Differential expression of soybean SLTI100 gene encoding translation elongation factor 1A by abiotic stresses

  • The translation elongation factor 1A, eEF1A, catalyzes the binding of aminoacyl-tRNA to the A-site of the ribosome by a GTP-dependent mechanism. By subtractive suppression hybridization technique, we have isolated a soybean low-temperature inducible gene, SLTI100 encoding translation elongation factor 1A. Multiple sequence alignments and phylogenic analysis showed that SLTI100 and other eEF1As originated from diverse organisms are highly conserved. RNA expression of SLTI100 was specifically induced by low temperature, high salt, ABA, or drought stress. Based on the subcellular localization of the corresponding gene product fused to GFP, we were able to confirm that SLTI100-GFP was restricted to the nucleus and cytoplasm. We propose that soybean eEF1A may play an important role in translational regulation during abiotic stress responses in plants.

Psychometric Properties of the Korean Translation of the Attention-Deficit/Hyperactivity Disorder Stigma Questionnaire

  • Objectives: This study evaluated the psychometric properties of the Korean version of the attention-deficit/hyperactivity disorder (ADHD) Stigma Questionnaire (ASQ) and the effect of the source of information about mental health on ADHD stigma. Methods: The Korean translation of the ASQ was prepared, and 673 participants, 20-64 years of age, completed the questionnaire using an online panel survey in South Korea. The participants also completed questionnaires detailing sociodemographic variables and the source of their mental health knowledge. Cronbach's alpha coefficient was used to explore the internal consistency of the ASQ. Factor analysis using Varimax rotation was conducted to investigate the structure of the ASQ. Results: The 26-item ASQ demonstrated excellent internal consistency (Cronbach's alpha=0.940). Factor analysis supported a three-factor structure, including Concerns with Public Attitudes, Negative Self-Image, and Disclosure Concerns. There were no significant differences in the total ASQ scores according to sociodemographic characteristics. Participants who reported the internet as their major source of information about mental health showed higher ASQ scores compared to those who used other sources for mental health information. Conclusion: The Korean translation of the ASQ has acceptable psychometric properties among Korean adults. Inaccurate information from the internet could increase the stigma toward ADHD.

  • 2016년 4월부터 6월까지 표고 원목재배 임가 4곳과 표고 톱밥재배 임가 1곳의 재배사 내 실내 공기에서 진균을 포집 및 분리 동정하였다. 총 6개의 국내 미기록 진균(Cenangium acuum, Periconia macrospinosa, Neopestalotiopsissurinamensis, Metarhiziummarquandii, Trichoderma petersenii, Trichoderma paratroviride)을 분리하였으며 이들 균류에 대하여 형태학적 특성 및 internal transcribed spacer rDNA region, large subunitregion, ${\beta}-tubulin$ 또는 translation elongation factor $1{\alpha}$유전자 염기서열에 기반하여 계통학적 분석 결과를 기술하였다.

Two distinct nodes of translational inhibition in the Integrated Stress Response

  • The Integrated Stress Response (ISR) refers to a signaling pathway initiated by stress-activated $eIF2{\alpha}$ kinases. Once activated, the pathway causes attenuation of global mRNA translation while also paradoxically inducing stress response gene expression. A detailed analysis of this pathway has helped us better understand how stressed cells coordinate gene expression at translational and transcriptional levels. The translational attenuation associated with this pathway has been largely attributed to the phosphorylation of the translational initiation factor $eIF2{\alpha}$. However, independent studies are now pointing to a second translational regulation step involving a downstream ISR target, 4E-BP, in the inhibition of eIF4E and specifically cap-dependent translation. The activation of 4E-BP is consistent with previous reports implicating the roles of 4E-BP resistant, Internal Ribosome Entry Site (IRES) dependent translation in ISR active cells. In this review, we provide an overview of the translation inhibition mechanisms engaged by the ISR and how they impact the translation of stress response genes.

AUA as a Translation Initiation Site In Vitro for the Human Transcription Factor Sp3

  • Sp3 is a bifunctional transcription factor that has been reported to stimulate or repress the transcription of numerous genes. Although the size of Sp3 mRNA is 4.0kb, the size of the known Sp3 cDNA sequence is 3.6kb. Thus, Sp3 functional studies have been performed with an artificially introduced start codon, and thus an amino-terminus that differs from the wild-type. Ideally, full-length cDNA expression vectors with the appropriate start codon should be utilized for these studies. Using 5'rapid amplification of cDNA ends, a full-length Sp3 cDNA clone was generated and the sequence verified in nine cell lines. No AUG initiation codon was present. However, stop codons were present in all three frames 5' to the known coding sequence. In vitro translation of this full-length cDNA clone produced the expected three isoforms-one at 100 kDa and two in the mid 60 kDa range. Electrophoretic mobility shift assays showed that the protein products had the ability to bind to the Sp1/3 consensus sequence. In vitro studies, using our Sp3 clone and site directed mutagenesis, identified the translation initiation site for the larger isoform as AUA. AUA has not been previously described as an endogenous initiation codon in eukaryotes.

