Trichophyton rubrum 및 Trichophyton raubitschekii에 의한 개 피부사상균증에 관한 연구 (Dermatophytosis by Trichophyton rubrum and T raubitschekii in dogs)

  • This study was carried out to determine the causative agent and the clinical features of dermatophytosis occurred in 3 dogs from Tae-gu, Korea between 1996 and 1997. Specimens of hair and scale, collected from skin lesions were inoculated on potato dextrose agar and mycobiotic agar supplemented with thiamine and inositol. The agar plates were incubated at $25^{\circ}C$ for 2 weeks. Growing fungi isolated were identified by the morphological and growth characterization on bromocresol purple-milk solids-glucose medium, Christensen urea broth and trichophyton media, and by hair perforation tests. Skin lesions were observed in legs, abdomen, ears, face and head with alopecia of various sizes accompanied by itching and/or inflammation. The causative agent of dermatophytosis in the 3 dogs was identified as Microsprum gypseum and Trichophyton rubrum (mixed infection, dog 1), T rubrum (dog 2) and T raubitschekii (dog 3). The dermatophytosis by T rubrum and T raubitschekii was first found on animals in Korea.

In vitro Antifungal Activity of Limonene against Trichophyton rubrum

  • In this study, the antifungal activities of limonene against Trichophyton rubrum were evaluated via broth microdilution and vapor contact assays. In both assays, limonene was shown to exert a potent antifungal effect against T. rubrum. The volatile vapor of limonene at concentrations above $1{\mu}l$/800 ml air space strongly inhibited the growth of T. rubrum. The MIC value was 0.5% v/v in the broth microdilution assay. The antifungal activity of limonene against T. rubrum was characterized as a fungicidal effect.

Genetic Variations of Trichophyton rubrum Clinical Isolates from Korea

  • Trichophyton rubrum is one of the well-known pathogenic fungi and causes dermatophytosis and cutaneous mycosis in human world widely. However, there are not an available sequence type (ST) classification methods and previous studies for T. rubrum until now. Therefore, currently, molecular biological tools using their DNA sequences are used for genotype identification and classification. In the present study, in order to characterize the genetic diversity and the phylogenetic relation of T. rubrum clinical isolates, five different housekeeping genes, such as actin (ACT), calmodulin (CAL), RNA polymerase II (RPB2), superoxide dismutase 2 (SOD2), and ${\beta}$-tubulin (BT2) were analyzed using by multilocus sequence typing (MLST). Also, DNA sequence analysis was performed to examine the differences between the sequences of Trichophyton strains and the identified genetic variations sequence. As a result, most of the sequences were shown to have highly matched rates in their housekeeping genes. However, genetic variations were found on three different positions of ${\beta}$-tubulin gene and were shown to have changed from $C{\rightarrow}G$ (1766), $G{\rightarrow}T$ (1876), and $C{\rightarrow}A$ (1886). To confirm the association with T. rubrum inheritance, a phylogenetic tree analysis was performed. It was classified as four clusters, but there was little significant correlation. Even so, MLST analysis is believed to be helpful for determining the genetic variations of T. rubrum in cases where there is more large-scale data accumulation. In conclusion, the present study demonstrated the first MLST analysis of T. rubrum in Korea and explored the possibility that MLST could be a useful tool for studying the epidemiology and evolution of T. rubrum through further studies.

The Antifungal Activity of Bee Venom against Dermatophytes

  • The antifungal activities of the bee venom against Trichophyton mentagrophytes and Trichophyton rubrum were determined by using modified broth dilution assay. The most common dermatophytes, named T. mentagrophytes and T. rubrum, were known to cause a variety of cutaneous infections in humans and animals. The bee venom exhibited prominent antifungal activities against the two dermatophytes tested in this investigation. Moreover, the antifungal activities of the bee venom were much stronger than that of fluconazole, one of the commercial antifungal drugs used in the treatment and prevention of superficial and systemic fungal infections. The result suggests that bee venom could be developed as a natural antifungal drug.

