The Surface Activities of Surfactant Mixtures (혼합 개면활성제의 개면활성에 관한 연구)

  • The changes of surface activities in the aquaous solutions of mixed surfactants composed of linear sodium dodecylbenznesulfoate (LAS), polyoxyethylene nonyl phenylether (PE, EO=10) and polyethylene glycol monolauryl ether (LE, EO=25) have been studied. Addition of nonionic surfactants to LAS reduces the surface tension, especially at the lower concentration than cmc. The interfacial tension of olive oil/LAS was lower than the other surfactant solutions. The removal of triolein from cotton fabrics by nonionic surfactants and mixtures is higher than by LAS. The addition of NaCI to surfactant solutions even though reduces surface tension smaller but enthances oil removal more than that of $CaCl_2$.

Studies on the Neutral Lipid Components of Korean Hazel Nut (한국산 개암종실의 중성지질 조성에 관한 연구)

  • Compositions of neutral lipid of the hazel nut (Corylus heterophylla Fischer var. thumbergii Blume, Betulaceae) produced in Korea were analyzed by thinchrography (Iatrosoan TH-10 type), which was equipped with a flame ionization detector. The neutral lipid components were nearly separated of the basis of their degree of unsaturation by employing the chromatography on silica gel sintered rod impregnated with 12.5% silver nitrate and it was noted that triglyceride was the major lipid components of Korean hazel nut. The triglyceride compositions were trilinolenin 10.63, trilinolein 18.01, triolein 42.24, tripalmitin 17.57 and tristearin 11.55% by thinchrographic analysis.

Effects of Egg Phospholipids on the Intestinal Absorption of Lipids

  • This study was conducted to determine the effects of egg phospholipids [(Phosphatidylcholine (PC) and sphingomyelin (SM)] on intestinal absorption of cholesterol and other lipids. Each rat with Iymph cannula was infused via a duodenal catheter at 3.0 mL/h for 8 h with a lipid emulsion containing triolein, cholesterol and PC in 24 mL PBS. The PC in the lipid emulsion was egg PC (EPC), hydrogenated egg PC (HPC), or soy PC (SPC). The EPC in the lipid emulsion markedly lowered the Iymphatic absorption of cholesterol, compared with SPC and a lipid emulsion containing no PC. The HPC further lowered the absorption of cholesterol. The phospholipid output was not affected by the source of PC infused. The total Iymphatic output of oleic acid (18: 1), the major fatty acid infused in the form of triolein, did not differ among the NPC, SPC and EPC groups, but was significantly lower in the HPC group. The findings provide the first evidence that EPC markedly lowers the Iymphatic absorption of cholesterol under in vivo conditions. The inhibitory effect of EPC appears to be due to the higher degree of saturation of its acyl groups relative to SPC, suggesting that the intestinal absorption of egg cholesterol may be reduced by the presence of PC in e99 yolk. Experiment 2 was designed to determine whether egg SM, structurally similar to PC, also inhibits the Iymphatic absorption of cholesterol. Egg SM lowered the Lymphatic absorption of cholesterol in a dose dependent manner. Likewise, SM lowered the Iymphatic absorption of oleic acid, whereas it had no effect on retinol absorption. SM at a high dose lowered the Iymphatic outputs of both PC and SM, whereas there was no such effect at a lower dose. These results also indicate that luminal egg SM has an inhibitory effect on the intestinal absorption of cholesterol and other lipids of relatively high hydrophobicity.

  • The conventional triolen absorption test has its defect in that the stool collection was cumbersome, time and energy-wasting. In the present study, the triolen absorption test was carried out using double tracer technique with $^{125}I-triolen\;and\;^{51}Cr_2O_3$ to determine if it can overcome the defect of the conventional method also with satisfactory results. Following were the results: 1. The clinical significance of this double tracer method was essentially the same with that previously done by radioactive. triolen alone. With the fractional fecal samples, the equation, y=0.626x+2.010 was substantiated, hence, this method appears to be clinically valuable if the appropriate correction is applied. With the mixed fecal samples, the equation y=0.642x+1.468 was substantiated (p<0.005) which appears to be also clinically valuable. When these two data were compared, the equation y=0.975x+0.090 (P<0.05) was substantiated, hence, $x{\fallingdotseq}y$. 2. The normal ranges of the fecal triolen excretion rate in this double tracer method were $3.46{\pm}1.69%$, namely, less than 6.9%. 3. The samplings were done from the first to third defecation in cases of clinically normal, and from the first to second defecation in cases of diarrhea or malabsorption. 4. The intestinal malabsorption of triolen was not observed in whom the triolen absorption was supposed to be clinically normal, however. a good number of suspicious malabsorptive cases showed the normal values.

