Effect of Proanthocyanidins on Dimethylnitrosamine-Induced Liver Damage in Rats

  • Proanthocyanidins, one of the major natural polyphenolic compounds of grape has been reported to exhibit a wide range of pharmacological properties. In this study, we investigated the hepatoprotective effect of proanthocyanidins on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of proanthocyanidins (20, 50mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase, and bilirubin levels. Study on the Effects of Podam-hwan on Brain GABA and Glutamate Levels in the Picrotoxin-induced Convulsion (포담환이 Picrotoxin-유도 경련시 뇌중 GABA 및 Glutamate 함량변화에 미치는 영향에 관한 연구)

  • Currently convulsion is considered to be a chronic central nerve disease characterized by involuntary and severe muscle contraction or spasm. In many recent studies, convulsion's mechanism is due to unbalance between stimulation and suppression of the central nerve system, such as GABA and glutamic acid. Objectives : This study was performed to examine the anticonvulsant effects of Podam-hwan on brain GABA levels and glutamate content in picrotoxin-induced convulsions and to determine the inhibitory activity on GABA transaminase. Methods : Brain GABA levels and glutamate content in the brains of picrotoxin-induced mice using reverse phase HPLC method, anticonvulsant effect in vivo, and the inhibitory effect on GABA transaminase activity in vivo have been investigated. Results : Podam-hwan significantly lengthened the onset time of picrotoxin-induced convulsion at a concentration of 15mg/kg, but did not show a dose-dependent pattern. Also, Podam-hwan shortened the duration of convulsion by 52.2% at a dose of 30mg/kg in comparison with the control group. Podam-hwan inhibited dose-dependently GABA transaminase activity by 35.5% at 30mg/kg, comparing with the control gmup. Podam-hwan also increased the brain GABA level by 38.7% and 68.8% at doses 15mg/kg and 30mg/kg, respectively. In addition, Podam-hwan decreased the brain glutamate level by 9.6% and 17.8% at doses 15mg/kg and 30mg/kg, respectively. Conclusions : Podam-hwan can be prescribed for the treatment of convulsion by enhancement of brain GABA level and inhibition of GABA transaminase activity.

  • The concentration of amino acids, total nitrogen, trehalose, lipids and the activities of respiratory, acid$\cdot$alkaline phosphatase, glutamic oxalozcetic transaminase and glutamic pyruvic transaminase during larval stage in Pine leaf gall midge, Thecodiplosis janensis Uchi. et Inouye were measured using Paper chromatographic method, micro-Kjeldahl method, Thin layer chromatographic method, Warburg's manometric method, Bessey-Lowry method and Reitman-Frankel method, respectively. Healthy specimens )yore chosen as samples of each larval stages; alrva in gall and larva in soil. Amino acids present in the alcoholic extracts were alanine, glutamic acid, glycine, histidine, methionine, proline, threonine, tryptophan and valine. The total nitrogen concentration reached to 31.348mg/g during the larva in gall and the larval stage in soil of the value was decreased to 29.027mg/g. The hemolymph sugar, trehalose value for larva in soil was about two times of the value for larva in gall. Total lipid, phospholipid,monoacylglycerol, triacylglycerol, sterol, free fatty acid and ester cholesterol were identified at larval stages in gall and soil. Triacylglycerol concentration reached high level in contrast with other lipid contents during larvae in gall and larva in soil. Free fatty acid, sterol except decreased lipids during larval stage in soil. Endogenous respiration, succinate of respiratory activities decreased at larval stage in soil compare with larva in gall. The activities of acid phosphatase decreased larval stage in soil but the activities of alkaline phosphatase increased remarkably. The activities of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase reached high level of the larva in gall.

  • Aromatic L-amino acid transaminase is an enzyme that is able to transfer the amino group from L-glutamate to unnatural aromatic ${\alpha}-keto$ acids to generate ${\alpha}-ketoglutarate$ and unnatural aromatic L-amino acids, respectively. Enrichment culture was used to isolate thermophilic Bacillus sp. T30 expressing this enzyme for use in the synthesis of unnatural L-amino acids. The asymmetric syntheses of L-homophenylalanine and L-phenylglycine resulted in conversion yields of >95% and >93% from 150 mM 2-oxo-4-phenylbutyrate and phenylglyoxylate, respectively, using L-glutamate as an amino donor at $60^{\circ}C$. Synthesized L-homophenylalanine and L-phenylglycine were optically pure (>99% enantiomeric excess) and continuously pre-cipitated in the reaction solution due to their low solubility at the given reaction pH. While the solubility of the ${\alpha}-keto$ acid substrates is dependent on temperature, the solubility of the unnatural L-amino acid products is dependent on the reaction pH. As the solubility difference between substrate and product at the given reaction pH is therefore larger at higher temperature, the thermophilic transaminase was successfully used to shift the reaction equilibrium toward rapid product formation.

