The Effect of Potassium Cyanate (KCN) on Radiation Treatment of the Colorectal Cancer Cell Line, HCT 116

  • Radiation is one of the major therapy for the removal of cancer cells. The results of the radiation therapy depend on the radio-resistance of cancer cells. For the effective treatment in these radio-resistant cancers, the use of chemicals that act on cancer cells is known to enhance the cytotoxic effects of radiation therapy. In this study, I investigated the effect of potassium cyanate (KCN) on the irradiated-colorectal cancer cell line, HCT 116 cells. KCN induces the carbamylation of proteins and can change the biological activity of various human cells. To understand the effect of KCN on the radiosensitivity of HCT 116 cells, I examined alteration of the cell cycle, generation of reactive oxygen species (ROS), cell viability, apoptosis and intracellular signaling proteins in the irradiated cells with/without KCN treatment. Combination treatment caused significant increase in sub $G_0/G_1$ and ROS generation in HCT 116 cells. KCN inhibited the proliferation and cell viability in irradiated HCT 116 cells. KCN-induced apoptosis of irradiated cells was processed via the activation of caspase 3 and caspase 9. Apoptosis-associated signal proteins, including Bax and Bcl-2 were regulated by irradiation with KCN treatment. Taken together, these results may indicate that KCN enhances the radiosensitivity of radio-resistant cell and then has a synergistic effect on radiation therapy in colorectal cancer.

  • Objectives: The purpose of this study is to evaluate the effect of Hwangryunhaedock-tang on Atopic Dermatitis Methods: This study population was 71 pediatric allergic rhinitis patients who had been treated in ${\bigcirc}{\bigcirc}$ hospital. All the patients were asked to answer the questionnaire consisting of 8 questions about symptoms of atopic dermatitis. Results: 1. The total scores from the same questionnaire about atopic dermatitis symptoms were significantly improved after the treatment compared to the scores before the treatment. 2. The score about papule/induration/edema, effusion/furuncle, and subjective symptoms at night were significantly improved in all groups after the treatment compared to the scores before the treatment. 3. The effect of Hwangryunhaedock-tang increase markedly with advancing age. The effect of Hwangryunhaedocktang increases markedly with advancing age. Conclusions Traditional Korean herbal medication; Hwangryunhaedock-tang is effective in treating pediatric atopic dermatitis.

  • Objectives : This study was designed to investigate the scavenging effect on Nitric Oxide of BUM. Methods : The scavenging effect on NO concentrations 2, 6, 12 and 24 hours after treatment with Vit C. and BUM at 1, 10, $100{\mu}g/ml$ were estimated. And those was compared with control group. Results : 1. The concentration of NO 2 hours after treatment with BUM at 1, 10, 1$100{\mu}g/ml$ were decreased compared with control group. 2. The conecntration of NO 6 hours after treatment with BUM at 1, 10, 1$100{\mu}g/ml$ were decreased compared with control group. 3. The concentration of NO 12 hours after treatment with BUM at 1, 10, $100{\mu}g/ml$ were decreased compared with control group. 4. The concentration of NO 24 hours after treatment with BUM at 1, $100{\mu}g/ml$ were decreased compared with control group. Conclusion : The scavenging effect of BUM on NO was dose-dependent.

Neuroprotective Effect of HyulBuChookAu-Tang on Focal Cerebral Ischemia of the Rats

  • Objectives; This study examined the neuroprotective effect of Hyulbuchookautang (血府逐瘀湯, HBCAT)against neural damage following focal cerebral infarction. Methods : Sprague-Dawley Rats were induced with focal cerebral infarction by temporal middle cerebral artery occlusion (MCAO). The rats were divided into 2 groups. We treated extract of HBCAT to one group after operation (sample group), and the other group wasn't treated after operation (control group). We observed neurological scores and TIC-stained infarct area, total infarct volume in brain sections and Bax-positive neurons, HSP70- positive neurons in brain regions. Results : HBCAT treatment at 3 days after MCAO reduced neurological scores induced by MCAO. HBCAT treatment at 5 days after MCAO reduced TTC-stained infarct area in brain sections induced by MCAO. HBCAT treatment at 5 days after MCAO reduced total infarct volume in brain sections induced by MCAO. HBCAT treatment after MCAO reduced Bax-positive neurons in cortex infarct core and cortex infarct penumbra and caudo-putamen of brain regions induced by MCAO. HBCAT treatment after MCAO reduced HSP70- positive neurons in cortex infarct penumbra of brain regions induced by MCAO. Conclusions : These results suggest that HBCAT has a neuroprotective effect against focal cerebral ischemia.

Differences in mechanical thrombectomy for acute ischemic stroke on weekdays versus nights/weekends in a Japanese primary stroke core center

  • Objective: The term "weekend effect" refers to an increase in the mortality rate for hospitalizations occurring on weekends versus weekdays. In this study, we investigated whether such an effect exists in patients undergoing mechanical thrombectomy for acute ischemic stroke with large vessel occlusion (currently the standard treatment for this condition) at a single center in Japan. Methods: We surveyed 151 patients who underwent mechanical thrombectomy for acute ischemic stroke with large vessel occlusion (75 and 76 patients were treated during daytime and nighttime, respectively) from January 2019 to June 2021. The items evaluated in this analysis were the rate of modified Rankin Scale ≤2 or prestroke scale, mortality, and procedural treatment time. Results: The rates of modified Rankin Scale ≤2 or prestroke scale and mortality at 90 days after treatment did not differ significantly between daytime and nighttime (41.3% vs. 29.0%, p=0.11; 14.7% vs. 11.8%, p=0.61, respectively). The door-to-groin time tended to be shorter during daytime versus nighttime (57 [IQR: 42.5-70] min vs. 70 [IQR: 55-82]) min, p=0.0507). Conclusions: This study did not reveal differences in treatment outcome between daytime and nighttime in patients undergoing mechanical thrombectomy for acute ischemic stroke with large vessel occlusion. Therefore, the "weekend effect" was not observed in our institution.

