폭약 TNT 분해세균 Stenotrophomonas sp. OK-5에서 분리된 NAD(P)H-nitroreductase의 정제 및 특성 연구 (Characterization of NAD(P)H-nitroreductase Purified from the TNT-degrading Bacterium, Stenotrophomonas sp. OK-5)

  • 2,4,6-Trinitrotoluene (TNT)을 분해할 수 있는 Stenotrophomonas sp. OK-5에서 분리한 NAD(P)H-nitroreductase의 특성을 조사하였다. 먼저 ammonium sulfate precipitation, DEAE-sepharose, 그리고 Q-sepharose 등의 일련의 정제 과정을 통하여 NAD(P)H-nitroreductase을 분리정제하였다. 분취기로부터 얻어진 시료로부터 NAD(P)H-nitroreductase의 효소활성을 가지는 3개의 다른 fractions (I, II 및 III)이 탐침되었다. NAD(P)H-nitroreductase의 fractions I, II, 그리고 III의 비활성(specific activity)은 각각 5.06 unit/mg, 4.95 unit/mg, and 4.86 unit/mg이었으며, crude extract와 비교하여 각각 10.5배, 9.8배, 8.9배 이상 농축되었다. 이 실험에서 이들 3개의 fractions 가운데, fraction I이 가장 높은 비활성을 나타내었다. NAD(P)H-nitroreductase (fractions I, II 및 III)의 효소활성에 영향을 미치는 몇 가지 요인을 조사하였다. 모든 NAD(P)H-nitroreductase (fractions I, II 및 III)의 최적 온도는 30$^{\circ}C$이었으며, 최적 pH는 약 7.5이었다. 4Ag^+, Cu_2^+, Hg_2^+$ 등의 금속이온은 약 80%의 효소활성을 저해하였으나, $Mn_2^+ 이나 Ca_2^+$의 첨가 시에는 약 30~40% 정도의 활성이 감소되었다. 그러나 $Fe_3^+$은 이들 효소의 활성을 증진시켰다. SDS-PAGE에 의해 측정된 NAD(P)H-nitroreductase의 fractions I, II 및 III의 분자량은 모두 약 27 kDa임이 확인되었다.

  • LOx와 Kerosene을 연료로 사용하는 LRE(Liquid Rocket Engine) Block에서 지상연소시험을 수행할 경우에 발생하는 위험도를 감소시키는 방법을 연구하였다. 추진제누출로 이어지는 위험한 상황에 대한 원인분석도와 추진제 유출시 사고로 이어지는 발전 양상 및 발전 프로세스, 사고시 손실을 줄이기 위한 기술적인 해결방법을 제시함으로써 기존의 확률적인 통계 값을 바탕으로 하는 것과는 또 다른 방법으로써 위험을 감소하기 위한 대안으로 사용하고자 한다.

오염물질 확산방지를 위한 운영중 군 사격장 환경관리방안에 대한 고찰 (An Environmental Management Protocol for the Mitigation of Contaminants Migration from Military Operational Ranges)

  • Pollutants such as heavy metals and explosives originating from the military operational ranges can be migrated to adjacent surface water body or offsite soil, and can affect to local residents and aquatic ecosystem. Therefore, Korea Ministry of the National Defense has established various guidelines for environmental management including the installation of pollutant migration prevention facilities (PMPFs) and monitoring methodologies for heavy metals in the operational range soil and effluent and sediment of PMPFs. However, current guidelines neither address the explosive compounds such as 2, 4, 6-trinitrotoluene (TNT) and hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) nor suggest detailed environmental investigation protocol. This paper introduces the new “Environmental Management Manual for Military Firing Ranges”, which includes the environmental criteria for explosives as well as the detailed investigation protocol for the affected environmental media including soil, effluent and sediment of PMPFs.

환경요인이 $Fe^0$ 에 의한 TNT의 환원 반응속도에 미치는 영향 (The Effects of Environmental Conditions on the Reduction Rate of TNT by $Fe^0$)

  • The effects of environmental conditions, initial dissolved oxygen concentrations, pH, and the presence of electron carrier vitamin B$_{12}$ , on the reduction rate of TNT by Fe$^{0}$ was Quantitatively analyzed using a batch reactor. In all experiments, TNT reduction was best described with a first order reaction and the reduction rate decreased with the increase in the initial DO concentration. However, the specific reaction rate did not decrease linearly with the increase in the initial DO concentration. In the presence of HEPES buffer 0.2 and 2.0 mM(pH 5.7$\pm$0.2), the specific reaction rate increased more than 5.8 times, which showed reduction rate is rather significantly influenced by the pH of the solution. To test the possibility of reaction rate enhancement, well-known electron carrier(or mediator), vitamin B$_{12}$ has augmented besides Fe$^{0}$ . In the presence of 8.0 $\mu\textrm{g}$/L of vitamin B$_{12}$ , the specific reaction rate increased as much as 14.6 times. The results indicate that the addition of trace amount of vitamin B$_{12}$ can be a promising rate controlling option for the removal of organics using a Fe$^{0}$ filled permeable reactive barrier.

Mass Spectrometric Analysis of Eight Common Chemical Explosives Using Ion Trap Mass Spectrometer

  • Eight representative explosives (ammonium perchlorate (AP), ammonium nitrate (AN), trinitrotoluene (TNT), 2,4-dinitrotoluene (DNT), cyclonite (RDX), cyclotetramethylenetetranitramine (HMX), pentaerythritol tetranitrate (PETN), and hexanitrostilbene (HNS)) were comprehensively analyzed with an ion trap mass spectrometer in negative ion mode using direct infusion electrospray ionization. MS/MS experiments were performed to generate fragment ions from the major parent ion of each explosive. Explosives in salt forms such as AP or AN provided cluster parent ions with their own anions. Explosives with an aromatic ring were observed as either $[M-H]^-$ for TNT and DNT or $[M]^{{\cdot}-}$ for HNS, while explosives without an aromatic ring such as RDX, HMX, and PETN were detected as an adduct ion with a formate anion, i.e., $[M+HCOO]^-$. These findings provide a guideline for the rapid and accurate detection of explosives once portable MS instruments become more readily available.

