Bioremediation of Heavy Metals from the Land Application of Industrial Sewage Sludge with Minari (Oenanthe stolonifer DC.) Plant

  • Laboratory experiments for the removal efficiency of heavy metals in land application of sludge, the accumulation and translocation of heavy metals in x plants after transplanting, and the responses of Minari growth with different ratio of land application of sludge were conducted to determine the potential ability of bioremediation with Minari plants. The removal rate and translocation of copper. zinc. lead. and cadmium in soil and plants were compared after transplanting the Minari plants to soil treated with different ratio of sludge. The removal efficiency of heavy metals in soil incorporated with sludge was different with application ratio, but increased with growing periods of Minari plants. The removal efficiency of Cu, Zn, Pb, and Cd ranged from 67 to 74% from 51% to 63%, from 37% to 71%. and from 15% to 25% after 45 days of transplanting. respectively. The amount removed the copper value. 65.9 mg/kg, observed to be highest in soil incorporated 3% sludge after 45 days. The translocation of Cu. Zn. Pb. and Cd from shoots to roots ranged from 18 to 53%, from 17 to 32%, from 14 to 49%, and from 23 to 38% over growing periods. respectively. In plant responses it appeared to be inhibited the plant growth in the treatment compared with the control at early stage of growth. However, the fresh weights of Minari plant increased from 12.5 to 62.5% in the sludge application after 45 days relative to the control. Therefore the Minari might play a useful role in bioremediation of Cu, Zn, Pb, and Cd in the land application of sludge.

Effects of Lime, Fly Ash & Rice Straw Ash on Cadmium and head Translocation from Soil to Radish (석회, Fly Ash 및 볏짚재가 토양에서 무우로 이행 축적되는 카드뮴과 납의 함량 감소에 미치는 영향)

  • Greenhouse pot experiments were conducted to evaluate the effects of lime, fly ash and ash(from rice straw) on the cadmium and lead translocation from soil to radish. The soils with low metal contents(Cd 1.52 ppd and Pb 25.37ppm) were prepared and high metal contents (Cd 8.99 rpm and Pb 50.81ppm) were prepared and amended with 0.25%, 0.5%, 1.095, 2.055 each of lime, fly ash and ash. Radishes(Raphanus satiuus) were cultivated and cropped on the soils during 25, 50 and 75 days after sprout, and then cadmium and lead contents of radishes were analyzed by roots and tops. The results obtained are as follows. 1. Lime and ash were effective in raising the soil pH, but fly ash was not effective. 2. The growth of radishes were not impaired by the cadmium and lead contamination but, impaired by soil pH 7.5 or more. 3. Cadmium was accumulated very strongly in radishes and the greater concentration was found in tops than roots, but lead showed no evidence of accumulation in radishes. 4. In general, when the concentrations of lime and ash in soils increased, the uptake of cadmium and lead by radishes decreased, and lime was more effective than ash, while fly ash revealed no effect of reducing the translocation of cadmium and lead from soils to radishes. 5. The uptake of cadmium by radishes decreased more effectively than lead and the uptake of Cd or Pb by radishes grown in the soils with high metal contents decreased more effectively than low metal con tents. 6. Cadmium and lead contents of radishes were negatively correlated with soil pH values and the relationship in cadmium content was stronger than that in lead content.

The first Korean case of a newborn with 3p26 microdeletion and 5q35 microduplication inherited from paternal balanced translocation

  • Genetic imbalances are a major cause of congenital and developmental abnormalities. We report the first case of a 3p26 microdeletion and 5q35.2q35.3 microduplication in a newborn with multiple congenital anomalies evaluated using chromosomal microarray analysis (CMA) and fluorescence in situ hybridization (FISH). The patient was born at 30 weeks and 2 days of gestation with a body weight of 890 g. He had symmetric intrauterine growth restriction, microcephaly, facial dysmorphism (hypertelorism, blepharophimosis, mild low-set ears, high-arched palate, and micrognathia), and right thumb polydactyly. Echocardiography revealed an atrial septal defect and patent ductus arteriosus. Furthermore, CMA revealed a concurrent microdeletion in 3p26 and a microduplication in 5q35.2q35.3. FISH analysis showed that these genetic changes resulted from a translocation mutation between chromosomes 3 and 5. The patient's mother had mild intellectual disability, short stature, and facial dysmorphism, while his father had a normal phenotype. However, parental FISH analysis revealed that the asymptomatic father carried a balanced translocation of chromosomes 3p26 and 5q35. CMA and FISH tests are useful for diagnosing neonates with multiple congenital abnormalities. Further parental genetic investigation and proper genetic counseling are necessary in cases of chromosomal abnormalities inherited from parental balanced translocations.

