  • Our purpose was to investigate the effect of a 5-week high-fat diet on the body weight, blood components [triglycerides (TGs), free fatty acids (FFAs), and glucose], and lipoprotein lipase (LPL) mRNA expression in the skeletal muscles of rats. Body weight increased overtime in experimental and control groups without significant differences. In terms of the blood components, the density of TG was significantly lower in the high-fat diet group compared to the control, whereas FFA and glucose levels were similar in the two groups. Although the levels of glyceraldehyde 3-phosphate dehydrogenase(GAPDH) mRNA expression were similar between the groups, LPL mRNA expression was significantly higher in the high-fat diet group than that in the control group. The results of this study suggest that a high-fat diet enhances LPL mRNA expression, as well as possibly increases fat metabolism. For a better understanding of the relationship between diet patterns and fat metabolism, further analysis of genes related to the fat metabolism is warranted.

  • In this study, we investigated in vitro inhibitory activity of wild-simulated ginseng (WSG) against non-alcoholic fatty liver disease using HepG-2 cells. T0901317 treatment increased the lipid accumulation in HepG-2 cells, but WSG treatment inhibited T0901317-mediated lipid accumulation. In addition, WSG downregulated T0901317-mediated expression of SREBP-1c, ACC, FAS and SCD-1 protein. In addition, WSG increased the phosphorylation level of LKB1 and AMPK. Compound C treatment blocked WSG-mediated downregulation of SREBP-1c protein. In conclusion, WSG is considered to inhibit the accumulation of lipids and triglycerides in HepG-2 cells by inducing the activation of LKB1 and AMPK successively, thereby reducing the expression of FAS, ACC, and SCD-1 through suppression of SREBP-1c expression.

  • In this study, the lipid components of three species of shellfish included oyster(Crassostrea gigas), top shell(Turbo cornutus) representing salt water shellfish and corb shell(Corbicula fluminea producta) representing flesh water shellfish were analysed and nutriontional significances were discussed. Analysed the total lipid composition, and the fatty acid and sterol composition of total lipid were determined. The lipid was fractionated into three lipid classes neutral, glyco and phospholipid by column chromatography. The fatty acid composition of each lipid class and sterols were determined by gas liquid chromatography. The lipid components of total lipid and neutral lipid were estimated by thin layer chromatography and TLC scanner. The results were as follows: Total lipid contents of shellfish were 1.8% in oyster, 0.4% in top shell and 4.0% in corb shell. The contents of total fatty acid in total lipid were 80.7, 71.2 and 73.2%; and the contents of unsaponifiable matters were 15.4, 18.1 and 23.1% respectively. Total lipids were mainly composed of triglycerides, polar lipid-pigments and sterols as major component, and hydrocarbon-esterified sterols were determined in each sample. The major fatty acids in total lipid were palmitic(37.0%), eicosapentaenoic(13.5%) and linoleic acid(11.2%) in oyster, Octadecatetraenoic(15.8%), palmitic(11.2%), oleic(8.6%) and linoleic acid(8.1%) in top shell, but palmitic(34.0%), linoleic(12.3%) and paimitoleic acid(9.8%) in corb shell. Particularly, the contents of eicosapentaenoic acid of oyster and top shell were higher than those of corb shell. Sterol composition from three species of shellfish were mainly consisted of cholesterol (42.7~64.0%), brassicasterol(15.6~24.7%) and 24-methylenecholesterol (4.7~21.9%). But sitosterol (5.3%) was detected only in oyster and 22-dehydrocholesterol(12.9%) was only in top shell. The contents of fractionated neutral lipid was commonly higher than that of polar lipid in each sample. Glycolipid and phospholipid in polar lipid showed similar in quantity. The neutral lipids were composed of triglycerides(33.0~36.7%), free sterols(25.7~31.2%), esterified sterol(12.4~23.7%) and free fatty acids(5.1~11.7%). The contents of triglycerides and free sterols were higher than those of free fatty acids and esterified sterols. The major fatty acids in neutral lipid were palmitic(28.4~26.4%) eicosapentaenoic(18.6~21.9%) and linoleic acid(9.0~5.4%) in oyster and corb shell but octadecatetraenoic(14.5%), eicosapentaenoic (13.5%) and palmitic acid(12.3%) in top shell. • Total lipids from the roe, muscle and viscus of L. carinata were analyzed for lipid composition by column chromatography, thin-layer chromatography and gas-liquid chromatography. The roe lipids were characterized by a high level of wax esters (63.1%) and a low proportion of trigiycerides (9.9%). The viscus lipids also contained wax esters (32.8%) as its main component, followed by free fatty alcohols and acids (23.5%). On the other hand, the muscle lipids were found to contain a large amount of triglycerides (66.1%) with a trace of wax esters. The main fatty alcohol component of roe and viscus wax esters was C16:0 alcohol (53.0%; 61.7%), accompanied by C18:1 alcohol (10.2%) in the former and by C15:0 alcohol (8.8%) in the latter. Considerable amounts of odd-numbered fatty alcohols were found in both wax esters. On the other hand, the fatty acids of the roe and viscus wax esters contained a high percentage of monounsaturated (49.7%-56.6%) consisting of C16:1, C18:1 and C17:1 acid, and a significant amount of polyunsaturated (41.2%-32.9%), particularly C20:5${\omega}$3. The fatty acid components of triglycerides and phospholipids were different among the tissues tested, especially between roe and muscle or viscus. The fatty acid compositions of free fatty acids from the muscle and viscus were characterized by a higher level of polyunsaturated fatty acids (46.0-34.3%) compared to those of triglycerides 'in the roe, muscle and viscus (28.4%, 19.4% and 19.2%).