TNT and Nutrients Removal in the Constructed Wetlands

  • This study was carried out to investigate the removal of 2,4,6-trinitrotoluene, nutrients such as nitrogen, phosphorous compounds in the wetland microcosms. Microcosm study indicated that TNT nutrients were more lastly reduced in the anaerobic condition. The major reductive transformation products included 2,4diamino-6-nitrotoluene (2,4-DANT) 2,6diamino-4-nitrotoluene (2,6-DANT) 4diamino-2,6-nitrotoluene (4-ADNT) 2diamino-4,6-nitrotoluene (2-ADNT). The experimental results for nitrogen removal showed that denitrification kom NO$\sub$3//$\^$-/ to NH$_3$ was dominant process at the bottom of marsh, but nitrification from NH$_3$ to NO$\sub$3//$\^$-/ was dominant on the top of marsh.

Effect of C/N Ratio on Composting Treatment of TNT-Contaminated Soil

  • This research was conducted to estimate the effect of C/N ratio control on composting of TNT (2,4,6 trinitrotoluene)-contaminated soil. Glucose or acetone was selected to control C/N ratio of the contaminated soil. The C/N ratios of the controlled experiment and no controlled one were 26.0 and 6.6, respectively. During 45days, the degradation efficiency (96.0 or 91.8%) of acetone or glucose C/N ratio controlled soil was higher than that (78.4%) of no C/N ratio controlled case. The first order degradation rate constant of glucose or acetone C/N ratio control was 0.0641 or 0.0820/day. This constant was over twice 0.0356/day of no C/N ratio control. The C/N ratio control with glucose or acetone also showed a rather high $CO_2$ evolution than that without C/N ratio control. It was proven that C/N ratio control for composting of TNT-contaminated soil improved the treatment efficiency.

Numerical analysis of tunnel in rock with basalt fiber reinforced concrete lining subjected to internal blast load

  • The present study focuses on the performance of basalt fiber reinforced concrete (BFRC) lining in tunnel situated in sandstone rock when subjected to internal blast loading. The blast analysis of the lined tunnel is carried out using the three-dimensional (3-D) nonlinear finite element (FE) method. The stress-strain response of the sandstone rock is simulated using a crushable plasticity model which can simulate the brittle behavior of rock and that of BFRC lining is analyzed using a damaged plasticity model for concrete capturing damage response. The strain rate dependent material properties of BFRC are collected from the literature and that of rock are taken from the authors' previous work using split Hopkinson pressure bar (SHPB). The constitutive model performance is validated through the FE simulation of SHPB test and the comparison of simulation results with the experimental data. Further, blast loading in the tunnel is simulated for 10 kg and 50 kg Trinitrotoluene (TNT) charge weights using the equivalent pressure-time curves obtained through hydrocode simulations. The analysis results are studied for the stress and displacement response of rock and tunnel lining. Blast performance of BFRC lining is compared with that of plain concrete (PC) and steel fiber reinforced concrete (SFRC) lining materials. It is observed that the BFRC lining exhibits almost 65% lesser displacement as compared to PC and 30% lesser displacement as compared to SFRC tunnel linings.

축척법에 의한 화약 폭풍과 분석 (Analysis of Blast Wave of Explosives by the Scaling Law)

  • To analyze a blast effect of developed explosives, three different kinds of aluminized tastable explosives and melted cast explosive TNT were used. Conventional explosive TNT was used as a reference. Each tested explosive charge of 340mm diameter spherical type was initiated at the charge center with DXD-65(${\sim}750g$) booster and RP-87 EBW detonator. Thirteen piezo type pressure sensors were located at a range from 4 to 50m away from the charge. From the blast wave profiles, we calculated a peak blast pressure and impulse of the explosion. The calculated pressures and in pulses were converted to TNT Equivalent Weight(TEW) factor by the scaling ]aw method. The average TEW factors based on the blast pressure of TX-01, TX-02, TX-03, TX-04 were 1.298, 1.05, 1.266, 1.274 and the average TEW factors based on impulse were 1.504, 1.686, 1.640, 1.679. From the results, we concluded that TEW factors based on blast pressure and based on impulse of aluminized explosives were superior to TNT. This results are owing to the high contents of aluminum in formulations.

Treatment of TNT and Parathion Using Constructed Wetland Microcosms

  • This study was carried out to investigate the removal of TNT (2,4,6-trinitrotoluene) and parathion in the batch and the continuous constructed wetland microcosms consisting of marsh and pond. The batch system study showed that TNT was almost reduced in the marsh and pond system within 20 days and parathion was within 8 days. The major reductive metabolites of TNT includes 2,4-diamino-6-nitrotoluene (24DANT) >2,6-diamino-4-nitrotoluene (26DANT) >4-diamino-2,6-nitrotoluene(4ADNT) > 2-diamino-4,6-nitrotoluene (2ADNT), and the concentrations of these metabolites were decreased during further operation. The generation rates of 4-nitrophenol, the major metabolite of parathion, were 82% and 15% in the bottom of marsh and pond system, respectively. In the continuous system study, although TNT/parathion degradation pattern was similar to the batch's, marsh-pond system showed the most stable TNT/ parathion removal among various continuous reactor combinations.
