Lactobacillus plantarum을 이용한 산양삼 추출물의 진세노사이드 Rg1 및 Rg5의 함량 증대 (Enhancement of Ginsenoside Rg1 and Rg5 Contents in an Extract of Wood-cultivated Ginseng by Lactobacillus plantarum)

  • 발효식품으로부터 분리한 젖산균 중 ${\alpha}$-rhamnosidase 효소활성을 보유한 젖산균 12점을 선별하였다. 효소활성이 우수한 Weissella confuse 1점, L. pentosus 1점과 효소활성이 우수하였던 4점의 L. plantarum 균주를 사용하여 진세노사이드 Rb1과 Re를 Rg1, Rg5로 각각 생물전환하였다. L. plantarum MBE/L2990 균주를 사용한 생물전환반응에서 Rg1과 Rg5의 함량이 각각 0.58 mg, 0.24 mg 가량 증가하였으며, 대조구로 사용한 L. plantarum KCTC21004 균주에 비해 약 56% 및 42% 우수한 생물전환 효율이었다. L. plantarum MBE/L2990 균주는 한국미생물자원센터에 KCTC18529P로 기탁하였다.

생강의 주성분인 6-Shogaol이 인체 약물대사효소인 Cytochrome P450에 미치는 영향 (Effects of 6-Shogaol, A Major Component of Zingiber officinale Roscoe, on Human Cytochrome P450 Enzymes in vitro)

  • Background : Ginger has been extensively used in foods and traditional medicines in Asian countries. Despite its frequent consumption in daily life, the mechanism of potential interactions between ginger components-drug has not been examined. To elucidate the mechanism of governing the effects of 6-shogaol, a primary constituent of dried ginger, on human cytochrome P450 (CYP) isoenzymes an incubation studies were carried out using pooled human liver microsome (HLM). Methods and Results : CYP isoenzyme specific substrate was incubated with multiple concentrations of inhibitor, HLM and cofactors. 6-shogaol showed a potent inhibitory effect on CYP2C9, CYP1A2 and CYP2C19 with half maximal inhibitory concentration ($IC_{50}$) values of 29.20, 20.68 and $18.78{\mu}M$ respectively. To estimate the value of the inhibition constant ($K_i$) and the mode of inhibition, an incubation study with varying concentrations of each CYP isoenzyme-specific probe was performed. 6-shogaol inhibited CYP2C9 and CYP2C19 noncompetitively ($K_i=29.02$ and $19.26{\mu}M$ respectively), in contrast, the inhibition of CYP1A2 was best explained by competitive inhibition ($K_i=6.33{\mu}M$). Conclusions : These findings suggest that 6-shogaol may possess inhibitory effects on metabolic activities mediated by CYP1A2, CYP2C9 and CYP2C19 in humans.

다시마를 첨가한 고추장의 숙성 중 화학적 성분 변화 (Changes of Chemical Components in Kochujang Added Sea Tangle Powder During Fermentation)

  • In order to improve functionality of kochujang which is one of the traditional foods of Korea, sea tangle powder(2, 4, 6 and 8% sea tangle powder on the glutinous rice weight basis) was added to the raw material of kochujang and then investigated the change of physiochemical properties with control kochujang during the fermentation at 3$0^{\circ}C$ for 120 days. During 120 days fermentation, moisture and crude protein contents were gradually decreased with fermentation time, whereas crude fat content NaCl were slightly increased with fermentation time. The contents of reducing sugar of sea tangle kochujang was rapidly increased until 60 days of fermentation after that it was increased slightly up to 90 days of fermentation to the highest value and then reduced slightly or not changed approximately until 120 days. pH was reduced up to 60 days of fermentation after that it remained 4.63~4.91 in 90~120 days. Acidity was increased with fermentation time, and it was the highest value of 11.5~12.4$m\ell$ in 120 days of fermentation. Viscosity was increased with increased with fermentation time. Especially a case of additional sample of 8% sea tangle powder was highly increased until 60 days. A case of color difference value, in initial time of preparation of kochujang was distinct difference of value for additional samples respectively but as the fermentation progressed, among the samples not found consistent change of color difference value. L value was gradually decreased during fermentation. A value was decreased up to 30 days of fermentation and it was increased again totally at 60 days of fermentation. B value was decreased totally at initial time of fermentation and a case of control was increased again at 90 days of fermentation, the other samples increased again 60 days earlier than 30 days.