6-Shogaol and 10-Shogaol Synergize Curcumin in Ameliorating Proinflammatory Mediators via the Modulation of TLR4/TRAF6/MAPK and NFκB Translocation

  • Extensive research supported the therapeutic potential of curcumin, a naturally occurring compound, as a promising cytokine-suppressive anti-inflammatory drug. This study aimed to investigate the synergistic anti-inflammatory and anti-cytokine activities by combining 6-shogaol and 10-shogaol to curcumin, and associated mechanisms in modulating lipopolysaccharides and interferon-γ-induced proinflammatory signaling pathways. Our results showed that the combination of 6-shogaol-10-shogaolcurcumin synergistically reduced the production of nitric oxide, inducible nitric oxide synthase, tumor necrosis factor and interlukin-6 in lipopolysaccharides and interferon-γ-induced RAW 264.7 and THP-1 cells assessed by the combination index model. 6-shogaol-10-shogaol-curcumin also showed greater inhibition of cytokine profiling compared to that of 6-shogaol-10-shogaol or curcumin alone. The synergistic anti-inflammatory activity was associated with supressed NFκB translocation and downregulated TLR4-TRAF6-MAPK signaling pathway. In addition, SC also inhibited microRNA-155 expression which may be relevant to the inhibited NFκB translocation. Although 6-shogaol-10-shogaol-curcumin synergistically increased Nrf2 activity, the anti-inflammatory mechanism appeared to be independent from the induction of Nrf2. 6-shogaol-10-shogaol-curcumin provides a more potent therapeutic agent than curcumin alone in synergistically inhibiting lipopolysaccharides and interferon-γ induced proinflammatory mediators and cytokine array in macrophages. The action was mediated by the downregulation of TLR4/TRAF6/MAPK pathway and NFκB translocation.

Clinical and Cytogenetic Analysis of Children with Maternal Chromosomal Balanced Translocation (모체의 염색체 균형전좌를 가진 환아들의 임상적 세포 유전학적 관찰)

  • Purpose : Parents' genetic information plays an important role in their children's genetic expression. Human chromosome has 23-paternal chromosomes and 23-maternal chromosomes. Parental chromosomal translocation can induce clinical problems in their children because of imbalance in genetic information. We intent to analyze the cytogenentic and clinical features about children with maternal balanced translocation between chromosome 15 and 18. Methods : We detected by one family's FISH study of chromosome 15. We have evaluated children born to clinically normal parents about peripheral bood analysis, endocrine, metabolic, radiologic study, electroencephalogram and social & intelligence scale. and We analysis their clinical manifestation by hospital records. Results : Patient's father and elder sister are normal clinically and genetically. Her mother's chromosome show balanced translocation, 46, XX, t(15;18)(p11.2;p11.3). One child has 46, XX, der(18) t(15;18)(p11.2;p11.3), mental retardation, growth retardation, speech & social developmental delay, recurrent infection and mild mitochondria dysfunction. Her young brother has 46, XY, der(15) t(15;18) (p11.2;p11.3), mental retardation, aggressive behavior, obesity and speech developmental delay. Conclusion : In this study we observed the children with developmental delay, dysmorphic facial features, mental retardation, growth retardation associated with growth hormone deficiency and aggressive behavior due to unbalanced translocation between chromosome 15 and 18.

Electrophoretic and Immunological Evaluation of Secalin in Rye, Triticale, and Wheat-Rye Translocation Wheat

