The Effects of High Temperature on Infection by Potato virus Y, Potato virus A, and Potato leafroll virus

  • We examined the effects of temperature on acquisition of Potato virus Y-O (PVY-O), Potato virus A (PVA), and Potato leafroll virus (PLRV) by Myzus persicae by performing transmission tests with aphids that acquired each virus at different temperatures. Infection by PVY-O/PVA and PLRV increased with increasing plant temperature in Nicotiana benthamiana and Physalis floridana, respectively, after being transmitted by aphids that acquired them within a temperature range of $10-20^{\circ}C$. However, infection rates subsequently decreased. Direct qRT-PCR of RNA extracted from a single aphid showed that PLRV infection increased in the $10-20^{\circ}C$ range, but this trend also declined shortly thereafter. We examined the effect of temperature on establishment of virus infection. The greatest number of plants became infected when N. benthamiana was held at $20^{\circ}C$ after inoculation with PVY-O or PVA. The largest number of P. floridana plants became infected with PLRV when the plants were maintained at $25^{\circ}C$. PLRV levels were highest in P. floridana kept at $20-25^{\circ}C$. These results indicate that the optimum temperatures for proliferation of PVY-O/PVA and PLRV differed. Western blot analysis showed that accumulations of PVY-O and PVA coat proteins (CPs) were lower at $10^{\circ}C$ or $15^{\circ}C$ than at $20^{\circ}C$ during early infection. However, accumulation increased over time. At $25^{\circ}C$ or $30^{\circ}C$, the CPs of both viruses accumulated during early infection but disappeared as time passed. Our results suggest that symptom attenuation and reduction of PVY-O and PVA CP accumulation at higher temperatures appear to be attributable to increased RNA silencing.

A study on Hepatitis B and knowledge about AIDS of dental hygiene student in Korea (전국 치위생과 학생들의 B형간염과 AIDS에 관한 지식도 조사)

  • The purpose of this was to evaluate the level of knowledge about Hepatitis B and AIDS among dental hygiene students at six dental hygiene educational programs in nationwide, compare the findings to those of Song's study. 1703 dental hygiene students surveyed by mail and answered a questionnaire, using the questionnaire used developed by DiClemente el al., on Hepatitis B vaccination, antibody production, experience of unintentional needle stick injuries, experience of surgical operation and blood transfusion, personal risk factors for HBV and HIV, management of HBV and HIV postexposure management et al. The data indicated that dental hygiene students (79.0%) in this study had more HIV vaccination than dental hygiene(74.3%) in Song's study. As for recognition of antibody production after Hepatitis B vaccination, only 34.6% of respondents answered they knew having antibody production and 95% of respondents knew not having antibody production. Over one-half of the respondents (55%) answered they didn't even recognize having antibody production. 52.3 percent of dental hygiene students experienced unintentional needle stick injuries. The findings on the knowledge about Hepatitis B and AIDS were that dental hygienists (76.5 points) received higher mean score than those of dental hygiene students (71.0 points) and that the distribution of right answer rate also showed wider range in dental hygiene students (27.7~97.5%) than those of dental hygienist (41.2~99.5%). There were no statistical differences among dental hygiene education programs. Finding of this study support that the curriculum of dental hygiene program should include instruction on sources and methods of transmission of infectious diseases, risk of virus transmission in the workplace and principles of infection control. Furthermore, infection-control practices should be utilized routinely and their application to dental personnel, management of HBV and HIV postexposure management. Furthermore, for preventing the further spread of infectious diseases (HBV, HIV and AIDS${\cdots}$) caused by bloodborne viruses, imposing an obligation rather than recommendation on Hepatitis B vaccination to all dental personnel and routinely utilizing infection-control guidelines for all dental patients in dental practices (include dental educational programs) should be accomplished by coordination the government agency.

Knowledge, Attitudes and Compliance Regarding Infection Preventive Behaviors for MERS among Staff in Long-term Care Hospitals (요양병원 종사자의 메르스에 대한 지식, 태도 및 감염예방행위 관계)

