Purification and Characterization of a Cyclohexanol Dehydrogenase from Rhodococcus sp. TK6

  • Activity staining on the native polyacrylamide gel electrophoresis (PAGE) of a cell-free extract of Rhodococcus sp. TK6, grown in media containing alcohols as the carbon source, revealed at least seven isozyme bands, which were identified as alcohol dehydrogenases that oxidize cyclohexanol to cyclohexanone. Among the alcohol dehydrogenases, cyclohexanol dehydrogenase II (CDH II), which is the major enzyme involved in the oxidation of cyclohexanol, was purified to homogeneity. The molecular mass of the CDH II was determined to be 60 kDa by gel filtration, while the molecular mass of each subunit was estimated to be 28 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The CDH II was unstable in acidic and basic pHs, and rapidly inactivated at temperatures above $40^{\circ}C$ . The CDH II activity was enhanced by the addition of divalent metal ions, like $Ba^2+\;and\;Mg^{2+}$. The purified enzyme catalyzed the oxidation of a broad range of alcohols, including cyclohexanol, trans-cyclohexane-1,2-diol, trans-cyclopentane-l,2-diol, cyclopentanol, and hexane-1,2-diol. The $K_m$ values of the CDH II for cyclohexanol, trans-cyclohexane-l,2-diol, cyclopentanol, trans-cyclopentane-l,2-diol, and hexane-l,2-diol were 1.7, 2.8, 14.2, 13.7, and 13.5 mM, respectively. The CDH II would appear to be a major alcohol dehydrogenase for the oxidation of cyclohexanol. The N-terminal sequence of the CDH II was determined to be TVAHVTGAARGIGRA. Furthermore, based on a comparison of the determined sequence with other short chain alcohol dehydrogenases, the purified CDH II was suggested to be a new enzyme.

  • Rate constants for complexing with thiocyanate after ring opening of molybdenum-oxalate in acid media were obtained spectrophotometrically at 460nm. The acid-assisted dissociation of molybdenum-oxalate and the reaction of thiocyanate with oxalatooxomolybdenum (V) complex in acid media were investigated with thiocyanate and hydrogen ion concentration. The kinetic data indicate that molybdenum-oxalate is protonated to a limited extent in acid media and the protonated complex is responsible for an increase in rate for the reaction of thiocyanate with oxalatooxomolybdenum (V) complex. Replacement of an oxalate ligand in acid media has been interpreted in terms of dissociative mechanism involving bond-breaking of the oxygen trans to the yl oxygen.

  • Hahoebyeolsingut Talnori, which is under permanent performance, has lost its relevance to folklore and rituals of the villagers and has secured the possibility of change depending on the actor's choice. This article seeks to expand the current stuck performance of Hahoe mask play, which has great significance as a play and play for the audience. To this end, We studied the possibility of expanding the performance of Hahoe Talnori by trans-identity of the Choragi. Chorangi character derides yangban, seonbi and monk, who are representatives of the establishment. Those who have a consciousness of potential transformation, those who passively represent the masses, and those who are tacitly authorized by the audience to ridicule the ruling class. The audience feels exhilarated by the super-cool, but at the same time reduces the limits. We transformed Chorangi who is clever but cannot overcome the limits of his status, into a character with a sense of transformation by applying the method of turnning identity and changing identity of the theory of trans-identity. And I applied the method of expansion of the media conversion storytelling theory to create a new Chorangi Madang that conforms to the audience's needs and the spirit of the times. In addition to the performance of Yangban Seonbi Madang(Act of Nobleman and Scholar) which ends with an ambiguous reconciliation, we wanted to complete it in a way that even today's audiences could accept by applying the function of the Hahoebyeolsingut Talnori ambassador and the principle of implementation. And We writed it in a script. This article is meant to be an experimental study that attempted to transform traditional performing arts in the context of new creation of tradition. Preservation and transmission of traditional performing arts are important, but it is necessary to move forward according to the trend and spirit of the times.

  • Nowadays we are not able to consider and imagine anything without taking into account what is called Artificial Intelligence. Even broadcasting media technologies could not be thought of outside this newly emerging technology of A.I.. Since the last part of 20th century, this technology seemingly is accelerating it's development thanks to an unbelievably enormous computational capacity of data information treatments. In conjunction with the firmly established worldwide platform companies like GAFA(Google, Amazon, Facebook, Apple), the key cutting edge technologies dubbed NBIC(Nanotech, Biotech, Information Technology, Cognitive science) converge to change the map of the current civilization by affecting the human relationship with the world and hence modifying what is essential in humans. Under the sign of the converging technologies, the relatively recently coined concepts such as 'trans(post)humanism' are emerging in the academic sphere in the North American and Major European regions. Even though the so-called trans(post)human movements are prevailing in the major technological spots, we have to say that these terms do not yet reach an unanimous acceptation among many experts coming from diverse fields. Indeed trans(post)humanism as a sort of obscure term has been a largely controversial trend. Because there have been many different opinions depending on scientific, philosophical, medical, engineering scholars like Peter Sloterdijk, K. N. Hayles, Neil Badington, Raymond Kurzweil, Hans Moravec, Laurent Alexandre, Gilbert Hottois just to name a few. However, considering the highly dazzling development of artificial intelligence technology basically functioning in conjunction with the cybernetic communication system firstly conceived by Nobert Wiener, MIT mathematician, we can not avoid questioning what A. I. signifies and how it will affect the current media communication environment.