Effects of Euphorbia Pekinensis RUPR. on Rats with Acute Renal Failure induced by Gentamicin Sulfate (대극전탕액(大戟煎湯液)이 Gentamicin sulfate로 유발(誘發)된 백서(白鼠)의 급성(急性) 신부전(腎不全)에 미치는 영향(影響))

  • This article is to investigate the effects of Euphorbia Pekinensis RUPR. on rats with acute renal failure induced by gentamicin sulfate. So this experimental research was focused on measuring (1)the levels of creatinine, blood urea nitrogen, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, total protein, sodium, potassium, chloride in the serum and (2)the levels of protein, blood, sodium, potassium, chloride, pH, specific gravity and volume in the urine and (3)intake water. The results were summarized as follows. 1. While the levels of creatinine and glutamic oxaloacetic transaminase, glutamic pyruvic transaminase in the serum were significantly increased, the level of BUN in the serum were partially increased and the levels of sodium and potassium in the serum were partially decreased on the Euphorbia Pekinensis $R_{UPR}$. group. 2. While the urine volume was significantly increased, the levels of protein, blood, sodium and chloride in the urine were partially increased and the specific gravity, pH in the urine were partially decreased. According to the above results, it is shown that Euphorbia Pekinensis $R_{UPR}$. without refinning had a diuretic effect on rats with acute renal failure induced by gentamicin sulfate, but aggravated the glomerular filtration rate and tubular reabsorption. So, it is supposed that Euphorbia Pekinensis $R_{UPR}$. without refinning needs to be used carefully on renal disease and futhermore research about the kinds of extracts form is necessary.

  • The isozymes of glutamic oxalacetic transaminase from young rice root were isolated by DEAE-cellulose column chromatography, and investigated some of their enzymic properties. Obtained results were summarized as follows: 1. 93 percent of GOT activity in rice root existed in supernatant fraction, and their specific activity exibited the same pattern. 2. The rice root contained two types of GOT isozyme; cationic and anionic GOT. It seemed that both of them were distributed in supernatant fraction. 3. GOT isozymes of rice root on the pH dependance showed their maximum activity at 7.4. 4. On the stability of GOT isozymes for temperature, anionic GOT was more unstable than cationic GOT. 5. Apparent Michaelis constant for L-aspartate of GOT isozymes from rice root showed 12-14mM in the cationic GOT and 16mM in the anionic GOT, respectively.

  • 1. L-Tyrosine-${\alpha}$-ketoglutaric transaminase and p-hydroxyphenylpyruvic acid oxidase are distributed in Aspergillus oryzae. 2. L-Tyrosine oxidation in extracts of acetone powder, cell free extract and culture liquid of Aspergillus oryzae cultivated in the shaking culture are considerably accelerated by the addition of ${\alpha}$-ketoglutaric acid and then formation of glutamic acid was identified by chromatography method. 3. The roles of ${\alpha}$-ketoglutaric acid and pyridoxal phosphate have been shown to be an amino group acceptor in a transamination reaction. 4. Enzyme systems of an extracts of acetone powder and cell free extract also rapidly oxidized L-tyrosine and p-hydroxyphenlpyruvic acid to homogentisic acid. 5. The optimum pH for L-tyrosine-${\alpha}$-ketoglutaric acid transaminase was pH values of 6.0 and 6.5, and that for p-hydroxyphenylpyruvic acid oxidase was at pH values of 7.5.