여주(Momordica charantia) 추출물이 생쥐의 지구력 운동수행능력 향상 효과에 미치는 영향 (Bitter Melon (Momordica charantia) Extract Enhances Exercise Capacity in Mouse Model)

  • Bitter melon (Momordica charantia) is used in traditional herbal medicine in many Asian countries for the treatment of several diseases such as diabetes, eczema, night blindness, psoriasis, and rheumatism. Especially, most reports concerning the biological activities of bitter melon have focused on its effects on diabetes and hyperglycemia. Also, bitter melon is regarded as a longevity food, suggesting that it has several beneficial effects on anti-aging and the maintenance of a healthy state. Thus, we investigated whether bitter melon could increase the capacity of exercise in this study. Interestingly, bitter melon fruit extract activated AMP-activated protein kinase (AMPK), which is important for regulating glucose homeostasis, mitochondrial content and exercise capacity. In addition, bitter melon extract increased the expression of enzymes involved in fatty acid oxidation such as mitochondrial uncoupling protein 3 (UCP3), carnitine palmitoyl transferase 1b (CPT1b), and pyruvate dehydrogenase lipoamide kinase isozyme 4 (PDK4). Moreover, exercise tolerance was much more enhanced in bitter melon treated animals compared to the non-treated control group. These results suggest that bitter melon is a promising candidate for the development of functional foods beneficial for physical strength and the enhancement of exercise capacity.

Isolation of Compounds with Antioxidative Activity from Quickly Fermented Soy-Based Foods

  • A bacterial strain, initially identified as B1-3, was isolated from cheonggukjang, a traditional Korean dish made from fermented soybeans. Using the Biolog system and 16S rRNA sequence analysis, we identified B1-3 as Bacillus mojavensis. We manufactured a quickly fermented soybean (QFS) food product using the B. mojavensis, and guided by their 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging ability. We isolated substances with antioxidative activity from it. Using mass spectrometry (MS) and nuclear magnetic resonance (NMR) techniques, we isolated 4 compounds from the ethyl acetate (EtOAc)-soluble neutral fraction of methyl alcohol (MeOH) extracts of the QFS food product (genistein, daidzein, 3R,4R-3-methyl-3,4-dihydroxy-2-pentanone, and 3S,4R-3-methyl-3,4-dihydroxy-2-pentanone) and 3 compounds from its acidic fraction (4-hydroxyphenylacetic acid, genistin, and daidzein). Two compounds from the neutral fraction (3R,4R-3-methyl-3,4-dihydroxy-2-pentanone and 3S,4R-3-methyl-3,4-dihydroxy-2-pentanone) were not detected in nonfermented soybeans (NFS) or in the filtrate of the LB broth used to culture B. mojavensis. However, they were detected in the filtrate of the same broth when it contained 2% glucose. These results suggest that these 2 compounds were derived from glucose (or other saccharides) in the soybean during fermentation. One compound that was found in the acidic fraction (4-hydroxyphenylacetic acid) was readily detected in NFS, but not in the culture broth. This suggests that 4-hydroxyphenylacetic acid was derived from NFS. We concluded that the antioxidative activity of cheonggukjang is a result of the interactions between soybean components and the microorganisms used in the fermentation of cheonggukjang.

  • In the 21th century, many people predict the change of agricultural conditions in our country because of the arrival of knowledge- information age. Now it is supposed that agricultural technology comes to $45%{\sim}50%$ level compare to advanced countries. So the development of new technology in agriculture are demanded all the more. Agriculture in the 21th century will be grown based on the connection of environment, knowledge and taffeta. The agricultural conditions in Kyongbuk has more profitablity and potentiality, because there are wide lands, distinguished 4 seasons, various main production crops and cultural inheritance etc. For the innovation of agricultural technology in Kyungbuk, province this plan presented 7 visions; such as foods-life, knowledge-brain, precisious environment, resouces-saving, export-market, traditional- specialization and development-participation agriculture and 7 basic strategies; such as environment-stabilization, speciality-discrimination, standard-modeliztion, demand-market, field-practicality, management-information, specialization-cooperation strategy. The major contents of this plan were presented to achieve strengthen -ment and advancement of competitive power on the agriculture of Kyongbuk; 21 superme technologies get to develop the highest technique, 21 major conventional projects will be mown continuously, 21 main clops snakes to intensify competitive power. Wheras, we will be propulsed 21 special projects for increasing a phase of Kyongbuk R D. A. and we inculuded 74 items of Si-Gym region for balanced development of area This plan consisted of 1,730 items in 84 fields and about 260 billion won will be invest every year. When it is finished successfully in 10 years, agricultural technical level of Kyungbuk will be not only promote to $80{\sim}90%$ level to advanced country, but also clanged to competitive industry, and farming villiages might be changed to a space of comfortable life.

Antibacterial Effect of Crude Extracts of Kaempferia parviflora (Krachaidam) against Cronobacter spp. and Enterohemorrhagic Escherichia coli (EHEC) in Various Dairy Foods: A Preliminary Study

  • Rhizomes of Kaempferia parviflora (Zingiberaceae) have been used in traditional Thai medicine for health promotion. In this study, the antibacterial activity of ethanol extract of K. parviflora against Cronobacter spp. and enterohemorrhagic Escherichia coli (EHEC) was investigated using paper disc dilution method. The results revealed that the ethanol extract exhibited antibacterial activity against Cronobacter spp. and EHEC. With an increasing concentration of K. parviflora ethanol extract, larger zones of inhibition of Cronobacter spp. and EHEC strains tested were observed. Therefore, its antibacterial activity suggested that K. parviflora could be used as a natural additive to ascertain food safety of various dairy products.

