  • (1) A potent lypolytic strain was selected by an extensive screening test of microorganisms isolated from air and various soils on the medium containing olive oil as sole source of carbon. (2) Morphological and physiological characteristics of the selected strain had been investigated, and it has been identified to Trichosporon cutaneum in the manual of LODDER.

On the CMCase Activity from Two species of Trichosporon (Trichosporon의 CMCase 활성에 관하여)

  • Dennis (1972) reported that Trichosporon cutaneum FRI-425 from the petioles of Pheum rhamponticum var, had showed the celluloytic activity. Chun (1977) also suggested that Trichoporon pullulons 225 isolated from the saline water of the Yeoung San River had a similar properties. However, the assay conditions for enzyme activity were not yet investigated. Thus, the present work was undertaken to examine some conditions for CMCase activity and at the same time to compare the activities of crude enzyme produce from above two species of Trichosporon pullulans. The results are as follows; 1. The maximum production of total reducing sugar by crude enzyme of Tr. pululans was after 30 minutes, whereas that of Tr. cutanuem FRI-425 was after 90 minutes. This fact showed that the reaction velocity of enzyme from Tr. pullulans 225 was more faster than that of Tr. cutaneum FRI-425. 2. Two species showed a similar trend to increase the production of reducing sugar in proportion to the increment in substrate concentration and to arrive at maximum level at lmg/ml of substrate concentration. However, Tr. pullulans 225 produced more $50{\mu}g$ of reducing sugar compared to Tr. cutaneum. 3. The optimum PH for CMCase activity is 5.0 for Tr. pullulans 225 as well as Tr. cutaneum FRI-425, and PH stability lie within the range of 6 and 8. In the activity and stability of enzyme on PH changes, enzyme of Tr. cutaneum FRI-425 was more unstable than that of TY. pullulans 225. 4. The optimum temperature for CMCase activity was $40^{\circ}C$, and enzyme activity from Tr. pullulans 225 was more sensitive to temperature changes compared with that of TY. cutaneum. The heat stability was within $40^{\circ}C$, but that was rapidly decreased above $40^{\circ}C$. In comparison of the heat stability for enzyme of Tr. cutaneum FRI-425 with that of Tr. pullulans 225 at the same temperature of $80^{\circ}C$, the former was some 10 percent more stable than the latter.

  • This study was conducted to clarify the mode of action and the positional specificity of Trichosporon cutaneum lipase during the course of hydrolysis of triolein and monoolein mixture by thin-layer chromatography. 1. The hydrolytic activity of the lipase to oleyl glycerides was in the order of triolein>diolein>monoolein. 2. Both of triolein and diolein were hydrolyzed by the lipase at high and almost the same rate. 3. The hydrolysis of monoolein by the lipase was very slow compared to the other two oleyl glycerides. 4. This lipase appeared to have a very low specificity toward the outer chains of triolein.

  • We report the first case of summer-type hypersensitivity pneumonitis(SHP) in Korea diagnosed by positive serum antibodies to Trichosporon cutaneum. Hypersensitivity pneumonitis(HP) has been commonly classified as an occupational respiratory diseases. However, evidence that sensitizing organisms can also contaminate and cause pulmonary diseases in home environment has been increasing. One such disease is SHP. In Japan, 75% of cases with HP are SHP. Even though there has been no known SHP case in Korea yet, there has been high possibility of SHPs in Korea because our country has areas which have hot and humid summer climate similar to Japan. This first case of SHP in Korea suggests that there may be another cases in Korea and nation-wide survey may be required. We report here the first confirmed case of SHP in Korea.

Effect of Sugars on Cell Growth and Lipase Production by Trichosporon cutaneum (Trichosporon cutaneum의 균체생육(菌體生育) 및 Lipase 생산(生産)에 미치는 당류(糖類)의 영향)

  • Inhibitory effect of sugars on lipase production by Trichosporon cutaneum was observed in the previous study (Kim, 1972), and inhibition was distinctive by the addition of glucose, fructose, mannose, xylose and arabinose to the soybean meal medium among various carbon sources. These experiments were carried out to study the effect of sugars on cell growth and lipase production by the strain using the soybean extracts liquid medium under a shaking culture system. Changes in color and pH of the medium were caused by heat sterilization when various sugars were added. To elucidate the possible effect of these coloring matters on lipase production and cell growth: changes in pH of the culture, cell concentration and level of the enzyme activities were determined when the culture was grown for 48 hours at $30^{\circ}C$ on a reciprocal shaker. The results obtained were as follows: 1. Density of brownish color which formed during heat sterilization was varied with the variety of sugar used, ie, strong in pentose such as xylose: weak in hexose such as galactose, mannose, glucose: very weak in disaccharide such as maltose, sucrose. When the color density was stronger, decrease in pH after sterilization was marked. 2. Cell growth and lipase production was not so effect by the coloring matters as by sugars. 3. The more the cell mass of the culture, the lower the level of lipase production in the culture supernatant. 4. Among the sugars which caused the distinctive inhibition of lipase production, a slight relief of inhibition was noticed by the addition of xylose, whereas the cell growth was repressed. 5. When cell growth was better, decrease in pH of the medium was greater during cultivation.

  • A mixed culture of Candida tropicalis and Trichosporon cutaneum was carried out using a n-paraffin medium. The growth of C. tropicalis was markedly enhanced by the mixed culture with T. cutaneum which did not grow on n-paraffin. C. tropicalis extracellularly excreted free fatty acids as metabolic products of n-paraffin in the culture medium. T. cutaneum appeared to assimilate these free fatty acids which were growth inhibitors for C. tropicalis, threreby enhancing the growth of C. tropicalis.

  • A microbial BOD sensor for the continuous estimation of BOD was been developed by immobilizing Trichosporon cutaneum, which was immobilized between a dialysis and a gas-permeable membrane, on an oxygen electrode. The optimum pH and temperature for BOD measurement using this sensor were pH 7.0 and $32{\sim}33^{\circ}C$, respectively. The best result was obtained at 2~3ml/min flow rate in 0.1M phosphate buffer solution. A linear relationship was observed between ${\Delta}DO$ and the concentration of standard GGA solution below 60mg/l(90ppm $BOD_5$). The reproducibility was found to be within 3% for the standard solution containing glucose 30mg/l and glutamic acid 30mg/l. The output DO value of this sensor was almost constant for 30 dalys. The response time and the recovery time were about 5 and 10 min, respectively. This sensor was employed for the BOD measurement of waste pollutants and was compared with $BOD_5$ method.

  • For the production of single cell protein from n-paraffin, yeasts utilizing n-paraffin and ethanol were isolated from oil deposit and oil field soils. The mixed cultivation between yeasts assimilating n-paraffin and ethanol was carried out to increase cell yield. Finally, selected strains were identified and suitable medium composition for mixed culture was compared with that of single cultures using flask and 5 l-jar fermentor. Yeasts grow on n-paraffin and ethanol were identified as Candida tropicalis var. KIST 76 and Trichosporon cutaneum KIST 76H respectively. By mixed cultivation under the suitable medium composition using 5 l-jar fermentor, maximum dry cell weight reached 20 g/l after 12 hrs. cultivation and it's protein content was 58%. Yield has been increased about 25% and protein content has been increased 6.7% compared to that of single culture, Candida tropicalis var. KIST 76, after 16 hrs. cultivation.