Hepatoprotecive Effects of Alnus japonica Extract on Experimental Liver Injury Models (오리나무 추출물(AI-1367)의 간질환 동물모델에서의 간 보호효과)

  • The protective effect of AI-1367 (Alnus japonica extract) on liver injury was investigated. Primary rat hepatocyte intoxication was induced by tert-butyl hydroperoxide (tBH), carbon tetrachloride ($CCl_4$), or D-glactosamine (D-GalN). Liver injury was induced by $CCl_4$, D-GalN or MCD (methionine choline deficient)-diet in mouse. The cellular leakage of lactate dehyrogenase and cell viability followed by the treatment of hepatotoxicants were significantly improved by AI-1367 treatment at a concentration range of 5~50 ${\mu}g/ml$ for tBH, 5~50 ${\mu}g/ml$ for D-GalN, and 5~100 ${\mu}g/ml$ for $CCl_4$, respectively. Treatment with AI-1367 (20, 10, 5 mg/kg, p.o.) on liver injury induced by subcutaneous injection of $CCl_4$ or D-GalN reduced significantly the levels of aspartate transaminase and alanine transaminase in serum. Histological observations revealed that fatty acid changes, hepatocyte necrosis and inflammatory cell infiltration in $CCl_4$ (D-GalN)-induced liver injury was improved by administration of AI-1367. AI-1367 treatment (10, 5, 2.5 mg/kg, p.o.) also significantly recovered the body weight change and serum levels of aspartate transaminase, alanine transaminase and triglyceride in liver injury induced by MCD diet. From these results, AI-1367 shows protective effects against tBH, $CCl_4$, D-GalN, or MCD diet-induced hepatotoxicity in vitro or in vivo.

A Comparative Study on Effects of Kyungohkgo and Kyungohkgo Ga Nokyong on Growth in Growth Deficiency Rat with Insufficient Nutrition Diet (성장장애 흰쥐에서 경옥고(瓊玉膏)와 경옥고가록용(瓊玉膏加鹿茸)의 성장효과 비교 연구)

  • Objectives This study was designed to evaluate the difference of between Kyungohkgo and Kyungohkgo Ga Nokyong effects of growth Methods We divided male Spraque-Dawley rats into 6 groups. They were Normal group, Growth deficiency rat with insufficient nutrition diet group, Growth deficiency rat with 0.1% Kyungohkgo group, Growth deficiency rat with 0.2% Kyungohkgo group, Growth deficiency rat with 0.1% Kyungohkgo Ga Nokyong group and Growth deficiency rat with 0.2% Kyungohkgo Ga Nokyong group. They were administered for 5 weeks. We measured body weight, serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, RBC, concentration of Hb and PCV ratio, total WBC and its composition, the values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities. Results 1. In body weight, Kyungohkgo 0.1%, Kyungohkgo Ga Nokyong 0.1%, 0.2% groups were showed sighnificantly different than control group. But That groups were not showed significantly different than each others. 2. In serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, we obtained the results of tendency to increase in Kyungohkgo Ga Nokyong group, however these values showed no significantly different. 3. In the counts of RBC, Kyungohkgo group and Kyungohkgo Ga Nokyong group were showed significantly different than control group. But these values showed no significantly different. In the concentration of Hb, Kyungohkgo Ga Nokyong 0.2%group was showed significantly different than control group. Kyungohkgo Ga Nokyong 0.2% group and Kyungohkgo 0.2% group were showed significantly different than control group. 4. The counts of total WBC and its composition showed no significantly different in all treatment groups. 5. The values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities showed no significantly different in all treatment groups. Conclusions So Kyungohkgo Ga Nokyong and Kyungohkgo have an effect of promoting growth of rats. And We know that the effect of Kyungohkgo Ga Nokyong is better than Kyungohkgo.

Developement of novel enzyme system for production of enantiomerically pure ${\beta}-amino$ acids : Kinetic resolution of racemic 3-amino-n-butanoic acid using transaminase from Alcaligenes denitrificans Y2k-2

  • (R,S)-3-amino-n-butanoic acid$(DL-\;{\beta}\;-homoalanine)$ has been kinetically resolved using Alcaligenes denitrificans Y2k-2 as a biocatalyst, which was isolated from soil by enrichment culture, which was carried out with minimal media containing (R,S)-3-amino-n-butanoic acid as a sole nitrogen source. The enzyme which peformed this kinetic resolution assumed to belong to the ${\omega}-transaminase$ family, because A. denitrificans used pyruvate as amino acceptor and its transaminase activity was inhibited by gabaculine, aminooxy acetic acid and hydroxylamine. In whole cell reaction, (R,S)-3-amino-n-butanoic acid was kinetically resolved to the corresponding (R)-3-amino-n-butanoic acid with excellent E (>100) in the presence of pyruvate as an amino acceptor at $37^{\circ}C$. (S-specific) We observed the substrate inhibition for pyruvate at 100mM. In this study, characteristics of transaminase activity of Alcaligenes denitrificans Y2k-2, such as substrate specificity and thermostability, are carried out for the development of (R)-3- amino-n-butanoic acid production system.