Depletion of PDCD4 Accelerates Stress Granule Assembly Through Sensitization of Stress Response Pathways

  • Programmed cell death 4 (PDCD4) is a novel tumor suppressor that function in the nucleus and the cytoplasm and appears to be involved in the regulation of transcription and translation. Stress granules (SGs) are cytoplasmic foci at which untranslated mRNAs accumulate when cells exposed to environmental stresses. Since PDCD4 has implicated in translation repression through direct interaction with eukaryotic translation initiation factor 4A (eIF4A), we here investigated if PDCD4 has a functional role in the process of SG assembly under oxidative stresses. Using immunofluorescence microscopy, we found that PDCD4 is localized to SGs under oxidative stresses. Next, we tested if knockdown of PDCD4 has an effect on the assembly of SG using PDCD4-specific siRNA. Interestingly, SG assembly was accelerated and this effect was caused by sensitization of phosphorylation of eIF2α and dephosphorylation of eIF4E binding protein (4E-BP). These results suggest that PDCD4 has an effect on SG dynamics and possibly involved in cap-dependent translation repression under stress conditions.

Phosphorylation of Elongation Factor-2 And Activity Of Ca2+/Calmodulin-Dependent Protein Kinase III During The Cell Cycle

  • Phosphorylation of the eukaryotic elongation factor 2 (eEF-2) blocks the elongation step of translation and stops overall protein synthesis. Although the overall rate of protein synthesis in mitosis reduces to 20% of that in S phase, it is unclear how the protein translation procedure is regulated during the cell cycle, especially in the stage of peptide elongation. To delineate the regulation of the elongation step through eEF-2 function, the changes in phosphorylation of eEF-2, and in activity of corresponding $Ca^{2+}$/calmodulin (CaM)-dependent protein kinase III (CaMK-III) during the cell cycle of NIH 3T3 cells, were determined. The in vivo level of phosphorylated eEF-2 showed an 80% and 40% increase in the cells arrested at G1 and M, respectively. The activity of CaMK-III also changed in a similar pattern, more than a 2-fold increase when arrested at G1 and M. The activity change of the kinase during one turn of the cell cycle also demonstrated the activation at G1 and M phases. The activity change of cAMP-dependent protein kinase (PKA) was reciprocal to that of CaMK-III. These results indicated: (1) the activity of CaMK-III was cell cycle-dependent and (2) the level of eEF-2 phosphorylation followed the kinase activity change. Therefore, the elongation step of protein synthesis might be cell cycle dependently regulated.

한국어판 영성측정도구(SAS)의 신뢰도와 타당도 검증연구 (Spiritual Assessment Scale;Psychometric Evaluation of the Korean Version)

  • Spirituality has been found as a factor that may influence an individual's health and response to illness and dying. The purpose of this study was to assess the reliability and validity of the Korean version of the Howden's Spiritual Assessment Scale(SAS). The SAS was developed based on the attributes of spirituality and constructed with 28 items, 4 subscales. The translation involved four steps : translation into Korean, checking agreement, back translation into English, and arriving at a consensus. Psychometric evaluation was done on 222 nurses from five hospitals. All responses were voluntary and anonymous. The Cronbach's alpa coefficient for internal consistency was .928 for the total 28 items and .700${\sim}$.805 for subdimensions. Item- total correlations ranged from .36${\sim}$.68. Principal Component Factor Analysis with Varimax Rotation yielded four factors with four or more items each loading at .40 or higher. These factors explained 51.3% of the total variance. The items clustered in this study were almost identical with initial scale. There was positive correlation(r=.648, p=.0001) between SAS and Ellison & Paloutzian's Spiritual Wellbeing Scale and negative correlation(r=-.418, p=.000) with Pines' Burnout Scale which indicate convergent and discriminant validity. In conclusion, this tool can be effectively utilized for assessing spirituality in Korea.

DETERMINING 3-D MOTION OF RIGID OBJECTS USING LINE CORRESPONDENCES

  • A linear method for determining three-dimensional motion of a rigid object is presented. In this method, two three-dimensional line correspondences are used. By using three-dimensional information of the features and observing that the rotation is unique regardless of the translation vector, the two components of motion parameters (rotation and translation) are computed separately. Also in this paper, the solution is given without a scale factor which is necessary in other methods that use only the two-dimensional projective constraints.