  • This study was conducted to identify the relationship between knowledge, attitude, and compliance regarding infection preventive behaviors among long-term care hospital staff during a Middle East Respiratory Syndrome (MERS) epidemic. The subjects were 211 staff members of a long-term care hospital in J province evaluated using a structured survey. Data were collected between June 15 and July 15, 2015 and analyzed using SPSS/WIN 24.0 based on an independent t-test, ANOVA, and Welch test, while post-hoc tests were conducted using the Scheffe test and the Grames-Hawell test. The MERS knowledge score of the staff at the long-term care hospital was high for nurses and nurse aids, who had experienced infection management when they received influenza vaccine in the previous year and were provided with easy access to hand sanitizers. Attitude towards MERS was more positive for nurses than nurse aids and caregivers. The degree of execution of infection preventive behaviors in terms of daily activity was high when influenza shots were received in the prior year; in terms of caring for patients, it was high when influenza shots were received in the previous year and they had easy access to hand sanitizers. The relationship between knowledge, attitude, and infection preventive behavior regarding MERS was such that high knowledge and positive attitude led to a higher degree of execution of infection preventive behavior. Therefore, during periods of high prevalence of newly infectious diseases such as MERS, it is important for the long-term care hospitals to conduct infection management education including the characteristics of the MERS disease, its transmission, and its prevention to enhance knowledge regarding MERS and induce positive change in attitude to improve the level of infection preventive behaviors.

Studies on the Soil Transmission of CGMMV and Its Control with Crop Rotation (오이녹반모자이크바이러스의 토양전염 생태 및 윤작에 의한 방제)

  • Cucumber green mottle mosaic virus (CGMMV) is one of major plant viruses infecting cucurbitaceous crops via soil or infected seeds. This study investigate ecology of infection of CGMMV in soil, and control tactics of this virus with soil hygiene and crop rotation. This virus was survival to 50% in soil without host plants for 17 months and had high vitality in debris of infected plant over 1 year. Infection rate of CGMMV was 1.0~3.6% in control soil and 12~36% in soil transplanted with wounded root of watermelon. It showed that wounded root may affect severity of soil infection. Rotation between rice and watermelon caused dramatical reduction from 76.8% in repeated cultivation to 7.3% of progeny infection by CGMMV. Therefore, it is suggested that crop rotation be effective for control of CGMMV.

Histopathological features of pacific whiteleg shrimp, Litopenaeus vannamei, infected with Infectious Myonecrosis Virus (IMNV) with an emphasis on micro-traumas and inflammatory responses in muscle tissues (전염성근괴사증바이러스(IMNV)를 인위감염 시킨 흰다리새우(Litopenaeus vannamei)의 근육에서 나타난 미세 손상과 염증반응에 대한 조직병리학적 특성 연구)

  • We injected infectious myonecrosis virus (IMNV) to pacific whiteleg shrimp, Litopenaeus vannamei, and observed closely with using light microscope and transmission electron microscope (TEM) for 4-8 days post infection (dpi). As clinical signs, abdominal bodies had mild opaque muscles at 5 dpi. And the mortality was shown at 6 dpi. At 8 dpi, most injected shrimps had severe opaque muscles and humped back that cause of movement disorder. As results of histopathological examinations, local parts of abdominal body muscle had muscle fiber hyalinization, muscle fiber atrophy, rounded muscle fibers, myofibrillar hypertrophy in size, a decrease in number of myofibrils and phagocytosis from the sarcolemmas by multiple hemocytes at 4 dpi. Especially, myofibrillar hypertrophy appeared at the whole or random part of single muscle fiber not in specific locations like the center or edge of muscle fiber. At 6-7 dpi, multiple muscle necrosis, muscle fiber segmentation, myofibril lysis ap- peared and a few hemocytes were infiltrated at lesions. At 8 dpi, extensive muscle necrosis, multiple myofibril lysis and muscle fiber atrophy were shown, and very few hemocytes were infiltrated. In early stage of infection, local viral myositis with zenker's degeneration were shown. These lesions appeared multiply after the early stage. In late stage of infection, extensive coagulative muscle necrosis appeared with few of inflammatory response such as hemocytes infiltration. The lack of hemocytes infiltration response at the late stage might be disadvantage for Litopenaeus vannamei to defense against IMNV and to recover, because hematocytes (granulocyte, semi-granulocyte) eliminate pathogen and damaged tissues from infection sites and help recover. As results of the TEM observation, IMNVs that had nonenveloped icosahedral capsid which was 30-40 nm diameter were in myofibril and beside tubules of sarcoplasmic reticulum and moved to the certain direction. The micro-tears and micro-trau- mas in myofibrils caused muscle fiber necrosis. And semi-granulocytes engulfed IMNVs to eliminate virus.

