean Patients

  • Although resistance of Helicobacter pylori to clarithromycin is a major cause of failure of eradication therapies, little information is available regarding gene mutations of clarithromycin-resistant primary and secondary H. pylori isolates in Korea. In the present study, we examined gene mutations of H. pylori 238 rRNA responsible for resistance to clarithromycin. DNA sequences of the 238 rRNA gene in 21 primary clarithromycin-resistant and 64 secondary clarithromycin-resistant strains were determined by PCR amplification and nucleotide sequence analyses. Two mutations of the 238 rRNA gene, A2143G and T2182C, were observed in primary clarithromycin-resistant isolates. In secondary isolates, dual mutation of A2143G+T2182C was frequently observed. In addition, A2143G+T2182C+ T2190C, A2143G+T2182C+C2195T, and A2143G+T2182C+A2223G were observed in secondary isolates. Furthermore, macrolide binding was tested on purified ribosomes isolated from T2182C or A2143C mutant strains with $[^{14}C]$erythromycin. Erythromycin binding increased in a dose-dependent manner for the susceptible strain but not for the mutant strains. These results indicate that secondary isolates show a greater variety of 238 rRNA gene mutation types than primary isolates, and triple mutations of secondary isolates are associated with A2143G+T2182C in H. pylori isolated from Korean patients.

『온역론(溫疫論)』 막원(膜原) 개념의 연원에 대한 소고(小考) (A Study on the Origin of Moyuan in Wenyilun)

  • Objectives : This paper contemplates how the special features found in Moyuan as revealed in Wenyilun has continued from the Moyuan concepts of past includig Huangdineinjing. The author of this paper attempted to distinguish common and different points between the two, and how these features changed in the following generations. Methods : The concept of Moyuan as revealed in Huangdineinjing is first reviewed before laying out the concepts of Moyuan in Wenyilun by different categories. These two concepts are then compared. A Chinese medicine literature database is used in continuing this study, and materials are selected with relation to the special features of Moyuan. Results : The special feature of half-exterior half-interior found in the Moyuan in Wenyilun is similar with that in Neijing in that they both thought Moyuan existed between the stomach, the lungs, and the five organs. The concealability of Moyuan is similar with the explanation of Moyuan in Suwen nuelun, and it is related with the story of Gaohuang in Chunqiu Zuozhuan. Compared to the fact that the concept of Moyuan in Neijing is explained as that pathogenic factor enters the surface of the body in sequence, the concept of Moyuan in Wenyilun is conveyed in that the pathogenic factor conceals itself first before being divided into exterior and interior. Moyuan is also related to upright qi, and Gaohuang-yuan: origin of Gaohuang, too, is connected Shenjiandongqi: the energy that moves between the two kidnets, and also the triple burner. Conclusions : The concepts of "Half-exterior Half-interior", concealability, and "divide it into several places in half-exterior", and upright qi contained within the Moyuan in Wenyilun are studied for their origins, and this effort is anticipated to be greatly helpful in both understanding the historical contexts of Wuyouke Wenyilun and conducting studies on clinical application of Moyuan theory.

의복디자인에서의 모듈성에 대한 연구 (A Study on the Modularity in Clothing Design)

  • Clothing design is one of design products for human, the end user and corresponds to artifacts. Sometimes artifacts with one mono module exist, and so does artifacts assembled and combined with multi modules of same shape and size or various shapes and sizes and thus all design products can be understood by modularity concept. The purpose of this study was to make a new suggestion for the clothing design and production of high uniqueness and creativity by reviewing and synthesizing the foundation of clothing modularity concept as the original clothing design figures from the history have shown various modularity concept evolution from mono module to multi module stages. The methods of this study were to identify clothing modularity and analyze the type, evolving direction, and category of clothing modularity, and the value of clothing modularization design through comprehensive literature reviews on topic-related books and theses. The original clothing figures with Significance from the clothing history were analyzed in evolution sequence for application direction and value of clothing modularity in flat pattern. Clothing modularity in ancient clothing figures was classified as three types of the fixed, drapery, and straight lined in evolving direction from clothing of mono module to flat patterned clothing. The direction of clothing modularity was identified as mono- dual-triple-multi into another level of multi modularity after intentional devolution. The categorization of clothing modularity was identified in terms of clothing flat construction, clothing design construction, and clothing form modeling. The value of clothing modularization design using clothing modularity was identified as economical efficiency, convenience, promptitude, adaptability, functionality, and creativity.

나로호 상단 발사관제시스템 개발 및 발사운용 결과 (Development and Operations of LV PACS-II for the Launch Vehicle NARO)

  • 지난 2009년과 2010년 2회 비행시험을 수행한 나로호는 나로우주센터에서 발사준비 및 발사작업이 진행되었다. 발사관제시스템은 우주센터의 발사패드에 총조립된 상태로 장착된 발사체에 대한 발사작업을 위하여 발사체, 발사대, 추진지상공급계 등 외부시스템에 대한 명령을 전달하고 데이터를 실시간 모니터링하여 발사준비 프로세스를 총괄 운용한다. 발사관제시스템의 서브시스템인 상단 발사관제시스템은 나로호 상단에 대한 지상 발사작업을 제어 및 감시할 수 있도록 개발되었으며 나로호 비행시험을 통해 발사준비 프로세스 자동 및 수동처리, RCS 충전, 상단 온도환경상태 감시 등의 요구 기능들이 모두 설계 기준에 부합하는 것으로 확인되었다. 본 논문에서는 상단 발사관제시스템의 핵심 기술과 특성을 기술하며 발사운용 결과를 분석하였다.