  • Production of cocoa butter equivalent fat (CBE) from palm oil and stearic acid by reverse micelle lipase reaction system was studied. Qualitative and quantitative analyses of triglycerides were performed by HPLC. The reaction conditions for maximum conversion from triolein and stearic acid to 1-stearoyl-2,3-dioleoyl glycerol(SOO) and 1,3-distearoyl-2-oleoyl glycerol(SOS) were as follows: a molar ratio of water/Aerosol OT, 10; triolein, 30 mM; stearic acid, 90 mM; pH, 7.5; and temperature, $50^{\circ}C$. By lipase in reverse micellar system containing palm oil and stearic acid, 1,3-dipalmitoyl-2-oleoyl glycerol(POP), 1-palmitoyl-2,3-dioleoyl glycerol(POO) and SOO decreased, but large amounts of 1-palmitoyl-2-oleoyl-3-stearoyl glycerol(POS) and SOS was formed.

Lipolytic Properties of Candida Cylindracea Lipase Toward Triacylglycerols with Different Fatty Acyl Chains

  • Lipolytic characteristics of candia cylindracea lipase was studied by various triacylglycerols with different fatty acyl chains as substrate. The substrate was emulsified with gum arabic and the rate of hydrolysis was determined by pH stat method. The effects of gum concentration, pH, temperature, and $Ca^{2+}$ ion on the enzyme activities were examined. The results show that the effect of these factors are markedly depending on the structurla nature of substrates. The triolein was the best substrate among tested. Present study demonstrates that for characterization of lipolytic enzymes, it is critically important to select proper substrate and activator.r.

  • The Korean Euodia daniellii $H_{EMSLEY}$ seed oil of triglyceride composition were analysed by thinchrography. Responses of sterol, triglyceride, sterol ester and acetate on a thin layer chromatographic apparatus, which was equipped with a flame ionization detector were determined. It was found that thinchrography is very superior and convenient means for the estimation of seed oil triglyceride compositions. The triglyceride compositions were tripalmitin 7.3%, tristearin 1.8%, triolein 21.1%, trilinolein 32.9%, trilinolenin 33.4% and unknown 3.5% in the case of Euodia daniellii seed oil in Korea.

Use of New Passive Sampler to Assess PAHs Contamination (새로운 생물농축 기법에 의한 PAHs의 오염도 확인)

  • A new passive sampler was developed, improving SPMD (Semipermeable Membrane Devices) that contained triolein in nonporous, low-density polyethylene layflat tubing. Experiments to measure PAHs concentration were carried out at four sites (Dukpo, Sochi, Sohwonggando, Yeonmok) on the southern coast of Korea that were contaminated by oil spills. Passive samplers were deployed at 4 and 8 week intervals at each site. Results showed that bioconcentration levels of PAHs were increased in proportion to exposure duration from all sample sites and accumulation in the passive sampler was much higher than in biota at the Dukpo site. Results of these tests suggest that new passive sampler is a useful tool for measuring bioconcentration organic compounds in aquatic environments.

Gene Cloning, High-Level Expression, and Characterization of an Alkaline and Thermostable Lipase from Trichosporon coremiiforme V3

  • The present study describes the gene cloning and high-level expression of an alkaline and thermostable lipase gene from Trichosporon coremiiforme V3. Nucleotide analysis revealed that this lipase gene has an open reading frame of 1,692 bp without any introns, encoding a protein of 563 amino acid residues. The lipase gene without its signal sequence was cloned into plasmid pPICZαA and overexpressed in Pichia pastoris X33. The maximum lipase activity of recombinant lipase was 5,000 U/ml, which was obtained in fed-batch cultivation after 168 h induction with methanol in a 50 L bioreactor. The purified lipase showed high temperature tolerance, and being stable at 60℃ and kept 45% enzyme activity after 1 h incubation at 70℃. The stability, effects of metal ions and other reagents were also determined. The chain length specificity of the recombinant lipase showed high activity toward triolein (C18:1) and tripalmitin (C16:0).