Enterococcus 속 박테리아의 안전성과 식품발효용 종균 개발의 방향성 (Safety of the genus Enterococcus and the development of food fermentation starters in Korea: Current status and future steps)

  • 발효식품의 우점종 Enterococcus 속 박테리아는 식품발효에 중요한 역할을 담당할 뿐만 아니라 사람과 가축의 프로바이오틱스로 사용되는 긍정적 측면을 가지고 있지만, 균혈증, 심내막염 등의 병원감염을 일으키는 병원균으로도 알려져 있다. 또한 여러 항생제에 대한 내성균주와 부착분자, 선모, 용혈소 등의 독성인자 보유 균주들이 발견되고 있어 식품용 미생물 및 프로바이오틱스로서의 적합성에 의문이 제기되고 있다. 본 총설에서는 우선 Enterococcus의 긍적적 및 부정적 측면에 대한 정보를 제공하여 논란이 되고 있는 문제점을 제시하였고, 유전체 연구를 통하여 부정적인 측면을 보유하지 않은 식품산업에서 활용할 수 있는 균주 선발 방향을 검토하였다. 또한 우리나라 전통발효식품용 종균개발 현황과 신규 식품용 미생물 인허가 제도를 검토하여 문제점을 파악하였다. 결론으로 Enterococcus 연구결과에 근거 우리나라 신규 식품용 미생물의 안전성 평가 방향성을 제시하였다.

물의 종류에 따른 오미자 추출액의 이화학적 품질특성 (Quality Characteristics of Omija (Schizandra chinensis) Extracts with Various Water Types)

  • 음청류에 이용할 수 있는 오미자 추출액을 얻기 위해 증류수, 수돗물과 Ca, Mg 등의 미네랄이 풍부한 청송약수를 이용하여 오미자 추출액을 제조하고 이화화적 특성을 측정하였다. 오미자 추출조건으로는 25배의 용매비로써 실온에서 6, 9, 12, 15시간, $65^{\circ}C$에서 2, 3, 6, 9시간 동안 추출하였다. 추출용액의 명도(L, Lightness), 적색도(a, redness), 황색도(b, yellowness), 가용성 고형분(solid content), 산도(acidity), pH, DPPH 라디칼을 소거능을 이용한 전자 공여능(Electron Donating Ability, EDA)으로부터 전반적으로 양호한 추출조건을 구한 결과는 다음과 같다. 실온에서는 물의 종류에 상관없이 12시간 추출하는 것이 적당하였으며, $65^{\circ}C$ 가온 상태에서는 6시간 추출함이 적합함을 알 수 있었다. 가온 상태에서 약수의 경우 증류수, 수돗물에 비해 빠른 추출이 가능하였으며, 추출 시간이 증가함에 따라 산도는 낮아지나, 고형분의 양과 DPPH 라디칼을 소거능은 증가하였다. 전반적으로 고려할 때에 오미자 추출을 위한 가장 적합한 물로는 미네랄이 풍부한 약수이며, 다음으로 수돗물 그리고 증류수 순서임을 알 수 있다.

숙성처리에 따른 지황 슬러리의 환원당 및 카탈폴 변화 (Changes in Reducing Sugar and Catalpol Contents of Rehmannia Root Slurry with Aging Treatments)

  • Rehmannia glutinosa root (R. root) has been used as an traditional medicine, and is important resource for natural medicines and functional foods. However, R. root contains catalpol which is bitter, and undigested sugars, including stachyose and raffinose, which can cause diarrhea. Therefore, this study was performed to identify the changes in reducing sugar from undigested sugars and in catalpol contents in R. root slurry induced by aging treatments. R. root slurry was treated at $10{\sim}70^{\circ}C$ for up to 72 hr; and extracted with a 50% ethanol solution. The catalpol content was analyzed using HPLC-UVD. Reducing sugar content generated from undigested sugars was measured by the Nelson-Somogyi methods, and the reaction rates were calculated from their variation according to aging time and temperature. During the aging treatment, reducing sugar increased and catalpol decreased. Their formation and degradation rates were highest at $50^{\circ}C$ and $30{\sim}40^{\circ}C$, and their rates were $2.05mg/g{\cdot}hr$ and 23.09 to 23.33%/hr, respectively. These results indicated that aging treatment can positively affect the sweetness and digestibility of R. root slurry. Therefore, an aging treatment could be considered for improving the taste and digestibility of R. root.