  • Seed storage proteins have been used for studying biochemical genetics and end-use quality aspects. We conducted enzyme-linked immunosorbent assay (ELISA) and one-dimensional SDS-PAGE (1D SDS-PAGE) to evaluate different cereal crop species and Korean wheat lines for rye secalin proteins. The antisecalin antibody showed consistent specificity for rye secalin with little cross-reactivity to gliadins. Immunological cross-reactivities measured by the ELISA technique using competition assay showed significant differences of absorbance among rye, triticale, wheat-rye translocated wheat and non-translocated wheat. The absorbance values were lowest in rye followed by triticale, translocated wheat and non-translocated wheat. The ELISA for discrimination of wheat-rye translocation on the basis of antigen-antibody reactivity showed that none of the Korean wheat lines possessed 1RS and secalin proteins. The competitive ELISA experiment demonstrated specific determination for secalin that was originated from rye chromosomal parts. The result of 1D SDS-PAGE for identifying rye secalin subunits showed all three rye specific secalin protein subunits (75 KDa, 45 KDa, and 40 KDa) for rye and triticale, and 1RS specific secalins (45 KDa and 40 KDa) for 1AL/1RS and 1BL/1RS translocated wheats. All Korean wheats were lacking 1RS of rye chromosome and secalin.

  • A surgery of penile translocation was performed on a seven-month-old male calf born in August 26, 2007. With a penis deviated, as to prevent proper coitus on a dairy farm, the calf would be introduced to the group of cows and heifers before sniffing around to detect which one is in heat and to indicate to a veterinarian the right time for artificial insemination or embryo transfer. This study showed how to make a teaser bull in his young age.

Growth and Physiological Properties of Wild Type and Mutants of Halomonas subglaciescola DH-l in Saline Environment

  • A halophilic bacterium was isolated from fermented seafood. The 16S rDNA sequence identity between the isolate and Halomonas subglaciescola AJ306801 was above 95%. The isolate that did not grow in the condition without NaCl or in the condition with other sodium (Na$\^$+/) or chloride ions (Cl$\^$-/) instead of NaCl was named H. subglaciescola DH-l. Two mutants capable of growing without NaCl were obtained by random mutagenesis, of which their total soluble protein profiles were compared with those of the wild type by two-dimensional electrophoresis. The external compatible solutes (betaine and choline) and cell extract of the wild type did not function as osmoprotectants, and these parameters within the mutants did not enhance their growth in the saline environment. In the proton translocation test, rapid acidification of the reactant was not detected for the wild type, but it was detected for the mutant in the condition without NaCl. From these results, we derived the hypothesis that NaCl may be absolutely required for the energy metabolism of H. subglaciescola DH-l but not for its osmoregulation, and the mutants may have another modified proton translocation system that is independent of NaCl, except for those mutants with an NaCl-dependent system.

Translocation and Accumulation of Assimilates after Heading under Different Fertilizer Application in Waxy and Non-Waxy Near Isogenic Lines of Rice

  • This experiment was designed to assess changes in yield difference between waxy (IWR) and non-waxy (INWR) near isogenic rice by evaluating the translocation and accumulation of assimilates in different parts of the rice plants after heading stage under different fertilizer application. Nitrogen content of culm in both IWR and INWR decreased rapidly from 35 days before heading to flowering time and after then it was maintained constantly, while the nitrogen content of leaf blade was continually reduced. Sugar content of leaf blade in IWR decreased steadly by 10 days after heading, but that in INWR increased 20 days after heading. There was no difference in starch content of culum between IWR and INWR from 35days before heading to heading, but that of culm of IWR increased again after heading. Dry weight of IWR plant was less reduced during ripening as compared with INWR. The dry weight of culm was not significantly different between two isogenic lines.

Inhibition of Apoptosis is Responsible for the Acquired Resistance of K562 Cells to Cisplatin

  • In all attempt to elucidate the role of apoptosis in drug resistance, cisplatin-resistant human chronic myelogenous leukemia (CML) K562 cells (K562/CDDP) were established and compared with drug sensitive parent cells (K562) in the induction of apoptosis. K562/CDDP cells were 5-fold more resistant to cisplatin compared to K562 cells. In addition, K562/CDDP cells were significantly more resistant to apoptois as judged by DNA fragmentation and DAPI staining. K562/CDDP cells exhibited decreased proleolytic activity of caspase-3 and this was further demonstrated by decreased cleavage of its substrate poly (ADP-ribose) polymerase (PARR- Western blot analysis showed that K562/CDDP cells had longer sustained levels of BCL-$X_L$ whereas no difference was noted in the level of Bcl-2. the translocation of Bax to mitochondria was significantly delayed in K562/CDDP cells. These results suggest that the reduced translocation of Bax and the sustained expression of BCL-$X_L$ may cause resistance to apoptosis through prevention of mitochondria release of cytochrome c, which subsequently induces reduction of caspase-3 activity and that this response is partly responsible for the acquired resistance to cisplatin ill K562 cells.