Effect of combination treatment of vitamin E and insulin in streptozotocin-treated rats and BB rats II. Effect on the fatty acid composition of phospholipid (BB 랫드 및 streptozotocin이 투여된 랫드에서 vitamin E와 insulin 병합 투여 영향 II. 인지질의 지방산 조성에 미치는 영향)

  • The present study was investigated fatty acid composition of the phospholipid in the RBC membrane, liver and microsomal fraction after vitamin E and/or insulin treatment to evaluate the effect of vitamin E on the oxidative stress in STZ-treated rat and BB rat. Results obtained through the experiments were summarized as follows; 1. Effect of vitamin E and/or insulin treatment in STZ-treated rat 1) In the insulin treated group and the combination treated groups of vitamin E with insulin, body weights were increased compared to STZ-treated rat(STZ control group). Especially it was more significantly increased in the combination treated group of high dose vitamin E with insulin. 2) The composition of fatty acids of the phospholipid in RBC membrane, liver and microsomal fractions was shown a decreased C16:1, C18:1, C20:4 and an increased C16:0, C18:0, C18:2 in STZ control group compared to normal control group. In RBC membrane, liver and microsomal fractions after vitamin E with insulin treatment in STZ-treated rat, effect on the composition of fatty acids of the phospholipid was shown the result of a decreased C16:0, C18:0, C18:2 and an increased C16:1, C18:1, C20:4. 3) Hemolysis rate of the RBC to $H_2O_2$ was increased in the STZ control group and it was decreased below the hemolysis level of normal control group by vitamin E treatment. 2. Effect of vitamin E and/or insulin treatment in BB rat 4) Only in microsomal fraction, fatty acid composition was different between insulin treatment group and vitamin E with insulin treatment group. It was increased C16:0 and C18:1, and decreased C18:0 and C18:2 in vitamin E with insulin treatment group: But C20:4 was not different in two groups. These results suggest that the combination treatment of vitamin E and insulin could prevent the oxidative change of fatty acids in P-lipid of the RBC membrane, liver and microsomal fraction in STZ-treated rats and BB rats.

  • In the present study, Bauschinger effect was investigated for the micro-alloying forging steel which has been developed for about 30 years ago to save energy consumption by eliminating the heat treatment processes in the forging industry. The micro-alloying steels used fur cold forging industry mainly aim to replace the usual carbon steel. With the conventional carbon steels, all the deformation history can be eliminated after the final heat treatment(quenching and tempering). In the case of micro-alloying forging steels, however, the prior deformation history should be taken into consideration to meet the mechanical property requirement since the microstructure of micro-alloying steels might exhibit the Bauschinger effect, which was not needed to consider in the case of conventional carbon steel having quenching and tempering treatment. In the present study, the reverse loading tests were carried out to determine the Bauschinger effect of micro-alloying steel which composed of ferrite and cementite phases.

  • The eight combined products composed of ranitidine.HCI, tripotassium dicitrato bismuthate and sucralfate were prepared with various ratios and studied in therapeutic effects of them on various gastrointestinal diseases. These were induced in rats with the porous ligation, ethanol-HCI, acetic acid and cysteamine method, etc. In all experimental setting, the effect of the combined treating was more pronounced than the effect of each drug alone. Specially, the combined treatment consisted of ranitidine : tripotassium dicitrato bismuthate sucralfate ratio of 1.5 : 2 : 6 showed the most powerful therapeutic effect on acute gastric ulcer model and revealed a significant acceleration of the healing on chronic gastroduodenal ulcer model. And that, therapeutic doses of ranitidine, tripotassium dicitrato bismuthate arid sucralfate given in combination had an additive or, in some case, synergistic effect. From the above results, this combined treatment may useful to heal the gastrointestinal diseases that aren't cured well by treatment of each them alone.

Effect of Solution Heat Treatment on Mechanical Properties in Incoloy 825 Alloy (Incoloy 825 합금의 기계적 성질에 미치는 열처리의 영향)

  • This study was carried out to investigate the effect of heat treatment on the microstructure and mechanical properties in 90% hot forged Incoloy 825 alloy. With increasing solution treatment temperature, the grain size increased and the volume fraction of total precipitates decreased, and the precipitates disappeared at $1,000^{\circ}C$. With increasing aging time at $700^{\circ}C$, the volume fraction of precipitate increased and the precipitates size increased. Most of the precipitates consist $Cr_{23}C_6$ carbide, and a small amount of TiC carbide was also observed. With decreasing solution treatment temperature and increasing aging time, tensile strength and hardness increased, and the elongation and impact value decreased. With increasing aging time, the impact value decreased sharply by the increased of the precipitate size.

The Effect of Resin Treatment for Improving of the Metal Thin Membrane Fastness on Polyester Fabrics by Sputtering (Sputtering에 의한 polyester 직물의 박막내구성 개선을 위한 수지처리 효과)

  • Functional properties are available with sputtering. But sputtering treatment alone cannot got a good fastness performance to washing, rubbing and light. This research was objected to investigate optimum condition by sputtering on polyester through various processing conditions such as ion current and treatment time, and then various resin treated onto metal coated polyester fabrics in order to increase washing fastness of metal membrane. As the results, the optimum conditions revealed 1500 mA of ion current, 2 min of treatment time in sputter, and suitable resin concentrations were 2% of o.w.s (on the weight of solution) in resin treatment. Therefore, we could get enhanced anti-static effect and flex stiffness as well as washing fastness in sputtered polyester fabric with various resin treatment, for example, melamine and polyurethane.