키토산의 Candida albicans와 Trichophyton rubrum에 대한 항진균 작용과 Apoptosis 유도작용 (Antifungal Activity of Chitosans on Candida albicans and Trichophyton rubrum and its Induction of Apoptosis)

  • 키토산및 올리고키토산의 C. albicans과 T. rubrum에 대한 항 진균작용에 대하여 조사하였다. 키토산은 C. albicans과 T. rubrum에 대하여 항 진균 효과를 나타내었으나 올리고키토산은 두 균주에 대한 성장억제를 나타내지 않았다. 키토산의 항 진균 양상은 살 진균 적 활성을 나타내었다. 키토산의 apoptosis유도에 대한 조사를 위하여 phosphatidylserine의 세포막 외부로의 노출을 FITC-Annexin 반응을 이용하여 관찰하였다. 실험결과 키토산은 C. albicans의 세포에 대하여 apoptosis를 유도하는 것을 보여주었다.

  • This experimental study was performed to investigate the antifungal effects of mixture of Sophorac Subprostratae Radix, Aconti Radix, and Hibisci Syriaci Cortex by the three types of extraction in the dermatophytes such as Trichophyton rubrum, Trichophyton mentagrophytes, and Epidermophyton floccosum. The following results were obtained by using Minimum Inhibition Concentration(MIC) and Inhibition Zone. 1. MIC on Trichophyton mentagrophytes in Sample S-2, Sample S-3, and Sample-Y were 10${\mu}l$, respectively, showing the highest antifungal effect. 2. MIC on Epidermophyton floccosum in Sample S-2, Sample S-3, and Sample-Y were 10㎕ respectively, showing the highest antifungal effect. 3. MIC on Trichophyton rubrum in Sample S-3, Sample-Y were 10${\mu}l$ respectively, showing the highest antifungal effect. 4. The size of inhibition zone on Trichophyton mentagrophytes for Sample S-3 were 22.3mm in 10${\mu}l$, showing the highest antifungal effect. 5. The size of inhibition zone on Epidermophyton floccosum for Sample S-3 was 34.3mm in 10${\mu}l$, showing the highest antifungal effect. 6. The size of inhibition zone on Trichophyton rubrum for Sample-Y was 26.5mm in 10${\mu}l$, showing the highest antifungal effect. Whereas. in extractions, that of Sample S-3 was 12.5mm in 10${\mu}l$, showing the highest antifungal effect. 7. From the above results, Sample S-3 on Trichophyton mentagrophytes, Epidermophyton floccosum and Sample-Y on Trichophyton rubrum showed the highest antifungal effects. This results suggests that vinegar extraction excels other extraction method using water or ethanol to measure the antifungal effect on dermatophytes. In addition, the result shows that the extract of herbal medicines can be used to tinea pedis and if further study is performed, the use of the extract of herbal medicines will be valuable and beneficial in the clinical medicines.   • Dermatophytes can invade in keratinized tissues and cause dermatophytosis [1] that rank among the most widespread and common infectious diseases world-wide. Although several systemically and topically administered drugs with activities against these fungi are available, still complete eradication of some of these infections, is difficult and relapses and remissions are often observed [2,3]. In addition, some people are allergic to many of the available drugs which add complications even more. Therefore, the search for novel, selective and more effective therapy is always required and it may help the clinicians to choose the correct treatment for their patients. Non-thermal plasmas primarily generate reactive species and recently have emerged as an efficient tool for medical applications including sterilization. In this study, we evaluated the ability of non-thermal dielectric barrier discharge (DBD) plasma for the inactivation of clinical isolates of Trichophyton genera, Trichophyton mentagrophytes (T. mentagrophytes) and Trichophyton rubrum (T. rubrum), which cause infections of nails and skin and, are two of the most frequently isolated dermatophytes [4]. Our results showed that DBD plasma has considerable time dependent inactivation potential on both T. mentagrophytes and T. rubrum in-vitro. Furthermore, the mechanisms for plasma based T. mentagrophytes and T. rubrum inactivation and planning for in-vivo future studies will be discussed.

변형 키토산의 항균효과 (The Anti-Microbial Activity of Modified Chitosan.)

