Effect of Ethanol on $Na^+-P_i$ Uptake in Opossum Kidney Cells: Role of Membrane Fluidization and Reactive Oxygen Species

  • This study was undertaken to examine the effect of ethanol on $Na^+ -dependent$ phosphate $(Na^+-P_i)$ uptake in opossum kidney (OK) cells, an established renal proximal tubular cell line. Ethanol inhibited ^Na^+-dependent$ component of phosphate uptake in a dose-dependent manner with $I_{50}$ of 8.4%, but it did not affect $Na^+-independent$ component. Similarly, ethanol inhibited $Na^+-dependent$ uptakes of glucose and amino acids (AIB, glycine, alanine, and leucine). Microsomal $Na^+-K^+-ATPase$ activity was not significantly altered when cells were treated with 8% ethanol. Kinetic analysis showed that ethanol increased $K_m$ without a change in $V_{max}$ of $Na^+-P_i$ uptake. Inhibitory effect of n-alcohols on $Na^+-P_i$ uptake was dependent on the length of the hydrocarbon chain, and it resulted from the binding of one molecule of alcohol, as indicated by the Hill coefficient (n) of 0.8-1.04. Catalase significantly prevented the inhibition, but superoxide dismutase and hydroxyl radical scavengers did not alter the ethanol effect. A potent antioxidant DPPD and iron chelators did not prevent the inhibition. Pyrazole, an inhibitor of alcohol dehydrogenase, did not attenuate ethanol-induced inhibition of $Na^+-P_i$ uptake, but it prevented ethanol-induced cell death. These results suggest that ethanol may inhibit $Na^+-P_i$ uptake through a direct action on the carrier protein, although the transport system is affected by alterations in the lipid environment of the membrane.

  • With the advent of the digital economy, the business eco-system has been changing from the competition system across individual companies to that of supply chains. Under the rapidly changing business environment, it becomes true that the competitive power of the steel maker depends on the shipbuilding company, an important customer of the steel maker. Accordingly, e-partnering between a steel maker and shipbuilding companies becomes important. Schemes of developing e-partnering are presented as follows: implementation of the inter-communication system, day/sequence order and supply, improvement of infrastructure such as transport, quay etc.

  • The transport phenomena of ten amino acid molecules through poly(2-hydroxyethyl methacrylate), P(HEMA) membrane have been investigated in various range pH solutions. It is found that the permeability and diffusivity of the amino acids through membrane depended on the different shape, size and the charge of them are changed by the pH. The permeabilities and diffusivities of amino acids have the largest value in the neutral solution. In this case, they are diffused through free water in the P(HEMA) membrane and the diffusion mechanism is the pore type. The basic solution have larger value than the acidic it. Whether the diffusion mechanism of the core type or the partition type, it is depended on the effect of side chain of the amino acid in basic and acidic solution.

Public Transportation Alighting Estimation Method Using Smart Card Data (교통카드데이터를 활용한 하차정류장 추정 방법론 연구)

  • Recently, there has been a growing interest in using smart card data. However, there are restrictions on the utilization of data in many areas outside the Seoul metropolitan area because the data does not contain alighting information. This paper presents a methodology for estimating alighting stops of smart card data. Estimation results were verified by smart card data from Seoul and Gwangju. The estimation rates were 78.2% and 81.6% in Seoul and Gwangju, respectively. The matching accuracy was 54.2% and 33.4%, respectively. However, if up to two stops of error are allowed, the accuracy values were 93.6% and 94.0%, respectively. We also discussed changes in estimation results due to adjusting the allowable walking distance, which is a key parameter of trip chaining methods. As the allowable walking distance increases, the estimation rate increases, while the accuracy decreases, and it is found that the estimation results change by around 500m.