  • The effects of Cordyceps militaris on weight gain, flood intakes, flood efficiency ratios, serum and hepatic lipid concentrations, serum protein levels and serum enzyme activities, were studied in adult male rats. Sprague-Dawley rats, 35 weeks old, were given four different types of diets for a succeeding period of five weeks: either a normal diet(5% com oil), a high fat diet(5% com oil + 15% lard). a CF diet (high fat diet + 3% Suiting body of cordyceps militaris), or a CM diet (high fat diet + 3% mycelium of cordyceps military). The body weight gains, hepatic weights and food efficiency ratios of the rats find (high fat) diets with CF and CM were significantly decreased compared with those find the high fat diet, but were similar to those find the normal diet. The concentrations of serum and hepatic triglycerides, and hepatic total lipids and cholesterol, of rats given CF or CM diets were significantly lower than those given the high fat diet. But the concentrations of total cholesterol, HDL-cholesterol, phopholipid and total protein in the serum of rats find the high fat, CF or CM diets were significantly higher than those find the normal diet. In the serum of the rats find the CF diet, the HDL-cholesterol/total cholesterol ratio was significantly higher but the atherogenic index was significantly lower than those fed the high fat diet. The alkaline phosphatase activity in the serum of the rats find the high fat, CF, and CM diets was more significantly decreased compared to rats on the normal dirt. No differences were noted in the weights of the pancreas, kidney and heart, the serum concentrations of glucose, hemoglobin and albumin, and the activities of GOT, GPT and ${\gamma}$-GTP, among the rats on all the experimental diets. In conclusion, the rats find the Cf or CM diets maintained normal body and hepatic weights. Despite of the high intake of fats in the CF and CM diets, the concentrations of total lipids, cholesterol and triglycerides were decreased in the liver, also, the concentration of triglycerides was decreased in the serum.

  • An experiment was conducted to study the performance, carcass traits, serum lipid profile and immune competence in commercial broilers (2 to 42 d of age) fed graded levels (25, 50, 75 and 100%) of finger millet (FM) (Elusine coracana) in place (w/w) of yellow maize (YM). Each diet was fed to eight replicates (five female Vencobb broilers/replicate) housed in stainless steel battery brooders. The estimated metabolizable energy content of FM was about 540 kcal less than the YM. FM contained more protein (10.42 vs. 9.05%) and fibre (9.52 vs. 2.24%) compared to YM. Body weight gain, ready to cook yield, relative weights of giblet, liver, intestine and length of intestine at 42 d of age was not affected due to replacing YM with FM. But, the feed efficiency decreased in broilers fed diets containing 75 and 100% FM in place of YM at both 21 and 42 d of age. The amount of fat deposited in abdominal area decreased and the relative weight of gizzard increased with increase in level of FM in the diet. The serum HDL cholesterol at 21 and 42 d of age and serum triglycerides at 42 d of age decreased with increase in level of FM in diet. The relative weight of spleen and antibody titers against sheep red blood cells (SRBC) at 5 d post inoculation (PI) decreased in broilers fed FM at 100% of YM. However, the relative weight of bursa, SRBC titers at 10 d PI, antibody titers against ND virus and mortality were not affected due to incorporation of FM in place of YM in diet. The fat content in thigh muscle and liver decreased, while the protein content in these tissues increased with increase in the level of FM in broiler diet. Based on the results, it may be concluded that YM can be replaced with FM up to 25% on weight basis without affecting weight gain, carcass yields and immunity in commercial broiler diet (up to 42 d of age). Further, inclusion of finger millet reduced the fat deposition in thigh muscle, liver and in abdominal area compared to those fed maize as the principal source of energy.