  • 게나 새우 등의 갑각류의 껍질에서 주로 얻어지는 생체적합성이 뛰어난 천연고분자인 chitosan에 음이온 수용성의 함인계 단량체인 mono(2-methacryloyl oxyethyl) acid phosphate를 그라프트 중합시켜 새로운 유도체인 chitosan-g-MAP을 합성하였다. 그라프트중합에서 개시제로는 ceric ammonium nitrate을 사용하여 개시제의 농도, 단량체의 농도, 반응온도, 반응시간에 따른 그라프트율, 단량체의 그라프트효율 및 단량체의 총전환율의 변화를 조사하여 최적조건을 규명하였다. 그라프트율은 개시제농도 3.5$\times$$10^{-3}$ M, 단량체농도 0.19 M, 반응온도 4$0^{\circ}C$에서 가장 높은 값을 나타내었고, 반응시간 4시간이 최적조건 이었다. Chitosan의 유리 아민의 함량과 그라프트된 단량체의 종류에 따른 항균성을 Candida albicans, Trichophyton rubrum 및 Trichophyton violaceum에 대하여 조사한 결과, pH 5.75에서 가장 우수한 항균능이 발현되고, 항균제의 농도에 따른 영향에서는 변형chitosan은 C. albicans와 T. violaceum에 대하여 100%의 우수한 항균력을 나타낸 반면, 탈아세틸화도가 다른 DA70과 DA-90인 chitosan은 T. rubrum균에 대해서만 변형 chitosan 보다 우수한 항균성을 나타내었다.

3종의 한국산 겨우살이 메탄올 추출물의 Tyromyces palustris, Endothia nitschkei 그리고 Trichophyton rubrum에 대한 항균활성 (Antifungal Activities of meOH Extracts from Three Korean Mistletose against Tyromyces palustris, Endothia nitschkei and Trichophyton rubrum)

  • 본 실험에서는 3종의 한국산 겨우살이 메탄올 추출물을 유기용매 분획과 컬럼크로마토그래피를 이용한 분획을 실시한 후, Tyromyces palustris, Endothia nischkei 그리고 Trichophyton rubrum에 대한 항균활성을 평가하여 겨우살이 추출물의 항균활성 자료로 활용하고자 하였다. 그 결과 모든 겨우살이 추출물은 갈색 불후균에 대한 활성은 매우 낮게 나타났다. 그러나 Viscum albumvar. coloratum 추출물중 부탄올 분획은 황색줄기 마름 병원균의 성장을 억제하는 것으로 나타났으며, 이 겨우살이의 EtOAC 분획은 족부백선균에 대하여 높은 포자 발아 억제효과를 나타내었다. 시험된 3종의 한국산 경우살이류 중 V. album var. Coloratum은 병원균에 대하여 황균활성 성분을 함유할 가능성이 가장 높을 것으로 제시되었다.

ITS-RFLP와 ITS1 염기서열 분석에 의한 피부사상균의 동정과 계통적 유연관계 (Identification and Phylogenetic Relationship of Dermatophytes Based on RFLP Analysis and Nucleotide Sequence of Internal Transcribed Spacer (ITS)1 in Nuclear Ribosome DNA)

  • ITSI-5.8S-ITSII rDNA region was amplified from the reference strains and clinical isolates with ITS1 and ITS4 primers. These primers amplified DNA fragments of 550 bp in Microsporum audouinii and Trichophyton violaceum, 700 bp in Microsporum gypseum, Trichophyton mentagrophytes, Trichophyton rubrum, and Trichophyton tonsurans, and 750 bp in Microsporum ferreugineum and Microsporum canis. The restriction enzyme patterns of PCR products digested with 13 restriction enzyme including PstI were distint among the genera, whereas identical in the same species. Examination of the ITS (Internal Transcribed Spacers)1 nucleotide sequence revealed that there was the genetic difference in each genera and species. Phylogenetic relationship among each species showed that the Trichophyton mentagrophytes was more closely related Trichophyton tonsurans than Trichophyton rubrum, and Microsporum gypseum was less related than Microsporum spp..