Congenital transmission of Theileria sergenti in cattle verified by immunohistochemistry (소 Theileria sergenti의 태반감염에 대한 면역세포화학적 증명)

  • The spleen and placenta from the aborted fetuses as well as lymphnodes and placenta of the corresponding dams naturally infected with T sergenti were used to localize the parasite antigens by immunohistochemical staining for the possible congenital transmission of theileriosis. Parasite-specific antigens were detected immunohistochemically by incubating the sections with specific monoclonal antibody prepared against 34KD surface antigen of T sergenti and visualized via the avidin biotin complex(ABC) method. Specific T sergenti antigen was detected in the sections of formalin or acetone-fixed fetal spleens and placenta. Similar antigens were also demonstrated in lymphnodes and placentas of the corresponding dams. It is concluded that this technique will eventually play an important role in specialized diagnostic laboratories in the verification/evaluation of congenital infection with T sergenti.

Non-Mandatory Influenza Vaccination Rates among Healthcare Workers during the 2017-2018 Influenza Season: a Multicenter Study in Korea

  • Purpose: Annual influenza vaccination is the best strategy to prevent healthcare-associated influenza transmission. Influenza vaccination rates among healthcare workers (HCWs) vary by country, region, and year. We investigated the influenza vaccination rates for HCWs during the 2017-2018 influenza season in South Korea, where a non-mandatory vaccination campaign was conducted. Methods: We retrospectively investigated factors affecting the influenza vaccination rate among HCWs during the 2017-2018 influenza season in three tertiary hospitals in Goyang City, where the non-mandatory influenza vaccination program is conducted. Results: Consequently, 6,994 of 7,180 HCWs (97%) were included, and the overall vaccination rate was 85%. Nurses had the highest rate with 92%, followed by health technicians (88%), physicians (84%), and non-medical HCWs (79%, P<0.001). Vaccination rates differed, depending on the frequency of contact with patients in the non-medical HCWs (frequent contact vs. less-frequent contact; 90% vs. 73%, P<0.001). Conclusions: The influenza vaccination rate among HCWs during the 2017-2018 influenza season in Korea was 85%, which is among the highest rates compared with previously reported non-mandatory vaccination rates in other countries. The vaccination rate may vary depending on the HCW's occupational characteristics, including the extent of contact with the patient. Therefore, a multifaceted strategy is needed to increase the vaccination rate of HCWs.

  • 젊은 말과 그 관리인에게 발생한 Microsporum canis 감염에 의한 백선에 대해 보 고하였다. 말과 사람에 있어서의 진단은 피부 병변의 KOH 표본에서 피부사상균을 직접 증명 하고,감염된 인설의 순수 배양물로부터 Microsporum canis를 분리한 것에 근거를 두었다. 말과 사람에 서 분리하고, PHOL 염색액으로 염색한 Microsporum canis는 현미경으로 자세히 관찰했을 때 형태학 적인 차이를 나타내지 않았다. 말에 있어서 옥도정기와 사람에 있어서 miconazole에 의한 국 소 요법은 효과적이었다. 병력은 관리인이 말로부터 감염된 것을 제시하였다.

  • Soil transmission ratio of Cucumber green mottle mosaic virus (CGMMV) was 0.2 to 3.5 % in watermelon growing fields naturally infested with the virus. Biological activities of CGMMV lost after 17 months in moist well-aerated soil but still continued more than 33 months in waterlogged soil. To inhibit the virus infection through soil, the roots of watermelon seedlings were soaked in 10% solution of skim milk prior to transplanting. The seedlings treated with skim milk solution were not infected, while 5.0 to 7.6% out of control seedlings were infected. The roots treated with skim milk were coated with membrane around the roots under scanning electron microscope.

  • This study is about a blockchain-based collective quarantine management system and its commercialization model. The configuration of this system includes a biometric information transmission unit that generates biometric information based on measured values generated from wearable devices, a biometric information transmission unit that transmits biometric information generated here from a quarantine management platform, and action information transmitted from the community server. is a system including an action information receiving unit for receiving from the quarantine management platform. In addition, a biometric information receiving unit that collects biometric information from the terminal, an encryption unit that encodes biometric information generated through the biometric information receiving unit based on blockchain encryption technology, and a database of symptoms of infectious diseases to store symptom information and an infection diagnosis database. The generated database includes a location information check unit that receives from the terminal of the user identified as a symptomatic person and determines whether the user has arrived in the community based on the location information confirmation unit and the location of the user after the location is confirmed. It includes a community arrival judgment unit that judges. And, the community server helps the interaction between the generated information. Such a blockchain based collective quarantine management system can help to advance the existing quarantine management system and realize a safer and healthier society.