  • Sulfonated poly(ether sulfone)(SPES) of various ion exchange capacity (IEC) was prepared by heterogeneous sulfonation with chlorosulfonic acid (CSA) to make hydrophilic ultrafiltration membrane for reducing fouling. The effects of CSA concentration, reaction temperature and reaction time has been investigated. The reaction was effective when the temperature is above 10$\circ$C and the CSA concentration is over 0.05 mol, although polymer chain has been significantly degraded. The substitution of sulfonic acid groups was characterized by FTIR and $^1$H-NMR. Transport properties and fouling test have been conducted to the modified SPES ultrafiltration rnembranrs by heterogeneors method. Membranes were obtained using DCM and PVP as a non-solvent and pore forming agent, respectively. Flux reduced and rejection increased with ion exchange capacity. Finger-like structure was disappeared and the thickness of top layer was increased. Dense membrane by non-solvent DCM and porous membrane by pore forming agent PVP was prepared. Fouling was reduced with increasing ion exchange capacity because of hydrophilicity.

Differential Responses of Three Cyanobacteria to UV-B and Cd

  • Interactive effects of UV-B and Cd on growth, pigment, photosynthesis, and lipid peroxidation have been studied in Anabaena, Microcystis, and Nostoc; all the tested cyanobacteria showed differential sensitivity to different dosage of UV-B and Cd alone as well as in combination. Phycocyanin was severely affected by UV-B and Cd by all the strains; the degree of pigment bleaching was most pronounced in Anabaena followed by Microcystis and Nostoc. $UV-B_2+Cd_2$ produced nearly 83, 78, and 65% inhibition of phycocyanin in Anabaena, Microcystis, and Nostoc, respectively. The above treatment also significantly decreased the contents of Chl ${\alpha}$ and carotenoid. Presence of capsule in Microcystis protected the phycocyanin bleaching as compared to decapsulated cells. Laboratory-grown Microcystis revealed about 75 and 80% inhibition, following $UV-B_2+Cd_2$ treatment, respectively. in capsulated and decapsulated cells. Damage caused by Cd was more pronounced than UV-B. Inhibition of photosynthesis did occur in all the test strains, being maximum in Anabaena. PS II was the most sensitive component of the electron transport chain, showing 84, 80, and 70% inhibition in Anabaena, Microcystis, and Nostoc, respectively. As compared to control, significant lipid peroxidation (6.5-fold higher) was observed in Anabaena with $UV-B_2+Cd_2$, $^{14}C-uptake$ was more susceptible to Cd and Uv-B than oxygen-evolution. Approximately 84, 80, and 76% inhibition of $^{14}C-uptake$ was observed in Anabaena, Microcystis, and Nostoc, respectively. Similarly, $UV-B_2+Cd_2$ inhibited APT content of Anabaena by 87%. This ,study suggests that inhibition of carbon fixation was due to decreased ATP content of the test cyanobacteria by UV-B+Cd, where Anabaena was the most sensitive and Nostoc the most tolerant.

Sequences and Phylogenic Analysis of Squid New Kinesin Superfamily Proteins (KIFs) (오징어과의 Kinesin Superfamily Proteins (KIFs)의 유전자분석 및 계통분석)

  • The movement of vesicles from the neuronal cell body to specific destinations requires molecular motors. The squid giant axon represents a powerful model for studies of the axonal transport mechanism because the axoplasm can readily be separated from the sheath by simple extrusion. In a previous study, vesicular movements in the axoplasm of the squid giant axon were inhibited by the kinesin antibody. In the present study, we cloned and sequenced the cDNAs for squid brain KIFs. Amplification of the conserved nucleotide sequences of the motor domain by polymerase chain reaction (PCR) using first-strand cDNAs of the squid optic lobe identified six new KIF proteins. Motif analysis of the motor domains revealed that the squid KIFs are homologous to the consensus sequences of the mouse KIFs. The phylogenetic tree generated by using the maximum parsimony (MP) method, the neighbor-joining (NJ) method, the minimum evolution (ME) method, and the maximum likelihood (ML) method showed that squid KIFs are closest to mouse KIFs. These data prove the phylogenetic relationships between squid KIFs and mouse ones.