  • This study investigated the effects of feeds naturally contaminated with mycotoxins on growth performance, serum biochemical parameters, carcass traits, and splenic heat shock protein 70 (Hsp70) mRNA expression levels in broiler chickens. The efficacy of yeast cell wall (YCW) adsorbent in preventing mycotoxicosis was also evaluated. Three hundred 1-d-old Arbor Acres broiler chicks were randomly allotted to 3 treatments in completely randomized design for 42 d. Each treatment group had 5 replicate pens with 20 birds. The treatments were as follows: i) basal diet (control), ii) naturally contaminated diet (NCD), and iii) NCD+0.2% YCW adsorbent (NCDD). The NCD decreased average daily gain (ADG) (p<0.01) of 0 to 21 d, 22 to 42 d, and 0 to 42 d, and increased feed conversion ratio (p<0.01) of 22 to 42 d and 0 to 42 d. Both the breast meat percentage and thigh meat percentage of the NCD group were significantly higher (p<0.01) than that of the control group on d 21. The NCD group showed significantly increased levels of triglycerides (p<0.05) and cholesterol (p<0.05) on both d 21 and d 42 compared to the control group. However, the NCD significantly reduced (p<0.01) the high-density lipoprotein (HDL) on d 42 compared to controls. Compared with the NCD, supplementation with YCW significantly improved (p<0.01) the ADG of 0 to 21 d and 0 to 42 d, and increased (p<0.01) concentrations of HDL on d 42, and on d 21, and triglycerides (p<0.05) on d 21 and d 42. Supplementation with YCW reduced (p<0.01) the breast meat percentage, the thigh meat percentage, the concentrations of cholesterol (p<0.01) and the low-density lipoprotein (p<0.05) on d 21, and improved (p<0.01) the splenic Hsp70 mRNA expression levels compared with the NCD group. The results of this study indicated that feeding NCD for 42 d had adverse effects on broiler chickens, and that YCW might be beneficial in counteracting the effects of mycotoxins.

  • Bokbunja (Rubus coreanus) is a Korean fruit and wild black raspberry that has antioxidant, anticancer, and beauty effects due to its abundant polyphenols and anthocyanins. The purpose of this study was to investigate the blood flow improvement effect of Bokbunja seed oil (BSO) in a high-fat diet-fed mouse model. We examined improvement of blood flow and its related biomarkers in vivo. Mice were divided into four groups; Control, high fat diet control (negative control, HFD), salmon oil control (positive control, HFD+commercial n-3 fatty acid), and BSO experiment groups (HFD+2 g/2,000 kcal, HFD+4 g/2,000 kcal). After the mice were sacrificed, plasma triglyceride, cholesterol, and blood flow-related biomarkers (coagulation factor 7, 12, serotonin, TXB2, PT, and aPTT) were measured in mouse blood and organs. BSO reduced blood viscosity through improvement of blood lipids (cholesterol and plasma triglycerides) as well as levels of blood coagulation factors and blood platelet activity. BSO also delayed blood coagulation time. Thus, we confirmed that BSO inhibits excessive blood clotting of blood vessels and improves blood flow. Taken together, these results suggest that BSO decreases plasma triglycerides and cholesterol and improves blood flow by regulating biomarkers.

  • This study was conducted to develop a pasture feeding program for reducing the Hanwoo feeding cost. Italian ryegrass (IRG) is a major plant which is abundantly produced in the southern area of South Korea. Locally produced Italian ryegrass has a 30 - 50% reduced pasture cost compared to imported pasture grasses. We hypothesized that Italian ryegrass feed could reduce the feeding cost but not change the physiological characteristics of the Hanwoo steers in the feedlot. Four treatments (Control, short fed IRG silage, long fed IRG silage, and IRG hay) in a completely random design were used to feed 27 Hanwoo steers. Three or four steers were fed in the same pen, and 8 pens were used for the treatments. Blood was drawn from each steer on the first week of every month from 11 to 30 months. Overall, the average daily gain and feed efficiency were not different among the treatments (p < 0.05) except for the final fattening diets (p < 0.05). The long fed IRG silage group had a low ADG compared to the other groups (p < 0.05). The serum non-esterified fatty acid and glucose concentrations were reduced during the fattening periods (p < 0.05); however, the serum albumin, total proteins, triglycerides, and phosphorus were increased during the fattening periods (p < 0.05). The carcass weight, longissimus muscle area and back-fat thickness were low in the long fed IRG silage group (p < 0.05). The IRG diet induced saturated fatty acids in the longissimus dorsi (p < 0.05). These results show that the IRG diet increased the serum triglycerides but reduced the glucose level at the final fattening diet and decreased the meat yield score in Hanwoo steers.