  • We consider the several plane sector domains which are bonded together along common edges with vertex at the origin. Such domains appear in electric conducting problem with multi-layered heterogeneous media. Our aim is to give a structure theorem of the singularities of the electric field at the corner. Also, we provide a regularity theorem for the electric field.

  • 트랜스 미디어 스토리텔링은 이용자의 적극적인 참여를 기반으로 디지털 컨버전스 시대에 새롭게 등장한 텍스트 유통 관습이자 미디어 경험 양식으로 영화, 드라마, 애니메이션 등의 콘텐츠 사업의 경우 다양한 트랜스 미디어 스토리텔링 시도가 활성화 되고 있지만 기업 브랜드의 경우 아직 기존의 OSMU의 범주를 크게 넘어서지는 못하고 있는 실정이다. 그러나 본 연구에서는 OSMU의 범주를 넘어 커뮤니케이션했던 브랜드의 트랜스 미디어 스토리텔링의 대표적 사례인 모닝케어 레이디 티안나걸스(T-ANNA Girls)의 사례를 통해 디지털 컨버전스에 적응하며 새로운 부가가치를 창출할 수 있는 브랜드 커뮤니케이션의 발전적 방안에 관해 논의코자 한다.

  • As the information society is advanced, the needs for mass information storage and retrieval grows. Digital image information is stored in retrieval systems, broadcasting in television transmission and exchanged over several kinds of telecommunication media. A major problem is that digital images are represented with large amount of data. The useful feature of image compression is that transmitting rapidly a lot of data in less time. Therefore we proposed a parallel Fractal trans-formation unit in Fractal Image compression system.

A Critical Review on the use of Media Platform in Dance: Focused on popularization strategy (무용의 미디어 플랫폼 활용에 대한 비판적 고찰: 대중화 전략을 중심으로)

  • This study examines the popularization strategy of domestic dance field through media platform and examines it based on the interaction of cultural participants. What does popularization of dance ultimately mean? What are the characteristics of media-based popularization strategies? Can all of these attempts be understood as popularization strategy with same attributes? The purpose of this study is to examine the characteristics of broadcasting media, which are used as the main medium of dance popularization strategy, and the online platform that recently attracts attention, and to focus on the communication process of cultural participants related to each media. Through this approach, media based activities turns out to be Mass Culture, controlled by businessman, capital, making it impossible for the creator to communicate directly with the audience, and producing the cultural products planned by the businessman. On the other hand, an online community platform is classified as Popular Culture since it allows creator to communicate and interact with audience without the direct involvement of the businessman and guarantees the rights of creation. Thereby it function as appropriate media platform and expand the discourse on popularization of dance in Korea.

Television Viewing in the Post-TV Era: An In-depth Interview Study of Young People's Television Experiences (포스트 TV 시대의 텔레비전 시청 경험에 관한 질적 연구: 20대들과의 심층 인터뷰를 중심으로)

  • Over the last ten years, media convergence and multiple platform expansion have affected the ways that people watch conventional television. In the post-TV era, the growing use of the Internet and mobile multi-media devices, such as smart phones, as well as the availability of abundant television content, allows television consumption to be more personalized, diversified, and linked with various media activities, especially social media uses. This study attempts to examine how television viewing experiences have been transformed with the development of the trans-media uses. Based on Walter J. Ong's concept of relation-ism, which posits that new media transform the meanings and relevance of old media rather than making old media obsolete, this study will pay particular attention to how the cultural meanings of television viewing have been redefined in the post-TV era. For the examination, this study has looked at concrete cases of the television viewing experiences of 29 young people in their twenties. Based on in-depth interview data, this study discusses the newly emerging characteristics of television viewing, its temporal and spatial experiences, and the significance of television as a medium and as a social place.

A Study on the Character of Media space in Responses viewd through the Digital poetics (디지털 시학 관점으로 본 뉴미디어 시대 매체공간의 특성 연구)

  • The purpose of this study had started from trying to look for a new standard in new media era and sounding out possibility of perspective of digital poetry, and had been extracting interpretational elements based on characteristics of otology in digital poetics research in technical method. Through examination of many documentary records, we suggested interpretive elements of medial space based on meaning and the theory of digital poetics. We deduced three perspectives based on relationship between the basic thesis in poetics and interpretive elements of medial space. We used them as tool for analysis of example. We chose as an example that change of program in space is clear through connection of media and space in where new form of creation happens through digital instruments and chose article as subject to which digital technology is applied and had been since 2000, The result of this study is as following: First, epistemological analysis about a space of digital media in new media era requires poetical approach unlike aesthetic approach. We can endow creative action and creation process with significance and pay attention to technical method of manufacturing, stating appropriateness of perspective in digital poetics. Second, the justice of digital poetics and its basic thesis help derive analytic elements from medial space and study purpose of production, technical method, ontological characteristics, which are three perspectives of digital poetics. Third, it analyzed characteristics of example according to three perspectives above and we can analyze characteristics of spacial expression in space of digital media into potential, trans-active, and semiosis space. It would be possible through analyzing examples in digital poetic perspective to overlook new methods of design in 21th and various perspectives and possibilities of medial space design.