A Study on Promotion of Value Added Logistics(VAL) Activities between Korea and China (한.중간 부가가치물류 활성화방안에 관한 연구 - 산동성을 중심으로 -)

  • The volume of trade between Korea and China has been growing significantly year by year since the diplomatic relations have been established in 1992. The economic and logistics cooperation between the two countries is crucial not only for the development of their economic relations but for promotion of value-added logistics. However there are still huge and serious impediments to logistics network and management between the two countries including bottlenecks in transport and customs procedures. The purpose of this study is to investigate how to promote value added logistics activities between Korea and Shandong Province of China. It is suggested the ways to increase VAL activities such as enhanced division of international labour within industries, provision of comprehensive logistics service and establishment of strategy for supply chain management between the two countries.

Radiation Dose from Computed Tomography Scans for Korean Pediatric and Adult Patients

  • Background: In recent events of the coronavirus disease 2019 (COVID-19) pandemic, computed tomography (CT) scans are being globally used as a complement to the reverse-transcription polymerase chain reaction (RT-PCR) tests. It will be important to be aware of major organ dose levels, which are more relevant quantity to derive potential long-term adverse effect, for Korean pediatric and adult patients undergoing CT for COVID-19. Materials and Methods: We calculated organ dose conversion coefficients for Korean pediatric and adult CT patients directly from Korean pediatric and adult computational phantoms combined with Monte Carlo radiation transport techniques. We then estimated major organ doses delivered to the Korean child and adult patients undergoing CT for COVID-19 combining the dose conversion coefficients and the international survey data. We also compared our Korean dose conversion coefficients with those from Caucasian reference pediatric and adult phantoms. Results and Discussion: Based on the dose conversion coefficients we established in this study and the international survey data of COVID-19-related CT scans, we found that Korean 7-year-old child and adult males may receive about 4-32 mGy and 3-21 mGy of lung dose, respectively. We learned that the lung dose conversion coefficient for the Korean child phantom was up to 1.5-fold greater than that for the Korean adult phantom. We also found no substantial difference in dose conversion coefficients between Korean and Caucasian phantoms. Conclusion: We estimated radiation dose delivered to the Korean child and adult phantoms undergoing COVID-19-related CT examinations. The dose conversion coefficients derived for different CT scan types can be also used universally for other dosimetry studies concerning Korean CT scans. We also confirmed that the Caucasian-based CT organ dose calculation tools may be used for the Korean population with reasonable accuracy.

Protective efficacy of attenuated Salmonella Typhimurium strain expressing BLS, Omp19, PrpA, or SOD of Brucella abortus in goats

  • Background: Attenuated Salmonella strain can be used as a vector to transport immunogens to the host antigen-binding sites. Objectives: The study aimed to determine the protective efficacy of attenuated Salmonella strain expressing highly conserved Brucella immunogens in goats. Methods: Goats were vaccinated with Salmonella vector expressing individually lipoprotein outer-membrane protein 19 (Omp19), Brucella lumazine synthase (BLS), proline racemase subunit A (PrpA), Cu/Zn superoxide dismutase (SOD) at 5 × 109 CFU/mL and challenge of all groups was done at 6 weeks after vaccination. Results: Among these vaccines inoculated at 5 × 109 CFU/mL in 1 mL, Omp19 or SOD showed significantly higher serum immunoglobulin G titers at (2, 4, and 6) weeks post-vaccination, compared to the vector control. Interferon-γ production in response to individual antigens was significantly higher in SOD, Omp19, PrpA, and BLS individual groups, compared to that in the vector control (all p < 0.05). Brucella colonization rate at 8 weeks post-challenge showed that most vaccine-treated groups exhibited significantly increased protection by demonstrating reduced numbers of Brucella in tissues collected from vaccinated groups. Real-time polymerase chain reaction revealed that Brucella antigen expression levels were reduced in the spleen, kidney, and parotid lymph node of vaccinated goats, compared to the non-vaccinated goats. Besides, treatment with vaccine expressing individual antigens ameliorated brucellosis-related histopathological lesions. Conclusions: These results delineated that BLS, Omp19, PrpA, and SOD proteins achieved a definite level of protection, indicating that Salmonella Typhimurium successfully delivered Brucella antigens, and that individual vaccines could differentially elicit an antigen-specific immune response.
